为探讨山豆根多糖(SSP)对体外培养的鸡脾脏淋巴细胞信号分子的影响,采用终浓度为50、100、200、400μg/mL的山豆根多糖分4、8、12、24 h 4个时间段刺激培养鸡脾脏淋巴细胞,测定培养上清或细胞内环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、6-酮-前列腺素F1α(6-keto-PGF1α)、血栓素B2(TXB2)和一氧化氮(NO)信号转导分子水平。结果表明,山豆根多糖在终浓度为100μg/mL时与鸡脾脏淋巴细胞共同培养8 h后能显著升高cAMP(P0.01)、6-keto-PGF1α(P0.05)、TXB2(P0.01)水平,4种浓度的山豆根多糖对NO分子水平随时间的变化有不同程度的升高(P0.05),影响cAMP/cGMP和6-keto-PGF1α/TXB2信号体系。提示山豆根多糖可通过影响免疫细胞内的信号转导而影响机体的免疫功能。  相似文献

为探讨NO/cGMP信号转导系统对小型猪特异性麻醉颉颃剂催醒分子机理的调控,144只Wistar大鼠随机分为对照组和试验组,对照组和试验组再分为早期催醒组、中期催醒组和晚期催醒组。用分光光度法测定脑NOS活性和NO产量,ELISA测定脑cGMP含量。结果显示,小型猪特异性麻醉颉颃剂能明显地激活大鼠相关脑区的NOS活性和NO及cGMP产量,并且大鼠不同脑区NOS活性、NO产量和cGMP含量的变化趋势不仅与大鼠行为学变化相平行,还与不同时期表现出的催醒效果基本吻合。这表明小型猪特异性麻醉颉颃剂的催醒可能与激活特定脑区的NO/cGMP信号转导系统相关。  相似文献

The genus Morbillivirus is classified into the family Paramyxoviridae, and is composed of 6 members, namely measles virus (MV), rinderpest virus (RPV), peste-des-petits-ruminants virus (PPRV), canine distemper virus (CDV), phocine distemper virus (PDV) and cetacean morbillivirus (CeMV). The MV, RPV, PPRV and CDV have been successfully attenuated through their serial passages in vitro for the production of live vaccines. It has been demonstrated that the morbilliviral virulence in animals was progressively attenuated with their consecutive passages in vitro. However, only a few reports were involved in explanation of an attenuation-related mechanism on them until many years after the establishment of a quasispecies theory. RNA virus quasispecies arise from rapid evolution of viruses with high mutation rate during genomic replication, and play an important role in gradual loss of viral virulence by serial passages. Here, we overviewed the development of live-attenuated vaccine strains against morbilliviruses by consecutive passages in vitro, and further discussed a related mechanism concerning the relationship between virulence attenuation and viral evolution.  相似文献

Osmotic-sensitive (os-1) mutant alleles in Neurospora crassa exhibit resistance to dicarboximides, aromatic hydrocarbons and phenylpyrroles. We have previously reported that the os-1 mutants can be classified into two groups based on their resistance to fungicides and osmotic stress: type I, which are highly resistant to iprodione and fludioxonil but moderately sensitive to osmotic stress, and type II, which are highly sensitive to osmotic stress but moderately resistant to fungicides. To explain the mechanism of resistance to these fungicides, we cloned and sequenced the mutant os-1 genes that encode putative osmo-sensing histidine kinase. Within the os-1 gene product (Os1p), the type I strains, NM233t and Y256M209, carried a stop codon at amino acid position 308 and a frameshift at amino acid position 294, respectively. These mutation sites were located on the upstream of histidine kinase and the response regulator domains of Os1p, strongly suggesting that type I strains are null mutants. The null mutants, NM233t and Y256M209, were highly resistant to iprodione and fludioxonil; thus Os1p is essential for these fungicides to express their antifungal activity. The amino acid changes in Os1p, 625Pro from Leu, 578Val from Ala, and 580Arg from Gly were found in the type II strains, M16, M155-1 and P5990, respectively. Os1p is novel in having six tandem repeats of 90 amino acids in the N terminal. Each amino acid change of the type II strains was located on the fifth unit of six tandem repeats. Type II strains with single amino acid changes were more sensitive to osmotic stress than the null mutants (type I), indicating that the amino acid repeats of Os1p were responsible for an important function in osmo-regulation.  相似文献

 荔枝霜疫霉(Peronophythora litchii)隶属于茸鞭生物界卵菌门疫霉属,由其引起的荔枝霜疫病是目前荔枝生产上一种最重要的病害,严重影响荔枝产量和鲜果品质。双组分信号途径在微生物中参与多种生命活动,但是在卵菌中尚未有相关研究。本研究通过生物信息学分析在荔枝霜疫霉中鉴定到2个杂合型组氨酸激酶(PlHK1、PlHK2)和1个响应调控蛋白(PlRR1)。其在卵菌中是保守存在的,并且与真菌在进化上相对独立。功能域分析表明,PlHK1和PlHK2的C端额外的融合了1个磷酸转移功能域(Hpt),这与真菌和植物的存在显著差异。转录分析表明3个基因在荔枝霜疫霉侵染阶段上调表达,并且响应渗透胁迫和氧化胁迫。以上结果揭示了双组分信号系统可能在疫霉致病过程中发挥重要作用。  相似文献

We conducted a retrospective study in presumed normal dogs to determine the adrenal gland attenuation and volume values. Multidetector computer tomography (MDCT 16) analysis of the gland was carried out in 48 adult dogs without evidence of adrenal gland disease that underwent CT examination for acute spinal injuries. The mean nonenhanced attenuation value +/- SD of the left adrenal gland was 36.0 +/- 5.3 HU (range: 22.0-42.0 HU). The mean nonenhanced attenuation value +/- SD of the right gland was 34.3 +/- 7.0 HU (range: 20.4-48.6HU). The mean enhanced attenuation value +/- SD were: left gland 101.5 +/- 10.6HU (range: 86.8-128.0 HU), and right gland 97.4 +/- 12.4 HU (range: 58.9-123.6 HU). The mean CT volume +/- SD were: left gland was 0.60 cm3 (range: 0.20-0.95; SD 0.17), and right gland (0.55cm3, range: 0.22-1.01; SD 0.19). Attenuation values and volume data were related to age, weight, and gender, using ANOVA. There was no statistically significant difference between the left and right side or in adrenal measurements, because of body weight class effects. The animal effect was the most important source of variation for all adrenal measurements. Based on our study, CT is an effective method for assessing adrenal characteristics in the dog. Normative CT data are provided to allow estimation of normal adrenal gland size and volume.  相似文献

利用SPF鸡胚对鸡传染性法氏囊病超强毒vvIBDV Gx株进行培育,通过鸡胚成纤维细胞对病毒进行传代致弱,对其结构蛋白VP3基因核苷酸及推导的氨基酸序列进行分析,从而揭示vvIBDV Gx从超强毒力向弱毒力转化过程中基因的序列变化规律。对不同代次细胞毒序列分析的结果表明,细胞毒在第8代以前,VP3基因序列没有改变,与标准超强毒HK46株氨基酸同源性达99%以上;细胞毒第9代核苷酸和氨基酸序列发生了改变;10代毒VP3基因与标准弱毒P2株氨基酸序列同源性达100%;细胞适应毒传至20代,其VP3基因序列不再改变。从而说明,vvIBDV Gx株在致弱过程中,VP3基因也随之改变。  相似文献
