根据伪狂犬病病毒 Rise毒株 g E基因序列 ,设计合成 1对引物 ,采用 PCR方法 ,从伪狂犬病病毒闽 A株 DNA扩增出 5 5 8bp片段。将该片段克隆到 p UC19质粒中 ,测序正确后再将其克隆到表达质粒 p ET- 2 8a中 ,以 BL 2 1(DE3)为宿主菌 ,在 IPTG诱导下获得了高效表达。经 SDS- PAGE及 Western- blotting鉴定分析 ,表达蛋白大小约 2 30 0 0 ,蛋白薄层扫描分析表明 ,该蛋白约占菌体总蛋白的 30 %。  相似文献

The poor adhesion of bamboo coatings is a serious issue in the bamboo industry. To develop a pretreatment method that improves the adhesion of films and increases the economic value of bamboo products, a study of hydrogen peroxide treatment with solutions of various pH (pH 4–9) on bamboo surfaces was conducted. Five-year-old ma bamboo (Dendrocalamus latiflorus Munro), and nitrocellulose lacquer were used in the study. Surface properties of the bamboo such as contact angle and color were evaluated, and 180° peel strength and shear strength tests for measuring adhesion of films were conducted. The results showed that the wettability of water droplets and the carbonyl group concentration on the bamboo surface were increased significantly by alkaline (pH 8 and 9) hydrogen peroxide treatments. There was only minor color variation and the outer wax layer of bamboo was etched to form additional recesses after hydrogen peroxide treatment. It was also found that all hydrogen peroxide treatments improved the adhesion of bamboo coating; bamboo treated with hydrogen peroxide at pH 7 showed the greatest improvement. The enhanced adhesion was attributed to the mechanical interlocking of the film on the treated bamboo rather than to surface activation.  相似文献

This experiment was conducted to study the effect of dietary supplementation of maggot dry on meat performance,nutritional quality and flavor compounds of meat in Qingjiao Ma chickens.A total of 1200 Qingjiao Ma cocks at 35 days of age were randomly allocated to 2 groups with 4 replicates per group and 150 chickens per replicate.The chickens in control group were only fed the basic diet (without maggot dry),and those in the experiment group were fed basal diet supplemented with 3% maggot dry.The chickens in two groups were raised in the same conditions.At 70 days of age,eight cocks were randomly selected and slaughtered in each group.The results showed as follows:①The meat performance in control group and experiment group were all good,and had no significant difference (P>0.05).② Compared with control group,the crude protein contents of chest and leg muscle in experiment group were increased by 8.8% and 4.4% (P<0.05),respectively,and the amino acids (especially essential amino acid) contents were increased by 11.0% and 2.3% (P<0.05),respectively;The relative contents of nutmeg acid and pentadecane acid in chest and leg muscle of experiment group were significantly higher than those of control group (P<0.05).③ Compared with control group,the delicious amino acid content in chest and leg muscle were increased by 11.9% and 3.5%,respectively;the inosine acid content in chest muscle of experiment group was increased by 21.1%,The four kinds of special volatile flavor substances (dipentene,methoxyacetone,3-nonanone and isobutyl lactate) of meat in experiment group were obtained,and the flavour components of aldehyde and mushroom alcohol were higher than those of control group.Therefore,dietary supplementation of 3% maggot dry could increase the contents of crude protein,amino acids,delicious substance and volatile flavour compounds,and helped to produce chickens with flavor and nutritious meats.  相似文献

目前肉苁蓉正处于野生种向栽培种驯化的初级阶段,肉苁蓉接种同步性差和接种成活率低是生产中的突出问题。本试验在不同起源的梭梭林内,使用不同质量的肉苁蓉种子,采用不同药剂处理、不同接种深度、不同接种方法、不同接种量等接种措施进行人工接种成功率对比试验。结果表明:用一、二级肉苁蓉种子、500ppmABT(3)号生根粉浸泡30分钟、4月上旬至5月中旬用营养诱导法接种、接种垂直深度与水平距离40-60cm之间接种成功率最高。这项研究将为肉苁蓉人工高效接种提供理论依据与技术支持。  相似文献

以提高荒漠肉苁蓉产量,改良其品质为目的,首次对肉苁蓉进行了多倍体诱导的研究.使用秋水仙素与0.1;琼脂和二甲基亚砜混合液处理植株生长点,分别处理1、2和3 d.处理3 d能得到多倍体植株,多倍体植株与原二倍体植株相比,在形态上,多倍体植株肉质茎变粗,子房体积增大,花粉粒明显变大,小孢子染色体数目为2 n=2 x=40,而原二倍体植株的花粉染色体数目为n=x=20,初步证明诱变植株为同源四倍体.用HPLC对肉苁蓉中松果菊苷进行定量测定,发现诱变株有效成分增加12.7;.  相似文献

通过简并引物和降落PCR方法结合RACE(rapid amplification of c DNA end)技术在香蕉果实c DNA文库中获得2个14-3-3基因,分别命名为Ma14-3-3d和Ma14-3-3h,把Ma14-3-3d和Ma14-3-3h与Ma14-3-3a、Ma14-3-3c、Ma14-3-3e、Ma14-3-3i进行多重序列比对和同源性比较。结果表明:Ma14-3-3d和Ma14-3-3h与Ma14-3-3a、Ma14-3-3c、Ma14-3-3e、Ma14-3-3i具有较高的同源性,同时具有一定的差异性。遗传进化分析结果表明,Ma14-3-3d与Ma14-3-3a、Ma14-3-3c、Ma14-3-3e、Ma14-3-3i同属于non-ε类14-3-3基因,Ma14-3-3h属于ε类14-3-3基因。RT-PCR分析结果表明,Ma14-3-3d和Ma14-3-3h在香蕉不同器官中差异表达,Ma14-3-3d在香蕉的根、茎、叶、花和果中均有表达,且在茎、叶和花中的表达量高于根和果;Ma14-3-3h在根、花和果中的表达量较高,而在茎和叶中的表达量较低。q RT-PCR分析结果表明,Ma14-3-3d基因的表达明显受乙烯的诱导,而Ma14-3-3h在正常成熟、乙烯处理与1-MCP处理的果实中表达量均很低,与果实成熟的相关性不明显。推测Ma14-3-3d可能与香蕉果实成熟密切相关,可能参与乙烯调控果实成熟过程中的生物合成与信号转导。  相似文献

本试验采用化学分析法和尼龙袋法测定了大叶麻竹笋壳的有机物(OM)、粗蛋白质(CP)、中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)、粗灰分(Ash)和钙(Ca)、磷(P)含量,以及在山羊瘤胃内干物质和有机物的降解率。结果表明:大叶麻竹笋壳各常规养分含量(绝干基础)为OM 95.7204%、CP 8.1185%、NDF 78.904 4%、ADF 45.742 3%、Ash 4.279 3%、Ca 0.119 0%、P 0.148 7%,山羊瘤胃内的干物质和有机物的有效降解率分别为DM 29.5577%、OM 25.9155%。  相似文献
