To determine whether senescing leaves provoke an active nitrogen (N) remobilization that results in the reduction of nitrogenase activity, 60% of Medicago truncatula lower leaves were either darkened or individually excised for two weeks. Background: Nitrogen losses is an economic problem for wheat production and a high risk to the environment. Therefore, improved N fertilizer management is a key to increasing the N efficiency and minimizing N losses. To increase N efficiency, enhanced fertilizers such as urea combined with urease inhibitor can be used. Aims: The aim of present study was to evaluate the effects of different N forms on grain storage protein subunits in winter wheat and to examine whether the observed changes correlate with parameters of baking quality. Methods: The investigation was performed over two consecutive years at two locations in Germany. Protein subunits were analyzed by SDS‐PAGE. Results: Protein concentrations were similarly increased after fertilization with ammonium nitrate and urea + urease inhibitor. Analysis of the individual storage protein fractions indicated that both fertilizers specifically enhanced ω‐gliadins and HMW glutenins, but the effect was more pronounced in the ammonium nitrate treatment. Application of urea + urease inhibitor had greater influence on the protein composition and resulted in higher specific baking volume as well as the best fresh keeping ability, in comparison with urea treatment. Conclusion: Considering that the urea + urease inhibitor treatment resulted in almost comparable improvements of NUE and baking quality, with the additional benefit of reduced N losses in combination with easy handling, urea + urease inhibitor can be recommended as a viable alternative to both urea alone and ammonium nitrate treatments. Aglime application can promote carbon dioxide (CO2) emissions from acid soils. However, the controlling mechanisms are still poorly understood, particularly the role of fertiliser-ammonium oxidation. This study therefore assessed the effects of aglime on soil inorganic C (SIC)– and soil organic C (SOC)–derived CO2 emissions from acid soils amended with ammonium.
Materials and methods
Ammonium at three N rates [0% (A0), 0.005% (A1), and 0.2% (A2) w/w] and labelled aglime (Ca13CO3,13C 5.94% aa) at three rates [0% (L1), 0.067% (L1), and 0.392% (L2) w/w] were applied to two contrasting acid soils (Nariva series, Mollic Fluvaquents; and Piarco series, Typic Kanhaplaquults) and incubated in 1-l media bottles for 23 days. A calcareous soil (Princes Town series, Aquentic Eutrudepts, carbonate δ13C of ??4.79‰) was included as a control that only received ammonium at the three rates.
Results and discussion
The application of ammonium at the A2 rate significantly (p?<?0.05) increased cumulative SIC-CO2 emissions by 15.8 and 27.1% in comparison to the A0 rate for the Nariva and Piarco soils, respectively, when they were limed at the L2 rate. The lower rate of ammonium (A1), however, had no effect on these emissions, which suggests that enough acidity may not have been generated at this rate to significantly enhance the release of SIC-CO2. Furthermore, no effect of ammonium rates was observed on SIC-CO2 emissions from the calcareous soil, which refutes the hypothesis that this amendment plays a greater role in regulating these emissions from calcareous soils compared with acid soils. Also, in contradiction to another hypothesis, the aglime-induced priming effect on SOC decomposition was more apparent in the low-C Piarco soil. This effect was also significantly (p?<?0.05) greater at the L2 rate (above the lime requirement for Piarco), which demonstrates the negative impact that over-liming could have on the sequestration of C in this soil. Our results also showed that ammonium addition may also help to reduce the magnitude of the aglime-induced priming effect in the Piarco soil when it is not over-limed.
Conclusions
Overall, the findings of this study suggest that ammonium fertiliser broadcast at conventional rates may not serve as a significant regulator of SIC-CO2 emissions from highly to moderately acidic soils amended with aglime. Our findings also indicate a need to consider nitrogen management as an important factor regulating the effects of aglime on SOC-CO2 emissions.

Cotton blue disease (CBD) is the most important disease present in cotton crops in South America and cotton leafroll dwarf virus (CLRDV) is the causal agent. The disease has been controlled by sowing cotton varieties resistant to CLRDV. However, in the 2009/10 growing season, an outbreak due to an atypical CLRDV isolate (CLRDV-at) occurred in northwest Argentina. Although CLRDV and CLRDV-at genomes are very closely related, the symptoms they produce in cotton plants are quite different. P0 is the most divergent protein between the isolates and in CLRDV is a silencing suppressor protein. This work characterized the silencing suppressor activity of the P0 protein encoded by CLRDV-at (P0CL-at) and evaluated its role in Cbd-resistance break in cotton plants. It was demonstrated that P0CL-at, despite having a mutation in the consensus of the F-box-like motif, was able to suppress local RNA silencing, but displayed lower activity than P0CL. P0CL and P0CL-at showed no differences in the interaction with Gossypium hirsutum SKP1 orthologue (GSK1) and Nicotiana benthamiana SKP1 and both P0 proteins triggered destabilization of ARGONAUTE1. However, when the ability to enhance PVX symptoms was evaluated, P0CL-at was shown to be a weaker pathogenicity factor than P0CL in N. benthamiana. Interestingly, trans-expressed P0CL-at enabled CLRDV to systemically infect CBD-resistant plants, and a chimeric CLRDV-P0CL-at infectious clone succeeded in establishing infection in CBD-resistant cotton varieties with symptoms resembling those produced by CLRDV-at. These results strongly suggest that P0CL-at is the avirulence (Avr) determinant involved in breaking cotton Cbd gene-based resistance. 相似文献

Efficient management of whitefly-borne diseases remains a challenge due to the lack of a comprehensive understanding of their epidemiology, particularly of the diseases tomato golden mosaic and tomato yellowing. Here, by monitoring 16 plots in four commercial fields, the temporal and spatial distribution of these two diseases were studied in tomato fields in Brazil. In the experimental plots these diseases were caused by tomato severe rugose virus (ToSRV) and tomato chlorosis virus (ToCV), respectively. The incidence of each virus was similar in the plots within a field but varied greatly among fields. Plants with symptoms for both diseases were randomly distributed in three of four spatial analyses. The curves representing the progress of both diseases were similar and contained small fluctuations, indicating that the spread of both viruses was similar under field conditions. In transmission experiments of ToSRV and ToCV by Bemisia tabaci MEAM1 (former biotype B), these viruses had a similar transmission rate in single or mixed infections. It was then shown that primary and secondary spread of ToCV were not efficiently controlled by insecticide applications. Finally, in a typical monomolecular model of disease progress, simulation of the primary dissemination of ToSRV and ToCV showed that infected plants were predominantly randomly distributed. It is concluded that, although the manner of vector transmission differs between ToSRV (persistent) and ToCV (semipersistent), the main dispersal mechanisms are most probably similar for these two diseases: primary spread is the predominant mechanism, and epidemics of these diseases have been caused by several influxes of viruliferous whiteflies. 相似文献
