The polymerase chain reaction–denaturing gradient gel electrophoresis (PCR‐DGGE) of 16S ribosomal RNA gene was used to investigate bacterial communities in the intestines of large yellow croaker at six different ages (12 d, 18 d, 26 d, 40 d, 3 mo, and 1 yr old) as well as within the corresponding feed and culture water. In addition, Illumina Miseq sequencing was utilized to compare intestinal microbiota between 12‐d‐old and 1‐yr‐old individuals. PCR‐DGGE results revealed that the culture water had the highest bacterial diversity, followed by the feed, while the intestines had the lowest diversity. The intestinal microbiota at six ages changed severely; however, the change did not follow any trend. The large yellow croaker intestines harbored specific bacterial communities that differed from those in both feed and water. Illumina Miseq sequencing results revealed that the diversity of intestinal bacteria in 12‐d‐old fish was higher than that in 1‐yr‐old fish, and the bacterial composition differed significantly between them. γ‐Proteobacteria and Pseudoalteromonas supplied the most abundant phylum and genus in the 12‐d‐old fish intestine. However, in the 1‐yr‐old fish intestine, Firmicutes and Clostridium were the most dominant, respectively. The study may contribute to a better understanding of gut microbiota and dynamics of the large yellow croaker and the relationship with their surrounding environment. 相似文献

The aim of this study is to investigate the composition of the fatty acids and phospholipids derived from large yellow croaker (Pseudosciaena crocea) roe. The composition of the total lipids and the molecular species of phospholipids were determined by gas chromatography–mass spectroscopy (GC–MS) and high performance liquid chromatography–evaporative light scattering detector (HPLC–ELSD), respectively. The results showed that large yellow croaker roe had high levels of the total lipid (19.6% ± 1.32%, w/w) and phospholipid (61.2% ± 1.22% of the total lipid). The phospholipid was rich in docosahexaenoic acid (31.0% ± 0.19% of the total phospholipids), and the major phospholipid molecular species was phosphatidylcholine (PC, 61.06% ± 0.02% of the total phospholipids, w/w). A comparative study on the volatile compounds in cooked muscle of wild and farmed large yellow croaker (LYC) was conducted. The two farmed LYC groups were fed with diets containing 44% (CF) and 25% (LF) of fish meal (FM) respectively. Results showed that 48 volatiles, including aldehydes, alcohols, ketones, hydrocarbons, aromatics, acids, esters, furans and miscellaneous compound, were detected in cooked fillets. The LF group had significantly lower amounts of total aldehydes and ketones, higher content of miscellaneous compound in cooked fillets than that in the CF and wild groups (p < .05). Compared with the wild group, the LF group had significantly lower amounts of total alcohols, acids and esters, while the CF group had significantly lower amounts of total aldehydes, higher content of total ketones in cooked muscle (p < .05). According to the principal component analysis (PCA), some volatiles (propanal, nonanal, etc.) could be considered as sensitive indicators to classify cooked muscle samples. In conclusion, differences in the volatiles in the cooked muscle between the wild and farmed LYC have been found. Low level of dietary FM (25%) changed the volatile profiles in cooked fillets of farmed LYC. A PCA may be useful to screen potential volatiles to classify cooked muscle samples in this study. 相似文献

为解决规模化猪舍废气排放造成的环境污染问题,设计了一种猪舍废气复合净化系统。该系统采用化学法与水洗法相结合,通过PLC(可编程逻辑控制器)控制系统实时采集净化系统内的pH值、电导率、液位和压差等动态环境数据,智能控制洗涤泵启停和电磁阀通断,自动完成供水、加酸、喷淋和排废4个工作环节。同时,控制系统采用MCGS触摸屏与PLC建立通讯,通过创建人机交互界面实现系统环境数据监测、运行状态流动显示、按需配置系统参数和报警信息输出等多种功能,实现了系统操作的人性化和过程的可视化。系统可根据实际应用中对净化效率和运行成本的要求,实现多种控制模式,均有效抑制了猪舍废气排放。试验结果表明,系统对主要污染成分氨气的平均去除率可达到85%,整体运行可靠、控制简单,经济成本量化清晰。该系统在江西某种猪场实施应用,成效显著,可为畜禽养殖环境废气净化处理的工程应用提供参考。 相似文献

探明夏玉米氮素营养生化指标(叶绿素a、叶绿素b、类胡萝卜素、叶片氮含量和叶片氮积累量)与叶片SPAD值垂直分布特征及两者间定量回归关系,确立基于叶绿素仪的夏玉米氮营养无损诊断敏感叶位和叶片部位,以实现氮营养时空变化的快捷和精准监测。利用2018-2019年连续2季不同氮营养水平下夏玉米关键生育期主茎各叶位(顶1叶~顶12叶,TL1~TL12)和叶片部位(每张叶片从叶片基部开始根据叶片长度每20%分为1个测试区间) SPAD值及氮营养指标数据,研究基于偏最小二乘(partial least square, PLS)回归模型的夏玉米不同位点SPAD值与氮营养指标间关系,确定可稳定指示夏玉米氮营养空间异质性变化的敏感叶位及叶片部位。结果表明,不同叶位间夏玉米叶片SPAD值和氮营养指标于植株间分布均呈典型的"钟型"特征,至TL5或TL6时达至峰值。同一叶位不同部位间SPAD值由20%至100%位点时则逐步升高,且80%~100%位点间无显著差异(P>0.05)。PLS分析结果显示,夏玉米不同叶位SPAD值与氮营养指标间模型精度决定系数(coefficient of determination, R2)和相对分析误差(relative percent deviation,RPD)范围分别为0.693~0.821和1.425~2.744。不同测试位点R2和RPD值范围则分别为0.660~0.847和1.607~2.451,满足模型精确诊断需求。此后,基于PLS模型中各叶位和叶片部位无量纲评价指标变量重要性投影值(variable importance for projection,VIP),确定顶4叶(TL4)完展叶60%~80%区间为夏玉米氮营养诊断的敏感区域,VIP值均高于临界值1.40,预测效果较为理想。研究可为实现氮营养的高效、快捷诊断和精准施氮提供参考。 相似文献

以成渝经济区为研究区,比较成渝地区农地经营方式转变的地域差异,评估农地经营方式转变的可持续性.选取成都平原区的广汉市和重庆山地丘陵区的江津区为研究样区,基于两个区域的1 102份调查数据,运用能值分析法分析两地农地经营方式转变的差异.由于资源禀赋、家庭特征、经营方式、流转市场、流转价格等因素的差异影响,江津区与广汉市农户农地流转呈现区域"悖反"现象;从能值分析来看,广汉市通过规模化、产业化、机械化的大生产模式提升农业科技装备水平,降低投入、提高农业生产率,实现了良性的农业集约化;而江津区通过高投入—高产出的高效农业产业模式大幅提升农业产出,但过度追加不可更新要素的投入方式却不可持续,引起了劣质的农业集约化.因此,就转变农地经营方式而言,成都平原区应着眼于"降本增效",发展规模化、产业化、机械化的大生产模式;重庆山地区应着眼于"提质增效",以农业科技、装备和设施更新为驱动发展生态农业. 相似文献

The objective of this case report is to describe the diagnostic and surgical techniques for removal of a metallic foreign body in the tongue of three adult horses. The three horses were presented for evaluation and treatment of dysphagia and marked hypersalivation of 3–5 days duration. Radiographs of the head revealed the presence of a metallic foreign body in the tongue of each horse. The foreign bodies could be precisely localised under general anaesthesia using palpation and lingual ultrasonography and/or lingual radiography in combination with a forceps as a marker. The foreign bodies were successfully removed using laparoscopic instruments creating minimal soft tissue trauma. The use of long (43 cm) small laparoscopic (5 mm) instruments enabled good visualisation of the surgical field, providing optimal conditions for successful minimally invasive surgical treatment of horses with foreign bodies in the tongue. The three horses made uneventful recoveries and 12 months after surgery were eating normally and could be ridden with a bit as per usual routine. It was concluded that using long laparoscopic instruments in combination with palpation, ultrasonography and/or radiography allowed removal of the foreign body creating minimal soft tissue trauma and allowing optimal conditions for a fast recovery. 相似文献
