ABSTRACT: Protoplast fusion between different species of Streptomyces was performed using a liquid regeneration method developed for a rapid and simple preparation of the fusants. Consequently, new clones, which could not be obtained using the conventional agar regeneration method, were obtained. In the crosses between S. griseus and S. durhamensis , and between S. californicus and S. catenulae , eight and two recombinants, respectively, were obtained using the liquid regeneration method. Conversely, in the case of crosses between S. ornatus and S. catenulae , and between S. ornatus and S. vendargensis , seven recombinants each were obtained using only the agar method. The physiological characteristics, such as the assimilation of carbohydrate and antibiotic resistance, of these fusants differed considerably from those of their parental strains. Using the proposed liquid regeneration method, a simpler and quicker procedure for protoplast fusion is described.  相似文献

We aimed to improve the purification of citrus Huanglongbing (greening) bacterium (HB), Candidatus Liberobacter asiaticum and to produce an antiserum against HB. Periwinkle plants Catharantus roseum L. graft-inoculated with HB were used to produce an antiserum. All young leaves of new shoots incubated at 20–25°C and 25–30°C, a few mature leaves incubated at 20–25°C, and all mature leaves incubated first at 25–30°C and later transferred to 20–25°C developed yellowing symptoms and were then used to prepare immunogen. The HB was partially purified from these leaves by an improved method that included a macerating enzyme treatment of the midribs of infected leaves and homogenization of infected phloem sieve tissues. An antiserum raised against partially purified HB reacted clearly at a dilution of 1/16 with HB-infected citrus extract prepared at a concentration of 40 times, but did not react with healthy or tristeza virus-infected citrus extract in microprecipitin tests. Feeding artificial micro diets instead of diatoms may allow a more constant and controlled form of nutrition, and thus results in lower mortality rate and increased growth. A 4-week trial was conducted to determine the growth and survival of postlarval Haliotis discus discus (Reeve.) postlarvae fed three experimental micro particle diets, and naturally occurring diatoms. A completely randomized design was adopted with each diet being replicated three times. All diets varied in terms of particle size and gross composition. Survival rate was significantly lower ( P  < 0.05) in the diatom diet (19.5 ± 2.1%) when compared with the K-1 diet (38.8 ± 8.9%), the K-2 diet (43.7 ± 5.6%) and the MD-Q diet (44.2 ± 14.8%). The size of postlarvae fed the four diets was not significantly different ( P  > 0.05). There was also little difference in the daily growth rate. Based on the results of this work for the preliminary implementation of artificial food as a replacement diet for diatoms, it seems that postlarval H . discus discus readily consumes artificial micro particle diets and has a superior survival rate when compared to that of diatoms.  相似文献
