Effects of remifentanil on monophasic action potential and transmural dispersion of repolarization in rabbit myocardium

AIM: To study the effect of remifentanil on monophasic action potential and transmural dispersion of repolarization (TDR) in the 3-layer myocardium of isolated rabbit hearts. METHODS: Adult rabbits (n=18, 2.0 ~ 2.5 kg) were used to isolate the hearts for preparing Langendorff perfusion model. The hearts were randomly divided into 3 groups after perfusion with K-H solution for 15 min: the perfusion in control group (C group) continued for 60 min; the hearts in remifentanil group (R group) were perfused with 12 μg/L remifentanil K-H solution for 60 min; the hearts in remifentanil+aminophylline group (RA group) were given 60-min perfusion of 12 μg/L K-H remifentanil+30 mg/L aminophylline. The HR and 3 layers of myocardial monophasic action potential (MAP) in the left ventricular anterior wall were recorded at time points after balanced infusion for 15 min (T₀), and continued perfusion for 15 min (T₁), 30 min (T₂) and 60 min (T₃). The monophasic action potential duration of repolarization at 90% (MAPD₉₀) and the transmural dispersion of repolarization (TDR) were calculated. The early afterdepolarization, delay afterdepolarization and arrhythmia were also observed. RESULTS: In R group, slower HR and prolonger MAPD₉₀ and TDR at T₁~T₃ were observed as compared with those at T₀ (P<0.05). R group showed slower HR and longer MAPD₉₀ and TDR than C group and RA group (P<0.05). CONCLUSION: Remifentanil slows the HR, extends the MAPD₉₀ and increases the TDR, thus being prone to induce reentry. Aminophylline makes HR faster and MAPD₉₀ shorter, thereby reducing the TDR.     

低温日灼对核桃腐烂病发生的影响

以"辽宁1号"等6个核桃品种为试材,对参试树进行低温日灼及腐烂病发生的方位、病级、发病率等统计分析,研究了核桃腐烂病的发生与低温日灼的关系。结果表明:核桃腐烂病主要发生在成龄结果树枝干南或西南方向,发病处均伴有低温日灼现象;核桃不同品种间抗逆性存在着较大差异,低温日灼发生率低的品种腐烂病发生率也较低,但发生率均与发病程度无必然联系;腐烂病病情与低温日灼伤情密切相关。     

Influence of paricalcitol on renal tubulointerstitial fibrosis in diabetic nephropathy

AIM: To investigate the effect of paricalcitol (P) on renal tubulointerstitial fibrosis and the underlying mechanisms in diabetic nephropathy (DN).METHODS: DN rat model was induced by a single intraperitoneal injection of streptozotocin after fasting. The animals were randomly divided into 2 groups:the DN rats in paricalcitol-intervened group (group P) were injected intraperitoneally with paricalcitol dissolved in propylene glycol after the day when the model was induced successfully at a dose of 0.4 μg/kg (3 times a week); the DN rats in DN group (group D) were given isopyknic propylene glycol. Normal control group (group C) was also set up. The samples of blood, urine and renal tissue were collected after intervention of paricalcitol for 12 weeks. The biochemical indexes were measured. The renal tissues were used for pathologic observation and determining the expression of transforming growth factor-β1 (TGF-β1), Wnt-4, β-catenin and Klotho by immunohistochemistry and Western blotting. In addition, the correlation among the above indexes was analyzed.RESULTS: (1) Scr, BUN and 24 h urine protein increased significantly in group D compared with group C, while decreased in group P compared with group D (P<0.05). (2) The area of renal tubulointerstitial fibrosis increased in group D compared with group C, while decreased in group P compared with group D (P<0.05). (3) The expression of Klotho decreased, while the expression of TGF-β1, Wnt-4 and β-catenin increased in group D compared with group C (P<0.05). Compared with group D, the expression of Klotho increased, while the expression of TGF-β1, Wnt-4 and β-catenin decreased in group P (P<0.05). (4) The expression of Klotho was negatively correlated with the fibrosis area, TGF-β1, Wnt-4 and β-catenin (P<0.05).CONCLUSION: Paricalcitol inhibits renal tubulointerstitial fibrosis in DN by promoting the expression of renal Klotho, and inhibiting Wnt/β-catenin signaling pathway activation and TGF-β1 synthesis.     

肠致病性大肠杆菌DPO-PCR快速检测方法的建立

为建立肠致病性大肠杆菌(EPEC)的快速检测方法,本研究以EPEC bfp A基因为靶基因设计了一对双启动寡核苷酸引物(DPO),建立了EPEC的DPO-PCR快速检测方法。结果显示,退火温度在45℃~65℃范围内均能够高效地扩增出目的基因,表明DPO引物对退火温度不敏感。该方法对其他菌株的扩增结果均为阴性,特异性强;其灵敏度为97 cfu/m L。利用该方法和国标法分别对采集的230份临床样品进行检测,均共计检出5份EPEC阳性样品,两者符合率为100%。本研究建立的DPO-PCR方法设计简单、特异性强、灵敏度高,具有良好的实用性,为快速准确检测EPEC提供新的检测手段。     

Expression,Purification and Activity Evaluation of Mb1230 Gene of Mycobacterium bovis

To explore the value of Mb1230 protein of Mycobacterium bovis in the diagnosis of bovine tuberculosis,we obtained the Mb1230 gene by PCR and constructed recombinant plasmid pET-22b-Mb1230.Recombinant Mb1230 protein was obtained by IPTG induction and purified by affinity chromatography.The activity of the recombinant protein was evaluated by TST test,IGRA test and indirect ELISA.The size of the recombinant protein matched with the theoretical value proved by SDS-PAGE;Western blotting result showed that the recombinant protein could react with mouse anti-His antibody,and had specific band;The results of TST test,IGRA test and indirect ELISA test also showed the recombinant protein had antigenic activity.The results indicated the recombinant protein Mb1230 had good B cell and T cell activity,so,it had the potential application in the diagnosis of bovine tuberculosis.

副猪嗜血杆菌单克隆抗体的制备及其B细胞抗原表位的鉴定

为筛选具有临床诊断价值的副猪嗜血杆菌(H.parasuis)单克隆抗体(MAb),本研究采用H.parasuis 5型强毒株(HPS-5)免疫BALB/c小鼠,利用常规细胞融合技术制备了一株稳定分泌MAb的杂交瘤细胞株。Western blot结果表明该MAb能够特异性识别所有H.parasuis标准菌株;免疫共沉淀技术与蛋白质谱分析显示该MAb所识别的蛋白为ATP结合盒转运蛋白(ABCT)家族的寡肽透过酶(OppA);利用MAb对噬菌体12肽库进行淘选,8个阳性噬菌体克隆展示有"KTPAE-R"保守序列,对应于H.parasuis SH29755株和SH0165株的469位~475位的氨基酸残基。对OppA氨基酸序列比对结果表明H.parasuis与其它10种猪常见细菌病病原序列一致性为9.1%~74.5%,本研究结果为进一步研究H.parasuis感染的病原学诊断建立了良好的基础。     

福州地区冬菠菜品种比较试验

冬季对7个菠菜品种进行植物学性状、产量、冬性以及抗病性进行比较试验,结果表明,BC2、越超2个品种具备长势强、叶色绿、抗病、冬性强、产量高等特点,可作为福州地区冬季主栽品种。
