双波长紫外分光光度法测定复方恩诺沙星可溶性粉中恩诺沙星和盐酸多西环素的含量

建立了复方恩诺沙星可溶性粉中恩诺沙星和盐酸多西环素含量的检测方法.采用双波长紫外分光光度法,直接测定两组分的含量.结果显示可同时测出复方恩诺沙星可溶性粉中恩诺沙星和盐酸多西环素的各自含量,且吸收度与各自的浓度成良好线性关系.恩诺沙星平均回收率为100.1%,RSD为0.04%(n=5).盐酸多西环素平均回收率为100.2%,RSD为0.14%(n=5).本方法简便、快速、可靠,可用于复方恩诺沙星可溶性粉的含量测定.     

Novel Canine Tumour Suppressor Gene p53 Mutations in Cases of Skin and Mammary Neoplasms

Exons 4 to 8 of the tumour suppressor gene p53 were analysed in 25 skin and 25 mammary tumours of 50 dogs. A 1 bp deletion (ACCAC) was detected in codon 89 in exon 4 in a squamous cell carcinoma. A missense mutation CGCCAC (argininehistidine) was present in codon 162 in exon 5 in a mammary adenocarcinoma. Moreover, a silent mutation occurred in codon 103 (serine) of exon 4 in a mammary adenoma. The somatic nature of the three mutations was demonstrated.     

Diagnosis of dehydratase inhibitors in melanin biosynthesis inhibitor (MBI-D) resistance by primer-introduced restriction enzyme analysis in scytalone dehydratase gene of Magnaporthe grisea

We have established a simple diagnosis method for rice blast fungus resistant to MBI-D. This involves the preparation of PCR templates directly from the lesions in combination with primer-introduced restriction enzyme analysis PCR (PIRA-PCR).     

被毛突变小鼠解剖及繁殖学特性观察

对一种新的具有个毛、无毛、稀毛３种表型的被毛突变小鼠的大体解剖学特性研究结果表明,其心肝、脾、肺、肾、肾上腺等脏器的位置、形态及组织学结构无表型间差异,但无毛小鼠被毛和皮肤结构异常,大多数脏器的重量和脏器系数表现出表型差异和性别差异（Ｐ〈０．０５或Ｐ〈０．０１）。繁殖学特性研究结果表明,虽然无毛小鼠雌雄生殖系统的器官组成、发情周期与其他２种表型之间无明显差异,但其雄性的初情期明显滞后（约２周左右）     

亚硝酸诱变选育黄霉素高产菌的研究

采用亚硝酸诱变方法对黄霉素产生菌进行诱变处理,利用菌株抗自身代谢物的特征,在添加高浓度黄霉素的分离培养基上选育获得优良高产突变株sg2-N2-2,比发酵菌株的发酵单位提高了7.2倍,而且变株的菌落形态与亲本菌株差异较大。     

多种提取方法比较野生与栽培蒲公英绿原酸含量

目的:比较野生与栽培蒲公英绿原酸含量。方法:利用紫外分光光度法,通过超声波法、乙醇回流法、甲醇回流法提取,分别测定野生蒲公英和栽培四倍体蒲公英的绿原酸含量。结果:超声波提取法:栽培蒲公英绿原酸含量0.41%,野生蒲公英绿原酸含量0.25%;乙醇回流法:栽培蒲公英绿原酸含量0.33%,野生蒲公英绿原酸含量0.23%;甲醇回流法:栽培蒲公英绿原酸含量0.31%,野生蒲公英绿原酸含量0.19%。结论:栽培蒲公英中绿原酸含量较高。     

Mutants in wheat showing multipathogen resistance to biotrophic fungal pathogens

Five fast-neutron-derived mutants were isolated from the wheat line Hobbit 'sib' that show enhanced field resistance towards Puccinia striiformis f.sp. tritici , the causal agent of yellow rust. Subsequent testing showed the yellow rust resistance phenotypes to differ between mutants, to be expressed at different growth stages and, in some cases, to show an isolate interaction. Three mutants, I3-48, I3-49 and I3-54, exhibited an enhanced yellow rust resistance phenotype from the third seedling leaf onwards, while mutants I3-27 and I3-30 did not show an altered yellow rust phenotype until later growth stages. Additional resistance for brown rust (causal agent Puccinia triticina ) was identified in mutants I3-27, I3-30, I3-48 and I3-49, and for powdery mildew caused by Blumeria graminis f.sp. tritici in mutants I3-27, I3-30, I3-48 and I3-54, although in some cases the resistance was isolate-specific.     

家蝇抗菌肽的研究进展马红霞

从家蝇抗菌肽的分类、结构特点、作用机理、诱变、活性检验以及分子生物学研究等方面介绍了家蝇抗菌肽的研究进展。家蝇抗菌肽独特的作用机理、成熟的诱导、分离纯化技术及其相关基因的克隆、表达等研究成果均为对家蝇抗菌肽进行更深入的研究奠定了基础。采用分子生物学技术有望筛选并制备大量的高效家蝇抗菌肽。     

Construction of Brucella ClpS Gene Mutant Strain and Its Effect on Virulence

The present study aimed to determine the role of ClpS gene,and to analyse the impact of ClpS mutation on the virulence of Brucella.A ClpS gene mutant strain,named ΔClpS was constructed by homologous recombination technology.The bacterial growth kinetics,the LPS synthesis ability and the survival ability of bacterial within macrophages as well as the virulence in mouse model were measured.In addition,the difference between parent strain 2308 and the mutant strain ΔClpS were compared.The results showed that under the same culture conditions,no difference in bacterial concentration was observed between 2308 and ΔClpS strains.The silver staining examination showed that the expression level of LPS extracted from two strains were similar,indicating ClpS gene mutation did not alter the growth rate and LPS synthesis ability of Brucella. In the cell infection assay,the survival ability of ΔClpS strain in cells was extremely significantly lower than that of 2308 strain at 72 h after infection (P<0.01).The results of mouse infection experiment showed that in the first week after infection,no significant difference in spleen weight and bacterial concentration between 2308 and ΔClpS strains infected mice was observed.However,at 4 weeks after infection,the bacterial concentration in spleen of ΔClpS infected mice was 10^3.93 CFU/g spleen,which was significantly lower than that of 2308 strain (10^6.68 CFU/g spleen,P<0.01).The spleen weight of ΔClpS infected mice was also remarkably lower than that of 2308 strain (P<0.01).In summary,the results suggested that the ClpS gene of Brucella did not play a role in Brucella growth rate and ability of LPS synthesis,whereas ClpS gene mutation decreased the ability of Brucella colonization in mouse spleen.     

苹果新品种‘天汪1号’

 ‘天汪1号’是从‘红星’的芽变中选育出的苹果短枝型新品种, 在天水地区9月中旬成熟,果实生育期148 d。果实圆锥形, 五棱突起, 果形指数0.90～0.98, 平均单果质量210 g, 果实全面浓红,色相片红; 果肉细而致密, 汁多, 味香甜, 可溶性固形物含量11.9%～14.1%。树势强健, 丰产, 稳产。
