秀珍菇原基形成相关基因PpFBD1的克隆与表达研究

前期研究中通过对秀珍菇经低温诱导后不同发育阶段样本进行转录组测序及差异表达基因分析发现,ID为Cluster-6377.64510的基因(命名为PpFBD1)在原基形成前期的样本中高表达。为进一步了解分析其表达特性及功能,根据转录组测序结果设计特异性引物,利用RT-PCR技术从秀珍菇中克隆获得了该基因的cDNA全长。该基因由342个核苷酸组成,编码一个由113个氨基酸组成的蛋白,蛋白相对分子质量约11 ku。进化树分析显示,PpFBD1编码蛋白与糙皮侧耳hydrophobin1编码的疏水蛋白亲缘关系最近。Gene Ontology功能分析表明,PpFBD1主要参与真菌类细胞壁的合成。荧光定量RT-PCR检测显示,该基因在菌丝体经低温诱导后恢复至室温阶段(原基形成前期)表达量最高,这表明其可能参与调控秀珍菇原基形成过程。本研究结果为阐明秀珍菇原基形成机制提供参考。     

不同贮藏温度和保鲜处理对黄秋葵贮藏品质及生理的影响

为探讨采后不同贮藏温度对黄秋葵贮藏品质的影响以及不同保鲜剂对黄秋葵抗氧化酶活性的影响，以台湾“五福”黄秋葵为试材，分别置于低温（（4±1）℃、（9±1）℃、（14±1）℃）和常温（（20±1）℃）条件下贮藏，研究温度对其贮藏品质的影响；采用1.0%壳聚糖、5.0 mmol/L水杨酸、0.5片安喜布、5%O2+8%CO2处理黄秋葵，研究保鲜处理对其抗氧化酶活力的影响。结果表明：（9±1）℃贮藏组的保鲜效果最好，显著改善了贮藏期间黄秋葵的失重率、硬度、叶绿素含量、VC含量、L值、呼吸强度、乙烯释放量、MDA含量、细胞膜相对电导率；4种保鲜处理均能显著提高黄秋葵的SOD、POD、CAT活性，并维持其活性在相对较高的水平；同时，显著抑制黄秋葵果实MDA含量的升高。通过研究发现，壳聚糖涂膜液处理组的效果最佳，水杨酸处理组次之并优于1-MCP处理组，气调处理组的效果相对较差。     

杂交旱稻丰产节水栽培关键技术研究与集成应用

针对安徽沿淮地区气候特点,通过试验筛选出适宜沿淮地区旱种旱管栽培,丰产性和稳产性较好的旱稻品种5个,节水耐旱型水稻品种7个;明确了沿淮地区以旱优73为代表的杂交旱稻的丰产节水栽培技术:播种期6月5-12日,播种量2.0 kg/667 m~2左右,氮肥用量10~14 kg/667 m~2;在施肥过程中适当降低基蘖肥用量、增施穗粒肥,基、蘖、穗肥比例以4∶4∶2较好。     

Verrucosamide,a Cytotoxic 1,4-Thiazepane-Containing Thiodepsipeptide from a Marine-Derived Actinomycete

A new cytotoxic thiodepsipeptide, verrucosamide (1), was isolated along with the known, related cyclic peptide thiocoraline, from the extract of a marine-derived actinomycete, a Verrucosispora sp., our strain CNX-026. The new peptide, which is composed of two rare seven-membered 1,4-thiazepane rings, was elucidated by a combination of spectral methods and the absolute configuration was determined by a single X-ray diffraction study. Verrucosamide (1) showed moderate cytotoxicity and selectivity in the NCI 60 cell line bioassay. The most susceptible cell lines were MDA-MB-468 breast carcinoma with an LD₅₀ of 1.26 µM, and COLO 205 colon adenocarcinoma with an LD₅₀ of 1.4 µM. Also isolated along with verrucosamide were three small 3-hydroxy(alkoxy)-quinaldic acid derivatives that appear to be products of the same biosynthetic pathway.     

鸡传染性贫血病毒VP1、VP2基因在杆状病毒表达系统中的表达

将鸡传染性贫血病毒M9905株VP1、VP2基因分别或同时克隆到杆状病毒转移载体pFastBacDUAL中，然后转化到DHIOBAC感受态细胞中与Bacmid杆状病毒穿梭载体进行转座重组，最后将重组子转染Sf9昆虫细胞，得到分别或同时含VP1、VP2基因的重组杆状病毒rBacVP1、rBacVP2及rBacVP1—2。PCR扩增结果证实VP1、VP2基因重组到杆状病毒基因组中；SDS-PAGE电泳分析和间接免疫荧光试验结果表明VP1、VP2基因在重组病毒中得到了表达。     

Impact of postovulatory food deprivation on the ova transport, hormonal profiles and metabolic changes in sows.

The effect of food deprivation on ova transport, hormonal profiles and metabolic changes was studied in 20 crossbred multiparous sows during their second oestrus after weaning. To determine the time of ovulation, transrectal ultrasonographic examination was performed. The sows were divided into 2 groups, one control group (C-group), which was fed according to Swedish standards, and one experimental group (E-group). The E-group sows were deprived of food from the first morning meal after ovulation until slaughter. Blood samples were collected every second hour from about 12 h before expected ovulation in the second oestrus after weaning until slaughter and were analysed for progesterone, prostaglandin F2 alpha-metabolite, insulin, glucose, free fatty acids and triglycerides. All sows were slaughtered approximately 48 h after ovulation and the genital tract was recovered. The isthmic part of the oviduct was divided into 3 equally long segments and flushed separately with phosphate buffered saline (PBS). Uterine horns were also flushed with PBS. A significantly greater number of ova were found in the first and second part of the isthmus in the E-group (p = 0.05) while in the C-group most of the ova were found in the third part of the isthmus or the uterus (p = 0.01). The level of prostaglandin F2 alpha-metabolite was significantly higher in the E-group compared with the C-group. The concentration of progesterone increased in both groups after ovulation but there were no significant differences between the groups. The other blood parameters showed that the food-deprived sows were in a catabolic state. The 48 h period of fasting results, directly or indirectly in an delayed ova transport, which may be due to a delayed relaxation in the smooth circular muscle layer of the isthmus.     

导入AGPase基因的转基因可育水稻及其经济性状的研究

以农杆菌介导法将淀粉合成关键酶AGPase(腺嘌呤二磷酸葡萄糖焦磷酸化酶）基因导入云南地方优质水稻合系35，38，39，41，42中，获得了大量的可育转基因植株。PCR扩增检测，外源基因已稳定整合进水稻基因组中。大量的农艺性状及淀粉含量研究表明，转基因水稻种子的千粒重比对照明显提高，但总淀粉含量没有显著变化。     

甘蓝型双低隐性核不育杂交种沪油杂1号的选育

沪油杂1号(原编号SH-19)是利用隐性上位互作核雄性不育系20118A育成的甘蓝型双低杂交油菜品种.20118A的育性由2对隐性重叠基因和1对隐性上位基因控制,基因型为aabbRfRf,临保系的基因型为aabbrfrf,恢复系的基因型为AA__或__BB__.沪油杂1号种子含油量42.43%,芥酸含量0.62%,硫甙含量24.23μmol/g.在上海市油菜区域试验中平均产量2 243.3kg/hm2,比双高对照汇油50增产11.9%,比双低对照沪油15增产3.8%;在上海市油菜生产试验中平均产量2 244.0kg/hm2,分别比对照汇油50增产33.6%,比对照沪油15增产15.9%.

同源四倍体水稻与狼尾草杂交结实的胚胎学证据

在以6份同源四倍体水稻为杂交母本与狼尾草进行属间杂交中发现,其结实率达到0.84%～2.06%,而在以二倍体水稻为母本的属间杂交中却没有获得结实种子.利用激光扫描共聚焦显微技术对属间杂交结籽的特殊生殖现象进行了观察研究,试图寻找其杂交结实的胚胎学证据.结果表明,狼尾草的花粉粒不能在二倍体水稻的柱头上萌发,说明二倍体水稻与狼尾草的生殖隔离很严格,两者很难杂交.然而,狼尾草的花粉粒在同源四倍体水稻的柱头上能萌发,花粉管能在花柱中伸长并能将雄配子送入胚囊内与雌配子融合成受精卵.同源四倍体水稻与狼尾草的受精作用比较特殊,包括精细胞与卵细胞的单受精作用、精细胞与次生极核的单受精作用和正常的双受精作用,总受精率和总成胚率分别为6.36%～11.68%和4.11%～9.44%.试验结果为解释同源四倍体水稻与狼尾草杂交结籽的特殊生殖现象提供了有力的胚胎学证据.
