利用常规肌肉营养分析方法测定了四倍体泥鳅、二倍体泥鳅和大鳞副泥鳅的肌肉营养成分.结果表明:四倍体泥鳅、二倍体泥鳅和大鳞副泥鳅肌肉中蛋白质含量(鲜重)分别为18.68%、18.69%和18.20%;脂肪含量分别为0.796%、0.106%和1.203%;水分含量分别为79.81%、79.52%和78.40%;灰分含量分别为0.951%、1.188%和1.162%.3种泥鳅肌肉中都含有常见的17种氨基酸,其中包括9种人体必需氨基酸.氨基酸总量(占鲜重)分别为17.57%、18.11%和17.47%;必需氨基酸总量分别为7.84%、7.98%和7.76%,必需氨基酸的构成比例符合FAO/WHO的标准.鲜味氨基酸含量分别为6.98%、7.33%和6.95%.经比较分析,四倍体泥鳅和二倍体泥鳅肌肉中蛋白质含量、17种氨基酸含量、氨基酸总量和必需氨基酸总量均很接近,但都明显高于大鳞副泥鳅.  相似文献

为建立大鳞副泥鳅(Paramisgurnus dabryanus)的科学投喂策略,在实验室内开展了大鳞副泥鳅的养殖模拟实验,通过72 h的观察,探讨了不同体质量和不同性别大鳞副泥鳅的昼夜摄食节律及日摄食量。结果表明,大鳞副泥鳅属于典型的夜间摄食类型,一天中有2个极明显的摄食高峰,集中在04:00—05:00和20:00—21:00。大鳞副泥鳅的昼夜摄食节律与个体大小和性别无显著相关性。小规格雌、雄群体均具有最大的日摄食量,且雌性群体的日摄食量随着个体大小的增加显著降低。15℃条件下,大鳞副泥鳅群体的日摄食量占群体总体质量的2.2%~4.8%,个体的日摄食量约占个体体质量的3.0%。建议大鳞副泥鳅的投饲频率为一天2次,以体质量的3%~5%进行饲料投喂,最佳投饵驯化时间集中在6:00—7:00和18:00—19:00。  相似文献

为探明三倍体和二倍体泥鳅的组织细胞学差异,运用血液涂片及组织切片方法,对三倍体、二倍体泥鳅的红细胞及其细胞核的长轴和短轴,血液中血细胞的组成及仔鱼眼部构造等进行了比较.结果表明,三倍体泥鳅红细胞及其核的长轴均比二倍体的要大,其中前者红细胞核的长轴约为后者的1.12倍,红细胞体积、核体积分别约为后者的1.62和1.44倍.三倍体泥鳅红细胞的相对数量明显少于二倍体泥鳅,而嗜中性细胞、单核细胞及淋巴细胞则多于后者.此外,三倍体泥鳅眼部色素层细胞数量少于二倍体.  相似文献

舟山群岛大鳞副泥鳅与台湾泥鳅的形态差异性分析   总被引:1,自引:0,他引:1  
台湾泥鳅(Misgurnus anguillicaudatus)是台湾地区选育的泥鳅新品种,虽具有明显优于大鳞副泥鳅(Paramisgurnus dabryanus)的养殖性状,但在实际生产过程中往往会将这2种泥鳅混淆,为了科学识别形态特征相似的台湾泥鳅和舟山群岛大鳞副泥鳅,采用单因素方差分析、主成分分析和判别分析等多元分析方法定量比较舟山群岛大鳞副泥鳅与台湾泥鳅人工养殖群体的形态特征差异。结果显示:(1)2种泥鳅在本研究所涉24项形态评价指标中存在显著差异的达8项,两者在形态特征上具较好的区分度;(2)经主成分分析,提取到5个特征值均大于1的主成分,累计贡献率达75.463%,其中第1主成分为以营养生理状况、体型匀称程度为主导的体质强壮因子,第2主成分为以头部大小和胸鳍长短为主导的觅食运动功能因子,后3个主成分是与眼睛发达程度为主导的视野功能因子;(3)采用逐步判别法,以判别贡献率较大的8项形态评价指标为自变量,建立分类函数方程组。大鳞副泥鳅:func_1=-681.190+4.992x1+498.305x2+1778.188x3;台湾泥鳅:func2=-860.382+5.373x1+650.034x2+1894.839x3;所建判别函数可清晰区分舟山群岛大鳞副泥鳅与台湾泥鳅人工养殖群体,且判别准确率为100%。研究表明,虽然台湾泥鳅和大鳞副泥鳅形态特征相似,但能利用形态指标建立判别函数对两者进行科学、快速的判别。  相似文献

二倍体泥鳅与大鳞副泥鳅杂交能够顺利获得杂交后代,其正反交核型都为(2n=49),但杂交后代精巢表现出明显的发育迟缓现象,杂交泥鳅自交与回交实验表明,其雄性不可育。为了解二倍体泥鳅和大鳞副泥鳅杂交F1雄性不育的原因,实验通过计算机辅助精子活力分析系统(CASA)、光学显微镜、扫描电镜、透射电镜、流式细胞技术分别对二倍体泥鳅(DD)、大鳞副泥鳅(PP)、其正反杂交F1(DP)和(PD) 4种泥鳅精子的形态结构与活力进行了分析。结果显示,激活30 s时,DD和PP的精子运动率分别为76.50%±0.70%和75.17%±8.60%,极显著高于DP (3.65%±1.75%)和PD (2.68%±0.63%),且杂交泥鳅的精子的平均运动速度(VAP)、平均曲线速度(VCL)、平均直线运动速度(VSL)等运动参数也极显著低于DD和PP,说明杂交泥鳅精子的活力极其低下。应用流式细胞技术对4种泥鳅精巢内细胞进行倍性鉴定,发现DD和PP精巢内大多为1N精子,而DP和PD精巢内除了正常单倍体精子外,还有大量的2N和4N以及少量的8N细胞。通过扫描电镜和透射电镜...  相似文献

为探明三倍体泥鳅的营养组成,对实验室培育2个月的三倍体与二倍体泥鳅幼鱼的氨基酸和脂肪酸含量进行了测定,发现三倍体泥鳅幼鱼的氨基酸总量、必需氨基酸总量、饱和脂肪酸和不饱和脂肪酸总量分别为:62.87%、26.23%、34%和61.5%,二倍体则分别为68.41%、29.16%、22.8%和69.7%;三倍体泥鳅幼鱼的4种主要鲜味氨基酸量及总量均低于二倍体,n-3系列高度不饱和脂肪酸C22:6(DHA)与C20:5(EPA)的总量则高于二倍体.与二倍体泥鳅相比较,三倍体泥鳅幼鱼在营养价值上并不具优势.研究结果为三倍体鱼类的营养评价提供了依据.  相似文献

大鳞副泥鳅群体线粒体DNA D-loop序列遗传变异分析   总被引:2,自引:0,他引:2  
为探明大鳞副泥鳅野生资源状况,利用线粒体DNA D-loop序列,对天津5个大鳞副泥鳅自然群体的遗传多样性、遗传结构及种群历史动态进行分析。5个群体72个个体的D-loop基因序列全长912~914bp,其中多态性位点30个,共检测到22个单倍型,平均单倍型多样性和核苷酸多样性分别为0.813和0.0015,呈现高单倍型多样性和低核苷酸多样性的模式。5个群体间的遗传分化指数为-0.0205~0.0795,总的遗传分化指数为0.0712,属中等程度的遗传分化。单倍型网络进化图和单倍型系统发育树均未显示出明显的地理分布格局,中性检验和核苷酸不配对分布分析均表明,大鳞副泥鳅经历过种群扩张,扩张事件发生在第四纪更新世晚期。研究结果表明,应加强5个湿地大鳞副泥鳅群体的保护,适时开展大鳞副泥鳅良种选育工作,减少对大鳞副泥鳅野生资源的破坏。  相似文献

To determine the genotype with better traits for aquaculture practices of the loach, factorial crosses were made among diploid (D), tetraploid (T) dojo loach, Misgurnus anguillicaudatus, and large‐scale loach, Paramisgurnus dabryanus (P), producing DD, DT, DP, TD, TT, TP, PD, PT, and PP genotypes (female listed first). The growth performance and survival of different genotypes in culture were evaluated through a 48‐wk rearing trail. The average fertilization rate in genotypes with P. dabryanus as female parental was significantly higher than those of M. anguillicaudatus as female parental. The average fertilization level of sperm from P. dabryanus was about the same as diploid M. anguillicaudatus but significantly higher than tetraploid M. anguillicaudatus. The highest survival rate was found in DD group (52.77%) and PP group (50.31%) (P > 0.05). The intergenetic crosses between P. dabryanus and M. anguillicaudatus, especially for PD (19.05%), DP (17.89%), and PT (15.44%) groups, exhibited poor survival rates. Under the similar rearing conditions, PP group showed significant growth advantage. Growth rates of the interspecific hybrid progenies were found to be intermediate in comparison with the parental species. 为对不同鳊鲂鱼类进行群体鉴定和遗传多样性分析,从60对微卫星标记中筛选出18对多态性高的引物,构建了6个鳊鲂鱼类群体的微卫星DNA指纹图谱。结果显示,东江三角鲂、钱塘江三角鲂、厚颌鲂、广东鲂、团头鲂和长春鳊6群体的平均等位基因数(N a)分别为5.17、6.11、3.50、6.56、5.22、5.22,平均期望杂合度(H e)分别为0.634 2、0.720 4、0.546 2、0.681 2、0.675 2、0.559 7,平均多态信息含量(PIC)分别为0.575 6、0.666 9、0.472 0、0.630 6、0.606 4、0.517 0,表明钱塘江三角鲂的遗传多样性最高,厚颌鲂的遗传多样性最低;聚类分析表明,钱塘江三角鲂和团头鲂首先聚为一支,遗传距离较近,为0.560 6;厚颌鲂与长春鳊的遗传距离最远,为1.759 2。引物Mam03和EST37产生的特异条带可将鲂属和鳊属鱼类区分,鉴定出鳊属鱼类长春鳊;引物TTF3、EST37、TTF2/TTF10、EST66依次组合可区分出鲂属东江三角鲂、厚颌鲂和广东鲂这3个群体。研究结果为我国鳊鲂鱼类种质资源保存、种群鉴定和良种选育奠定了基础。  相似文献

Exotic animals can cause genetic contamination of indigenous species if they escape and reproduce in wild populations. The loach Misgurnus anguillicaudatus and species related to it have been imported into Japan for commercial use. We collected live loach specimens from the central wholesale market in Tokyo. For the 451 specimens collected, ploidy status was examined by DNA content flow cytometry, and polyploid loaches with triploid, tetraploid, and other higher ploidy ranges were detected. Hypertriploid and hypertetraploid individuals were easily detected by flow cytometry using standard eudiploid, eutriploid, and eutetraploid controls, and the reproductive capacities of these hyperpolyploid males were then examined. Sperm of hypertriploid males did not exhibit active progressive motility, and major populations of spermatozoa or spermatozoon-like cells were detected in the triploid and hexaploid ranges. Motile haploid spermatozoa were very rare in sperm from hypertriploid males. Therefore, hypertriploid males were sterile, whereas hypertetraploid males produced fertile hyperdiploid spermatozoa with active progressive motility after contact with ambient water. Viable progeny occurred in crosses between normal wild-type diploid females and hypertetraploid males, but androgenotes induced by the fertilization of UV-irradiated eggs with the sperm of hypertetraploid males were inviable hyperdiploids. Cytogenetic analyses of those androgenotes indicated that hypertetraploid males should produce hyperdiploid spermatozoa with 2n?=?54 (i.e., four supernumerary microchromosomes).  相似文献

Tetraploid fish, which are considered as key resources of diploid gametes for further breeding and ploidy manipulation, can be artificially induced by inhibition of the mitotic cell division with hydrostatic pressure or temperature treatments. Although many attempts have been made to induce artificial tetraploid strains, successful establishment of viable and fertile tetraploid strains are rare. In pond loach, Misgurnus anguillicaudatus, natural tetraploid individuals are distributed in wild populations and diploid gametes from the tetraploid fish have been used for the induction of polyploid individuals, but artificially induced tetraploid strains have not been established yet. In the present study, we optimised starting timing of the heat-shock treatment (41 °C for 2 min) to inhibit a mitotic cell division in fertilised eggs of the normal diploid pond loach between 21 and 51 min after insemination at 20 °C. After the treatment, we observed external appearance of hatching larvae and flow cytometrically determined ploidy status of the resultant larvae. Although tetraploid and diploid/tetraploid mosaic larvae were obtained, the optimum timings for induction of tetraploidy varied amongst crosses. Various kinds of ploidy such as haploidy, diploidy, triploidy, pentaploidy, hexaploidy, aneuploidy and mosaic were detected in non-optimum heat-shock timings for tetraploidisation. Survivors, a tetraploid and a diploid/tetraploid mosaic male, matured at the age of 1-year-old, but they produced functional haploid spermatozoa.  相似文献

3种泥鳅微卫星标记和D-Loop部分序列遗传变异分析   总被引:2,自引:2,他引:0  
为进一步了解中国当前主要养殖鳅类种质资源现状,实验采用7个微卫星标记和线粒体D-Loop部分序列,对泥鳅(Misgurnus anguillicaudatus,MA)、大鳞副泥鳅(Paramisgurnus dabryanus,PD)和台湾大泥鳅(未见种属分类,TW)3种泥鳅进行群体遗传变异分析.结果显示,6个微卫星位点在3种泥鳅中均能获得有效扩增,1个微卫星标记(Mac239)只在MA中获得特异性扩增,而在PD和TW中未能获得有效扩增条带.在3种泥鳅共90尾个体的D-Loop 部分序列中发现32个单倍型,仅在PD和TW间存在1个共享单倍型.实验中共检测到65个变异位点,其中MA与PD和TW间存在29个特异性位点,而PD和TW间未检测到特异性位点.瓶颈效应和中性检验显示,TW近期可能发生有效群体数量的减少.基于微卫星标记和D-Loop部分序列的遗传变异分析显示,TW和PD间的Nei's遗传距离和K2P遗传距离最近(0.297和0.006),明显小于两者与MA间的遗传距离(1.011~1.899和0.095~0.099);分子方差分析(AMOVA)显示,3种泥鳅的遗传分化极显著(P<0.01).群体间遗传结构和单倍型网络分析显示,3种泥鳅的遗传结构相对独立,仅在TW和PD间存在一定程度的遗传结构混杂.研究表明,泥鳅和大鳞副泥鳅在遗传结构上存在明显差异,可采用分子标记进行有效鉴别;台湾大泥鳅可能是大鳞副泥鳅的生态种群或遗传改良群体,而非有效物种.  相似文献

归纳了白鲟、长江鲟、中华鲟近40年自然资源量变化情况,统计了长江鲟和中华鲟人工增殖放流数据,评估了增殖放流成效。因物种特性和分布的差异,放流无效的原因迥异。过度捕捞使人工放流长江鲟在放流后6个月之内难逃被“误捕”,而中华鲟放流数量不足及长江和近海过度捕捞导致中华鲟人工增殖放流的贡献甚微,其结果是长江鲟和中华鲟增殖放流均无法达到自然繁殖群体的补充水平。本文探究了近40年来白鲟、中华鲟和长江鲟在保护、管理和决策上存在的误区和不足,提出在生态大保护的背景下,(1)应编制和实施《长江鲟拯救行动计划》优先项目;(2)应以《中华鲟拯救行动计划》为指引,在就地保护、迁地保护和遗传多样性保护方面提出更具有可操作性的优先计划;(3)应设立中华鲟和长江鲟物种拯救行动计划专项,对现有涉栖息地或保护区生态补偿项目进行优化整合。唯有以恢复长江鲟和中华鲟自然繁殖为核心,才有望延续和恢复其自然种群,实现人与自然的和谐共处,推动长江流域社会经济绿色可持续发展。  相似文献

以二倍体、三倍体、四倍体长牡蛎(Crassostrea gigas)闭壳肌和精子为研究对象, 采用石蜡组织切片、透射电镜和扫描电镜方法, 比较了 3 种倍性长牡蛎闭壳肌和精子的组织学特性及超微结构的差异。研究结果显示, 二、三、 四倍体闭壳肌组织的肌纤维都包含粗肌丝、细肌丝、肌膜、肌束膜、肌内膜、线粒体、囊泡、小管、细胞核、肌浆网等结构, 但三者的形态及超微结构不同, 其中三倍体闭壳肌线粒体直径、肌纤维直径、粗肌丝密度显著大于二倍体和四倍体, 肌纤维密度显著小于二倍体和四倍体。不同倍性精子的扫描电镜和透射电镜结果显示, 二、三、四倍体长牡蛎精子在组成结构上相似, 分别由头部、中段和尾部构成, 但不同倍性精子中各结构大小和线粒体数目不同。在长牡蛎精子头部长度、宽度, 顶体高度、宽度, 线粒体长度的测量分析中, 四倍体精子的头部、顶体以及线粒体显著大于二、三倍体。三倍体精子的头部和顶体显著大于二倍体, 但三倍体精子的线粒体大小与二倍体相比无显著差异。此外, 二、三倍体精子均含 4 个线粒体, 但在本研究中发现了二倍体精子含有 5 个(占比 1%)线粒体的特例。四倍体长牡蛎精子的线粒体数目不同, 含有 4 个(占比 46%)、5 个(占比 52%)、6 个(占比 2%)这 3 种情况。 该研究阐明了不同倍性长牡蛎闭壳肌及精子的组织学特性和超微结构, 为深入研究长牡蛎肌纤维生物学特性和精子繁育生物学提供了基础资料。  相似文献

This study determined biometric and weight parameters and relevant indices of diploid and triploid tench. Altogether, 137 siblings of tench were studied. The effect of ploidy level appeared in significantly better growth of triploids (P<0.001) as to biometric [total length (TL), standard length (SL), body height (BH), body width (BW)] and weight [fish weight (FW), carcass weight (CW)] parameters of T3 of both sexes and of T3+ females. The effect of ploidy level also appeared as significantly higher dressing percentage (DP; P<0.001) of triploid T3 females compared with other groups, significantly higher gonad weight (GW) and gonadosomatic index (GSI; P<0.001) of diploid T3 females, as well as GSI and hepatosomatic index of diploid T3+ females. The effect of sex appeared in significantly higher (P<0.001) biometric (TL, SL, BH, BW) and weight (FW, CW) parameters of T3 females of both ploidy levels, as well as of triploid T3+ females. The effect of sex also appeared as significantly higher DP (P<0.001) of males in diploid T3 fish, as well as of males of both ploidy levels in T3+ fish and significantly higher GW and GSI (P<0.001) of females in diploid T3 fish, as well as of females of both ploidy levels in T3+ fish. This study shows evidence for faster somatic growth and bigger final weight of triploid populations of tench compared with diploids in both age categories T3 and T3+.  相似文献

为了研究副溶血弧菌对斑节对虾非特异性免疫酶活性的影响,分别对斑节对虾注射生理盐水和副溶血弧菌,于不同时间点测定肝胰腺和鳃中的总抗氧化能力(T-AOC)、酸性磷酸酶(ACP)、碱性磷酸酶(ALP)和溶菌酶(LSZ)的活性变化。结果显示,与对照组相比,感染副溶血弧菌后,肝胰腺和鳃中T-AOC活性分别于12和6 h达到最大值(P0.05),随后逐渐降低;肝胰腺中ACP活性于3 h显著升高,随后逐渐降低,并于24 h达到最小值(P0.05),而鳃中ACP活性于12 h达到最大值,随后逐渐降低,并于48 h显著低于对照组(P0.05);肝胰腺和鳃中ALP活性整体均呈现出先升高后下降的趋势,但未出现显著低于对照组的现象;肝胰腺和鳃中LSZ活性均于3 h显著升高至最大值(P0.05),随后逐渐恢复至初始水平。研究表明,副溶血弧菌感染对斑节对虾非特异性免疫酶活有显著影响,对其机体免疫防御系统有明显的破坏作用。  相似文献

Experimental crossings were made between tetraploid loach (Misgurnus anguillicaudatus, T for short) and diploid large‐scale loach (Paramisgurnus dabryanus, P for short), resulting in T×T, T×P and P×P crosses (female before male). Meanwhile, a part of T×P cross was cold shocked to inhibit second meiosis (T×P‐0 for short). Fertility rate, hatching rate and survival rate of T×T, T×P, P×P and T×P‐0 were measured. Flow cytometry and chromosome count were taken to identify ploidy statuses of the four crosses. Results showed that T×T, T×P and P×P were, respectively, tetraploidy (4n = 100), triploidy (3n = 74) and diplody (2n = 48), while 89.19 per cent of T×P‐0 of 12 months old were pentaploidy (5n = 124, T×P‐0‐5n for short) and the others were triploidy. Morphological data of the four crosses of 12 months old showed that morphological characteristics of T×P and T×P‐0‐5n loach were between those of their parents, while T×P‐0‐5n loach presented a smallest head length/body length. Total lengths of T×P and T×P‐0‐5n were significantly larger than those of T×T and P×P, showing hybrid vigour.
