Sweet potato (Ipomoea batatas L.) is the fifth most important crop in the developing countries after rice, wheat, maize and cassava. The amplified fragment length polymorphism (AFLP) method was used to study the genetic diversity and relationships of sweet potato accessions in the germplasm collection of Sokoine University of Agriculture, Morogoro and Sugarcane Research Institute, Kibaha, Tanzania. AFLP analysis of 97 sweet potato accessions using ten primer combinations gave a total of 202 clear polymorphic bands. Each one of the 97 sweet potato accessions could be distinguished based on these primer combinations. Estimates of genetic similarities were obtained by the Dice coefficient, and a final dendrogram was constructed with the un-weight pair-group method using arithmetic average. AFLP-based genetic similarity varied from 0.388 to 0.941, with a mean of 0.709. Cluster analysis using genetic similarity divided the accessions into two main groups suggesting that there are genetic relationships among the accessions. Principal Coordinate analysis confirmed the pattern of the cluster analysis. Analysis of molecular variance revealed greater variation within regions (96.19%) than among regions (3.81%). The results from the AFLP analysis revealed a relatively low genetic diversity among the germplasm accessions and the genetic distances between regions were low. A maximally diverse subset of 13 accessions capturing 97% of the molecular markers diversity was identified. We were able to detect duplicates accessions in the germplasm collection using the highly polymorphic markers obtained by AFLP, which were found to be an efficient tool to characterize the genetic diversity and relationships of sweet potato accessions in the germplasm collection in Tanzania.  相似文献

为了解福建省稻曲病菌群体的遗传多样性和遗传组成,应用随机扩增多态性RAPD (random amplified polymorphic DNA)技术分析了来自福建省不同水稻种植区的102个稻曲病菌(Ustilaginoidea virens)的遗传多样性.从100条随机引物中筛选出10条扩增带清晰、重复性好的引物进行稻曲病菌多态性扩增,共扩增出157条带,多态性条带比率为82.17%,遗传距离变化范围为0.02~0.67.遗传多样性分析表明,福建省稻曲病菌具有丰富的遗传多样性,相对于其它地区而言,福建闽西地区分离的菌株遗传多样性水平最高PPB=76.43,H=0.2212,I=0.3383),晚稻分离的菌株群体遗传多样性(PPB=91.08,H=0.2402,I=0.3655)高于早稻群体(PPB=63.06,H=0.1892,I=0.2870).聚类分析显示,在遗传距离0.349水平上,供试的所有菌株可被划分成7个遗传聚类组(R1~R7),聚类组R1为优势聚类组,包含有80个菌株,其内又存有一些亚组.这些菌株的聚类与菌株的地理来源及水稻品种无明显相关性.但是在遗传距离0.330水平上,来自宁化的10个菌株可被明显划分成早稻群体和晚稻群体.初步分析认为菌株的地理来源、水稻品种及其生长季节是影响福建省稻曲病菌遗传多样性的主要因素,在稻曲病菌的遗传变异以及该病的发生和流行中可能起重要的作用.  相似文献

This paper describes a method for achieving qualitative identification of four rice varieties from two different Italian regions. To estimate the presence of genetic diversity among the four rice varieties, we used polymerase chain reaction-randomly amplified polymorphic DNA (PCR-RAPD) markers, and to elucidate whether a relationship exists between the ground and the specific characteristics of the product, we studied proximate composition, fatty acid composition, mineral content, and total antioxidant capacity. Using principal component analysis on genomic and compositional data, we were able to classify rice samples according to their variety and their district of production. This work also examined the discrimination ability of different parameters. It was found that genomic data give the best discrimination based on varieties, indicating that RAPD assays could be useful in discriminating among closely related species, while compositional analyses do not depend on the genetic characters only but are related to the production area.  相似文献

Knowledge of the genetic diversity in the germplasm accessions is important for the efficient germplasm management. We studied 45 colored rice accessions, which had been earlier collected from peninsular India. The accessions were evaluated at two diverse locations for 12 morpho-agronomic traits and genotyped using 50 simple sequence repeat (SSR) markers. Significant genotypic variability was observed for all morpho-agronomic traits studied. Dendrogram and principal component analysis based on morpho-agronomic traits separated the accessions into three clusters. The first two principal components accounted for 82% of the total variation. The most discriminatory traits were number of grains per panicle, biomass yield, and days to flowering and maturity. The SSR analysis revealed high polymorphic information content value of 0.84. Though, Mantel test did not show a significant correlation, yet the classification based on phenotyping and genotyping data showed good agreement for the expression of morpho-agronomic traits in the formation of clusters. Five accessions had high concentration (> 100 μg/g) of Fe and one of Zn. Our study revealed the presence of large genetic variation among the colored rice accessions evaluated and moderate agreement between morpho-agronomic and SSR–based classifications and of these with geographic diversity.  相似文献

为了利用分子标记方法评价青钱柳种质资源的遗传多样性,本文对青钱柳DNA的提取、SRAP扩增体系重要参数进行了优化,运用优化体系筛选多态性引物,并用1对引物对青钱柳9个种源进行了遗传多样性初步分析。研究结果建立了适于青钱柳SRAP的扩增体系;从110对SRAP引物中筛选出了13对多态性引物,运用1对引物组合Me7+Em2扩增获得21个多态性位点,多态率达100%。遗传多样性分析表明有效等位基因数(Ne)为1.3429,平均Shannon's信息指数(I)为0.3687,Nei's基因多样性指数(H)为0.2267,种源的遗传分化指数Gst为0.1983;聚类分析结果表明9个种源在遗传距离0.100处聚为3类,聚类结果和地理距离之间呈现较高的相关性。本文的研究结果表明所建立的青钱柳种质资源库具有广泛的遗传多样性,为进一步的开发利用提供了条件。  相似文献

The pattern of genetic diversity among 92 genotypes of soybean from 5 different origins/sources (Pakistan, the USA, Asian Vegetable Research Development Centre (AVRDC), Japan and North Korea) was analyzed using randomly amplified polymorphic DNA markers. Out of 20 random primers 6 tested, 10 were polymorphic among genotypes and they yielded 107 markers, with an average of 10.7 markers per primer. The proportion of polymorphic bands within genotypes ranged from 0.47 to 0.71 with an average of 0.59. Pakistani and US genotypes exhibited the highest number of polymorphic bands (95%), while North Korean genotypes revealed the lowest (60%). The mean band frequency of the primers among genotypes was 0.57 with a range of 0.08–0.99. The Shannon’s index and Nei’s genetic diversity index revealed that primer OPF-06 showed maximum genetic diversity among the genotypes. Dendrogram constructed using Unweighted Pair Group Mean Average (UPGMA) method divided the genotypes into 5 main groups consisting of 13 clusters. The results of cluster analysis indicated that the genetic diversity between Pakistani and US or AVRDC genotypes is much larger than that between Pakistani and North Korean or Japanese genotypes. The Pakistani genotypes had distinct bands from plant introductions. Therefore, the Pakistani genotypes may be useful to soybean breeders.  相似文献

用SSR和SRAP两种分子标记方法研究51份甘蓝型油菜杂交种亲本系的遗传多样性,并对两种分子标记研究结果进行比较。结果发现,在51份材料中,45对SSR引物共扩增出194条多态性条带,平均每对引物为4.3条,25对SRAP引物共扩增出197条多态性条带,平均每对引物为7.9条。UPGMA聚类分析表明,SSR和SRAP标记都可将51份亲本材料划分为五大类群,本所选育的玻里马细胞质雄性不育系(Polima CMS)的主要保持系和恢复系都聚在同一类群的不同亚群中。根据系谱资料分析发现,SRAP标记划分的类群与系谱资料更为接近,SRAP标记更适用于遗传关系较近材料的遗传多样性分析。SRAP标记揭示的亲本间遗传距离要大于SSR标记揭示的遗传距离。两种不同标记方法揭示出油菜亲本遗传多样性的差异主要是由不同的标记方法揭示的标记位点等位基因变异数不同造成的。  相似文献

本研究的目的在于筛选合适的RAPD随机引物,应用RAPD技术对药用植物绞股蓝进行遗传多样性分析,并构建DNA指纹图谱。研究结果表明,我们利用生物信息学方法挑选出的20条引物中有19条引物的扩增条带清晰且多态性好;在清晰稳定出现的354条带中,294条具有多态性;其中有3条引物的扩增条带可清楚区分绞股蓝与混淆品种乌蔹莓,可建立其DNA指纹图谱。按UPGMA法进行聚类分析,计算其遗传相似系数,结果显示,8份绞股蓝供试材料聚为两类,聚类结果与其地理区域远近和生长环境一致。本研究中筛选出的19条引物适用于绞股蓝遗传多样性分析,且获得的DNA指纹图谱可用于鉴别绞股蓝。  相似文献

为研究广西地方食用木薯材料的遗传多样性,以48份广西地方食用木薯为试验材料,分析其变异系数、遗传多样指数、聚类分析、多态性和遗传相似性系数。结果表明,收集的木薯材料的平均表型变异系数为37.9%,平均遗传多样性指数为0.807,其中主茎分叉角度的变异系数最大,为86.7%,块根内皮颜色的遗传多样性指数最大,为1.842。13对SSR引物共扩增出118个条带,其中多态性条带为106个,多态性比率为86.23%;分子标记聚类结果发现,遗传相似性系数在0.415~1.000之间;通过对比发现表型聚类和分子聚类结果不一致,其中表型聚类发现类群划分与地理来源之间没有关联,但与株型性状有一定的关联;在遗传相似系数为0.62时,SSR分子标记聚类为两类材料,第一类材料地理分布无规律,第二类材料大多分布在桂南。本研究结果表明,广西地方食用木薯资源遗传多样性具有一定的丰富度,可为创制优异食用木薯种质资源和新品种选育奠定基础。  相似文献

Genetic diversity is an area of concern for sustaining crop yield. Information on genetic relatedness/diversity among Gossypium arboreum L. cultivars/genotypes is scanty. We have used random amplified polymorphic DNA (RAPD) analysis to assess the genetic divergence/relationship among 30 genotypes/cultivars of G. arboreum. Of 45 primers surveyed, 63% were polymorphic. Out of the total number of loci amplified, 36% were polymorphic. The calculated genetic similarity between the cultivars/genotypes was in the range of 47.05–98.73%. Two genotypes, HK-244 and Entry-17, were the most distantly related. The average genetic relatedness among all the genotypes was 80.46%. However, most of the cultivated varieties showed a close genetic relationship, indicating a narrow genetic base in comparison to the non-cultivated germplasm. The calculated coefficients were used to construct a dendrogram using the unweighted pair group of arithmetic means (UPGMA) algorithm, which grouped the genotypes/cultivars into two major and three smaller clusters. The study is the first comprehensive analysis of the genetic diversity of G. arboreum germplasm and identifies cultivars that will be useful in extending the genetic diversity of cultivated varieties and future genome mapping projects.  相似文献

Genetic diversity and relationships among 48 safflower accessions were evaluated using 22 inter-simple sequence repeats (ISSR) primers. A total of 429 bands were amplified, and 355 bands (about 82.7%) were polymorphic. Five to forty-one polymorphic bands could be amplified by each primer, with an average of 16.1 polymorphic bands per primer. The results showed that the polymorphism of the safflower germplasm was higher at the DNA level. All the 48 accessions could be distinguished by ISSR markers and were divided into 9 groups based on ISSR GS by using UPGMA method. The genetic relationships among the accessions from different continents were closer. Comparatively, the genetic diversity of the accessions originated from Asia was higher, from Europe assembled. The results also showed that the genetic variation of accessions from Indian and Middle Eastern safflower diversity centers were relatively higher. ISSR is an effective and promising marker system for detecting genetic diversity among safflower and give some useful information on its phylogenic relationships.  相似文献

Oryza latifolia is the most abundant and widespread wild relative of rice in the lowlands of Costa Rica. It also has the most diverse morphology. However, little is known about the genetic diversity and mating system of this allotetraploid species. Genetic analyses of nine populations from different life zones from the Pacific and Atlantic slopes were performed using six isozyme loci. Differences in genetic variability were observed among populations, of which Palo Verde and Santa Rosa were the most diverse. Most Atlantic populations clustered together, and a similar result was obtained with the Pacific populations. High levels of interpopulation diversity were observed while most populations were monomorphic for at least one genotype, suggesting little genetic flow within populations. Even in polymorphic populations no variation was observed within progeny groups, combined with Hardy-Weinberg disequilibria in most populations, commonly observed in autogamous species or in species with clonal reproduction. However, the high frequency of heterozygous-like patterns may suggest that the reproductive system of O. latifolia might be more complex. Future research may explain the genetic patterns as well as the reproductive biology of this species. The knowledge of the genetic diversity and mating system of O. latifolia could contribute to the implementation of genetic resource conservation strategies and gene flow analyses, as well as of breeding programs for rice improvement.  相似文献

Breeding progress in black raspberry (Rubus occidentalis L.) has been limited by a lack of genetic diversity in elite germplasm. Black raspberry cultivars have been noted for showing very few phenotypic differences and seedlings from crosses between cultivars for a lack of segregation for important traits. Despite these challenges, little molecular work has been done to explore genetic diversity and relationships in wild and cultivated black raspberry germplasm. Microsatellite, or simple sequence repeat (SSR), markers are highly polymorphic codominant markers useful for studying genetic diversity, population genetics, genetic fingerprinting and other applications. We examined genetic diversity in 148 wild and cultivated black raspberry accessions using 21 polymorphic SSR markers. Black raspberry cultivars clustered tightly and showed higher than expected heterozygosity while that of wild accessions was low. Relationships between wild black raspberry accessions were poorly resolved and regional clusters were mostly absent from our analysis. Our results indicated that wild black raspberry germplasm is a relatively untapped resource available for future breeding.  相似文献

Genetic variation within and among several Sorghum populations from different agroecological zones in Malawi were investigated using random amplified polymorphic markers (RAPDs). DNA samples from individual plants were analyzed using 35 oligonucleotides of random sequence. Twenty five of these primers allowed amplifications of random polymorphic (RAPD) loci. Overall, 52% of the scored loci were polymorphic. Every accession was genetically distinct. The analysis of molecular variance revealed that the within-region (among accessions) variations accounted for 96.43% of the total molecular variance. Observed variations in allelic frequency was not related to agroecological differences. The degree of band sharing was used to evaluate genetic distance between accessions and to construct a phylogenetic tree. Further analysis revealed that the sorghum accessions analyzed were genetically close despite considerable phenotypic diversity within and among them. It is suggested that all the sorghum landraces currently available in Malawi should be conserved both ex situ and in situ to maintain the current level of genetic diversity.  相似文献

Weedy rice has been becoming a notorious weed in the paddy field of China in recent decades due to its increasing damage to rice yield and rice quality. In this study, a microsatellite technique with 21 pairs of SSR markers was utilized to estimate the genetic structure of two biotypes of weedy rice with Japonica and Indica rice characteristics, collected from Liaoning and Guangdong provinces, respectively. The genetic diversity of the weedy rice in the two provinces was relatively low (Liaoning h = 0.086; Guangdong h = 0.160), and distinctly large genetic differences existed between these two provinces (Gcs = 0.623). The genetic diversity was found primarily within populations, and genetic differentiation was relatively low within the same province. Both cluster analysis (UPGMA) and principle component analysis (PCA) showed that weedy rice had a closer relationship with the cultivated rice collected from the sample field than with other cultivated rice and common wild rice varieties in China. Thus, the results of this study on samples from the Liaoning and Guangdong provinces in China support the de-domestication hypothesis that weedy rice most probably originated from local cultivated rice.  相似文献

As an oilseed crop, the cultivation of Ethiopian mustard (Brassica carinata) is restricted only to Ethiopia. Even though geographic diversity is a potent source of allelic diversity, the extent of genetic diversity among germplasm material of Ethiopian mustard from different countries has not been assessed. Forty-three accessions, comprising 29 accessions from eight different geographic regions of Ethiopia and 14 exotic accessions from Australia, Pakistan, Spain, and Zambia were analysed for their genetic diversity using random amplified polymorphic DNA (RAPD) technique. A set of 50 primers yielded a total of 275 polymorphic bands allowing an unequivocal separation of every Ethiopian mustard accession. The usefulness of the 50 RAPD primers in measuring heterozygousity and distinguishing accessions was variable such that polymorphic information content (PIC) varied from 0.05 to 0.40, band informativeness (BI) from 0.05 to 0.65 and primer resolving power (RP) from 0.15 to 6.83. Jaccard's similarity coefficients ranged from 0.44 to 0.87 indicating the presence of a high level of genetic diversity. On the average, Australian and Ethiopian accessions were the most similar while, Spanish and Zambian accessions were the most distant ones. Cluster analysis grouped the 43 accessions into four groups, which has quite a high fit (r = 0.80) to the original similarity matrix. With no prior molecular information, the RAPD technique detected large genetic diversity among the 43 accessions from five different countries and their grouping by dendrogram and principal coordinate analysis (PCoA) was inclined towards geographic differentiation of RAPD markers. Conversely, RAPD differentiation along geographic origin was not apparent within the Ethiopian accessions.  相似文献

For adding the hulless barley resources of Shangri-la region to the global barley resource library, a basic work was done by us to assess their genetic diversity of this region. The genetic diversity of 60 hulless barley samples collected from three counties in Shangri-la region of Yunnan Province, were studied using SSR (simple sequence repeats) and AFLP (amplified fragment length polymorphism) markers. A total of 70 alleles were detected for 19 pairs of SSR primers, and 525 band containing 464 polymorphic bands were revealed for 5 pairs of AFLP primers. The value of polymorphism information content (PIC) ranged from 0.03 to 0.86 for SSR primers. The total numbers of alleles were 51, 55, 43 in three populations and the polymorphic bands were 188, 205 and 141. The genetic distances and genetic identity among the three populations showed their close relationship. The gene diversity among populations relative to the total population diversity (Gst) was 0.13 for SSR markers and 0.02 for AFLP markers and indicated that just 13 and 2% variations were among populations, respectively. The UPGMA cluster analysis revealed that all of the samples grouped randomly rather than clustered into distinct groups corresponding to their populations, row types and spring/fall types. We concluded that there was high genetic diversity in the population of Shangri-la region and the formation of diversity was related to complex environment and inhabitants’ traditional practices.  相似文献

Guinea yam (Dioscorea cayenensisD. rotundata complex) is an important tuber crop that highly contributes to food security and poverty alleviation in Benin. The knowledge and understanding of the extent of genetic variation of Guinea yam germplasm is important for planning of the genetic conservation, and the utilisation of this resource. The objective of this study was to assess the genetic diversity and relationships of 64 yam landraces whose dried chips are considered as resistant to insect attacks using 41 simple sequence repeat. Among these primers, 13 were found to be polymorphic, giving 113 polymorphic alleles. The number of alleles per locus ranged from 4 (Ym50) to 13 (Ym29), with an average of 8.69. Unique allele was observed with some landraces (Singou and Tchakatchaka) and can be considered as unique gene and use in yam breeding program. The mean polymorphic information content values for all markers used was 0.76 and ranged between 0.58 and 0.91 in loci YM3 and YM32 respectively. The genetic distance of yam landraces ranged from 0.45 (Yasoubagarou) to 0.04 (Assinapeira and Alahina), indicating that the yam germplasm has a high degree of genetic diversity supported by an averagely observed heterozygosity of 0.78. Cluster analysis using unweighted pair group method with arithmetic average grouped the 64 yam landraces into two distinct clusters. This tendency was also observed in the principal coordinate analysis. The analysis of molecular variance revealed that 96 % of the variation was found within the population and only 4 % between the populations. Genetic diversity and relationship assessments among the 64 yam landraces of Benin could provide useful information for efficient use of these materials, especially for genetic improvement.  相似文献

The genetic diversity of 70 Houttuynia Thunb. accessions from Sichuan, Chongqing, Guizhou and Jiangsu province in China were tested by using random amplified microsatellite polymorphism (RAMP) markers. All of the 43 primer combinations were found to amplify polymorphic products. A total of 304 products were amplified. Of which, 97.7 products were polymorphic. 6.9 polymorphic bands were amplified by each primer combination on average. The genetic similarity (GS) between the accessions within H. emeiensis and H. cordata were 0.660 and 0.575, respectively. The GS between two species was 0.525. The GS values between H. emeiensis and the H. cordata cytotype with the chromosome number of 36 was 0.559, higher than that between H. emeiensis and the cytotypes of H. cordata with other chromosome numbers. Within the species H. cordata, the genetic variation between cultivated and wild accessions was insignificant. The results of cluster analysis by using UPGMA method showed that all the tested accessions could be differentiated by RAMP markers, and classified into 11 groups. Many accessions with the same chromosome numbers could be classified together. The genetic diversity was more plentiful in mountainous and margin areas of the Sichuan Basin than at the bottom of the Basin and the highlands or hills surrounding it. It was concluded that there existed higher genetic diversity at the molecular level (RAMP markers) among the germplasm resources of the genus Houttuynia. The genetic relationships and phylogeny of the germplasm resources of Houttuynia were also discussed.  相似文献

A total of 242 accessions of common wheat (Triticum aestivum L.) released in China since the 1940s were evaluated with AFLP (amplified fragment length polymorphism) technique. Genetic diversity was analyzed using five pairs of polymorphic primer combinations with 245 polymorphic bands. The highest genetic diversity was found in the accessions of the 1950s, and in the next place was that in the 1940s. The genetic diversity began to descend in the 1960s, and fell to the lowest in the 1970s. After that, the genetic diversity came back to some extent in the 1980s, however, it became much lower in the 1990s compared with that in the 1940–1950s. Landraces and introduced accessions from foreign countries showed greater genetic diversity in comparison to improved varieties. In addition, greater genetic diversity was observed in winter wheat. It was emphasized that great attention should be paid on further exploration of genetic diversity in wheat breeding program.  相似文献
