空肠弯曲菌共同抗原单克隆抗体的研制及其在细菌检验中的应用 Ⅱ.杂交瘤细胞株的建立及特性鉴定

应用B-淋巴细胞杂交瘤技术,建立了5株稳定分泌抗空肠弯曲菌共同抗原(CA)单克隆抗体(McAb)的杂交瘤细胞株(4A7,9D6、9C2、11A1、11D4)。在5株杂交瘤分泌的McAb中,有4株(4A7、9D6、11A1、11D4)针对CA中的耐热抗原成分,对其中4A7、9D6两株分泌的McAb进行免疫印迹分析表明,其与CA中大约为7.7×104u的蛋白移行带出现特异反应;一株(9C2)针对CA中的不耐热抗原成分。对杂交瘤细胞的核型分析表明,其染色体数在93-108之间。体外传代4个月,仍保持稳定分泌抗体的特性。这种McAb具有较强的特异性,除与幽门弯曲菌出现弱阳性反应外,与参试的其它31种非弯曲菌属的细菌均为阴性反应。对不同地区、不同来源的516株空肠弯曲菌的鉴定表明,制备的McAb与现有空肠弯曲菌均出现阳性反应,与常规鉴定法的符合率为100％。     

夹心酶联免疫吸附试验检测包虫病循环抗原

建立了检测包虫病循环抗原的夹心酶联免疫吸附试验。对１３０份阳性血清（猪６５，羊６５）作了检测，总阳性率为７９．２％，抗原最小检出量为５０μｇ／Ｌ。     

Cloning, characterization and expression of cDNA containing major histocompatibility complex class I, IIα and IIβ genes of Japanese flounder Paralichthys olivaceus

ABSTRACT:   The nucleotide sequences of Japanese flounder Paralichthys olivaceus , major histocompatibility complex (MHC) cDNA, classical MHC class Iα, non-classical MHC class Iβ, MHC class IIα and IIβ, were determined. The domain structures and antigen binding motifs of vertebrate MHC are conserved in the Japanese flounder MHC. A phylogenetic analysis supports the classification of these genes into class I and class II MHC. Classical MHC class Iα was ubiquitously expressed, whereas the non-classical MHC class Iβ was expressed mainly in lymphoid organs, gills, intestine and stomach. The MHC classes IIα and IIβ were also ubiquitously expressed.     

呋喃妥因代谢物化学发光酶联免疫检测方法的研究

对呋喃妥因代谢物1-氨基乙内酰脲(AHD)进行半抗原改造,采用活化酯法将半抗原与卵清蛋白(OVA)偶联为包被原,与标准品竞争AHD单克隆抗体的抗原结合位点,加入辣根过氧化物酶标记羊抗鼠二抗,建立了AHD间接竞争化学发光酶联免疫(CLEIA)检测法,并对化学发光液、包被原与抗体最优稀释度、包被条件、封闭液和竞争时间五项参数进行优化。结果表明:该方法具有良好的特异性,IC50为0.753 ng/m L,在鸡肉组织中的检测限为0.028μg/kg,添加回收率在80.54%~102.64%之间,变异系数均小于10%。与国标中液相色谱-串联质谱法(LC-MS/MS)和我国出入境行业标准中酶联免疫检测法(ELISA)相比,不仅降低了检测限,而且操作简便,为动物源性食品中呋喃妥因代谢物的残留提供了准确、便捷的分析检测手段。     

Identification of novel tumour‐associated antigens in canine mammary gland tumour

Canine mammary gland tumour (MGT) is the most common neoplasm in female dogs and has similar biological characteristics to human MGT. Spontaneous canine MGT is a more attractive clinical model in oncological research than that of the murine experimental model. Tumour‐associated antigens (TAAs), which are produced in tumour cells, are applied as tumour markers, tumour vaccine antigens and molecular targets of therapeutic drugs. In this study, we have primarily identified 13 different TAAs of canine MGT by serological immunoscreening of cDNA expression library. The results of serological mini‐arrays of identified antigens showed that CCDC41 antigen specially reacted with 35% of sera from MGT‐dogs and did not react with control sera. We also found that HSPH1 mRNA expression levels increased significantly in MGT tissues. These findings will contribute to the development of diagnostic technologies and translational target therapies for dogs. Clinical relevance : HSPH1, which is strongly expressed in the tumour tissue, will be a possible vaccine antigen of canine MGT.     

副猪嗜血杆菌外膜蛋白P5的B细胞抗原表位鉴定

副猪嗜血杆菌外膜蛋白P5能诱导机体产生免疫保护反应,同时可以用于特异性的血清学诊断,本试验选取P5蛋白进行抗原表位鉴定。首先通过PCR扩增P5F1（1～204aa）,P5F2（170～296aa）及P5F3（280～371aa）3个片段,PCR产物分别定向克隆到表达载体pET32a（＋）中表达纯化。根据ELISA和Western blotting结果确定P5F3片段（280—371aa）是OmpP5的免疫优势决定区。为了进-步对该免疫优势决定区进行抗原表住鉴定,设计了-套11个部分重叠的短肽,这些短肽覆盖全部280～371aa片段。每-个短肽合成1对寡核苷酸链,退火后插入表达载体pGEX-6p-1,与GST进行融合表达。用HPS阳性血清进行ELISA和Western blotting扫描,鉴定出其表位位于”。TGNTCDAVKGRKALIT351。通过序列分析证实该抗原表位在不同的HPS菌株中高度保守。本试验确定了位于HPSOmpP5上的-个抗原表位,为建立-种方便、快捷、适用于现地大规模样品检测的鉴别诊断方法奠定了基础,同时也为HPS新型亚单位疫苗的研制,以及研究病原茵感染和机体免疫过程中P5蛋白与宿主体内相应分子之间的相互作用提供了有用信息。     

猪繁殖与呼吸综合征病毒胶体金检测试纸卡的研制

为快速检测猪繁殖与呼吸综合征病毒,应用抗猪繁殖与呼吸综合征病毒单克隆抗体PRRSV-3D10株和4H11株分别作为胶体金标记物和诊断抗体,羊抗鼠IgG作为质控线,制备胶体金免疫层析抗原检测试纸。该试纸卡具有良好的特异性、敏感性、重复性和稳定性,与PCR方法对比总符合率为93．3％。研究显示,本试纸卡能够快速、准确检测猪繁殖与呼吸综合征病毒,可为临床诊断提供参考。     

Determination of carcinoembryonic antigen and cancer antigen (CA 15‐3) in bitches with tumours on mammary gland: preliminary report

The aim of this work was to determine levels of carcinoembryonic antigen (CEA) and cancer antigen (CA 15‐3) in the blood serum of 45 bitches. A modified procedure was used to determine the CEA and CA 15‐3 markers with the human kits using the radioimmunoassay method. Samples collected from extirpated tumour of mammary glands were histologically processed and classified as per WHO guidelines. The average age of animals with tumour was 10.00 ± 2.2 years; for healthy bitches average age was 4.2 ± 3.2 years. Values of CEA and CA 15‐3 were considered positive, if they exceeded 0.23 ng mL^?1 and 7 IU mL^?1, respectively. Average levels of CEA in the tumour group were 0.25 ± 0.06 versus 0.20 ± 0.03 in healthy bitches (P = 0.0001). The average CA 15‐3 value in bitches with tumour was 8.58 ± 1.27 versus 5.14 ± 1.34 in healthy animals (P < 0.0001).     

刺激隐核虫抑动抗原生物学信息及抗原特性分析

对刺激隐核虫抑动抗原基因进行生物学信息分析,利用抗原表位多肽制备了鼠源抗抑动抗原抗体,并研究其抗原特性.试验结果表明：利用表位串联多肽所制备的鼠源抗抑动抗原抗体包含有针对刺激隐核虫抑动抗原的特异性抗体,为刺激隐核虫单克隆抗体和疫苗的制备奠定了基础.
