小麦-滨麦易位系M8657-1抗条锈病基因遗传分析和分子标记   总被引:3,自引:0,他引:3  
 M8657-1, one of the wheat translocation lines derived from Leymus mollis Trin. Hara, is possessed of effective resistance at all stages to Su-ll and other dominant races of Puccinia striiformis f. sp. tritici in China. Seedlings of the parents, F1, and F2 progeny derived from the cross of M8657-1 (resistant) Mingxian169 (susceptible) were inoculated with Su-ll in greenhouse to identify and map the probable new stripe rust resistance gene. The results suggested that the stripe rust resistance in M8657-1 was conferred by a pair of recessive genes. Simple sequence repeat (SSR) technique was used to detect molecular marker associated with the resistance gene:208 pairs of wheat SSR primers were used to screen the two parents, as well as resistant and susceptible bulks and then three SSR markers were selected for genotyping the F2 population. The geue, temporarily designated as YrLml, was found to be located on the chromosome 7DL and flanked by three SSR markers GDM67, WMC150 and WMC671, with the genetic distance of 5.0, 9.7 and 11.8cM, respectively.  相似文献

 Sumai 3, a wheat variety resistant to Fusarium head blight(FHB), was crossed with Neimai 9, a commercial wheat cultivar with the resistance to powdery mildew.The SCAR(sequence characterized amplified region) markers of powdery mildew resistance gene Pm21 and four SSR(simple sequence repeats)markers flanking the major FHB resistance QTL(Qfhs.ndsu-3BS) in Sumai 3 were used to detect the resistance loci by marker assisted selection(MAS) in the plants of the F2 population.Identification of resistance to both powdery mildew and FHB in field showed that 12 plants resistant to both diseases were obtained.In addition, the agronomic traits of these plants were better than those of Sumai 3, and are perhaps the excellent parental materials for wheat breeding.  相似文献

Streamflow forecasting in drylands is challenging.Data are scarce,catchments are highly humanmodified and streamflow exhibits strong nonlinear responses to rainfall.The goal of this study was to evaluate the monthly and seasonal streamflow forecasting in two large catchments in the Jaguaribe River Basin in the Brazilian semi-arid area.We adopted four different lead times:one month ahead for monthly scale and two,three and four months ahead for seasonal scale.The gaps of the historic streamflow series were filled up by using rainfall-runoff modelling.Then,time series model techniques were applied,i.e.,the locally constant,the locally averaged,the k-nearest-neighbours algorithm(k-NN)and the autoregressive(AR)model.The criterion of reliability of the validation results is that the forecast is more skillful than streamflow climatology.Our approach outperformed the streamflow climatology for all monthly streamflows.On average,the former was 25%better than the latter.The seasonal streamflow forecasting(SSF)was also reliable(on average,20%better than the climatology),failing slightly only for the high flow season of one catchment(6%worse than the climatology).Considering an uncertainty envelope(probabilistic forecasting),which was considerably narrower than the data standard deviation,the streamflow forecasting performance increased by about 50%at both scales.The forecast errors were mainly driven by the streamflow intra-seasonality at monthly scale,while they were by the forecast lead time at seasonal scale.The best-fit and worst-fit time series model were the k-NN approach and the AR model,respectively.The rainfall-runoff modelling outputs played an important role in improving streamflow forecasting for one streamgauge that showed 35%of data gaps.The developed data-driven approach is mathematical and computationally very simple,demands few resources to accomplish its operational implementation and is applicable to other dryland watersheds.Our findings may be part of drought forecasting systems and potentially help allocating water months in advance.Moreover,the developed strategy can serve as a baseline for more complex streamflow forecast systems.  相似文献

 The activity change of defensive enzymes and PR-1a gene expression of tobacco (Nicotiana tabacum) seedling induced by chito-oligosaccharides were studied. The results showed that high level systemic acquired resistance (SAR) was expressed in tobacco plants treated with chito-oligosaccharides solution at the concentration of 50 μg/mL. PAL activity increased greatly with 2 peaks, the activity of SOD decreased initially followed by an increase with higher increment, and the activity of POD peaked early followed by a gentle fall in chito-oligosaccharide treated plants. The PR-1a gene was strongly expressed in tobacco due to systemic acquired resistance induced by chito-oligosaccharides. At 168 h after inoculation the expression quantity (co-pies/2 μL) of PR-1a gene was increased to 2 469.6 in treated tobacco leaf, reached 392.6% than that at 0 h after inoculation, it was increased 3.05 times of that in untreated control.  相似文献

 The growth and sporulation of Clonostachys rosea strain 67-1 in PD broth was observed and figured out. A large amount of submerged spores were obtained in shake flask with proprietary Czapek me-dium. Meanwhile, the colonies of strain 67-1were cultured in PDA plate and aerial spores were collected by elution. Resistances of submerged spores and aerial spores to high temperature, dry condition and UV treatment were determined. The results showed that the survival of the submerged spores kept in 60℃ for 30 min was 76.7%, while the aerial spores were hardly germinated in the same condition. After two weeks' drying treatment, the spore activities were 89.2% and 29.3%, respectively. Similarly, the activity of submerged spores was 72.6% and that of aerial spores was 19.7% when exposing to UV for 1 min. It is illuminated that deep submerged fermentation is more efficient than solid culture for strain 67-1 to produce chlamydospores, which act as main component in biopesticide mass production.  相似文献

Scarce and scattered precipitation in arid regions is detrimental for newly planted seedlings. It is essential to provide required water storage for seedlings in restoration projects in the first year of their establishment. The subsurface irrigation can be much more effective than the surface irrigation because of the regulation of water availability and reduction in water evaporation. We studied the effect of surface and subsurface irrigation methods on the growth and survival of four common tree species including heaven tree(Ailanthus altissima(Mill.) Swingle), China berry(Melia azedarach L.), white mulberry(Morus alba L.), and black locust(Robinia pseudoacacia L.) by installing underground clay reservoirs with different permeabilities in Isfahan City, Iran. Different amounts of animal manure and wheat straw were mixed with clay fraction and cooked in a pottery kiln at 900°C to produce reservoirs with different permeabilities. The experimental treatments consisting of irrigation and tree species were considered with a factorial arrangement in a completely randomized design with three replications in 2016 and 2017. Leaf water potential of seedlings, which is indirectly related to drought resistance, was measured by a portable pressure chamber. The results showed that saplings height, basal diameter, number of leaves, chlorophyll content and stomatal conductance were significantly(P0.05) higher in the subsurface irrigation with low permeability than in the surface irrigation, but the number of branches of the studied species were not significantly(P0.05) affected by the irrigation methods and different permeabilities of clay reservoirs. The clay reservoirs with low and medium permeabilities constantly provide better conditions for plant growth, and water with lower pressure and longer time intervals to the plant roots as compared with the reservoirs with high permeability. Analysis of variance of the data showed that year and interaction between year and permeability of reservoir had significant effects(P0.05) on all growth parameters, except for the chlorophyll content. In addition, the highest percentage of survival was 100% associated with the subsurface irrigation and the control treatment had the lowest survival percentages of 60%, 70%, 80% and 100% for M. alba, M. azedarach, A. altissima and R. pseudoacacia, respectively. Finally, the values of leaf water potential showed that R. pseudoacacia was the most drought resistant species.  相似文献

Stomatal behavior is a central topic of plant ecophysiological research under global environmental change. However, the physiological mechanism controlling the response of stomata to vapor pressure deficit(VPD) or relative humidity(RH) has been inadequately understood till now. In this study, responses of stomatal conductance(gs) to VPD in two species of trees(Fraxinus chinensis Roxb., Populus alba L. var. pyramidalis Bge.) in three different climate zones(Jinan with typical warm humid/semi-humid climate, Urumqi with temperate continental arid climate and Turpan with extreme arid desert climate) were measured. Levels of two phytohormones(abscisic acid, ABA; indole-3-acetic acid, IAA) in the leaves of the two tree species at these three sites were also measured by high performance liquid chromatography. The results showed that the responses of gs to an increasing VPD in these two tree species at the three sites had peak curves which could be fitted with a Log Normal Model(gs=a·exp(–0.5(ln(D/c)/b)2). The VPD/RH values corresponding to the maximum gs can be calculated using the fitting models for the two tree species in the three sites. We found that the calculated gs-max-VPD correlated negatively with relative air humidity in the three sites during the plant growth period(April to October 2010), which showed the values of gs-max-VPD were related to the climate conditions. The prevailing empirical stomatal model(Leuning model) and optimal stomatal behavior model could not properly simulate our measured data. The water use efficiency in the two tree species did not show obvious differences under three very different climatic conditions, but the highest gs, photosynthetic and transpiration rates occurred in P. alba var. of Turpan. The sensitivity in response of gs to VPD in leaves of the two trees showed positive correlations with the concentration of ABA, which implied that ABA level could be used as an indicator of the sensitivity of stomatal response to VPD. Our results confirmed that the prediction of the response of gs to VPD might be incomplete in the two current popular models. Therefore, an improved gs model which is able to integrate the results is needed. Also, the stomatal response mechanism of single peak curves of gs to VPD should be considered.  相似文献

 为探索核基质结合区(matrix attachment regions,MARs)对RNA介导的病毒抗性的影响,我们将从烟草中克隆到的核基质结合区TM2构建在包含马铃薯Y病毒全长非翻译CP基因的植物表达载体pRPVYCPN的表达盒的两侧,构建了植物表达载体pRTM2CPNTM2。采用农杆菌介导基因转化法,将表达载体pRPVYCPN和pRTM2CPNTM2转入烟草品种NC89中,分别获得了144株和344株转基因烟草。抗病性检测发现,核基质结合区的存在能明显提高RNA介导抗性的产生效率。在含MARs转基因植株中,抗病植株的比率为15.1%,而不含核基质结合区的转基因植株的抗病比率则为8.3%。这一研究结果对抗病毒植物的分子育种和转基因表达调控有指导意义。  相似文献

 以马铃薯Y病毒坏死株系(PVYN)的外壳蛋白(coat protein,CP)基因3'端50bp片段为hpRNA的茎,以pUC19不同长度的序列为hpRNA的环,构建了茎环比例分别为4:1、2:1、1:1、1:2、1:4和1:8的植物表达载体。利用农杆菌介导法转化烟草品种NC89,获得了多种转基因植株。室内抗病性检测发现:不同茎环比例的hpRNA介导的病毒抗性效率不同;茎环比例为4:1、2:1和1:1时效率较高,抗性植株的比例达60%左右;随着环长度的逐渐增加,抗性植株的比例逐渐降低;当茎环比例为1:8时,抗性植株的比例仅为9.52%。Southern blot分析结果表明:外源基因已整合于烟草的基因组中,且转基因植株的抗病性与转基因的拷贝数之间无明显的相关性。Northern blot分析结果表明:目的片段转录产物的积累量与植株的抗病性呈负相关,证明所获得的抗病性是RNA介导的。  相似文献

为获得兼抗马铃薯X病毒(Potato virus X,PVX)、马铃薯Y病毒(Potato virus Y,PVY)、马铃薯卷叶病毒(Potato leaf roll virus,PLRV)和马铃薯潜隐花叶病毒(Potato virus S,PVS)4种病毒的转基因马铃薯新材料,分别以这4种病毒全长CP基因为模板,通过设计PCR引物和亚克隆获得4种病毒CP基因相对保守区段的基因片段,并将其拼接成融合基因,以载体pHANNIBAL和pBI121为基础,构建RNA干扰(RNA interference,RNAi)载体,利用农杆菌介导的转基因体系进行马铃薯遗传转化,并对获得的转基因马铃薯进行病毒抗性检测。结果表明,所获得的融合基因片段RH1和RH2,酶切鉴定分别得到长度为1 200 bp的条带,与预期片段相符;构建了含pdk内含子和RH1、RH2融合基因的RNAi植物表达载体,经Bam H I/Sac I双酶切,获得长度约3 200 bp的片段,表明RNAi植物表达载体pBI121-pRH构建成功;转化易感病毒马铃薯品种陇薯11号,PCR检测和PCRSouthern杂交分析表明融合基因已整合到陇薯11号马铃薯基因组中;抗病性检测显示4株转基因马铃薯植株对4种病毒均免疫。表明利用RNAi可筛选出抗多种病毒的转基因马铃薯新种质。  相似文献

为探究靶序列位置对RNA介导的病毒抗性产生的影响,利用聚合酶链式反应(polymerase chain reaction,PCR)技术扩增马铃薯Y病毒(Potato virus Y,PVY)复制酶基因(nuclear inclusion b,NIb)不同位置的cDNA区段,反向插入双元载体pROKII中,构建了发夹RNA(hairpin RNA,hpR-NA)结构的植物表达载体。将构建的植物表达载体采用冻融法转入农杆菌LBA4404,叶盘法转化烟草NC89,获得转基因植株。攻毒试验表明:PVYNIb基因不同位置cDNA区段介导的对PVY的抗性存在显著差异;3′端1/2处和中间位置的序列可介导高水平的病毒抗性,抗性植株的比例在50%以上,而5′端、5′端1/2处和3′端的序列介导的抗性效率较低,抗性植株的比例仅为10%~30%。Northern杂交显示:抗病植株中RNA的积累量明显低于同类型的感病植株,抗性与RNA积累量呈负相关;抗病转基因植株中有siRNA存在,表明病毒抗性是由RNA介导的。  相似文献

Transgenic plants expressing double-stranded RNA (dsRNA) of virus origin have been previously shown to confer resistance to virus infections through the highly conserved RNA-targeting process termed RNA silencing or RNA interference (RNAi). In this study we applied this strategy to soybean plants and achieved robust resistance to multiple viruses with a single dsRNA-expressing transgene. Unlike previous reports that relied on the expression of one long inverted repeat (IR) combining sequences of several viruses, our improved strategy utilized a transgene designed to express several shorter IRs. Each of these short IRs contains highly conserved sequences of one virus, forming dsRNA of less than 150 bp. These short dsRNA stems were interspersed with single-stranded sequences to prevent homologous recombination during the transgene assembly process. Three such short IRs with sequences of unrelated soybean-infecting viruses (Alfalfa mosaic virus, Bean pod mottle virus, and Soybean mosaic virus) were assembled into a single transgene under control of the 35S promoter and terminator of Cauliflower mosaic virus. Three independent transgenic lines were obtained and all of them exhibited strong systemic resistance to the simultaneous infection of the three viruses. These results demonstrate the effectiveness of this very straight forward strategy for engineering RNAi-based virus resistance in a major crop plant. More importantly, our strategy of construct assembly makes it easy to incorporate additional short IRs in the transgene, thus expanding the spectrum of virus resistance. Finally, this strategy could be easily adapted to control virus problems of other crop plants.  相似文献

表达dsRNA的细菌提取液可抑制黄瓜花叶病毒对烟草的侵染   总被引:7,自引:0,他引:7  
 利用RT-PCR分别克隆了CMV P3613株系的RNA2片段、MP(movement protein)基因片段及CMV AN株系的CP(coat protein)基因片段。以CP基因为中间间隔序列,分别构建了含有RNA2片段和MP基因反向重复片段的原核表达载体。体外转录试验表明:两个载体转录后都能形成预期大小的dsRNA。经过IPTG诱导,在大肠杆菌HT115(DE3)菌株中可表达产生预期大小的核酸片段,经DNase和RNaseA消化处理,证实为dsRNA。将表达病毒基因dsRNA的细菌超声破碎后处理烟草,进行保护和治疗试验,结果表明:表达CMV MP基因和RNA2片段dsRNA的细菌破碎液能够诱导烟草对CMV产生抗性。接种病毒60d后,保护效果试验病株率分别为45%和60%,治疗效果试验病株率分别为75%和85%,而其他对照发病率均为100%。本研究结果证明了利用RNA沉默的原理,构建具有反向重复序列的原核表达载体,用细菌表达dsRNA的粗提取物可防治CMV对烟草的侵染。  相似文献

 本研究克隆了来源于马铃薯Y病毒坏死株系(PVYN)外壳蛋白(CP)基因(PVYN CP)5'端和3'端的反向重复cDNA序列,并构建了植物表达载体pROKⅡ-5'IR和pROKⅡ-3'IR,利用农杆菌介导的叶盘法转化烟草NC89,分别获得166和126株转基因烟草。抗病性试验表明,转化PVYN CP基因3'端茎50 bp(环50 bp) hpRNA(hairpin RNA)的烟草抗病率达69%,而转化PVYN CP基因5'端相同片段长度的hpRNA的烟草却全部发病。Southern blot结果表明,转基因植株的抗病性与转基因的拷贝数之间无明显的相关性;Northern blot结果表明,目的片段转录产物的积累量与植株的抗病性呈负相关,证明所获得的抗病性是RNA介导的抗病性。研究结果证明PVYN CP基因的3'端比5'端更能有效地诱发基因沉默,并且hpRNA茎部的长度可减少到50 bp。该结果为探索利用CP基因的最短长度和有效部位的基因片段来培育多抗病毒转基因植物提供了依据。  相似文献

瞬时表达靶向TMV外壳蛋白基因的siRNA能干扰病毒侵染   总被引:10,自引:0,他引:10  
 RNA干扰(RNA interference,RNAi)是与内源性mRNA编码区某段序列同源的双链RNA导入细胞后,该mRNA发生特异性降解,从而导致该基因表达沉默的现象。小干扰RNA(small interfering RNA,siRNA)作为RNAi途径的重要中介,已被广泛应用于动、植物抗病毒治疗研究。本文以烟草花叶病毒(Tobacco mosaic virus,TMV)外壳蛋白基因为靶位,设计合成表达小干扰RNA的寡核苷酸,亚克隆到植物双元表达载体pBI121中,直接转化根癌农杆菌。通过根癌农杆菌介导的瞬时表达法,研究了同源于TMV外壳蛋白的siRNA对TMV侵染的干扰作用。结果表明,瞬时表达的siRNA能够特异性干扰TMV侵染。含有重组表达载体pBI121/siRNA的根癌农杆菌渗入普通烟植株,在TMV接种后14d其上部叶片没有表现典型的花叶症状。对这些叶片进行Northern杂交试验也没有检测到TMV病毒的RNA积累或仅有很少量的积累。在枯斑寄主心叶烟上,siRNA的瞬时表达可使TMV侵染后的枯斑数明显减少,甚至不产生枯斑。此外,同源于TMV外壳蛋白的siRNA瞬时表达对非同源的黄瓜花叶病毒(Cucumber mosaic virus,CMV)没有抑制作用,表明siRNA的干扰作用具有高度的同源依赖性。  相似文献

基因沉默是广泛存在于各种生物中的一种古老现象,是生物抵抗外来入侵者的保护机制。RNA干扰(RNA interference,RNAi)则是近年来发现的一种重要基因沉默现象。此策略已在植物抗病毒育种等研究中应用,如对水稻、大麦、大豆、玉米、马铃薯、番茄、辣椒、木瓜、南瓜、李、烟草等的抗病毒研究。在转基因抗病毒展现出诱人的前景时,对转基因抗病毒植物释放的安全性问题的关注也越来越多。本文介绍了RNAi的作用机制,在转基因抗病毒育种中的应用,并探讨了以RNAi为基础的转基因抗病毒作物的食用安全性和环境安全性等问题。  相似文献

MicroRNAs (miRNAs) are negative regulators of gene expression via mRNA degradation or translational repression. The potential of artificial miRNAs (amiRNAs) as antiviral agents has been used in plant biotechnology. In this study, we designed eight amiRNAs derived from Potato Virus Y (PVY) Coat Protein RNA sequences and generated transgenic tobacco plants to express these amiRNAs to confer virus resistance against PVY. Together with transient assays, the hairpin amiRcp precursors that mimic natural miRNA precursor molecules proved to be effective in expressing amiRcps and silencing the target gene. Virus resistance assay revealed that not all amiRcps targeting viral CP sequence are equally effective in preventing PVY infection. Plants with amiRcp-8 targeting the 3′ end (nt735–nt754) region exhibited high virus resistance up to 64.69%. The amiRcp-6 harboring RNA sequence (nt567–nt586) induced the lowest percentages with only 17.05%. Besides, northern blots showed that there was a correlation between the resistance level and the accumulation of amiRNA expression. Furthermore, we used bioinformatics approach to predict the mRNA structure and found that targeting sequences of loose structure benefit to improve the virus resistance level. Our results indicate that the selection of appropriate target sequence is crucial for transgenic plant against virus and provide useful guideline for the design of pathogen-derived amiRNAs.  相似文献
