Both environmental and climatic changes are known to influence soil microbial biomes in terrestrial ecosystems. However, there are limited data defining the interactive effects of multi-factor environmental disturbances, including N-deposition, precipitation, and air temperature, on soil fungal communities in temperate forests. A 3-year outdoor pot experiment was conducted to examine the temporal shifts of soil fungal communities in a temperate forest following N-addition, precipitation and air temperature changes. The shifts in the structure and composition of soil fungal communities were characterized by denaturing gradient gel electrophoresis and DNA sequencing. N-addition regimen induced significant alterations in the composition of soil fungal communities, and this effect was different at both higher and lower altitudes. The response of the soil fungal community to N-addition was much stronger in precipitation-reduced soils compared to soils experiencing enhanced precipitation. The combined treatment of N-addition and reduced precipitation caused more pronounced changes in the lower altitude versus those in the higher one. Certain fungal species in the subphylum Pezizomycotina and Saccharomycotina distinctively responded to N fertilization and soil water control at both altitudes. Redundancy discrimination analysis showed that changes in environmental factors and soil physicochemical properties explained 43.7% of the total variability in the soil fungal community at this forest ecosystem. Variations in the soil fungal community were significantly related to the altitude, soil temperature, total soil N content (TN) and pH value (P < 0.05). We present evidence for the interactive effects of N-addition, water manipulation and air temperature to reshape soil fungal communities in the temperate forest. Our data could provide new insights into predicting the response of soil micro-ecosystem to climatic changes.  相似文献

本文以3个种源、14个家系的细叶桉苗木为试验材料,研究了其种源、家系叶脉密度的差异以及各级叶脉密度之间的相关性。结果表明:种源间总叶脉密度和次级叶脉密度的差异极显著,2°主叶脉密度差异不显著,次级叶脉密度与总叶脉密度的比值在88.07%以上;种源地 China Dongmen 家系间的总叶脉密度、次级叶脉密度、2°主叶脉密度之间差异极显著,2°主叶脉密度与次级叶脉密度和总叶脉密度呈极显著负相关,次级叶脉密度与总叶脉密度呈极显著正相关;其他种源内家系间叶脉密度的差异较小。  相似文献

Soil compaction impacts growing conditions for plants: it increases the mechanical resistance to root growth and modifies the soil pore system and consequently the supply of water and oxygen to the roots. The least limiting water range (LLWR) defines a range of soil water contents within which root growth is minimally limited with regard to water supply, aeration and penetration resistance. The LLWR is a function of soil bulk density (BD), and hence directly affected by soil compaction. In this paper, we present a new model, ‘SoilFlex‐LLWR’, which combines a soil compaction model with the LLWR concept. We simulated the changes in LLWR due to wheeling with a self‐propelled forage harvester on a Swiss clay loam soil (Gleyic Cambisol) using the new SoilFlex‐LLWR model, and compared measurements of the LLWR components as a function of BD with model estimations. SoilFlex‐LLWR allows for predictions of changes in LLWR due to compaction caused by agricultural field traffic and therefore provides a quantitative link between impact of soil loading and soil physical conditions for root growth.  相似文献

The response of the soil food web structure to soil quality changes during long-term anthropogenic disturbance due to farming practices has not been well studied. We evaluated the effects of three tillage systems: moldboard plow/rotary harrow (MP), rotary cultivator (RC), and no-tillage (NT), three winter cover-crop types (fallow, FL; rye, RY; and hairy vetch, HV), and two nitrogen fertilization rates (0 and 100 kg N ha−1 for upland rice, and 0 and 20 kg N ha−1 for soybean production) on changes in nematode community structure. Sixty-nine taxa were counted, total nematode abundance (ALL), bacterial feeders (BAC), predators (PRD), omnivores (OMN), and obligatory root feeders (ORF) were more abundant in NT than in MP and RC, but fungal feeders and facultative root feeders (FFR) were more abundant in RC than in NT and MP. Cover crop also influenced nematode community structure; rye and hairy vetch were always higher in ALL, BAC, FFR, ORF, and OMN than fallow. Seasonal changes in nematode community structure were also significant; in particular, as soil carbon increased, nematode abundance also increased. The relationship between nematode indices and soil carbon was significant only in NT, but not in MP and RC. In NT, with increasing soil carbon, enrichment index and structure index (SI) were positive and significant and channel index was negative. Bulk density was significantly negatively correlated with FFR and ORF. Seasonal difference in nematode community between summer and autumn was larger in an upland rice rotation than in a soybean rotation. Over the nine-year experiment, SI increased not only in NT but also in MP and RC, suggesting that repeated similar tillage inversions in agroecosystems may develop nematode community structures adapted to specific soil environmental conditions. Because NT showed the highest values of both SI and soil carbon, the increase of soil carbon in NT is expected to have a great impact on developing a more diverse nematode community structure.  相似文献

Consistent spray coverage that is evenly distributed throughout the canopy is necessary to control pest populations that can negatively affect yield. As applicators are switching to Coarser spray quality nozzles to reduce risk and liability of pesticide spray drift, concerns about efficacy loss are growing. Previous research has indicated that small droplets are the most effective at penetrating through crop canopies, but newer nozzle technologies have improved the effectiveness of larger droplet or Coarser sprays. Research was conducted to assess the canopy penetration of nozzles that produce Coarse, Very-Coarse and Extremely-Coarse spray qualities compared to nozzles that produce Fine and Medium spray qualities. Kromekote collectors were positioned in four configurations in an oat (Avena sativa L.) var. ‘Yarran’ (AusWest Seeds, Forbes, NSW, Australia) crop to quantify the coverage and droplet number densities (droplets cm−2) across three application carrier volume rates: 50, 75 and 100 L ha−1. Applications were made in the field in 30 cm tall, tillering oats, with collectors arranged in a randomised complete block design with three replications. The entire study was repeated on the following day. Results showed that droplet number densities were inversely related to the droplet size produced by the nozzles, yet coverage was increased more by application volume rate than droplet size. Thus, both spray drift reduction and improved canopy penetration can be achieved with proper nozzle selection and operation parameters for the control of agronomic pests.  相似文献

Aquaculture production relies on controlled management of gametogenesis, especially in species where assisted reproduction is needed for obtaining gametes in captivity. The present study used human chorionic gonadotropin (hCG) treatments to induce and sustain spermatogenesis in European eel (Anguilla anguilla). The aim was to evaluate effects of strip-spawning timing (12 vs. 24 hr) after weekly administration of hCG and the necessity of a primer dose (in addition to weekly hormonal treatment) prior to strip spawning (primer vs. no-primer) on sperm quality parameters. Sperm parameters included milt production (weight), density and sperm kinematics at Week 9, 11 and 13 after onset of treatment. Spermiation commenced in 11.5% of males in Week 5 and by Week 9, and all males produced milt. Male weight, milt production, sperm density and spermatocrit did not differ among hormonal treatments during the experimental period. Overall, male weight decreased from 106.3 to 93.0 g, milt weight increased from 3.5 to 5.4 g, sperm density counts decreased from 11.7 × 109 to 10.5 × 109 cells/ml, and spermatocrit decreased from 46.5% to 40.5%. Furthermore, spermatocrit was positively related to haemocytometer counts (R2 = .86, p < .001), providing a reliable indicator of sperm density. Differences in sperm kinematics were observed depending on strip-spawning timing after hormonal injection (12 vs. 24 hr) but with no consistent pattern. These sperm quality parameters also did not consistently differ between the no-primer and primer treatments. Considering that each male may be stripped 4–5 times over the 2–3 months spawning season, omitting the primer would reduce animal handling, material costs and labour intensity, while sustaining high-quality sperm production.  相似文献

研究旨在从水牛犊牛血浆中分离外泌体,并对分离的外泌体进行分子生物学特征分析和鉴定。采用差速离心法分离3头水牛犊牛的血浆外泌体,并在透射电子显微镜下观察其形态,使用纳米粒度及Zeta电位仪对提取的外泌体进行粒径大小分析,应用Western blotting方法检测外泌体标记蛋白钙联蛋白(Calnexin)、肿瘤易感基因101(TSG101)、CD9、CD81在3头水牛犊牛血浆外泌体中的表达情况。结果显示,从水牛犊牛血浆中分离的外泌体形态多为圆形和椭圆形,直径在30~150 nm;Western blotting检测结果表明,在水牛血浆外泌体表面,特异性蛋白Calnexin、TSG101和CD81阳性表达,CD9不表达。本研究从形态学和分子生物学特征等方面证实研究获得的提取物为外泌体,由此说明,差速离心法可以成功分离提取水牛犊牛血浆中的外泌体,为水牛外泌体的后续研究提供了重要技术基础和参考。  相似文献

[目的] 研究去势术对雄性中华田园犬脂代谢的影响。[方法] 将12只年龄、体重相近的中华田园犬分为非手术组和去势组,对去势组中华田园犬进行去势手术,观察并测定术前第1天、术后第14天、术后第21天血清中甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)含量,以非手术组作为对照,分析去势术对雄性犬脂代谢的影响。[结果] 去势组与非手术组相比,血清中TG含量在第14天到第21天显著上升;血清中TC含量相比非手术组呈下降趋势;HDL第14天和第21天都低于非手术组;LDL都高于非手术组,整体呈上升趋势。[结论] 去势手术会影响雄性中华田园犬机体脂类代谢,会导致去势犬甘油三酯升高,总胆固醇和高密度脂蛋白下降,低密度脂蛋白上升。  相似文献
