Intermediary metabolic response and gene transcription modulation on the Sub-Antarctic notothenioid Eleginops maclovinus (Valenciennes, 1930) injected with two strains of Piscirickettsia salmonis

Pathogen interactions with cultured fish populations are well studied, but their effects on native fishes have not been characterized. In Chile, the disease caused by bacterial species Piscirickettsia salmonis represents one of the main issues and is considered to be one of the important pathogens in the field of aquaculture. They have been found to infect native fish. Therefore, it is necessary to understand the impact of P. salmonis on native species of local commercial value, as well as the potential impact associated with the emergence of antibiotic-resistant strains of P. salmonis. Due to this purpose, the native fish Eleginops maclovinus was used in our study. Fish were randomly distributed in tanks and intraperitoneally inoculated with two strains of P. salmonis. No mortality was recorded during the experiment. Cortisol, glucose and total α-amino acid levels increased in fish injected with AUSTRAL-005 strain compared to sham-injected and LF-89-inoculated fish. Moreover, results showed an increase in the activity of carbohydrates and lipids metabolism in liver; and an increase in the carbohydrates, lipids and total α-amino acid metabolism in muscle after injection with AUSTRAL-005. Our results suggest that P. salmonis modulates the physiology of E. maclovinus and the physiological impact increase in the presence of the antibiotic-resistant strain AUSTRAL-005.     

Anaesthetic and antioxidant effects of Myrcia sylvatica (G. Mey.) DC. and Curcuma longa L. essential oils on tambaqui (Colossoma macropomum)

Anaesthetic substances are necessary to reduce fish stress during aquaculture activities. The objectives of this study were: (i) to determine the efficacy of essential oils (EOs) of Myrcia sylvatica (EOMS) and Curcuma longa (EOCL) as anaesthetics for Colossoma macropomum and (ii) to evaluate the effects of rapid anaesthesia and long‐term sedation (6 h) with these oils. Therefore, the main primary stress indicator (cortisol) and secondary factors (biochemical indices, hepatic metabolism, oxidative biomarkers) were measured. Sedation with the EOCL resulted in lower cortisol levels compared to control group. Total cholesterol levels were lower in fish sedated with EOMS than in control. Lactate levels were higher in fish anaesthetized with both EOs and sedated with EOCL compared to control. Both EOs increased hepatic glycogen levels after anaesthesia and EOMS increased this parameter after sedation compared to control. Anaesthesia and sedation with EOs resulted in lower levels of lipid peroxidation (LPO) compared to control. In turn, the activity of some antioxidant enzymes evaluated (superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase and glutathione‐S‐transferase), the content of non‐protein thiols and total reactive antioxidant potential were higher in tissues of fish anaesthetized and sedated with EOs compared to control. This induction of antioxidant capacity in the tissues could be due to the antioxidant property exerted by these EOs. Thus, EOMS and EOCL are recommended for anaesthesia and sedation of fish because in spite of inducing anaerobic metabolism, these EOs did not alter most biochemical parameters, reduced the LPO and increased the antioxidant capacity in vital tissues.     

Histomorphological changes between short and long days in ovary guanacos (Lama guanicoe)

This is the first morpho-histological comparison of guanaco ovaries between reproductive (long-days) and non-reproductive (short-days) seasons, and oestrogen receptor-alpha (ERα) and beta (ERβ) detection. Different stages of follicle development were found in the cortical area, but no corpus luteum was detected. The size and frequency of antral follicles and large atretic follicles were higher in long-day ovaries than short-days, consistent with ovarian activity in this season. Differential expression of ERα and ERβ was observed in follicles at different stages of development between short and long days. These data reveal histological and molecular differences between reproductive and non-reproductive seasons of guanaco ovaries.     

Combining fishers’ knowledge and cost‐effective monitoring tools in the management of marine recreational fisheries: A case study of the squid and cuttlefish fishery of the Ría of Vigo (NW Spain)

A new methodology based in the use of fishers’ knowledge and cost‐effective tools to obtain information about marine recreational fisheries (MRF) is presented. The squid and cuttlefish fishery of the Ría of Vigo (NW Spain) was selected because it is managed in a data‐poor environment. In‐depth interviews (57) were conducted with fishers, collecting ecological and socio‐economic information. A cartography of fishing grounds based on their knowledge was obtained, while the intensity of effort and catches was mapped by the monitoring of two vessels with low‐cost GPS data loggers. The 102 shore anglers and 248 recreational boats catch 8 t/year of European squid Loligo vulgaris and 11 t/year of common cuttlefish Sepia officinalis (11% of total catches on these species in the area). Shore anglers fish from 11 ports, while boat fishers use 14 fishing grounds (covering 30 km²). Most of the catches (86%) are landed by boats, and their CPUE is higher in the outer part of the Ría of Vigo. The use of fishers’ knowledge and cost‐effective monitoring is encouraged to obtain information for the management of MRF. Given the economic contribution of MRF (260,000 €/year in direct expenses), this activity should be considered in the regulations.
