Chemical Synthesis and Structure-Activity Relationship Study Yield Desotamide a Analogues with Improved Antibacterial Activity

Desotamides A, a cyclohexapeptide produced by the deep-sea-derived Streptomyces scopuliridis SCSIO ZJ46, displays notable antibacterial activities against strains of Streptococcus pnuemoniae, Staphylococcus aureus, and methicillin-resistant Staphylococcus epidermidis (MRSE). In this study, to further explore its antibacterial potential and reveal the antibacterial structure-activity relationship of desotamides, 13 cyclopeptides including 10 new synthetic desotamide A analogues and wollamides B/B1/B2 were synthesized and evaluated for their antibacterial activities against a panel of Gram-positive and -negative pathogens. The bioactivity data reveal that residues at position II and VI greatly impact antibacterial activity. The most potent antibacterial analogues are desotamide A4 (13) and A6 (15) where l-allo-Ile at position II was substituted with l-Ile and Gly at position VI was simultaneously replaced by d-Lys or d-Arg; desotamides A4 (13) and A6 (15) showed a 2–4-fold increase of antibacterial activities against a series of Gram-positive pathogens including the prevalent clinical drug-resistant pathogen methicillin-resistant Staphylococcus aureus (MRSA) with MIC values of 8–32 μg/mL compared to the original desotamide A. The enhanced antibacterial activity, broad antibacterial spectrum of desotamides A4 and A6 highlighted their potential as new antibiotic leads for further development.

利用雷州林业局刚果12号桉W5无性系林分中胸径8～18 cm径阶，树高11～23 m的样木1 207株和尾叶桉U6无性系林分中胸径8～18 cm径阶，树高10～21 m的样木784株，按材种规格标准造材实测材积，且分别建立刚果12号桉W5无性系和尾叶桉U6无性系胶合板材立木出材量二元回归方程，经检验精度可靠，可供参考使用。     

Prediction of bending properties for structural glulam using optimized distributions of knot characteristics and laminar MOE

This study established a prediction model for bending properties of glued-laminated timber (glulam) using optimized knot and modulus of elasticity (MOE) distributions of lumber laminate as the main input variables. For this purpose, knot and MOE data were investigated for all pieces of lumber that were prepared for glulam manufacturing, and statistical distributions of knot size, knot number in one lumber, and MOE of each laminate were optimized as distribution functions. These knot and MOE data were used as input variables in the prediction model for bending properties, and were also used in generating virtual glulam using the inverse transform method. Prediction of bending properties for glulam was carried out using the transformed section method, which is partially provided in ASTM D 3737 (Annex A4). Predicted values were compared with those from full-scale four-point bending tests for 60 six-layered glulams with 10 different laminar combinations. Finally, the allowable bending properties of glulam for each specific laminate combination were determined by calculating the fifth percentile of the modulus of rupture and the average modulus of elasticity from virtual test results of more than 1000 virtual glulams. From the results of this study, predicted bending properties for glulam and their distributions could be used for structural design in both allowable stress design and limit state design.

应用荧光定量RT-PCR检测J亚群禽白血病病毒

针对J亚群禽白血病病毒（ALV-J）基因序列保守区域设计一对特异性引物和一条特异性探针,通过构建重组阳性标准质粒作为阳性标准品,建立了检测ALV-J核酸的荧光定量PCR方法。优化反应体系和条件后进行特异性、敏感性、重复性试验。结果显示,该检测方法特异性强,与其它禽源病毒如A亚群禽白血病病毒（ALV-A）、B亚群禽白血病病毒（ALV-B）、新城疫病毒（ NDV）、禽流感病毒（AIV）、鸡传染性贫血病毒（CIAV）和马立克病病毒（MDV）均不发生交叉反应;该方法可检测到3.2×102拷贝/μL的病毒核酸,与常规RT-PCR相比,敏感性高100倍;重复性试验的变异系数小于2%。研究结果表明,建立的Real-time RT-PCR 检测方法特异性强、灵敏度高、可重复性好,可用于ALV-J的定量检测。     

大棚丝瓜长季节栽培密度试验

为充分挖掘丝瓜长季节栽培技术的增产潜力,以品种、密度2个因素进行大棚设施丝瓜长季节栽培高产优质技术与经济效益比较研究。试验结果表明,在种植密度为1 000~3 500株/667 m^2范围内,产量以3 000株/667 m^2最高,经济效益以2 000株/667 m^2(绿王子)、1 500株/667 m^2(三比2号)最好。
