选取8个常见小麦品种,通过室内限菌试验筛选出对于田菁茎瘤固氮根瘤菌Azorhizobium caulinodans ORS571 (A.caulinodans) 侵染响应敏感的品种,研究了浸种侵染、菌液浓度和添加葡萄糖对接菌小麦幼苗生长的影响,并结合荧光显微镜检测绿色荧光蛋白(GFP)标记A.caulinodans在小麦幼苗根系内的分布与定殖规律。结果表明:小偃22为响应敏感品种;在室内限菌条件下,浸种侵染小偃22幼苗平均根长和平均株高分别较对照增加了17.04%和8.37%;最适菌液浓度为1.0108个/mL;在菌液中添加1g/L葡萄糖有助于A.caulinodans浸种侵染和定殖。荧光显微镜检测结果显示,GFP标记A.caulinodans从小麦幼苗根毛和侧根裂隙处进入,定殖于根维管组织等部位。田间试验结果进一步表明A.caulinodans浸种侵染对不同小麦品种均具较明显的促生作用。  相似文献

为探究Toll-6基因在小菜蛾Plutella xylostella先天免疫中的功能,对克隆鉴定的Toll-6基因序列进行生物信息学分析,利用实时荧光定量PCR检测Toll-6基因的时空表达模式及微生物侵染响应模式,体外验证其在微生物结合、病原相关分子模式(pathogen associated molecular pattern,PAMP)结合和细菌凝集等方面的功能。结果表明:Toll-6基因全长2 313 bp,编码770个氨基酸,预测Toll-6蛋白具有富含亮氨酸重复序列(leucine-rich repeats,LRR)胞外区、跨膜区和Toll/白介素-1受体同源(Toll/interleukin-1 receptor homologous,TIR)胞内等典型Toll受体家族特征;Toll-6基因在不同发育阶段及不同组织中均有表达,其中成虫期表达量最高,4龄幼虫期次之,在4龄幼虫中肠表达量最高;此外,苏云金芽胞杆菌Bacillus thuringiensis菌株Bt8010侵染小菜蛾4龄幼虫6 h后,Toll-6基因表达被显著抑制,而黏质沙雷氏菌Serratia marcesc...  相似文献

Effect of the eye fluke Diplostomum spathaceum on fish growth has remained somewhat unclear because 1) the question has not been subjected to experimental examination with treatment-control setup and 2) growth has not been related to the coverage of parasite-induced cataracts in a quantitative manner. We examined effects of the parasite on growth and competitive ability of whitefish (Coregonus lavaretus) in experimental conditions resembling those at fish farms by maintaining groups of exposed and control fish, as well as mixed groups from both treatments, under optimal conditions for 8 weeks. Contrary to our expectations, we did not observe differences in fish growth between the treatments. However, at the level of individual fish, parasite-induced cataracts had a negative effect on weight of the exposed fish but this took place only when the cataract coverage reached 100% in both eyes. No effect of the infection on competitive ability of fish was observed in mixed treatments which may be related to patterns of food distribution in the tanks or cues received by the fish from less infected conspecifics. Overall, the results suggest that effects of D. spathaceum on fish growth become apparent only in cases of very high cataract coverage, and that feeding regimes and the shape of the cataract coverage distribution in a fish group are important determinants of the magnitude of these effects.  相似文献

 通过测定热处理对潜伏侵染于芒果果实中的胶胞炭疽菌(Colletotrichum gloeosporioides Penz.)和香蕉果实中的芭蕉炭疽菌[C.musae(Berk.& Curt.)Arx.]离体培养菌的生长、繁殖和致病性的影响,结果表明:当温度达55℃和60℃,时间20 min时,对菌体的生长和孢子萌发可起明显抑制或杀伤作用,并降低其致病性。2种炭疽菌中,芭蕉炭疽菌比胶胞炭疽菌对热更敏感。作者认为,果实采后热处理时,应根据果实种类和不同菌,采用不同的处理温度和时间,才能得到更好的效果。  相似文献

The study comprises 136 strains of Corynebacterium pyogenes originating from cattle (105), swine (20), sheep (1), and insects (10). For comparison 2 strains of human origin and 1 strain of Gorynebacterium hemolyticum were examined.One of the bovine strains was atypical, being gelatinase-negative, otherwise the strains of Cb. pyogenes were found to be biochemically identical apart from minor deviations in fermentation patterns (Table 1). Neither were antigenic differences demonstrated (gel diffusion analyses, Figs. 1 and 2).Both of the human strains agreed biochemically with Cb. pyogenes (Table 1). By gel diffusion cross analyses one of them was found to be identical with Cb. pyogenes, the other not, though anti-genically related to it (Fig. 2).Gb. hemolyticum deviated biochemically as well as serologically from Gb. pyogenes, but the 2 organisms shared antigenic determinants (Fig. 3).  相似文献

为探明意蜂来源的微孢子虫对不同日龄中蜂成年工蜂的交叉侵染性,将纯化自意蜂的微孢子虫分别接种无微孢子虫感染的2日、5日、8日、11日、14日龄的中蜂工蜂,当每只蜂经口接种5.12×104个孢子,在30±1℃,45%~60%RH,黑暗环境中饲养12天后,2~14日龄的中蜂工蜂的感染率为46.7%~72.6%,平均感染率为65.8%;其中2日龄的感染率显著(P<0.05)低于5~14日龄的感染率。  相似文献

为探讨中蜂来源的微孢子虫对意蜂的侵染性,本试验将纯化自中蜂的微孢子虫,以每只蜂1×105个和5×105个孢子的接种量,分别给2日、5日、8日、11日龄无微孢子虫感染的意蜂成年工蜂接种,在30℃±0.5℃,65%~80%RH,黑暗中隔离饲养10日后,各日龄工蜂的平均感染率达86.3%~100%,每只蜂中肠的平均产孢量为5.60×105~3.39×106个。在两接种量下,2日、5日龄意蜂工蜂的产孢量均显著(P<0.05)高于8日、11日龄的产孢量。试验首次证实了中蜂来源的微孢子虫对意蜂成年工蜂具较强的侵染性。  相似文献

为了更好地进行日本对虾抗病良种的选育,本文用浓度为每毫升1×105个病毒粒子的对虾白斑综合征病毒(white spot syndrome virus,WSSV)粗提液注射感染抗白斑病日本对虾子三代和普通日本对虾,观察其死亡率,同时测定WSSV感染后0,4,24,48和96 h血淋巴酚氧化酶(PO)、酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、超氧化物歧化酶(SOD)活力和蛋白含量.结果显示:注射WSSV 14 d内抗病子三代和普通对虾的平均死亡率分别为15%和85%,前者的抗病保护率达82.4%;感染前前者的各项免疫指标均高于后者,且PO,AKP和蛋白含量存在极显著差异(P<0.01);感染后两者血淋巴检测的5项指标在96 h内的变化趋势基本相似,除ACP以外,抗病子三代的其他指标都比普通日本对虾的高.研究结果表明,所选育的抗白斑病日本对虾子三代能够有效地抵抗对虾白斑综合征病毒的侵染,其免疫指标也高于普通日本对虾.  相似文献

SUMMARY: Experimental infection of Tetrahymena pyriformis was conducted on ornamental fishes with skin wounded by acetic acid treatment. Among fishes used in this experimental infection, guppy Poecilia reticulata , pristella Pristella maxillaris , neontetra Paracheirodon innesi and cherry barbs Puntius titteya proved to be sensitive to challenge, while medaka Oryzias latipes , dwarf gourami Colisa lalia , goldfish Carassius auratus , platy Xiphophorus maculatus and angelfish Pterophyllum scalare were resistant. In catfish Corydoras aeneus infection was unsuccessful. Exposure to Tetrahymena at ≥100 cells/mL at 25 to 30°C and at pH of 6.0 to 8.0 resulted in the successful infection of some sensitive fish species. Histological and bacteriological observations suggest that deeply destroyed skin tissues from the acid-treated method are the primary factors in successful infection.  相似文献

The prevalence of bovine mastitis has been reduced over the past 25 years due to the implementation of a five-point control plan aimed at reducing exposure, duration and transmission of intramammary infections by bacteria. This has markedly reduced the incidence of bovine mastitis caused by bacteria which show a contagious route of transmission, but has had little effect on the incidence of mastitis due to bacteria which infect the gland from an environmental reservoir. Streptococcus uberis is one such bacterium which is responsible for a significant proportion of clinical mastitis worldwide.The inadequacies of the current methods of mastitis control have led to the search for additional measures, particularly vaccines to prevent intramammary infection by this bacterium. Such an approach requires detailed knowledge of the pathogenesis of intramammary infection. Our understanding of this area has grown in recent years but a lack of information still hampers disease control. Both live vaccines and, recently, crude sub-unit vaccines have shown promise against bovine mastitis due to S. uberis. Vaccines against mastitis must, however, be able to control infection without the participation of a marked inflammatory response. This review provides an overview of the recent advances which have been made in our understanding of host-pathogen interactions which promote infection and disease and highlights areas for strategic research aimed at controlling this bacterial infection.  相似文献
