The characteristics of arbuscular mycorrhizal fungal (AMF) community structure in various soil depths and growing seasons of watermelon (Citrullus vulgaris) grown in commercial greenhouses in Daxing of Beijing and Weifang and Laiyang of Shandong, China were investigated using both morphological identification and denaturing gradient gel electrophoresis. The sampled soils had been used for continuous greenhouse production of watermelon for 0, 5, 10, 15, or 20 years. Glomus claroideum was the dominant species in the greenhouse soils planted for 5, 10, and 15 years in Laiyang, while Glomus mosseae and Glomus etunicatum were dominant in the nearby open farmland soil. Sorenson’s similarity index of AMF community composition ranged from 0.67 to 0.84 in the soils planted for 5 years, and from 0.29 to 0.33 for 20 years among the three locations. Spore abundance, species richness, and the Shannon index were highest near the soil surface (0-10 cm) and decreased with soil depth, and higher in June and October than in August and December. Canonical correspondence analysis showed that available P and the number of years that soil had been used for greenhouse production were the main factors contributing to the variance of AMF community composition. It was concluded that the community structure of AMF was mainly influenced by soil available P and planting time of watermelon as well as by soil depth and seasonal variation in the commercial greenhouse.  相似文献

Andrographis paniculata is a medicinal plant of immense therapeutic value. The present study was aimed to elucidate its genetic diversity based on morphochemical and RAPD markers from 53 accessions belonging to 5 eco-geographic regions. Analysis of variance and D 2 statistics revealed significant differences in all the metric traits and sufficient inter-cluster distances indicating considerable diversity among the accessions. The complementary approach of RAPD was used to evaluate the genetic dissimilarities among all the accessions using 6 highly polymorphic primers. The average proportion of polymorphic loci across primers was 96.28%. The molecular genetic diversity based on Shannon index per primer averaged 5.585 with values ranging from 3.08 to 8.70 indicating towards wide genetic base. RAPD based UPGMA and D 2 cluster analysis also revealed that various accessions available in different eco-geographic regions might have originated from native places of wild abundance. Similarity matrices were generated for molecular markers and morphometric data to determine the degree of congruence between the two. A highly significant but low correlation (r = 0.547, P < 0.001) was obtained thus implying the correspondence between the two. The species is hermaphroditic and a habitual inbreeder. The present study yielded a typical triangular congruence between its breeding system, morphometric traits and RAPD markers thus elucidating the usefulness of complementary approaches to make diversity analysis more explanatory and purposeful for optimum genetic amelioration and effective conservation of its genotypic variability.  相似文献

Pot experiments were conducted to investigate the effect of soil water regimes on the formation of iron (Fe) plaque on the root surface of rice seedlings (Oryza sativa L.) and on the microbial functional diversity in a paddy soil. The rice seedlings were subjected to three moisture regimes (submergence, 100%, and 60% water‐holding capacity [WHC]), and were grown for 5 and 11 weeks. Aerobic lithotrophic Fe(II)‐oxidizing (FeOB) and acetate‐utilizing Fe(III)‐reducing bacteria (FeRB) in the rhizosphere and non‐rhizosphere soil were determined at 5 weeks using the most probable number (MPN) method. The carbon substrate use patterns of the microbial communities in the rhizosphere and non‐rhizosphere soil samples were determined at 11 weeks using Biolog‐GN2 plates. The amount of Fe plaque (per unit dry root weight) was much higher under submerged conditions than at lower soil moisture contents and decreased with plant age. There was a positive correlation between the amount of Fe plaque and phosphorus accumulated in the Fe plaque at both sampling times (r = 0.98 and 0.92, respectively, n = 12). Numbers of FeOB and FeRB in the submerged soil were lower than in aerobic soil, but by two orders of magnitude higher in the rhizosphere than in the bulk soil. On the other hand, the functional diversity of the rhizosphere microbial communities was much higher than that of the non‐rhizosphere soil, irrespective of soil water regimes. We conclude that soil flooding results in a decreased number and diversity of Fe‐oxidizing/reducing bacteria, while increasing the Fe‐plaque formation.  相似文献

Understanding population genetics and evolution within species requires recognition of variation within and between populations and the ability to distinguish between the potential causes of an observed distribution of variation. For this aim several established indices of diversity, and a novel one, were applied to population samples of the barley powdery mildew pathogen,Erysiphe graminis f. sp.hordei. Random spore samples were obtained from the air along transects through regions of interest across large parts of Europe in 1990. Significant geographical differences in diversity of virulence genotypes occurred among regional sub-samples. Diversity was highest in the samples from eastern Germany, Denmark and Austria, whereas the lowest values were found in the samples from Italy, southern France and parts of western Germany. Diversity in the pathogen population was generally related to the degree of diversification of host resistance in time and space, although there was considerable variation in ranking among different measures of diversity. Sensitivity to sample size proved to be the major problem with the use of several established indices of diversity. Working with very large sample sizes we used multiple random subsamples of various smaller sizes to determine how the mean index values changed with changing sample size. The Shannon index proved to be considerably affected by sample size, in contrast to the Simpson index that was therefore used as a global measure of diversity. Limits of confidence were estimated for the Simpson index using the bootstrap method of numerical resampling. The two aspects contributing to global diversity, richness and evenness, were considered separately to allow meaningful interpretation of the Simpson index. Random sub-sampling was used to reduce the influence of sample size differences for these measures. Dissimilarity, a novel measure of diversity for use in plant pathology, indicates the average number of major genes for host resistance against which pathotypes in a sample respond differently. It is thus able to account for the genetic relationship among pathotypes, which is not considered by any other index. The approaches developed in this study help to compare major forces driving evolution of large-scale populations of the barley mildew pathogen.  相似文献

基于Shannon信息熵理论,分析研究了甘肃省土地利用结构信息熵的动态变化,并对影响信息熵值大小的土地利用类型做了初步探讨.结果表明,1997~2004年,甘肃省土地利用结构信息熵较全国水平低,熵值呈现单调递增趋势,说明该区土地利用系统朝相对无序状态演变.在8个土地利用一级类型中,面积总体呈上升趋势的有园地、林地、居民点及工矿用地、交通用地和未利用地,它们与土地利用结构信息熵呈正相关;面积总体呈下降趋势的有耕地、牧草地和水域,它们与土地利用结构信息熵呈负相关.其中,林地和耕地的面积与该区土地利用结构信息熵的变化显著相关,是信息熵值变化的主要影响因子,然后依次为园地、交通用地、牧草地、居民点及工矿用地、未利用地和水域.  相似文献

以南京市为例,将多样性理论与方法应用于土壤和土地利用相关性评价中,对该市1986年和2003年的遥感数据进行了土地利用分类,计算了各土壤类型和各土地利用类型的构成组分多样性指数以及多网格尺度下的空间分布多样性指数,并提出一种用于评价土壤和土地利用相互关系的关联系数,求算了5 km网格尺度下南京市典型土种单元同9种土地利用类型之间的关联系数。研究结果表明,1986年至2003年这十余年间,南京市土地利用的构成组分变化明显,其中1986年的构成组分多样性为0.361,2003年为0.444。2003年南京市土地利用的空间分布离散性更高,5种主要土地利用类型的空间分布多样性均有明显增加。南京市典型土种与各土地利用类型关联系数的计算结果表明,多组关联系数的变化趋势相同,由此推断相比于1986年,2003年南京市土壤与土地利用之间的相互联系更为复杂多变。  相似文献

调查了印度西高止山脉南北Goal区内25个科的36种药用植物的丛枝菌根真菌多样性。30个物种带有丛枝菌根真菌,其余6个物种没有丛枝菌根真菌定植。在根际士壤中,共发现了来自5个菌属(球囊霉属、无梗囊霉、盾巨孢囊霉、巨孢囊霉属和Ambispora)的42种丛枝菌根真菌。本研究区内,球囊霉属是优势属,聚线绣球菌是优势种。定植百分比和孢子密度间存在显著负相关。南、北Goa区内,Simpson和ShannonWeiner多样性指数差异不显著,说明该区具有稳定、多样的植物群落。  相似文献

The pattern of genetic diversity among 92 genotypes of soybean from 5 different origins/sources (Pakistan, the USA, Asian Vegetable Research Development Centre (AVRDC), Japan and North Korea) was analyzed using randomly amplified polymorphic DNA markers. Out of 20 random primers 6 tested, 10 were polymorphic among genotypes and they yielded 107 markers, with an average of 10.7 markers per primer. The proportion of polymorphic bands within genotypes ranged from 0.47 to 0.71 with an average of 0.59. Pakistani and US genotypes exhibited the highest number of polymorphic bands (95%), while North Korean genotypes revealed the lowest (60%). The mean band frequency of the primers among genotypes was 0.57 with a range of 0.08–0.99. The Shannon’s index and Nei’s genetic diversity index revealed that primer OPF-06 showed maximum genetic diversity among the genotypes. Dendrogram constructed using Unweighted Pair Group Mean Average (UPGMA) method divided the genotypes into 5 main groups consisting of 13 clusters. The results of cluster analysis indicated that the genetic diversity between Pakistani and US or AVRDC genotypes is much larger than that between Pakistani and North Korean or Japanese genotypes. The Pakistani genotypes had distinct bands from plant introductions. Therefore, the Pakistani genotypes may be useful to soybean breeders.  相似文献
