用不同浓度的5-氮胞苷溶液配合不同时间处理了发生在转录水平的转基因cry1Ab沉默水稻。结果表明,5-氮胞苷处理能使8%~30%转基因水稻植株的转基因cry1Ab恢复表达活性,复活转基因表达的Cry1Ab蛋白含量最高可达0.147%,45mg/L5-氮胞苷处理1d可使转基因cry1Ab的复活率及Cry1Ab蛋白含量达最高。同时,5-氮胞苷处理使转基因水稻植株出现矮化,分蘖数、单株粒重下降,穗长减小及开花期延迟等发育异常现象。5-氮胞苷作为一种弱诱变剂在克服转基因沉默及创造具优良农艺性状的转基因植物中具有一定的应用价值。  相似文献

转Bt基因水稻Cry1Ab杀虫蛋白在水稻土中的降解   总被引:8,自引:0,他引:8  
室内研究了转 Bt基因水稻克螟稻 2号和华池B6 叶片中的Cry1Ab杀虫蛋白在稻田水稻土中的降解动态。分别将克螟稻 2号的粉碎叶片和华池B6 的整张叶片埋入 3种土壤 (即老黄筋泥田、青紫泥田和黄松田 )中 ,此后每隔 6~ 10d测定土壤或叶片中的Cry1Ab含量。克螟稻 2号粉碎叶片中的Cry1Ab均以处理后的前 36d,尤其是前 6d的降解较快 ,其中在青紫泥田中最快 ,黄松田中最慢。在处理后 6~ 36d内 ,不同土壤中的Cry1Ab残留量有显著差异 ,黄松田中的明显最高 ,老黄筋泥田中的次之 ,青紫泥田中的为最低 ;此后土壤间的残留量差异逐渐缩小 ,至处理后 78d差异已不明显。土壤淹水可显著加快华池B6 叶片中Cry1Ab的降解 ,且淹水后其降解动态在不同土壤之间十分相似 ;淹水对Cry1Ab降解的促进作用仅发生在前 12d之内 ,此后多数时间内残留量在淹水与非淹水处理间无显著差异。建立了各处理中Cry1Ab降解的动力学指数方程 ,相应地得出了其降解的半衰期。还讨论了土壤有机质和微生物等因子对土壤中Cry1Ab降解的影响。  相似文献

抗虫抗除草剂转基因水稻B1C893的获得与鉴定   总被引:2,自引:1,他引:1  
利用农杆菌介导法将Cry1Ca#和Bar基因转化水稻恢复系R893,获得6株再生植株。经过分子鉴定,发现Bar基因和Cry1Ca#基因成功整合到水稻基因组并稳定表达。测定其中单拷贝且可育的转基因株系苗期不同器官中Cry1Ca蛋白的表达量,发现根系中最低而叶片中最高,其中最高的第6号株系叶片中含量达到35.07μg/g。获得的转基因株系对除草剂草铵膦和稻纵卷叶螟均具有良好的抗性。  相似文献

分子改造Cry蛋白基因的转基因玉米创制及其抗虫功能评价   总被引:1,自引:0,他引:1  
利用结构域交换和密码子优化方法对苏云金芽孢杆菌(Bt)杀虫晶体蛋白Cry1Ab进行合理化设计改造,获得新型Bt蛋白编码基因cry FLIa。利用农杆菌介导法将该基因转入玉米Hi II中,对转基因后代开展分子鉴定和抗性评价等相关工作。结果表明,cryFLIa基因已整合进玉米基因组中,连续3代自交材料检测显示,转基因玉米目标基因正常表达并稳定遗传;蛋白在植株叶片中发育各时期的表达各异,在灌浆期蛋白表达量较高(471.883 ng/g)。室内生测和田间人工接虫试验表明,转cryFLIa基因玉米具有抗亚洲玉米螟的能力。  相似文献

转Cry30Fa1基因抗褐飞虱水稻的获得及鉴定   总被引:1,自引:1,他引:0  
将编码高效杀褐飞虱蛋白的苏云金芽胞杆菌基因Cry30Fa1密码子改造后,通过农杆菌介导法转入蜀恢818(R818),并最终获得46个转基因植株。通过定量PCR及Western Blot鉴定了Cry30Fa1在转录水平和蛋白水平的表达,并通过分子检测固定稳定表达的抗性基因,并结合传统育种系谱法选择具有优良农艺性状株系。对选育的株系在室内和大田环境下进行了抗虫性鉴定,选育的R818-Cry30Fal株系抗性明显优于亲本材料并达到抗的水平;在单株抗虫试验中,观察到了转基因株系对褐飞虱具有致死作用。说明转入Cry30Fa1基因使水稻产生了对褐飞虱抗性。培育出了具有抗褐飞虱蛋白的新型恢复系R818-Cry30Fal,为三系杂交育种提供了新的抗性材料并丰富了抗褐飞虱水稻种质资源。  相似文献

采用田问小区试验,研究了不同氮、磷、钾配比及氮用量对水稻植株氮的吸收、转运,籽粒全氮、蛋白氮含量及蛋白质产量的影响,结果表明:推荐配比及氮低量配比处理水稻植株N向籽粒转运的能力比对照要强,籽粒全氮、蛋白氮含量以及蛋白质产量明显高于对照。  相似文献

转铁蛋白基因增强水稻对氧化胁迫与稻瘟病菌的抗性   总被引:13,自引:2,他引:11  
对转豌豆铁蛋白(pea ferritin,Fer)基因水稻T1代的53个株系进行PCR检测,52个株系能扩增出阳性PCR产物。通过测定光合作用过程中最大光化学通量(Fv/Fm值)分析了由百草枯处理引起的T1代水稻叶片的氧化损害。与未转基因水稻相比,转Fer基因水稻的叶片对氧化胁迫的耐受能力有不同程度的增强。百草枯处理后转基因植株叶片叶绿素含量与水处理叶片相比没有明显下降,而未转基因植株叶片叶绿素含量降低至水处理叶片的20%左右。选取9株对氧化胁迫耐受能力较强的水稻进行了Northern blot分析和子代的稻瘟病抗性测定,其中5株转基因植株Fer mRNA 积累增强。病原菌接种后T2代转基因植株的病斑数量明显少于非转基因植株。表明转Fer基因水稻对氧化胁迫和病原菌有较好的抗性。  相似文献

转cry1Ab基因水稻对拟水狼蛛捕食作用间接影响的评价   总被引:12,自引:0,他引:12  
室内评价了拟水狼蛛(Pirata subpiraticus)对取食转[i]cry1Ab[/i]基因水稻的稻纵卷叶螟([i]Cnaphalocrocis medinalis[/i])幼虫和褐飞虱([i]Nilaparvata lugens[/i])若虫的捕食量及功能反应。结果表明,拟水狼蛛对这2种猎物的捕食功能反应均符合HollingⅡ型圆盘方程。该蜘蛛对取食转[i]cry1Ab[/i]基因水稻的稻纵卷叶螟幼虫的捕食量和处理时间低或短于取食对照的,但差异未达显著水平,而瞬时攻击率显著低于对照。该蜘蛛对以转[i]cry1Ab[/i]基因水稻为食的褐飞虱若虫的捕食量、瞬时攻击率和处理时间与取食对照的相比有一定的变化,但差异均不显著。综合考虑,认为转[i]cry1Ab[/i]基因水稻对拟水狼蛛捕食作用无明显的负作用。  相似文献

BRC-ZLL4是一株从鱼尾葵(Cargota mitis Lour)上分离到的,对柑橘凤蝶幼虫具有毒性的苏云金芽胞杆菌(Bacillus thuringiensis)菌株。本试验采用PCR-RFLP技术鉴定该菌株中含有cry1Ib基因。设计一对特异引物,分别引入SalⅠ和BamHⅠ酶切位点,以BRC-ZLL4的总DNA为模板,扩增cry1Ib全长基因。生物信息学分析表明,该基因全长2160bp,编码一个由719个氨基酸组成的、相对分子质量为81.39ku、等电点为6.425的蛋白。该基因已在GenBank上登录(登录号为EU233027),并被Btδ-内毒素国际命名委员会正式命名为cry1Ib2。信号肽预测显示,Cry1Ib2蛋白没有信号肽,为胞内蛋白。二级结构预测显示,在Cry1Ib2蛋白二级结构中,25%为α-螺旋,28%为β-片层,18%为β-转角,29%为无规则线圈。此外还对Cry1Ib蛋白的功能区和三级结构进行了预测和分析。  相似文献

研究Cry2Ab蛋白对棉花黄萎病菌、枯萎病菌和红腐病菌生长速率及对黄萎病菌分生孢子产量、分生孢子大小和粗毒素分泌量的影响。结果发现,当Cry2Ab蛋白质量浓度为1.0 μg·mL-1时,显著降低黄萎病菌分生孢子的大小;当其质量浓度大于5.0 μg·mL-1时,显著抑制棉花黄萎病菌、枯萎病菌和红腐病菌在PDA培养基上生长及黄萎病菌分生孢子的大小,促进黄萎病菌产孢和粗毒素分泌。  相似文献

Three transgenic rice varieties namely Khazar, Neda and Nemat, all containing a cry1Ab gene, were evaluated through PCR analysis and field examinations for their resistance at natural infestation of insect pests during 2007. The results showed that all transgenic varieties produced 1.2 kb PCR product derived from application of cry1Ab gene. In field conditions, transgenic varieties exhibited high levels of resistance against natural infestation of stem borer and the damaged plants based on dead heart or white heat for them were less than 1%. Moreover, in stem-cut bioassay 100% of released larvae died within four days after infestation. These results demonstrate that expression of cry1Ab gene in the genome of transgenic varieties provided season-long protection from the natural infestation of lepidopteran insects.  相似文献

 对转Bt基因水稻与常规品种的杂交后代进行了GUS活性的组织化学分析和农艺性状考察。结果表明,报告基因gus与抗虫基因 cry1Ab是连锁遗传和协同表达的,利用GUS组织化学染色法可快速筛选抗虫杂种植株;无论是籼粳交,还是粳粳交后代,抗虫和感虫的两类植株间在株高、穗长、单株分蘖数、播始历期和千粒重等主要农艺性状上无显著差异,这无疑为利用Bt水稻培育优良的抗虫品种提供了科学依据。  相似文献

对筛选到的转[WTBX][STBX]Xa21[WTBZ][STBZ]基因培矮64S后代纯合株系抗性与育性的研究表明,在第5代杂种中,该外源基因仍能稳定遗传表达;以其作父本、培矮64S为母本所配制的F1植株, Xa21基因PCR检测均呈阳性,且绝大多数表现抗白叶枯病,说明该外源基因能够通过常规杂交方式转移利用。这些杂交组合F2群体中抗病与感病株出现3∶1分离,表明转Xa21基因在杂交后代的传递属单基因显性遗传。长日高温下,(培矮64S/转Xa21基因培矮64S)F1雄性完全可育,F2群体可育与不育株出现27∶1分离。  相似文献

Abstract

Mechanism(s) of gene transformation and integration in rice (Oryza sauva L.) is/are not currently well understood. This research was conducted to determine whether a transgene is inserted into the rice genome specifically or randomly. Seven homozygous transgenic Taipei (T) 309 and Nipponbare plants with the bar transgene from different rice transformation events were crossed. The segregation of F2 and F3 populations from a total of 21 crosses was studied in a greenhouse and field to determine if the genes were allelic or non-allelic. Five genomic locations appeared to be involved among the seven transgenic plants. An additional 20 homozygous transgenic T309 plants, with the bar transgene from different transformation events, were crossed reciprocally with the previous seven plants. One hundred and fifteen crosses made during 1999 and 2000 were analyzed for allelism. In some combinations, the genes were allelic, but most of them were non-allelic, with two or more pairs of genes being expressed. Twenty loci among the 27 transgenic plants were involved and some plants had several inserted genes expressed. Genes in nine out of 27 transformed plants were allelic. We concluded that the functional foreign (bar) gene was restrictively/preferentially inserted into the rice genome in some cases and was not completely randomly inserted and expressed in the rice genome. If the mechanism(s) for preferential insertion were identified, rice researchers could possibly control insertion sites of transgenes to optimize gene expression.  相似文献

Transgenic of Indian potato cultivar Kufri Badshah expressing synthetic, modified cry1Ab gene were developed against potato tuber moth (Phthorimaea opercullela Z.) a destructive pest. The cry1Ab gene was in spatial and temporal expression under the control of tuber-specific GBSSi promoter. The transformation vector pBinCG1 was developed harbouring transgene expression cassette comprising cry1Ab gene under the control of potato GBSSi promoter, castor bean catalase intron (5′UTR) and OCS termination signals. Selected kanamycin-resistant putative transformed potato lines were evaluated by positive PCR amplification of 713 bp, 1206 bp and 700 bp with npt II, GBSSi promoter and cry1Ab gene specific primers, respectively. Gene integration in transformed potato plants was elucidated through Southern hybridization and in planta transgene expression analysis. Transgene expression was highest in stolons, followed by tubers matured leaves and young leaves. Insect bioassay of stored transgenic tubers resulted in significant retardation and mortality in neonate tuber moth larvae. RT-PCR and northern hybridization confirmed stable expression in stored tubers. These transgenic lines; thus can represent an effective resistance management strategy which can significantly reduce the selection pressure on target and non-target insect pests to a threshold level.  相似文献

 就转cry1Ac+CpTI双基因抗虫水稻不同生育期对二化螟Chilo suppressalis和大螟Sesamia inferens的室内致死特性及田间螟虫的构成进行了研究。室内测定结果表明,不同生育期转基因水稻对二化螟、大螟都表现明显的致死效应,但水稻生长后期的致死效果降低。转基因水稻对大螟的致死效应显著弱于对二化螟的,其中,二化螟除在齐穗期和成熟期有少量幼虫(0.5%~6.4%)存活到第4天外,其余均在第4天死亡;大螟在两种转基因水稻上的存活率高于二化螟,且少量个体(<1.6%)还能化蛹、羽化,但化蛹率和羽化率均明显低于在非转基因对照上的。早、晚两季水稻的田间调查结果表明,转基因水稻上两种螟虫虫口数量均显著低于相应的非转基因对照品种,转基因水稻上二化螟虫口减退率>99%;大螟虫口减退率相对较低,早、晚稻上有所不同,其中早稻 >93%,晚稻仅44%~64%。转基因水稻上残存螟虫中,大螟所占比例明显上升,推测转基因水稻对两种螟虫致死效应差异可能是其主要原因。  相似文献
