The present study aimed at analyzing the genetic variability of indigenous goat breeds (Capra hircus) using the MHC-associated microsatellite markers BF1, BM1818, BM1258, DYMS1, and SMHCC1. The following breeds were included: Chinese Xuhuai, Indian Changthangi and Pashmina, Kenyan Small East African (SEA) and Galla, and Albanian Vendi. To examine genetic variability, the levels of heterozigosity, degrees of inbreeding, and genetic differences among the breeds were analyzed. The mean number of alleles ranged from nine in the Galla to 14.5 in the Vendi breed. The mean observed heterozygosity and mean expected heterozygosity varied from 0.483 in the Vendi to 0.577 in the Galla breed, and from 0.767 in the SEA to 0.879 in the Vendi breed, respectively. Significant loss of heterozygosity (p < 0.01) indicated that these loci were not in Hardy-Weinberg equilibrium. The mean FIS values ranged from 0.3299 in the SEA to 0.4605 in the Vendi breed with a mean value of 0.3623 in all breeds (p < 0.001). Analysis of molecular variance indicated that 7.14% and 4.74% genetic variation existed among the different breeds and geographic groups, whereas 92.86% and 95.26% existed in the breeds and the geographic groups, respectively (p < 0.001). The microsatellite marker analysis disclosed a high degree of genetic polymorphism. Loss of heterozygosity could be due to genetic drift and endogamy. The genetic variation among populations and geographic groups does not indicate a correlation of genetic differences with geographic distance.  相似文献

This study was aimed to investigate the genetic diversity of swamp buffalo breeds in China.The analysis of genetic diversity was performed in 40 buffalo individuals from 8 buffalo breeds(Dechang buffalo,Dehong buffalo,Wenzhou buffalo,Guizhou buffalo,Xilin buffalo,Fuzhong buffalo,Murrah buffalo and Nili-Ravi buffalo)by 30 microsatellite loci and LabChip chip test method.The results showed that 332 alleles at 30 microsatellite loci were found in 8 buffalo breeds,the average values of gene diversity and PIC were 0.7808 and 0.7554,respectively.Cluster analysis indicated that Dechang buffalo and Dehong buffalo firstly clustered together,followed by Fuzhong buffalo,Guizhou buffalo,Wenzhou buffalo and Xilin buffalo.Moreover,Murrah buffalo and Nili-Ravi buffalo clustered together.Our findings revealed that the 30 microsatellite loci could be used as an effective genetic marker for the analysis of genetic diversity among the buffalo breeds,which enriched the current SSR marker resources in buffalo.  相似文献

为探究中国沼泽型水牛品种或类群的遗传多样性,本研究采用了沼泽型水牛30个微卫星标记结合LabChip芯片检测法对德昌水牛、德宏水牛、温州水牛、贵州水牛、西林水牛、富钟水牛及两个引进品种摩拉水牛、尼里-拉菲水牛等40个样本进行了检测和遗传多样性分析。结果表明,在30个微卫星DNA标记上共发现332个等位基因,平均基因多样性和PIC值分别为0.7808和0.7554。聚类分析结果显示,德宏水牛和德昌水牛先聚为一类,随后与富钟水牛、贵州水牛、温州水牛、西林水牛聚为一类;摩拉水牛与尼里-拉菲水牛聚为一类。本研究证实了所选的30个沼泽型水牛微卫星DNA标记可作为有效的遗传标记用于水牛品种间的遗传多样性分析,同时也丰富了当前的水牛微卫星标记资源。  相似文献

The genetic and demographic bottleneck analysis of Indian camel breeds was carried out utilizing 40 microsatellite markers. Allelic polymorphism was observed at 20 loci in the Indian dromedary breeds. A total of 66 alleles were scored. The average number of alleles, expected heterozygosity and polymorphic information content were, respectively, 3.25?±?0.27, 0.56?±?0.04 and 0.49?±?0.04 in Bikaneri; 3.25?±?0.25, 0.53?±?0.03 and 0.46?±?0.03 in Jaisalmeri; 3.0?±?0.21, 0.53?±?0.04 and 0.45?±?0.03 in Kachchhi and 3.1?±?0.19, 0.51?±?0.03 and 0.44?±?0.03 in Mewari breed. Higher genetic variation was observed in most numerous Bikaneri breed. Genetic distances were least between the breed pair Bikaneri and Jaisalmeri which was closely placed with the Kachchhi breed. The Mewari camels had relatively higher genetic distance from the other three Indian dromedary breeds. The bottleneck analysis revealed the presence of genetic bottleneck in all four breeds of Indian dromedary. However, the qualitative graphical method resulted in normal L-shaped distribution of allele frequencies in Jaisalmeri breeds and shifted mode in Bikaneri, Kachchhi and Mewari breeds. The demographic bottleneck analysis revealed minimum reduction (?9.65 %) in the population of camels in Jaisalmeri breeding tract as compared to that of Bikaneri (?14.18 %), Kachchhi (?27.78 %) and Mewari (?32 %) breeding tracts. Conclusively, the genetic bottleneck analysis could explain the demographic bottleneck in the Indian dromedary populations. Therefore, appropriate conservation and improvement efforts are needed in all four dromedary breeds with immediate attention on Mewari and Kachchhi breeds. The present study is the first report in demonstrating the genetic basis of demographic bottleneck in the Indian dromedary populations.  相似文献

利用27个微卫星DNA标记对12个地方乌骨鸡品种进行遗传多样性分析,通过计算等位基因频率、多态信息含量、杂合度、有效等位基因数和遗传距离,评估各品种内遗传变异和各品种间遗传相关,并根据遗传距离对12个鸡品种进行了聚类分析。结果表明,各乌骨鸡群体的遗传多样性较为丰富,并具有较高的选择潜力。平均杂合度最高的是竹乡鸡,为0.670;最低的是江山乌骨鸡,为0.581。27个微卫星标记中4个为中度多态座位,1个低度多态座位,22个为高度多态座位;LEI0234与LEI0192分别检测到了10.3与12.2个等位基因。以标准遗传距离为准,遗传距离最近的为沐川乌骨鸡与略阳鸡,为0.1002,而乌蒙乌骨鸡与略阳鸡最远,为0.2546。通过UPG—MA法聚类后,略阳鸡、沐川乌骨鸡、兴文乌骨鸡、盐津乌骨鸡、竹乡鸡首先聚为1类,江山乌骨鸡、郧阳白羽乌鸡、余干乌骨鸡聚为1类;金湖乌凤鸡、丝绒乌骨鸡和丝毛乌骨鸡聚为第3类;乌蒙乌骨鸡独为一类。  相似文献

福安水牛是福建省地方优良品种之一,为了进一步阐明其群体遗传结构和遗传多样性,采用8对微卫星引物对福建福安、闽清、古田及南靖等4个福安水牛群体,共计130头样本进行检测分析。结果表明:福安水牛4个群体8个微卫星座位共有78个等位基因,其中4个群体共享的等位基因为50个,非共享等位基因28个。各等位基因频率介于0.015 2~0.527 8之间;共检测到基因型6~29种;4个群体的平均多态信息含量、杂合度、有效等位基因数分别为0.817 2±0.023 0、0.820 4±0.029 4和5.85±0.84;根据奈氏标准遗传距离进行UPGMA聚类分析,结果表明:遗传距离最远的是闽清群体与南靖群体(0.351 8),遗传距离最近的是福安群体与闽清群体(0.183 2)。  相似文献

The Lanyu pig is an indigenous miniature pig breed on Lanyu Islet near Taiwan, with a mitochondrial DNA genetic lineage remote from Asian and European pig breeds. The unknown population genetic structure and increased inbreeding among the small population of conserved Lanyu pigs is now of great conservation concern. Additionally, the presence for more than a century of exotic pig breeds in Taiwan has made gene introgression from exotic pig breeds into Lanyu pigs very possible. The present study thus aimed to investigate nuclear genetic variation within the conserved Lanyu pigs and the phylogenetic relationship and possible genetic introgression between Lanyu and exotic pig breeds by determining the polymorphism of 19 microsatellite loci. In the neighbor-joining tree constructed from 7 pig breeds based on Cavalli-Sforza and Edward chord genetic distances, 3 major clades were recognized, in which the Asian and European breeds were separately clustered into 2 clades with a 93.0 and 99.9% bootstrap confidence value, respectively. All individuals of the Lanyu breed formed a unique subclade within the Asian clade based on the distance of the proportion of shared alleles, -ln(ps), suggesting that the Lanyu breed possesses a unique nuclear genetic structure and that no nuclear gene introgression from exotic breeds into the conserved Lanyu pigs has occurred in recent history. Fifteen of 19 microsatellite loci deviated from Hardy-Weinberg equilibrium (by Wright's statistic), suggesting a significant loss of heterozygosity in the conserved population. The valuable nuclear genetic structure and phylogenetic information should assist future conservation and population management of Lanyu pigs.  相似文献

为分析福建黄兔、新西兰兔、日本大耳白兔群体内的遗传变异情况和群体间的遗传相关性,本试验选取3个品种兔各25只,分别耳静脉采血并采用试剂盒抽提全血基因组,运用15个微卫星标记,结合荧光PCR技术和毛细管电泳方法获得目的片段,通过计算等位基因数、有效等位基因数、杂合度、多态信息含量和遗传距离,分析3个品种兔的遗传多样性。结果显示,3个品种兔在15个座位共检测到110个等位基因,平均等位基因数为7.3个,其中福建黄兔、新西兰兔和日本大耳白兔分别检测到95、95和88个等位基因,且分别在11、12和11个微卫星座位表现为高度多态,表明3个品种兔在筛选的15个微卫星座位均呈现较丰富的遗传多态信息。Hardy-Weinberg遗传平衡检验结果表明,福建黄兔、新西兰兔和日本大耳白兔分别有8、8和6个座位处于Hardy-Weinberg不平衡状态。Nei氏遗传距离分析显示,福建黄兔与新西兰兔、福建黄兔与日本大耳白兔、新西兰兔与日本大耳白兔的Nei氏遗传距离分别为0.1761、0.2347和0.0432。遗传距离越小时,相似度越高,亲缘关系越近,因此,新西兰兔与日本大耳白兔间的亲缘关系最近,福建黄兔与日本大耳白兔间的亲缘关系最远。  相似文献

利用30个微卫星座位,对中国6个群体山羊(阿尔巴斯、二狼山、乌珠穆沁、阿拉善、辽宁绒山羊和陕南白山羊),共计240个个体的遗传多样性进行了分析。计算了各群体的等位基因频率,并以其为基础获得了各群体的平均杂合度(He)、平均多态信息含量(PIC)和平均有效等位基因数(Ne),分别为0.367~0.467、0.533~0.639、2.571~4.915。分析了群体内和群体间的遗传变异,并根据遗传距离,进行了NJ聚类,结果将6个山羊群体分为两大类。为进一步对山羊品种的保存和利用提供科学的依据。  相似文献

The distribution of genetic diversity and the genetic relationships among western Mediterranean horse breeds were investigated using microsatellite markers. The examined sample included seven Spanish and three Italian local horse breeds and populations, plus a Spanish Thoroughbred outgroup. The total number of animals examined was 682 (on average 62 animals per breed; range 20–122). The microsatellite marker set analysed provided 128 alleles (10.7 alleles per locus). Within‐breed genetic diversity was always high (>0.70), with breeds contributing about 8% of the total genetic variability. The mean molecular coancestry of the entire population examined was 0.205, Losino being the breed that contributed most. In addition to Nei's standard and Reynolds’ genetic distances, pair‐wise kinship distance and molecular coancestry were estimated. Remarkably similar breed rankings were obtained with all methods. Clustering analysis provided an accurate representation of the current genetic relationships among the breeds. Determining coancestry is useful for analysing genetic diversity distribution between and within breeds and provides a good framework for jointly analysing molecular markers and pedigree information. An integrated analysis was undertaken to obtain information on the population dynamics in western Mediterranean native horse populations, and to better determine conservation priorities.  相似文献

In this study, genetic analyses of diversity and differentiation were performed on five horse breeds raised in Algeria (Barb, Arab‐Barb, Arabian, Thoroughbred and French Trotter). All microsatellite markers were highly polymorphic in all the breeds. A total of 123 alleles from 14 microsatellite loci were detected in 201 horses. The average number of alleles per locus was the highest in the Arab‐Barb horses (7.86) and lowest in the thoroughbred breed (5.71), whereas the observed and expected heterozygosities per breed ranged from 0.71 (Thoroughbred) to 0.752 (Barb) and 0.71 (Thoroughbred) to 0.77 (Arab‐Barb), respectively. The genetic differentiation between the breeds was significant (p < 0.01) based on the infinitesimal model (FST). Three different approaches for evaluating the genetic relationships were applied. Genetic distances, the factorial correspondence analysis and structure analysis showed that a significant amount of genetic variation is maintained in the native horse populations and the other breeds. The Barb and Arab‐Barb breeds seem to be the most genetically related and support the decision to consider the breeds as same population.  相似文献

Polymorphisms for seven microsatellite loci in three red deer subspecies (9 populations) found in XinJiang were detected by polymerase chain reaction (PCR), 12% nondenaturation polyacrylamide gel electrophoresis and the Sanguinetti silver staining method. Numbers of alleles, average effective numbers of alleles (E) and the average rate of homozygosity, allelic frequencies of seven microsatellite loci, polymorphism information content (PIC), mean heterozygosity (H) and genetic distances among the populations were calculated for each population. Dendrograms were constructed based on genetic distances by the neighbor‐joining method (NJ), utilizing molecular evolutionary genetics analysis software PHYLIP (3.6). The phylogenetic tree was constructed based on allelic frequencies using maximum likelihood (ML); the bootstrap value was estimated by bootstrap test in the tree. Lastly, phylogenesis was analyzed. The results showed that four of the seven microsatellite loci were highly polymorphic, but BMS2508 and Celjp0023 showed no polymorphism and BM5004 was a neutral polymorphism. It is our conclusion that the four microsatellite loci are effective DNA markers for the analysis of genetic diversity and phylogenetic relationships among the three red deer subspecies. The mean PIC, H and E‐values across the microsatellite loci were 0.5393, 0.5736 and 2.64, which showed that these microsatellite loci are effective DNA markers for the genetic analysis of red deer. C.e. songaricus populations from Regiment 104, 151 and Hami are clustered together. C.e. yarkandensis populations from Regiment 35, Xaya and Alaer are clustered together. These two clusters also cluster together. Lastly, C.e. sibiricus populations from Burqin, Regiment 188 and the first two clusters were clustered together. The phylogenetic relationship among different red deer populations is consistent with the known origin, history of breeding and geographic distributions of populations.  相似文献

The objective of this study was to evaluate the genetic structure of Mexican Criollo cattle populations using microsatellite genetic markers. DNA samples were collected from 168 animals from four Mexican Criollo cattle populations, geographically isolated in remote areas of Sierra Madre Occidental (West Highlands). Also were included samples from two breeds with Iberian origin: the fighting bull (n = 24) and the milking central American Criollo (n = 24) and one Asiatic breed: Guzerat (n = 32). Genetic analysis consisted of the estimation of the genetic diversity in each population by the allele number and the average expected heterozygosity found in nine microsatellite loci. Furthermore, genetic relationships among the populations were defined by their genetic distances. Our data shows that Mexican cattle populations have a relatively high level of genetic diversity based either on the mean number of alleles (10.2-13.6) and on the expected heterozygosity (0.71-0.85). The degree of observed homozygosity within the Criollo populations was remarkable and probably caused by inbreeding (reduced effective population size) possibly due to reproductive structure within populations. Our data shows that considerable genetic differentiation has been occurred among the Criollo cattle populations in different regions of Mexico.  相似文献

本研究利用10个微卫星标记分析了鲁西黄牛、布莱凯特肉牛、渤海黑牛和日本和牛4个品种185个个体的品种内遗传变异情况及各品种间的亲缘关系,共检测到58个等位基因,每个位点平均观察等位基因数为5.8个,从4(BM1818)到8个(ETH225和TGLA53)不等;有效等位基因数为2.5417~3.5849个;观察杂合度、期望杂合度和多态信息含量变异范围分别为0.2270~0.5459、0.6082~0.7230和0.5482~0.6791,均属高度多态性位点。群体间遗传分化明显,有34.49%的遗传分化来自群体间的变异。以Nei氏遗传距离(DA)构建UPGMA系统进化树,聚类结果表明,渤海黑牛与鲁西黄牛首先聚为一类,布莱凯特肉牛和日本和牛聚为一类。布莱凯特肉牛的3个微卫星座位10个克隆的PCR扩增片段测序获得的序列已提交GenBank,登录号分别为:GQ368896、GQ368897、GQ368898、GQ368899、GQ368900、GQ368901、GQ368902、GQ368903、GQ368904、GQ368905。10对微卫星座位可作为有效的遗传标记用于4个肉牛品种的遗传多样性和系统发生关系分析。  相似文献

利用10个微卫星DNA标记对七个家兔品种进行了遗传检测。利用变性聚丙烯酰胺凝胶电泳检测微卫星的PCR扩增产物,计算了七个家兔品种(9个家兔群体)样本在10个微卫星基因座上的等位基因频率、微卫星的多态信息含量(PIC)、各群体遗传杂合度、群体间的遗传距离、并根据遗传距离对这几个群体进行了聚类分析。结果表明:所选择的10个微卫星位点在所检测群体中均表现了较好的多态性,平均多态信息含量为0.569905。平均杂舍度范围在0.5885~0.6826。表明所研究的家兔品种的遗传多样性较丰富,还可以做进一步的选择利用。根据遗传距离所做的聚类分析图表明吉戎-Ⅰ系和吉戎-Ⅱ系、哈尔滨大白兔、塞北兔的亲缘关系依次较近,与它们的培育历史相吻合。  相似文献

利用微卫星DNA多态性预测引进肉用绵羊品种杂种优势   总被引:4,自引:2,他引:2  
旨在利用微卫星DNA在不同绵羊群体中的多态性预测引进肉用绵羊品种与地方绵羊品种的杂种优势。利用23个微卫星基因座对4个国外引进肉羊品种及3个地方绵羊品种的群体多态信息含量、有效等位基因数、杂合度和遗传距离进行遗传检测,并测定引进肉羊与地方肉羊品种的杂交效果。结果,23个微卫星座位在7个绵羊群体中均呈现高度多态,共检测到348个等位基因,平均每个座位等位基因数为15个;多态信息含量在0.532 1~0.936 1,有效等位基因数在2.572 8~9.345 8,平均杂合度在0.549 2~0.936 0,品种平均杂合度在0.714 2~0.829 5,可以用于绵羊遗传多样性的评估。从不同品种看,无角陶赛特的遗传变异最大,而小尾寒羊的遗传变异相对较小;经遗传关系分析,在引进的4个肉羊品种中,与小尾寒羊、蒙古羊及滩羊杂交优势由大到小依次为白萨福克、波德代、无角陶赛特、特克赛尔,与实际杂种优势测定结果一致。利用微卫星DNA多态性预测引进肉用绵羊品种与小尾寒羊、蒙古羊及滩羊的杂种优势是可行的,这将对今后绵羊育种具有重要的应用价值和指导意义。  相似文献

通过筛选的28个多态性较好的微卫星标记,检测了我国24个地方鸭品种的遗传多样性。利用等位基因频率计算了各群体的遗传参数和群体间的遗传距离,采用邻接法构建了聚类图,并进行了系统发生分析。结果表明:28个微卫星座位在我国家鸭群体中的多态信息含量除APL23和APL79为中度多态外,其他座位均为高度多态座位,可以作为有效的遗传标记用于鸭种之间遗传多样性分析和系统发生关系的分析;我国所有家鸭群体平均杂合度(0.569)低于国内的家鸡和家鹅,其遗传多样性相对贫乏;聚类结果分析表明了各家鸭品种的分子系统发生关系与其育成史、分化及地理分布比较一致。  相似文献

Seven microsatellite markers were used to study genetic diversity of three Egyptian (Egyptian Baladi, Barki and Zaraibi) and two Italian (Maltese and Montefalcone) goat breeds. The microsatellites showed a high polymorphic information content (PIC) of more than 0.5 in most of the locus-breed combinations and indicated that the loci were useful in assessing within- and between-breed variability of domestic goat (Capra hircus). The expected heterozygosity of the breeds varied from 0.670 to 0.792. In the geographically wider distributed Egyptian Baladi breed there were indications for deviations from random breeding. Analysis of genetic distances and population structure grouped the three Egyptian goat breeds together, and separated them from the two Italian breeds. The studied Mediterranean breeds sampled from African and European populations seem to have differentiated from each other with only little genetic exchange between the geographically isolated populations.  相似文献

利用29对微卫星引物对我国的12个地方鸡种进行群体遗传结构及遗传多样性评估,结果表明:每个座位的等位基因数从2到23不等,所有的12个群体均显示较高水平的杂合度,固始鸡最低,为0.4532,大骨鸡最高,为0.6271。鸡种间存在较大的遗传分化,16.7%的遗传变异源于品种间的差异。基于Reynolds’遗传距离的邻近(NJ)聚类图显示,12个群体可以分为2个主要的类群:重体型的鸡种首先聚在一起,然后轻体型鸡种加入。structure程序同时也被用于评价鸡种间的遗传关系,结果与NJ聚类法相似,重体型的鸡种首先与轻体型鸡种分成两类。  相似文献
