Piscine orthoreovirus genotype 1 (PRV‐1) is widespread in farmed Atlantic salmon (Salmo salar L.) populations in northern Europe, Canada and Chile. PRV‐1 occurs in wild fish in Norway and Canada; however, little information of its geographical distribution in wild populations is currently available, and the effect of PRV‐1 infection in wild populations is currently unknown. In this study, we present the findings of a survey conducted on 1,130 wild salmonids sampled in Denmark, Sweden, Ireland, Faroe Islands, France, Belgium and Greenland between 2008 and 2017. PRV‐1 is reported for the first time in wild salmonids in Denmark, Sweden, Faroe Island and Ireland. The annual PRV‐1 prevalence ranged from 0% in France, Belgium and Greenland to 43% in Faroe Islands. In total, 66 samples tested positive for PRV‐1, including Atlantic salmon broodfish returning to spawn and Atlantic salmon collected at the feeding ground north of Faroe Islands. The phylogenetic analysis of S1 sequences of the PRV‐1 isolates obtained in this survey did not show systematic geographical distribution. This study sheds light on the spread and genetic diversity of the virus identified in populations of free‐living fish and provides rationale for screening wild broodfish used in restocking programmes.  相似文献

对15尾大鳞副泥鳅的线粒体细胞色素b基因进行PCR扩增和正反测序,获得长度为1140 bp的细胞色素b基因同源片断。所有序列中共检测到15种单倍型,序列中共出现81个变异位点,总变异率为7.1%。各单倍型的变异全部是转换或颠换,无插入和缺失,转换/颠换比为6.36。变异位点在3位密码子中的分布呈现偏倚,密码子第3位的变异占总变异的85.18%,而第1和第2位均只占7.41%。A、T、C、G碱基的平均含量分别为27.4%、30.8%、26.5%、15.3%,A+T>C+G。在所得序列中,密码子第1位上4种碱基使用较为均衡;第2位上碱基T的使用率高达41%,碱基G的使用率低至13.2%;第3位上碱基A的使用率为37.9%,而碱基G的使用率仅为7.0%。以红尾副鳅为外群用邻接法构建分子系统进化树。结果显示,在系统地位上大鳞副泥鳅与黑龙江泥鳅有较近的亲缘关系。  相似文献

Genetic variability in melon based on microsatellite variation   总被引:8,自引:0,他引:8  
A set of 18 simple‐sequence repeat (SSR or microsatellite) markers was used to study genetic diversity in a collection of 27 melon (Cucumis melo L.) accessions, representing a broad range of wild and cultivated melons. The materials studied were highly polymorphic for SSRs and a total of 114 alleles were detected (average of 6.3 alleles per locus). Cluster analysis suggests the division of these accessions into two major groups, largely corresponding to the division of C. melo in the two subspecies agrestis and melo. The assignment of the accession to the subspecies was generally in agreement with published reports, except for those corresponding to the ‘dudaim’ and ‘chito’ cultivar groups, which, according to the observed SSR variability, should be included in subspecies agrestis. Based on cluster analysis, five groups of accessions were defined. The two most divergent groups include mainly accessions from the Mediterranean which form one group, and accessions from China, Japan, Korea and India forming the other. Both groups shared a low level of intra‐accession variation compared with the other groups, which suggests an erosion of their genetic variability because of drift and/or inbreeding. The remaining accessions, mainly from Central Africa and India, were more variable and may be an important source of genetic variation for melon breeding.  相似文献

Three apple (Malus×domestica) cultivars and 11 Malus accessions have been investigated by the probe hybridization method on their mitochondrial DNA (mtDNA). The gene probes used were: coxI, coxII, atpA, atp6, and atp9. Our results revealed enough variation to characterize ten mtDNA haplotypes among the Malus genotypes examined. The taxonomic and phylogenetic implications of mtDNA polymorphism are also discussed.  相似文献

Variability of acid phosphatase (ACP), esterase (EST), and superoxide dismutase (SOD) isoenzymes among 660 accessions of wild diploid wheats Triticum boeoticum (475) and T. urartu (185) and among 35 accessions of wild tetraploid wheats T. araraticum (21) and T. dicoccoides (14) was studied with the use of polyacrylamide gel electrophoresis. The wild diploids are differentiated by the electrophoretic variants of ACP-B, ACP-C, EST-A, EST-B, EST-C, EST-D, and SOD-B. Triticum urartu matches the A genome of wild tetraploid wheats T. araraticum and T. dicoccoides by allozymes of ACP-B, ACP-C and SOD-B, but differs distinctly by the monomorphic allozyme EST-A66. Triticum boeoticum fits the A genome of polyploid wheats by allozymes of EST-A and EST-B, but differs in allozymes of ACP-B and SOD-B. Thus, none of the two contemporary diploid wheats corresponds exactly to the A genome of wild tetraploid wheats by all diagnostic isoenzymes. Triticum boeoticum and T. arararicum share EST-C46 and EST-D71, whereas T. urartu and T. dicoccoides are fixed for silent EST-C0 and share EST-D73, suggesting independent origin of the two tetraploid wheats with a contribution from ancestral forms of both diploids.  相似文献

Summary The occurrence of chromosomal interchanges was studied in F1 hyrids in two diallel schemes, including nine accessions of Triticum aestivum subsp. spelta, originating from Iran, Europe and North Africa, and the common wheat variety Chinese Spring as a reference. Seven accessions of subsp. spelta differed in one translocation from Chinese Spring. The European accession Grey differed in one or perhaps two interchanges from the reference variety, and Iran 417a showed a difference of three interchanges with Chinese Spring. In total six or perhaps seven interchanges were observed. Thus the number of translocation events in subsp. spelta appeared to be low. One translocation was unique and occurred only in the accession from North Africa. Also one accession from Europe, the landrace Schwarzwald, possibly carried a unique translocation. In both diallels an interchange with a relatively low coefficient of multivalent realisation at first meiotic metaphase was observed in most accessions. The two extra translocations in Iran 417a are considered to be the result of more recent translocation events, whereas the possible second translocation in accession Grey showed such a low coefficient of multivalent realisation that its existence could be doubted.  相似文献

Mitochondrial DNA RFLP in genus Oryza and cultivated rice   总被引:1,自引:0,他引:1  
Summary Ninety-three accessions representing 23 species from the genus Oryza were surveyed for restriction fragment length polymorphism (RFLP) in mitochondrial (mt) DNA by probing total DNA with 15 known mt sequences cloned in plasmids from higher plants, and five mt genomic cosmid clones from maize. Very low levels of intra-specific and even intra-cytologically-defined nuclear genome mt DNA RFLP were found. High between-genome differentiation appeared, suggesting phylogenetic relationships consistent with data from previous nuclear and chloroplast (cp) DNA studies. Parallel inheritance of cp and mt DNA was found. There was one major exception: the mt DNA of the allotetraploid CD genome is apparently equally related to two putative diploid progenitors, which is suggestive of an interspecific recombination.RRLP in mt DNA was also probed in 82 cultivars, with four plasmid probes. Some bands not seen in the wild species appeared in O. sativa, with intra-specific polymorphism relatively higher than in the wild species. The pattern of variation paralleled that at the cp DNA level between the indica and japonica subspecies.  相似文献
