The outer membrane proteins (OMPs) of Brucella (B.) abortus have been extensively studied, but their immunogenicity and protective ability against B. abortus infection are still unclear. In the present study, B. abortus Omp28, a group 3 antigen, was amplified by PCR and cloned into a maltose fusion protein expression system. Recombinant Omp28 (rOmp28) was expressed in Escherichia coli and was then purified. Immunogenicity of rOmp28 was confirmed by Western blot analysis with Brucella-positive mouse serum. Furthermore, humoral- or cell-mediated immune responses measured by the production of IgG1 or IgG2a in rOmp28-immunized mice and the ability of rOmp28 immunization to protect against B. abortus infection were evaluated in a mouse model. In the immunogenicity analysis, the mean titers of IgG1 and IgG2a produced by rOmp28-immunized mice were 20-fold higher than those of PBS-treated mice throughout the entire experimental period. Furthermore, spleen proliferation and bacterial burden in the spleen of rOmp28-immunized mice were approximately 1.5-fold lower than those of PBS-treated mice when challenged with virulent B. abortus. These findings suggest that rOmp28 from B. abortus is a good candidate for manufacturing an effective subunit vaccine against B. abortus infection in animals.  相似文献

An enzyme-linked immunosorbent assay has been developed for the detection of antibodies against Brucella ovis using serum from control rams (Con-S), naturally infected rams (Inf-S), rams inoculated intravenously with B. ovis (IV-S) and rams vaccinated intramuscularly (IM-S). The serum was titrated by serial double dilutions from 1/25 to 1/25,600 against whole bacteria, B. ovis lipopolysaccharide and a detergent-extracted component of the outer membrane complex of B. ovis as antigens immobilised on microtitre plates. Sheep antibodies bound to antigen were assayed with rabbit anti-sheep gammaglobulin and alkaline phosphatase conjugated protein A. A high level of antibody activity against intact B. ovis cells was detected in Inf-S and IM-S. When lipopolysaccharide was the immobilised antigen, only IM-S yielded significant antibody activity. The component from detergent extracts of the outer membrane complex of B. ovis reacted best with serum (up to 1/6,400) from field-infected rams, while serum from vaccinated and intravenously inoculated rams registered significant titres up to a serum dilution of 1/800 and 1/200 respectively. These results indicate that ELISA is a very sensitive test but its value as a serodiagnostic procedure is dependent upon the choice of antigen used in the assay.  相似文献

Enzyme-linked immunosorbent assay (ELISA) profiling was used to identify the immunoreactive membrane antigens of Brucella ovis. Immunoreactive membrane antigens obtained after detergent extraction of the bacterial membrane complex (inner and outer membranes) were resolved into five peaks (A, B, B1, C and D) by gel permeation chromatography. Aliquots from each of the chromatographed fractions were coupled to 96-well microtitre plates and immunoreactive fractions identified with sera from two rams. Serum from ram 1 which had been vaccinated with a single injection of formalin-killed B ovis emulsified in incomplete Freund's adjuvant identified A and B as the major immunoreactive peaks. Serum from ram 2, which had been successfully infected with B ovis, reacted mainly against peaks A, B1, C and D. This observation facilitated the use of A, B, B1, C and D peak antigens as test reagents to examine the serological response of 12 other rams exposed to B ovis by vaccination or intraconjunctival or intravenous inoculation. Sera from rams which developed productive infections reacted strongly against peaks A, B1, C and D while vaccinated rams had preferential antibody activity against peaks A and B.  相似文献

To aid in the development of novel efficacious vaccines against brucellosis, Omp25 was examined as a potential candidate. To determine the role of Omp25 in virulence, mutants were created with Brucella abortus (BA25), Brucella melitensis (BM25), and Brucella ovis (BO25) which contain disruptions in the omp25 gene (Deltaomp25 mutants). Western immunoblot analysis and PCR verified that the Omp25 protein was not expressed and that the omp25 gene was disrupted in each strain. BALB/c mice infected with B. abortus BA25 or B. melitensis BM25 showed a significant decrease in mean CFU/spleen at 18 and 4 weeks post-infection, respectively, when compared to the virulent parental strain (P<0.05, n=5). Mice infected with B. ovis BO25 had significantly lower mean CFU/spleen counts from 1 to 8 weeks post-infection, at which point the mutant was cleared from the spleens (P<0.01, n=5). Murine vaccination with either BM25 or the current caprine vaccine B. melitensis strain Rev. 1 resulted in more than a 2log(10) reduction in bacterial load following challenge with virulent B. melitensis (P<0.01, n=5). Vaccination of mice with the B. ovis mutant resulted in clearance of the challenge strain and provided 2.5log(10) greater protection against virulent B. ovis than vaccine strain Rev. 1. Based on these data, the B. melitensis and B. ovis Deltaomp25 mutants are interesting vaccine candidates that are currently under study in our laboratory for their safety and efficacy in small ruminants.  相似文献

Cellular as well as humoral immune responses were detected in six rams experimentally infected with Brucella ovis. Specific antibodies were detectable by enzyme-linked immunosorbent assay by day 11 after infection in all the rams. The levels of IgM antibodies and total antibodies in the serum rose until 33 and 41 days after infection respectively, then levelled off. Antigen-induced blastogenic responses by lymphocytes developed as early as five days after infection in all rams but had decreased to low levels by day 63 in most. Blastogenesis induced by phytohaemagglutinin and concanavalin A varied among infected rams and did not differ significantly (P greater than 0.05) from control rams. All rams had developed delayed-type skin hypersensitivity by day 63 after infection. One ram which did not become infected as a result of exposure had low levels of B ovis serum antibodies and a detectable antigen-induced lymphocyte blastogenic response before infection, suggesting the involvement of cell-mediated immunity in protection against B ovis.  相似文献

The potential diagnostic usefulness of antibodies to the ribosome recycling factor of Brucella melitensis (CP24) was assessed in sheep by an indirect ELISA with purified recombinant CP24. Sera from uninfected animals from the UK (n=44) and from local flocks (n=42), from sheep naturally infected with B. melitensis (n=12) or B. ovis (n=12), and from lambs (n=7) or pregnant ewes (n=6) vaccinated with B. melitensis Rev-1, were assayed. High specific optical densities (OD(with antigen) - OD(without antigen)) were obtained with both the groups of normal sera, which resulted in high cut-off values (1.414 and 1.267, respectively). Only two infected sheep yielded specific OD higher than these cut-off values. No significant difference was found between mean specific OD from B. melitensis- or B. ovis-infected sheep (0.574 and 0.472, respectively), those from vaccinated animals (0.396 and 0.400 for pregnant ewes and lambs, respectively), and those from Brucella-free animals. An inhibition ELISA with soluble CP24 confirmed the specificity of the antibodies detected in normal sera by the indirect ELISA; these antibodies belonged to the IgG class as revealed by the use of a specific conjugate. Sera from infected sheep were all positive for antibodies against lipopolysaccharides and lumazine synthase from Brucella. These results show that anti-CP24 antibodies have no diagnostic role in ovine brucellosis.  相似文献

A Brucella ovis surface protein antigen (P-II), obtained by gel filtration with Sepharose 4B of a hot saline extract was characterized. The analysis of P-II over gradient sodium dodecylsulfate electrophoresis yielded an 18.5 and a 20 kDa band. In a radioimmunoprecipitation assay using P-II labeled with 125I, the antigen reacted specifically only with sera from rams experimentally infected with a naturally occurring rough strain of B. ovis and did not react with sera from rams experimentally infected with other smooth Brucella strains (B. abortus and B. melitensis).  相似文献

An enzyme-linked immunosorbent assay (ELISA), using an autoclave-extracted soluble antigen, for the detection of serum antibodies to Brucella ovis in sheep was developed. The test seemed to be both sensitive and specific, on the basis of the control groups studied. The antigen showed no deterioration in prepared plates stored at -70 C for up to a year. The ELISA was used in conjunction with palpation of rams for epididymal lesions as a means to detect and control B ovis infection in a naturally infected flock. All rams were evaluated by the ELISA. At the time that the first blood sample was obtained, all positive and suspicious reactors were removed. With subsequent blood sample collections, only the positive reactors were removed. Brucella ovis was not isolated from any rams during the following year, and none of the mature breeding rams developed epididymal lesions.  相似文献

An ELISA for the detection of antibodies to Histophilus ovis was used to evaluate the association of epididymal lesions in rams with serologic response to His ovis. Comparison of ELISA results for His ovis in groups of rams with epididymal lesions with ELISA results of clinically normal rams (control group) revealed a significant difference (P less than 0.01) between the control group and those rams from which His ovis was isolated. A significant difference (P less than 0.01) was noticed between the control group and rams with lesions from which an organism other than His ovis or Brucella ovis was isolated. Additionally, a significant difference (P less than 0.01) was noticed in ELISA results between the control group and affected rams from which no organism was recovered and in which the epididymal lesion was not limited to the head of the epididymis. A difference was not detected in the His ovis ELISA results between control rams and rams with lesions associated with a B ovis infection or rams from which no organism was recovered and in which the epididymal lesion was limited to the head of the epididymis. The serologic findings in our study suggest that His ovis is more important in the development of epididymitis in rams than culture results alone would indicate.  相似文献

AIMS: To investigate the persistence of infection and serum antibody titres after infection of red deer (Cervus elaphus) stags with Brucella ovis, and compare these with those of rams. To assess the effects of recent and chronic infection on semen characteristics of stags. METHODS: Fourteen stags and eight rams were artificially infected with B. ovis by intravenous inoculation. Semen and blood samples were collected at approximately monthly intervals for 649 days. Semen samples were subjected to bacterial culture, and sera were tested for B. ovis antibodies using a complement fixation test (CFT) and an enzyme-linked immunosorbent assay (ELISA). At the end of the study, animals were slaughtered and reproductive organs subjected to bacterial culture. During the first and second breeding seasons, three and five semen samples, respectively, were evaluated from each stag for sperm motility and morphology. RESULTS: Twelve of 14 (86%) stags and 6/8 (75%) rams developed a patent B. ovis infection and shed the organism in semen. All six infected rams continued to shed B. ovis in semen throughout the 649-day study period, and at slaughter B. ovis was isolated from the reproductive tract and urinary bladder. In contrast, 10/12 (83%) infected stags stopped shedding B. ovis in semen 103-342 days after inoculation, and the organism could not be isolated from their reproductive tracts at slaughter. The remaining two infected stags shed B. ovis in semen throughout the study period and the organism was isolated from their reproductive tracts at slaughter. All inoculated animals initially developed serum antibody titres detectable using the B. ovis CFT and ELISA. For infected stags, the diagnostic sensitivity of these tests was 100% for the first 166 days, but decreased to 50-90% after this. The diagnostic sensitivity for the infected rams was 100% throughout the study period. Infection in stags resulted in variable effects on semen characteristics. Eight of 12 (67%) infected stags had a mean sperm motility of < 50%, and < 60% mean normal sperm in the first year of infection. Seven of these stags had resolved the infection by the following breeding season, and there was a significant improvement in sperm motility and morphology. CONCLUSIONS: Stags are as susceptible as rams to experimental B. ovis infection. However, the majority of infected stags resolved the infection within a year, whereas rams remained infected for at least 649 days (22 months). Serology, using CFT and ELISA, was effective at detecting infection during the first 166 days in both species, but after this time was less effective at detecting infection in stags than in rams. Infection with B. ovis had variable but generally deleterious effects on the semen characteristics of stags, which resolved following resolution of the infection. Differences in the characteristics of the disease in stags compared with rams mean that different control methods are warranted for the two species. CLINICAL RELEVANCE: Most stags infected with B. ovis are likely to resolve the infection within a year, and semen characteristics return to levels acceptable for breeding. Serology is useful for detection of infection in the early stages of the disease, but once disease has been present in the herd for some time false-negative reactions are likely to occur in individual stags.  相似文献

To investigate the value of the BP26 protein in the serological diagnosis of ovine brucellosis caused by Brucella ovis, recombinant BP26 protein was produced in Echerichia coli and purified for use in an indirect enzyme-linked immunosorbent assay (I-ELISA). The majority of the recombinant protein was recovered from the supernatant of sonicated recombinant E. coli cells in a soluble form. This facilitated the purification of the recombinant BP26 protein which was achieved by using ion-exchange chromatography. After one step of purification, the purity of the recombinant BP26 protein was analyzed by using SDS-PAGE, Coomassie blue staining, and Western blot with a monoclonal antibody (MAb) directed against the BP26 protein. The degree of purity appeared satisfactory so that it could be directly used in I-ELISA. Although the recombinant BP26-ELISA appeared less useful than I-ELISA using the B. ovis hot saline (HS) extract as antigen, the high number of sera from B. ovis infected rams found positive (90%) in the recombinant BP26-I-ELISA indicated that the BP26 protein may be an additional suitable antigen for serological diagnosis of B. ovis infection in rams.  相似文献

An enzyme-linked immunosorbent assay (ELISA) for antibody to Brucella ovis was compared with a standard complement fixation test. Sera of 176 rams from uninfected flocks gave 175 negative and one suspect ELISA reaction (diagnostic specificity 99.4%) whereas the complement fixation test yielded 167 negative, seven suspect and two anticomplementary reactions (diagnostic specificity of 96.0%). Diagnostic sensitivity was evaluated on sera of 79 rams from which B. ovis had been isolated. The ELISA showed 75 positive and four suspect reactions, while complement fixation test revealed 64 positive, 13 suspect and two negative results. Considering both positive and suspect reactions, the diagnostic sensitivity was 100% for ELISA and 97.5% for complement fixation test. The ELISA method was considered more specific, more sensitive and technically more advantageous than complement fixation test as a serodiagnostic test for B. ovis infection in rams.  相似文献

Brucella spp. are Gram-negative, facultative, intracellular coccobacilli that are pathogenic to a variety of mammals, including ruminants and humans. The conventional serological test for diagnosing brucellosis in cattle in Korea is the standard tube agglutination test. However, agglutination tests sometimes give false-positive results due to cross-reactions with other pathogens. The outer membrane proteins of Brucella species have been extensively studied for their immunogenicity and serodiagnostic applications. However, an application of B. abortus OMPs for serodiagnosis has not been successfully established. In this study, cloning and expression of B. abortus Omp28, a group 3 antigen, were accomplished by PCR amplification cloning into a pMAL expression system, and purification of a recombinant Omp28 (rOmp28). The immunogenicity of rOmp28 was confirmed by Western blot analysis with Brucella-positive bovine serum. To determine whether rOmp2 has a potential benefit for use in the serodiagnosis of bovine brucellosis, rOmp28-based ELISA and latex bead agglutination test were performed. B. abortus positive (n=122) or negative (n=88) from TAT were positive (118/122, 96.7%) or negative (84/88, 95.4%) in ELISA and were positive (94/122, 77%) or negative (71/88, 81.7%) in that the latex bead agglutination test, respectively. The sensitivity, specificity and accuracy were 96.7, 95.4, 96.2% in ELISA and 77, 80.6, 78.5% in latex bead agglutination test, respectively. These findings suggest that the rOmp28 of B. abortus might be a good candidate for developing serological diagnostic tools for bovine brucellosis.  相似文献

The protective anti-B. abortus monoclonal antibody ISS/32 (Ab1) was used as an immunogen to induce anti-idiotypic antibodies (Ab2) in rabbits. The purpose was to produce and characterize anti-idiotypic antibodies that share conformational similarity with the corresponding bacterial epitope recognized by Ab1. The rabbit anti-IdAb so induced was isolated and affinity-purified. Its specificity for the paratope of Ab1 was determined by evaluating its ability to compete withB. abortus for binding to Ab1 in a competitive ELISA assay. The anti-idiotypic ISS/32 antibodies were able to compete withB. abortus for binding to Ab1 in a dose-dependent manner. 研制抗羊种布鲁菌脂多糖抗原的单克隆抗体,并应用其建立检测布病的双夹心ELISA方法。本研究采用热酚水法提纯羊种布鲁菌(16M菌株)的脂多糖抗原,并经SDS-PAGE鉴定。用脂多糖和灭活的羊种布鲁菌16M作为免疫抗原,交替免疫6~8周龄BALB/c雌鼠,第1次免疫用羊种布鲁菌标准菌16M全菌加等量弗氏完全佐剂;第2次免疫用脂多糖加等量弗氏不完全佐剂。将脂多糖作为包被抗原建立间接ELISA方法,筛选针对抗羊种布鲁菌(16M菌株)脂多糖的单克隆抗体杂交瘤细胞株。筛选出3株能稳定分泌抗羊种布鲁菌脂多糖单克隆抗体的杂交瘤细胞株,分别命名为5H3、6B8和3H7,细胞培养上清的ELISA效价在1∶1 000~1∶5 000,小鼠腹水单克隆抗体ELISA效价在1∶10 000~1∶160 000;抗体亚类鉴定表明:5H3、6B8属于IgM亚类,3H7属于IgG3亚类;特异性试验结果显示:3株杂交瘤细胞分泌的抗体不与大肠杆菌O157裂解抗原、鸡白痢沙门氏菌裂解抗原、鸭源鸡杆菌脂多糖抗原以及福氏志贺菌裂解抗原反应,仅与灭活的羊种布鲁菌(16M)发生反应。布鲁菌虎红平板凝集试验和试管凝集试验检测结果显示,获得的单抗可与标准检测抗原形成明显的颗粒凝集物和伞状凝集物。利用所建立的单克隆抗体细胞株,建立了一种检测布鲁菌的双夹心间接ELISA方法,并进行了特异性和敏感性检测。对模拟样品和临床样品进行检测,准确性均很高。  相似文献

为制备苯乙醇胺A(phenylethanolamine A,PA)单克隆抗体,建立一种针对苯乙醇胺A的快速、简便、灵敏度高的检测方法,本试验采用重氮化法将制备的苯乙醇胺A衍生物分别与牛血清白蛋白(BSA)和卵清白蛋白(OVA)进行偶联作为免疫原和包被原。利用弗氏佐剂充分乳化免疫原(PA-BSA),按常规免疫程序免疫6~8周龄的雌性BALB/c小鼠,选择血清抗体效价较高的小鼠,取其脾细胞与SP2/0骨髓瘤细胞以PEG法进行细胞融合。结果显示,经3次细胞亚克隆筛选后,最终筛选出一株可稳定分泌抗苯乙醇胺A单克隆抗体的杂交瘤细胞株,命名为D6H8,利用此细胞以小鼠体内诱生法制备抗体。经鉴定,D6H8腹水抗体亚型为IgG1,轻链为κ链;纯化后的抗体与沙丁胺醇、盐酸克伦特罗、盐酸异丙肾上腺素和盐酸去氧肾上腺素均无明显的交叉反应(CR<0.18%),表明该抗体特异性良好。利用此抗体建立针对苯乙醇胺A药物残留检测的间接竞争ELISA方法,结果表明,腹水抗体效价为1∶12 800,猪肉中苯乙醇胺A添加浓度在5~1 000 ng/mL时线性关系良好,标准工作曲线为y=0.3861x-0.1845 (R^2=0.990),其IC50为58.88 ng/mL,LOD为3.83 ng/mL,回收率在85.96%~104.32%之间。综上所述,本试验成功建立了检测苯乙醇胺A药物残留的间接竞争ELISA方法,该方法灵敏度高、稳定性较好。  相似文献

The complement fixation test (CFT), the enzyme labelled immunosorbent assay (ELISA) and the gel diffusion precipitin test (GD) were compared, for the diagnosis of Brucella ovis infection in rams. The sensitivities of the tests in 109 rams which were shedding B. ovis in their semen were: CFT 96.3%; ELISA 97.2%; GD 91.7%. The specificities of the tests in 141 rams from non-infected flocks were: CFI 99.3%; ELISA 98.6%; GD 100%. Predictive values of the three tests were measured in 285 rams from infected flocks. Thirty-eight percent of these rams were shedding B. ovis in their semen. Predictive values of positive tests were: CFT 75.5%; ELISA 66.7%; GD 72.5%. Predictive values of negative tests were: CFI 97.1%; ELISA 97.6%; GD 93.8%.  相似文献

A commercial flock of Suffolk and Suffolk-cross breeding rams was monitored for 5 years in an effort to control epididymitis caused by Brucella ovis. Scrotal palpation, semen evaluations, and vaccination against B ovis were used the first 3 years. Serologic evaluation (complement fixation and ELISA) was added the fourth year, and bacteriologic culturing was added to the program the fifth year. Semen culturing in the fifth year revealed 9 (37.5%) of 24 rams were actively excreting B ovis; 6 of those 9 rams were seronegative. Neither semen quality nor the presence of WBC in the semen were dependable criteria to detect these seronegative carriers. In spite of the high percentage of B ovis excretors, few clinical signs of epididymitis were detected in the flock during the last 3 years of the study. It was hypothesized that vaccination protected rams against the clinical disease but not the carrier state. The importance of culturing semen for assessment of a control program was emphasized.  相似文献

为获得禽脑脊髓炎病毒(Avian Encephalomyelitis virus,AEV)VP1蛋白的单克隆抗体,通过原核表达AEV VP1蛋白,纯化后作为免疫原免疫BALB/c小鼠,并按常规方法制备杂交瘤细胞。经ELISA方法筛选阳性杂交瘤细胞,经过3次亚克隆获得2株杂交瘤细胞株,命名为4#、19#,并进行了抗体亚类的鉴定、Western-blot和IFA检测。结果显示:制备的株单克隆抗体亚型分别为IgG2b、IgG2a,Western-blot和IFA试验结果表明单克隆抗体均能与AEV发生特异性反应而与其他禽病常见病毒均无交叉反应。运用建立的IFA对单抗进行了初步运用,在外源病毒检测方面与经典方法符合率高。本研究成功制备了AEV单克隆抗体,为进一步建立AEV检测方法和深入研究AEV的生物学特性奠定了基础。  相似文献

Brucellosis causes serious economic losses to goat farmers by way of reproductive losses in the form of abortions and stillbirths. Nucleic acid vaccines provide an exciting approach for antigen presentation to the immune system. In this study, we evaluated the ability of DNA vaccine encoding the omp31 protein of Brucella melitensis 16M to induce cellular and humoral immune responses in mice. We constructed eukaryotic expression vectors called pTargeTomp31, encoding outer membrane protein (omp31) of B. melitensis 16M. pTargeTomp31 was injected intramuscularly three times, at 3-week intervals in groups of mice 6 weeks of age. pTargeTomp31 induced good antibody response in ELISA . pTargeTomp31 elicited a T-cell-proliferative response and also induced a strong gamma interferon production upon restimulation with either the omp31 antigen or B. melitensis 16M extract. We also demonstrate that animals immunized with this plasmid elicited a strong and long-lived memory immune response. Furthermore, pTargeTomp31 elicited a typical T-helper 1-dominated immune response in mice, as determined by immunoglobulin G isotype analysis. This vaccine also provided the moderate degree of protection to the mice. This study for the first time focuses on DNA immunization of a gene from B. melitensis. These results may lead to the development of a DNA-based vaccine for the control of brucellosis in goats.  相似文献
