为探究不同施肥和保水措施对油茶土壤N_2O排放的影响,采用静态暗箱-气相色谱法,设置对照(B0CK)、氮肥(N,0.13 g N·kg~(-1))、磷肥(P,0.065 g P·kg~(-1))、氮磷肥(NP,0.13 g N·kg~(-1)+0.065 g P·kg~(-1))、低复合保水材料(生物炭和聚丙烯酰胺,B1,每盆13.65 g炭+1.35 g聚丙烯酰胺)、高复合保水材料(生物炭和聚丙烯酰胺,B2,每盆27.30 g炭+2.70 g聚丙烯酰胺)、低复合保水材料和N(NB1)、高复合保水材料和N(NB2)、低复合保水材料和P(PB1)、高复合保水材料和P(PB2)、低复合保水材料和NP(NPB1)、高复合保水材料和NP(NPB2),共12个处理,进行不同施肥和保水措施下土壤N_2O排放的差异比较。结果表明,N、P添加均显著增加土壤N_2O的累积排放量,NP添加与对照无差异。施加复合保水材料抑制土壤N_2O的排放,随着复合保水材料施用量的增加,土壤N_2O的排放显著降低,与对照相比,B1和B2处理N_2O减排50%以上。N添加条件下,与对照相比,添加复合保水材料NB1、NB2的N_2O累积排放显著降低。P与复合保水材料无交互作用。N、P和复合保水材料对土壤N_2O累积排放量具有显著作用,在NP同施时,与对照相比,添加复合保水材料NPB1、NPB2的N_2O累积排放分别降低了1.18%、30.69%。因此,高复合保水材料类型的施肥措施对减少油茶土壤N_2O排放具有重要意义,从而对缓解全球气候变化具有重要影响。  相似文献

Excessive fertilization combined with unreasonable irrigation in farmland of the Hetao Irrigation Area (HIR), China, has resulted in a large amount of nitrogen (N) losses and agricultural non-point source pollution. Application of soil amendments has become one of the important strategies for reducing N losses of farmland. However, there is still no systematic study on the effects of various soil amendments on N losses in the HIR. In this study, three types of soil amendments (biochar, bentonite and polyacrylamide) were applied in a maize-wheat rotation system in the HIR during 2015-2017. Yields of maize and wheat, soil NH3 volatilization, N2O emission and NO3- leaching were determined and soil N balance was estimated. The results showed that applications of biochar, bentonite and polyacrylamide significantly increased yields of maize by 9.2%, 14.3% and 13.3%, respectively, and wheat by 9.2%, 16.6% and 12.3%, respectively, compared with the control (fertilization alone). Applications of biochar, bentonite and polyacrylamide significantly reduced soil N leaching by 23.1%, 35.5% and 27.1%, soil NH3-N volatilization by 34.8%, 52.7% and 37.8%, and soil N surplus by 23.9%, 37.4% and 30.6%, respectively. Applications of bentonite and polyacrylamide significantly reduced N2O-N emissions from soil by 37.3% and 35.8%, respectively, compared with the control. Compared with application of biochar, applications of bentonite and polyacrylamide increased yields of maize and wheat by 5.1% and 3.5%, respectively. Our results suggest that soil amendments (bentonite and polyacrylamide) can play important roles in reducing N losses and increasing yield for the maize-wheat rotation system in the HIR, China.  相似文献

A method for fractionating sorghum proteins using extraction solvents and techniques designed to obtain polymeric protein structures (especially disulfide linked) was developed. Extraction and separation conditions were optimized in terms of completeness of protein extraction, sample stability, and analytical resolution. After pre-extraction of albumins and globulins, a 3-step sequential procedure involving no reducing agents was applied to ground whole sorghum flour. The three fractions obtained represented proportionally different protein polymer contents and molecular weight distribution as evidenced by comparative size exclusion chromatography. Protein composition also varied among the extracts with differences in kafirin composition and non-kafirin proteins detected in the fractions by RP-HPLC and SDS-PAGE analysis. The ability to quantify and further characterize sorghum polymeric protein complexes will be useful for additional studies linking protein structures with functionality and digestibility and variations for these properties within diverse sorghum germplasm.  相似文献

采用人工模拟降雨溅蚀方法,通过采样制作土壤切片,研究聚丙烯酰胺(PAM)对黄土结皮形成的影响。结果表明,PAM能改善土壤结构状况,显著提高土壤团聚体的稳定性,从而有效地抑制土壤结皮形成;但是随着降雨历时的延长,这种抑制作用逐渐被雨滴破坏;试验筛选出PAM-1抑制土壤结皮形成效果较好。  相似文献

Background   Several types of organic polymers with various molecular weights and surface charges are produced for various industrial and agricultural purposes, and recent studies have shown that high molecular weight anionic polymers are the most effective in reducing runoff and soil loss. However, anionic polyacrylamide (PAM) is marketed under various commercial names with variable molecular weight and length according to its chemistry (Barvenik 1994). Consequently, its interactions with soil and efficiency in reducing soil erosion are expected to vary. Moreover, types of PAM other than those recommended for erosion control may also be effective in reducing soil erosion. Objectives   The objective of this research is to test the influence of 9 types of polyacrylamide (PAM) on water infiltration and soil loss of an arid silt loam soil under simulated rainfall conditions. Method  logy. Nine types of organic polymers varied in surface charge, ranging from +20 to –75, and molecular weight ranging from very low to very high were used in this study. Soil plots with dimensions of 60 cm by 120 cm having moderate slope of3 to 5% were constructed in the field and equipped with runoff collection installations. A specific type of PAM was spread on soil plots at a rate of 10 kg/ha. Rainfall was supplied from a drip type rainfall simulator at constant intensity and duration of 80 mm/h and 50 min, respectively. Runoff, infiltration and soil loss were measured for each plot subjected to a specific type of PAM and compared to control plots. Results and Conclusions   Land application of PAMs significantly increased infiltration prior to runoff but had insignificant effect on total infiltration volume and final infiltration rate under the present experimental conditions. Experimental results confirmed that anionic polymers were more effective in reducing soil loss than that of non-ionic or cationic polymers. In fact the cationic, low molecular weight and low surface charge non-ionic PAMs had insignificant effect on soil loss. High molecular weight anionic PAM with 30% surface charge (A-130) was more effective(46% reduction) than those having a high molecular weight and a lower surface charge of 20% (A-110 and A-120) with an aver-age soil loss reduction of 24%. The PAM A-836, described assoil erosion polymer with 20% surface charge and high molecular weight, reduced soil erosion by 41%. Unexpectedly, the anionic PAM with low molecular weight but very high surface charge was also effective in reducing soil loss (47% reduction). Recommendations and Outlook   Land application of PAM improves water infiltration and highly reduces soil erosion thus improving agricultural production. Application of PAM could be the only viable way to improve arid land farming. It seemed that various types of PAMs, other than those specifically produced for erosion protection can be used to combat soil erosion. This means that farmers in poor countries can use any type of PAM that is available to them to improve agricultural production.  相似文献

该文研究土壤结构改良剂对土壤水分动力学参数的影响。通过对试验数据进行初步分析后得出:加入土壤结构改良剂后,土壤饱和导水率有所提高;各处理的非饱和扩散率与对照相比,施加土壤结构改良剂的处理,在水平土柱试验中,远水端同一距离处土壤含水率要低于对照处理的含水率;土壤结构改良剂具有良好的吸水和保水性能,使得土壤的持水能力增强。在水势相同的情况下,与对照相比,施加土壤结构改良剂的土壤可保持更多的水分,并增加土壤中有效水含量。施加部位不同,土壤结构改良剂对土壤所持水分的含量也有较大差别,说明在实际应用中土壤结构改良剂的施用方法和施用深度也是影响土壤水分状况的一个较为重要的因素。  相似文献

为改良DNA聚丙烯凝胶电泳(PAGE)银染方法,采用PCR方法扩增鸡基因组微卫星DNA序列后,经PAGE分离扩增产物和pBR322 DNA/BsuRI标准分子质量,利用2种银染法对微卫星位点进行检测,分析其灵敏度和可行性。结果表明,2种方法都能染出Marker的所有条带,灵敏度均为25 ng,但在染色所需时间和可操作性方面,改进的染色方法优于Sanguinetti等所建立的方法。改进的银染方法染色过程较快,仅需25 min左右,且染色液和显影液可回收利用。该方法能显著提高检测分辨率,在实际应用中提高了染色效率。  相似文献

Digestive enzymes of Cirrhinus mrigala (Ham.) were studied during ontogenic development. Specific amylase activity was detected in first feeding fish. The enzyme activity decreased up to day‐18 and then it increased with the age of fish to reach the highest level on day‐34. Protease activity was 28.61 ± 8.90 mU mg protein?1 min?1 on day‐4 and increased with the age throughout the study period. Trypsin activity was 31.86 ± 1.12 mU mg protein?1 min?1 on day‐4. The activity decreased up to day‐10 and from day‐12 onwards increased up to day‐26. Chymotrypsin activity was 14.56 ± 2.74 mU mg protein?1 min?1on day‐4 and constantly increased up to day‐26. A significant increase in lipase activity was observed between days‐24 and 34. SDS‐PAGE and substrate SDS‐PAGE showed the diversity of protein (17.4–127.8 kDa) and protease activity bands (16.6–88.8 kDa) during ontogenesis. Soybean trypsin inhibitor, phenyl methyl sulphonyl fluoride, N‐α‐p‐tosyl‐l ‐lysine chloromethylketone and N‐tosyl‐l ‐phenylalanine chloromethylketone inhibited the protease activity up to 79.72–97.21, 65.55–94.83, 45.41–75.31 and 40.78–64.72%, respectively. Inhibition study in substrate SDS‐PAGE revealed the abundance of serine proteases and the presence of isoforms of trypsin and chymotrypsin. Ethylenediamine‐tetraacetate showed 5.56–22.78% inhibition of metal ion‐specific enzyme activity.  相似文献

Abstract. The stability of aggregates can be improved by amending the soil with charged synthetic polymers. We have evaluated the effects of addition of low rates of anionic polyacrylamide (PAM) on the stability of aggregates from three predominately kaolinitic soils, and investigated whether the applied PAM penetrated into the aggregates or was adsorbed onto exterior surfaces only. Aggregates (6.3–9.5 mm) from the Ap horizons of a Cecil sandy loam and a Cecil loamy sand (Typic Kanhapludult) and from the Bt horizon of a Davidson clay (Rhodic Kandiudult) were treated with a high molecular weight(2 × 107Da), negatively charged (20% hydrolysis) PAM at rates of 0, 10 or 20 kg/ha. The treated aggregate were dried and exposed to 20 mm of high kinetic energy (23.0 kJ/m3) simulated rain. Some of the PAM-treated Cecil loamy sand aggregates (5 or 10 kg/ha) were broken in half to expose untreated aggregate interiors to the rain. Percentage stable aggregates (weight/weight) in the PAM treatments ranged from 52 to 97%, vs. 20-32% in the control. PAM addition significantly increased the percentage of >4 mm sized aggregates compared with the untreated aggregates. PAM was more effective at stabilization in the light to medium textured Cecil soils than in the clayey Davidson. When internal surfaces of PAM treated aggregates were exposed, the percentage of stable aggregates exceeded the amount expected if the polymer was only adsorbed on to external surfaces. This suggested that some PAM had penetrated to some degree into aggregate interiors, thereby stabilizing both external and internal aggregates surfaces. Based on these results it is envisaged that use of PAM could be a viable alternative to the commonly used soil and water conservation practices (e.g. mulching, dyking, contour tillage).  相似文献
