To investigate the impact of different dietary lipid sources on fillet composition and lipid transport, we conducted a feeding trial and evaluated the proximate composition of muscle tissue, fatty acid profiles, total cholesterol (in muscle and plasma), triglycerides, and lipoprotein concentrations in Nile tilapia, Oreochromis niloticus. Five semi‐purified diets, containing different oils (soybean – SO, corn – CO, linseed – LO, fish – FO, and olive – OO), were supplied to tilapia for 160 d. Fish fed with LO and FO diets had a lower percentage of total lipids in muscle compared with the others (P < 0.05). The highest percentage of protein was found in fish fed with FO diet (P < 0.05). The muscle fatty acid profile was influenced differently by diets (P < 0.05). The group supplemented with SO and CO had a higher concentration of 18:2n‐6, whereas the fish fed with LO diet had a higher level of 18:3n‐3 and those that received the FO diet had more 22:6n‐3 in comparison with those supplemented with vegetable oils. Plasma lipid transport was also affected by the diets: the fish fed with FO diet had higher total cholesterol and high‐density lipoprotein and lower very‐low‐density lipoprotein concentrations (P < 0.05). 相似文献

Mediterranean ecosystems are global hotspots of biodiversity threaten by human disturbances. Growing evidence indicates that regeneration of Mediterranean forests can be halted under certain circumstances and that successional stages can become notoriously persistent. The Mediterranean sclerophyllous forest in central Chile is been largely transformed into savannas dominated by the invasive legume tree Acacia caven as result of interacting management and ecological factors. We used multi-temporal satellite imagery to study the transition dynamics of these major vegetation types over the last four decades (1975-2008). Vegetation changes were related to indicators of resource availability (topography, water availability, solar radiance), potential propagule availability (distance to forest remnant patches) and disturbance regimes (grazing, fire occurrence and distance to roads and cities). During this study period, forests were mostly converted into Acacia savannas (46.1%). Acacia savanna was the most persistent natural vegetation type. The probability of sclerophyllous forest degradation into Acacia savanna increased on drier northern-exposed slopes, close to roads and further away from forest remnants. In contrast, forest regeneration from Acacia savanna was higher on moister southern-exposed slopes and closer to forest remnants. Acacia savannas are increasingly being converted into cultivated land on the moister locations or switching into a bare soil state in locations close to cities and further away from forest remnants. These results highlight the vulnerability of diverse sclerophyllous forests and its increasing conversion into persistent Acacia savannas in the Mediterranean region of central Chile and identify the ecological conditions for successful conservation and restoration of the native sclerophyllous forest vegetation that can be used for sensible land use planning. 相似文献

A protein-binding technique was employed to visualize, using scanning electron microscopy, the soy protein as well as the association between HMP and soy protein fractions. Image analysis indicated that at pH 7.5 and 3.5 soy protein isolate showed a bimodal distribution of sizes with an average [ d(0.5)] of about 0.05 microm, but at pH 3.8 the proteins formed larger aggregates than at high pH. Addition of HMP at pH 3.8 changed the surface charge of the particles from +20 to -15 mV. A small addition of HMP caused bridging of the pectin between soy protein aggregates and destabilization. With sufficient HMP, the suspensions showed improved stability to precipitation. The interactions of high-methoxyl pectin (HMP) and soybean-soluble polysaccharide (SSPS) with sodium caseinate-stabilized emulsions were investigated using a multitechnique approach, including dynamic light scattering (DLS), electrophoretic mobility measurements, transmission diffusing wave spectroscopy (DWS), and ultrasonic spectroscopy (US). At pH 6.8, both polysaccharides are negatively charged and did not adsorb onto caseinate-coated droplets due to electrostatic repulsion; however, SSPS showed a different behavior compared to HMP in the turbidity parameter 1/l* and sound attenuation parameters measured by DWS and US, respectively. The present study brought the first evidence of the stabilization effect of SSPS in acidified sodium caseinate-emulsions. While destabilization occurred at low polysaccharide concentrations, probably via bridging flocculation, acid-induced aggregation of the oil droplet was completely prevented by 0.2% SSPS or HMP. However, the interaction behavior of SSPS during acidification was different from that of HMP. The aim of this research was to validate the results obtained previously by purge and trap (PT) and to investigate the ability of solid phase microextraction (SPME), a more rapid and less expensive technique, to discriminate drinking milk subjected to different heat treatments (i.e., pasteurization, ultrahigh temperature, "in-bottle" sterilization) and produced at different factories. The data obtained by both methods were processed by multivariate statistical analysis. PT and SPME showed comparable repeatability, although with different performances for the yield of extraction, and allowed the three milk categories to be distinguished. Within the chemical class of methyl ketones, 2-heptanone was found to be the most discriminating compound, and the possibility of using the concentration of this volatile as a marker for heat treatment was investigated. 相似文献

The identification of fish species in transformed food products is difficult because the existing methods are not adapted to heat-processed products containing more than one species. Using a common to all vertebrates region of the cytochrome b gene, we have developed a denaturing high-performance liquid chromatography (DHPLC) fingerprinting method, which allowed us to identify most of the species in commercial crab sticks. Whole fish and fillets were used for the creation of a library of referent DHPLC profiles. Crab sticks generated complex DHPLC profiles in which the number of contained fish species can be estimated by the number of major fluorescence peaks. The identity of some of the species was predicted by comparison of the peaks with the referent profiles, and others were identified after collection of the peak fractions, reamplification, and sequencing. DHPLC appears to be a quick and efficient method to analyze the species composition of complex heat-processed fish products. 相似文献

An Agrobacterium-mediated transformation procedure for soybean [Glycine max L. Merrill] proliferating somatic embryos is here described. The Agrobacterium tumefaciens LBA4404 strain harboring pTOK233, pCAMBIA1390-olp or pH7WG2Dwrky plasmids was used to mediate gene transfer into the plant genome. Prior to Agrobacterium inoculation, proliferative soybean embryogenic clusters were microwounded by DNA-free tungsten particle bombardment. Three
independent transformation experiments were performed. In Experiment I, 26 transgenic plants were obtained from a unique clone
of cv Bragg, while 580 plants were recovered from 105 clones of cv IAS5. In Experiment II, a single hygromycin-resistant clone
of cv BRSMG68 Vencedora was recovered and gave rise to five plants. In Experiment III, 19 plants of cv Bragg and 48 plants
of IAS5 were recovered, representing five and 14 independent transformation events, respectively. PCR and Southern analyses
confirmed the transgenes’ integration into plant genomes. Transgenic plants were fertile. They flowered, set pods and seeds.
Transgene segregation in two T1 progenies fits the Mendelian pattern (3:1 transgenic:non-transgenic plants). This is the first report of transgenic fertile

Four agronomic traits were analysed including dry matter concentration (DMC) and dry matter yield (DMY) for stover, plant height (PHT) and days from planting to silking (DPS). We mapped quantitative trait loci (QTL) in three populations with doubled haploid lines (DHL), one RIL population and two testcross (TC) populations derived from crosses between two of the four populations mentioned above to elite tester lines, based on field phenotyping at multiple locations and years for each; 146–168 SSRs were used for genotyping of the four mapping populations. Significant high phenotypic and genotypic correlations were found for all traits at two locations, while DMC was negatively correlated with the other traits. A total of 42, 41, 54, and 45 QTL were identified for DMC, DMY, PHT, and DPS, respectively, with 9, 7, 12, and 7 major QTL for each trait. Most detected QTL displayed significant interactions with environment. Major QTL detected in more than two populations will contribute to marker‐assisted breeding and also to fine mapping candidate genes associated with maize agronomic traits. 相似文献

Purpose
Concentrations and transformations of mercury were measured in river, estuarine, and marine sediments to determine factors affecting the fate of mercury entering the northern Adriatic Sea.Materials and methods
Radiotracer methodology was used to compare rates of mercury methylation (203Hg), MeHg demethylation (14C), and sulfate reduction (35S) in sediment depth profiles to concentrations of total and dissolved mercury species in the lower freshwater region of the Isonzo River, the coastal lagoons, and in the Gulf of Trieste, northern Adriatic Sea.Results and discussion
Mercury was readily methylated and demethylated in all sediments, but the relative activity of these processes varied greatly with location. Methylation activity increased greatly from freshwater to the marine regions; however, demethylation was extremely high in the estuarine and lagoon sites. Ratios of methylation to demethylation were low in these coastal sites but increased further offshore in the gulf, which agreed with increased ratios of MeHg to total Hg (%MeHg) in gulf sediments. Comparisons of microbial activities indicated that sulfate reduction strongly controlled both methylation and demethylation. However, Hg methylation in coastal lagoon sediments was controlled by rapid demethylation and the bioavailability of Hg that was affected by Hg adsorption and precipitation. Methylation in offshore marine sites correlated with sulfate reduction but not the partitioning of Hg between pore water and solid phases. The decrease in sulfide production offshore exacerbated Hg methylation.Conclusions
The freshwater to marine gradient in the Idrija/So?a/Isonzo/Adriatic region is dynamic, exhibiting horizontally variable rates of microbial activities and Hg transformations that create “hot spots” of MeHg accumulation that are controlled differently in each region.10.

Summary Functional markers (FMs) are derived from polymorphic sites within genes causally involved in phenotypic trait variation (Andersen,
J.R. & T. Lübberstedt, 2003. Trends Plant Sci 8: 554–560). FM development requires allele sequences of functionally characterized
genes from which polymorphic, functional motifs affecting plant phenotype can be identified. In maize and other species with
low levels of linkage disequilibrium, association studies have the potential to identify sequence motifs, such as a few nucleotides
or insertions/deletions, affecting trait expression. In one of the pioneering studies, nine sequence motifs in the dwarf8 gene of maize were shown to be associated with variation for flowering time (Thornsberry, J.M., M.M. Goodman, J. Doebley,
S. Kresovich, D. Nielsen & E.S. Buckler, 2001. Nat Genet 28: 286–289). Proof of sequence motif function can be obtained by
comparing isogenic genotypes differing in single sequence motifs. At current, the most appropriate approach for this purpose
in crops is targeting induced local lesions in genomes (TILLING) (McCallum, C.M., L. Comai, E.A. Greene & S. Henikoff, 2000.
Nat Biotechnol 18: 455–457). In central Europe, maize is mainly grown as forage crop, with forage quality as major trait,
which can be determined as proportion of digestible neutral detergent fiber (DNDF). Brown midrib gene knock out mutations have been shown to be beneficial for forage quality but disadvantageous for overall agronomic performance.
Two brown midrib genes (bm1 and bm3) have been shown to be involved in monolignol biosynthesis. These two and additional lignin biosynthesis genes have been
isolated based on sequence homology. Additional candidate genes putatively affecting forage quality have been identified by
expression profiling using, e.g., isogenic bm lines. Furthermore, we identified an association between a polymorphism at the COMT locus and DNDF in a collection of European
