The begomovirus Tomato severe rugose virus (ToSRV) and the crinivirus Tomato chlorosis virus (ToCV), in single and co-infections, are very common in tomato crops in Brazil. Both viruses are transmitted by the whitefly Bemisia tabaciMEAM1 (biotype B). The objective of this study was to analyse the interaction between ToSRV and ToCV in tomato plants of cultivars Santa Clara and Kada. Plants at 15, 30 and 45 days after emergence were inoculated with 30 viruliferous B. tabaci per plant. The following treatments were compared: plants inoculated with ToSRV, ToCV, ToSRV + ToCV, and healthy (control). The interaction between these viruses was analysed by measuring the virus titre by qPCR and the fresh and dry weights of the aerial parts of the tomato plants. Based on two independent assays, no significant effects for co-infection of ToSRV and ToCV on virus titres and plant development were observed compared to single infections. The dry weight of tomato plants of both cultivars infected with ToSRV, ToCV, or co-infected did not differ significantly. However, the dry weight of Santa Clara tomato plants infected with ToSRV, ToCV and ToSRV + ToCV showed mean reductions of 21.5%, 25.5% and 32%, respectively, compared to healthy plants, and mean reductions for Kada were 31.7%, 37.5% and 38%, respectively.  相似文献

Bemisia tabaci is important in agriculture worldwide, mainly because it is a vector of numerous plant viruses, probably the most important of which are members of the genus Begomovirus. Dozens of begomoviruses have been reported to infect tomato plants in Brazil, although tomato severe rugose virus (ToSRV) predominates in tomato crops. ToSRV, found so far only in Brazil, is efficiently transmitted by B. tabaci MEAM1. However, no studies have assessed the occurrence of vertical and horizontal transmission of the virus in the insect, which may have epidemiological consequences affecting disease management. This study evaluated the possibility of transmission of ToSRV between whiteflies during copulation and transovarial transmission from viruliferous females of B. tabaci MEAM1 to their progeny. Transmission of ToSRV did not occur during mating between males and females of B. tabaci MEAM1. Aviruliferous males and females confined with viruliferous insects of the opposite sex were also unable to transmit the virus to tomato plants. ToSRV was detected, by PCR, in the ovaries of viruliferous females of B. tabaci MEAM1 but not in eggs, nymphs, or adults of the progeny of viruliferous females. Adult progeny of viruliferous females also did not transmit ToSRV to tomato plants. Together, the results indicate that vertical and horizontal transmission of ToSRV by B. tabaci MEAM1 is unlikely. Sustainable management of the tomato golden mosaic disease caused by ToSRV should continue to focus on using resistant varieties, managing sources of inoculum around tomato fields, and rational chemical control of the vector.  相似文献

番茄褪绿病毒Tomato chlorosis virus(ToCV)是一种由烟粉虱Bemisia tabaci传播的正义单链RNA病毒,在田间常与番茄黄化曲叶病毒Tomato yellow leaf curl virus(TYLCV)复合侵染而造成番茄生产上重大的经济损失。为了明确ToCV与TYLCV的复合侵染对烟粉虱传播ToCV所造成的影响,本文采用RT-PCR以及qRT-PCR检测了复合侵染的番茄对烟粉虱获取和传播ToCV的影响。研究表明,烟粉虱取食复合侵染的番茄后对ToCV的传播效率显著提高,仅25头烟粉虱的传毒率即可达到100%,ToCV在烟粉虱以及番茄体内的累积量均显著提高。说明这种复合侵染促进了烟粉虱对ToCV的传播,在田间应当及时防控烟粉虱,警惕病毒与烟粉虱的蔓延。  相似文献

Tomato chlorosis virus (ToCV) is a plant virus that is mainly propagated by Bemisia tabaci in a semi-persistent and non-circulative manner.It has a wide range of host plants,and has been reported in many countries,causing serious economic losses in vegetable production.In 2019,we investigated about 10 fields,one ha each in Shouguang,Shandong province (China),and in each field we observed symptoms of interveinal chlorosis on lower leaves of the Solanum torvum Swartz,and a large number of B.tabaci gathered on the back of its leaves.To determine the presence of ToCV,total RNA of S.torvum was extracted followed by RT-PCR.The 1 074 (GenBank accessions number MN545620) and 466 bp (GenBank accessions number MN545621) fragments were gel purified and sequenced.The sequences shared 99.44% and 99.57% similarity with ToCV reference sequence tomato chlorosis virus segment RNA1 (AY903447) and RNA2 (AY903448).The results of insect transmission test confirmed that ToCV can spread from S. torvum to tomato.This study confirms S.torvum as a newly reported host of ToCV.  相似文献

Tomato chlorosis virus (ToCV) and Tomato infectious chlorosis virus (TICV) are two criniviruses inducing similar yellowing symptoms in tomato. An approximately 4 kb central region of the genomic RNA2 of French ToCV and TICV isolates was sequenced. TICV, for which no other sequences were available, appeared as a distant species in the genus, being close only to LIYV ( Lettuce infectious yellows virus ) for some, but not all, proteins. ToCV has more than 98% nucleotide identity with isolates from the US and Spain, and sequencing the CP gene of several isolates collected in different regions in southern France during 2 years suggested a unique origin. Polyclonal antisera were produced using capsid proteins of both viruses expressed in Escherichia coli . DAS-ELISA assays were developed for routine diagnosis and conditions for preparing samples for an optimized detection were determined. No cross-reactions were observed. However, some false-negative results, corresponding to samples giving ELISA readings close to the detection limit were regularly detected, particularly for ToCV (approximately 5% of the samples). A triplex RT-PCR assay was thus developed, which allowed detection of both viruses in a one-step protocol. An internal PCR control was included, which in addition showed that it could be used as a control for the entire RT-PCR procedure. Finally, combining DAS-ELISA in a first round, and triplex RT-PCR for doubtful samples, appeared the best way to achieve a reliable diagnosis of these viruses.  相似文献

Tomato chlorosis virus (ToCV) and Tomato infectious chlorosis virus (TICV) are two criniviruses that are emerging worldwide, and induce similar yellowing diseases in tomato crops. While TICV is transmitted only by Trialeurodes vaporariorum , ToCV is transmitted by three whitefly species in two genera Trialeurodes vaporariorum , T. abutilonea and Bemisia tabaci . The efficiency of transmission by T. vaporariorum from plants infected by one virus or by both was compared, and the probability of virus transmission by a single whitefly was derived from group testing experiments. The estimated transmission probabilities ranged from 0·01 to 0·13, and were not significantly different between ToCV and TICV, or between single and mixed infections. Experiments using B. tabaci as a vector and source plants infected by TICV and ToCV did not reveal any functional trans-complementation for transmission of TICV by ToCV, suggesting that if this phenomenon occurs in nature, it is at a very low frequency. Possible reasons why TICV did not establish in southern France while ToCV is now endemic are discussed.  相似文献

番茄褪绿病毒tomato chlorosis virus (ToCV)是我国蔬菜生产的重要新发病毒,其寄主范围逐年扩大。2019年10月项目组在山东寿光西葫芦温室调查时发现部分叶片呈现黄化、脉间褪绿,类似于ToCV侵染症状,并伴有烟粉虱发生。利用特异性引物检测发现,扩增的目的片段与GenBank中登录的侵染番茄的ToCV基因序列(登录号:KC887998.1)同源性高达99.58%,充分说明西葫芦植株已被ToCV侵染。通过对病毒病发生规律和烟粉虱虫口数量调查发现,西葫芦定植后1个月表现侵染症状的植株为2.0%,定植后2个月达到4.2%,定植后4个月后高达68.2%,病毒发生呈指数增长,而烟粉虱虫口数量却维持较低密度。从济南和德州采集的西葫芦疑似病叶和烟粉虱中也检出ToCV病毒,说明该病毒可能已经在山东省设施西葫芦主要种植区普遍发生,并经烟粉虱广泛传播,需引起高度重视。  相似文献

为明确沼泽红假单胞菌Rhodopseudomonas palustris PSB-06菌剂对番茄褪绿病毒(tomato chlorosis virus,ToCV)的抑制作用,连续3年在田间进行沼泽红假单胞菌PSB-06菌剂对ToCV的药效试验,并在室内研究其抗病毒的作用机制。田间试验结果显示,PSB-06菌剂处理能降低ToCV的发生,植株发病率均低于50%,均显著低于清水和氨基寡糖素处理;室内试验结果显示,PSB-06菌剂处理能抑制病毒传播,喷施PSB-06菌剂后的感病番茄的SA含量达532.67 mg/g,PI IINPR1基因表达量也相对上升;同时,健康番茄与感病番茄的叶绿素含量均明显升高;经PSB-06喷施后番茄植株上的烟粉虱Bemisia tabaci生存数量降低。蛋白组学分析结果表明,PSB-06处理促使植物代谢及防御的相关蛋白均上调。表明沼泽红假单胞菌PSB-06菌剂能增强植物免疫和防御烟粉虱的能力,从而有效地控制ToCV的发生及传播。  相似文献

在广东番茄上发现一种新病害,病株表现为叶片褪绿,叶脉颜色变深及叶片增厚等症状。利用番茄褪绿病毒Tomato chlorosis virus(ToCV)HSP70基因的两对特异引物对番茄病样进行RT-PCR检测,结果表明,从所采集的6份病样中均扩增到预期大小的DNA特异片段。对其中1份样品的扩增片段进行克隆与序列分析,结果表明,扩增片段包括1个长度为1 665个核苷酸的完整病毒基因,其核苷酸序列与已报道的ToCV HSP70基因有较高的同源性,表明广东番茄受到了ToCV的侵染。但ToCV广东番茄分离物的HSP70序列与国内外已报道的各分离物的同源性均低于82%,存在较大差异,其中与塞浦路斯tomato、约旦JU_20分离物的同源性最高,为81.8%。这是ToCV在广东发生的首次报道,也是该病毒HSP70基因序列存在显著差异分离物的首次发现。  相似文献

Tomato severe rugose virus (ToSRV) is the most important begomovirus transmitted and spread by the whitefly Bemisia tabaci in tomato crops in Brazil. Cultural practices are being adopted, along with insecticides, for controlling this virus. However, little is known about the importance of weeds in the pathosystem, which can contribute to the failure of these practices. This work aimed to evaluate the role of Datura stramonium and Nicandra physaloides as alternative hosts of ToSRV and verify the viral influence on the biological performance of Bemisia tabaci Middle East-Asia Minor 1 (MEAM1) and Mediterranean (MED) cryptic species. N. physaloides was a better alternative host for ToSRV when combined with MEAM1 whiteflies, while D. stramonium was mostly a good host for whitefly reproduction. Viral infection improved MEAM1 performance on both host plants but affected MED performance negatively. These data suggest that both weeds can be of some importance for the pathosystem, and their control should be included in management programmes.  相似文献

ABSTRACT Multiple viral infections frequently are found in single plants of cultivated and wild hosts in nature, with unpredictable pathological consequences. Synergistic reactions were observed in mixed infections in tomato plants doubly infected with the positive-sense and phloem-limited single-stranded RNA (ssRNA) crinivirus Tomato chlorosis virus (ToCV) and the negative-sense ssRNA tospovirus Tomato spotted wilt virus (TSWV). Synergism in a tomato cultivar susceptible to both viruses resulted in a rapid death of plants. A pronounced enhancement of ToCV accumulation mediated by TSWV co-infection was observed with no evident egress of ToCV from phloem tissues. No consistent alteration of TSWV accumulation was detected. More remarkable was the synergism observed in tomato cultivars which carry the Sw-5 resistance gene, which are resistant to TSWV. Pre-infection with ToCV resulted in susceptibility to TSWV, whereas co-inoculations did not. This suggested that a threshold level or a time lapse is needed for ToCV to interfere or downregulate the defense response in the TSWV-resistant plants.  相似文献

ABSTRACT Tomato chlorosis virus (ToCV) is the second whitefly-transmitted, phloem-limited, bipartite closterovirus described infecting tomato. ToCV is distinct from tomato infectious chlorosis virus (TICV), based on lack of serological and nucleic acid cross-reactions and differences in vector specificity. TICV is transmitted only by the greenhouse whitefly (Trialeurodes vaporariorum), whereas ToCV is transmitted by the greenhouse whitefly, the banded-wing whitefly (T. abutilonea), and Bemisia tabaci biotypes A and B (B. argentifolii). Double-stranded (ds) RNA analyses of ToCV show two prominent dsRNAs of approximately 7,800 and 8,200 bp, with several small dsRNAs. Digoxigenin-11-UTP-labeled riboprobes derived from cDNA clones representing portions of RNAs 1 and 2 were used in Northern blot hybridizations to detect two large nonhomologous dsRNAs and a subset of smaller dsRNAs. These probes were used in dot blot hybridizations to detect ToCV in infected tomato. Inclusion bodies and cytoplasmic vesicles were consistently observed in phloem tissues of ToCV-infected Nicotiana clevelandii. Computer-assisted sequence analysis showed significant homology between ToCV clones that hybridize specifically with RNAs 1 and 2 and the lettuce infectious yellows virus methyltransferase of RNA 1 and the HSP70 heat shock protein homolog of RNA 2, respectively. Thus, ToCV is another member of the growing subgroup of bipartite closteroviruses transmitted by whiteflies.  相似文献

研究烟粉虱传播番茄褪绿病毒Tomato chlorosis virus (ToCV)的发生规律,建立其预测预报模型,能够指导田间早期有效防治。本研究于2014年-2018年每年采集山东寿光蔬菜基地大棚番茄和杂草的植株叶片,并收集植株上携带的所有烟粉虱,以健康番茄、杂草叶片和室内饲养的健康烟粉虱为阴性对照;实验室ToCV侵染性克隆接种的感病叶片以及从感病叶片上获毒的烟粉虱为阳性对照,将采集的样品带回实验室进行检测和鉴定等试验。根据病情发生规律,建立了番茄褪绿病毒病的预测预报模型,模拟所得方程为:Y=2.570+0.089X4-7.548X7,其中Y为11月份番茄褪绿病毒病发生率,X4为采集的样品上平均每株所携带的烟粉虱数量,X7为采集的田间杂草的带毒率。预测模拟结果显示,烟粉虱数量以及杂草的带毒率与番茄褪绿病毒病的发生率极显著正相关,回归检测结果历史符合率为96.8%以上。明确了影响番茄褪绿病毒病发生的影响因子,基于烟粉虱数量以及杂草的带毒率,构建了病害预测预报模型。研究结果有助于及时发现番茄褪绿病毒病并采取相应预防措施。  相似文献

 番茄黄化曲叶病毒(Tomato yellow leaf curl virus, TYLCV) 及番茄褪绿病毒(Tomato chlorosis virus, ToCV)是危害番茄的主要病毒。2016~2017年,在河北省18个番茄主产区调查番茄病毒病危害情况,采集262份植株矮化、叶片黄化、褪绿、卷曲的番茄样品,利用分子生物学技术对病原进行鉴定。结果表明:2016~2017年河北省番茄病毒病发生普遍,所检样品中TYLCV的侵染率为68.66%,其中与ToCV复合侵染率为19.5%;除栾城、滦南、乐亭、永年4地样品暂未检测到ToCV外,其他14个采样点的样品均检测到ToCV,侵染率为19.5%,且全部表现为与TYLCV复合侵染,河北省番茄主产区暂未发现ToCV单独侵染的样品。  相似文献

近年来在陕西番茄生产区出现了一种新病害,病株表现为叶片褪绿,叶脉颜色变深及叶片增厚,果实变小发白,严重影响番茄产量及经济效益,2016年发病极为严重,部分产区甚至绝收。该病疑似由番茄褪绿病毒Tomato chlorosis virus(ToCV)引起。利用ToCV特异引物对感病番茄样品进行RT-PCR检测,结果从所采集的4份病样中均检测到ToCV预期大小的特异片段。对ToCV外壳蛋白CP基因和类热激蛋白HSP70h基因进行克隆与序列分析,ToCV-YL1 CP基因774个核苷酸序列与已报道的ToCVCP基因有较高的一致性,与山东青岛和山西晋中分离物同源性为100%。ToCV-YL1 HSP70h基因1 665个核苷酸序列与已报道的ToCV HSP70h基因有较高的同源性,与中国、韩国、日本、美国、希腊分离物同源性达到99%以上。研究表明ToCV已传播至陕西地区。  相似文献

病毒病对番茄生产造成严重危害, 近年来在番茄种植新区发生严重, 疑似为种子带毒传播。本研究通过对云南省怒江州番茄种植新区的番茄病毒病样品采用RNA-seq高通量测序, RT-PCR验证的方法检测病毒种类;对番茄病果种子进行超薄切片制样透射电子显微镜观察, 将病果种子播种后对种苗进行RT-PCR带毒检测。结果表明, RNA-seq高通量测序及RT-PCR检测到的病毒有番茄环纹斑点病毒(tomato zonate spot orthotospovirus, TZSV)、番茄黄斑驳相关病毒(tomato yellow mottle-associated virus, TYMaV)、辣椒脉斑驳病毒(chili veinal mottle virus, ChiVMV)、南方番茄病毒(southern tomato virus, STV)。透射电镜观察到种胚细胞及胚乳细胞中分布典型的正番茄斑萎病毒属Orthotospovirus病毒粒体。病果种子播种28 d后的种苗具有病毒病症状, 通过RT-PCR检出TZSV、ChiVMV、STV, 检出率分别为60%、100%、80%。上述研究结果为TZSV通过种子传播提供了有利的证据, 并为源头防控番茄病毒病提供了依据。  相似文献

The name Tomato yellow leaf curl virus (TYLCV) has been applied to a group of virus species of the genus Begomovirus in the family Geminiviridae that cause a similar tomato disease worldwide. In 1995, TYLCV was first reported in Algarve (southern Portugal) as responsible for an epidemic outbreak of a severe tomato disease. Molecular data have shown that this Portuguese TYLCV isolate was distinct from those previously reported in Europe, as it belonged to the TYLCV-Israel species 1 . Since then, TYLCV epidemics have occurred annually, being a limiting factor mainly for autumn/winter glasshouse tomato crops. In 1998, TYLCV was also found associated with the emergence of a novel disease of Phaseolus vulgaris in Algarve. The affected bean plants were severely stunted and gave no marketable yield. However, the disease occurs only sporadically, even in conditions of high TYLCV infection pressure. Recently, Tomato chlorosis virus (ToCV), a whitefly-transmitted bipartite closterovirus (genus Crinivirus , family Closteroviridae ), was found associated with an unusual tomato yellow leaf syndrome, in single or mixed infection with TYLCV. The impact of this new pathosystem on tomato production has yet to be determined. Surveys are in progress in mixed cropping systems infested with whiteflies. So far, TYLCV and ToCV diseases are limited to the Algarve region.  相似文献
