In this study, the changes in vitamin C, l-ascorbic acid (AA) and l-dehydroascorbic acid (DHA) levels in broccoli flower buds were examined during pre-storage and storage periods, simulating refrigerated transport with wholesale distribution and retail, respectively. Broccoli heads were pre-stored for 4 or 7 days at 0 °C or 4 °C in the dark and then stored for 3 days at 10 °C or 18 °C. During storage the broccoli heads were exposed for 12 h per day to three different levels of visible light (13, 19 or 25 μmol m−2 s−1) or a combination of visible light (19 μmol m−2 s−1) and UV-B irradiation (20 kJ m−2 d−1), or they were stored in the dark. The vitamin C content in broccoli flower buds during storage was significantly affected by pre-storage period and temperature. Higher vitamin C levels in flower buds after storage were observed for broccoli heads pre-stored for 4 days or at 0 °C as compared to those pre-stored for 7 days or at 4 °C. Storage temperature also affected vitamin C in broccoli flower buds, with higher levels observed for broccoli stored at 10 °C than at 18 °C. Hence, vitamin C in broccoli flower buds was demonstrated to decrease together with increasing pre-storage period, pre-storage temperature and storage temperature. AA in broccoli flower buds was influenced mainly by storage temperature and to a minor extent by pre-storage temperature. The DHA level and DHA/AA ratio were stable in flower buds of broccoli pre-stored for 7 days, whereas increasing tendencies for both DHA level and ratio were observed after pre-storage for 4 days. These results indicate a shift in the ascorbate metabolism in broccoli flower buds during storage at low temperatures, with its higher rate observed for broccoli pre-stored for shorter time. 为研究肉桂酰辅酶A还原酶在青花菜生长发育中的作用,采用RT-PCR技术克隆青花菜Bo CCR基因全长c DNA序列,并利用q RT-PCR检测其在不同器官中的表达模式。结果表明:Bo CCR基因开放阅读框为987 bp,推导编码328个氨基酸。预测蛋白分子量为36.86 ku,PI值为6.04。该蛋白亲水性氨基酸多于疏水性氨基酸,为亲水性蛋白。高级结构预测表明青花菜Bo CCR蛋白具有完整的二级结构,三级结构的α-螺旋主要位于外部,β-折叠位于内部。分子进化分析显示CCR蛋白在植物进化过程中,各属形成单独的分枝,可以区分其亲缘关系。荧光定量技术检测到青花菜的Bo CCR基因在花蕾发育早期表达最高,推测该基因可能与青花菜花粉发育相关。 相似文献

为探究宁夏南部山区自然降水条件下露地青花菜的最佳补灌灌溉方案,采用大田田间对比试验,设置5个补灌水平,在宁夏原州区冷凉蔬菜基地开展补灌水平对土壤水分分布、青花菜产量、品质、水分利用效率的试验研究。结果表明,青花菜全生育期自然降雨主要集中在7月份,且占全生育期降雨量的78.49%,此月份无需补灌即可满足生育后期对水分的需求;不同处理青花菜生育期内土层深度20 cm处土壤含水率总体表现为W5(27mm·667m-2)>W4(22.5mm·667 m-2)>W3(18 mm·667 m-2)>W2(13.5 mm·667 m-2)>W1(9 mm·667 m-2),补灌后各处理土壤含水率均达到田间持水量的85%以上;补灌灌溉水平对青花菜花球产量性状、营养品质均有显著影响,W3处理经济总产量最高,达1524.10 kg·667 m-2,显著高于其他处理6.28%~33.24%;可溶性糖和可溶性蛋白含量也最高,W3可溶性糖含量显著高于W1、W4和W5处理24.46%、12.20%、21.41%,W3可溶性蛋白含量显著高于W135.78%,且可溶性固形物和维生素C质量分数也较高,分别为6.26 g·100 g-1和9.57 mg·100 g-1。基于主成分分析的青花菜膜下滴灌最优补灌方案为补灌灌溉定额为69 mm,苗期-莲座期补灌灌水2次,灌水量为33mm,莲座期-球前期补灌灌水2次,灌水量36mm,结球前期-采收期补水主要依靠自然降雨。该结果可为本区露地冷凉青花菜夏季栽培产量的积累与水资源的合理应用提供理论指导。 相似文献

Fifty-nine Brassica oleracea cultivars, belonging to five botanical varieties, were evaluated for microsatellite (SSR) polymorphisms using 11 database
sequence derived primer pairs. The cultivars represented 12 broccoli (Brassica oleracea var. italica), ten Brussels sprouts (B. o. var. gemmifera), 21 cabbage (B. o. var. capitata, including the groups white and red cabbage), six savoy cabbage (B. o. var. sabauda), and ten cauliflower (B. o. var. botrytis) cultivars from 13 seed suppliers. The 11 primer pairs amplified in total 47 fragments, and differentiated 51 of the cultivars,
whereas the remaining eight cultivars were differentiated from the rest in four inseparable pairs. All SSR markers, except
one, produced a polymorphic information content (PIC value) of 0.5 or above. The average diversity for all markers within
the tested material was 0.64. There was no major difference in the diversity within botanical varieties and groups. The cluster
analysis and the resulting dendrogram showed that the cultivars tended to group within these taxonomic units. The present
study substantiates the use of microsatellite markers as a powerful tool for cultivar differentiation and identification in

[目的]建立一种简单、快速、经济的鉴定蔬菜中硫代葡萄糖苷的方法。[方法]用70%甲醇从西兰花籽中提取硫代葡萄糖苷,以甲酸作为离子调节剂,采用梯度洗脱的方法对提取物进行分离,并采用液相色谱-质谱-质谱联用技术对提取物成分进行鉴定。[结果]从西兰花籽中成功鉴定出8种硫代葡萄糖苷,其保留时间分别为5.9、7.7、8.8、9.5、13.6、16.0、19.3、28.6min,分子量分别为422、468、438、436、372、406、420、402;该鉴定方法所测样品不需经过脱硫等预处理,大大节省了试验时间,且样品需要量较少,试验效率较高。[结论]该研究建立了一种鉴定蔬菜中硫代葡萄糖苷的新方法。 相似文献

本研究对50条甘蓝SSR引物在其近缘种青花菜中的通用性进行了分析,结果表明,38对引物在青花菜上可有效扩增,扩增产物分子量在100~1500bp,有效扩增比率为76%,其中18%具有较好的多态性,揭示了两种作物基因组间存在一定的相似性。同时利用获得的多态性较好的9对引物对青花菜进行基因型鉴定和遗传多样性分析,20份青花菜基因型中共检测到51个基因位点,平均每个引物组合可扩增出5.67个条带,多态性为66.7%。多引物组合可鉴别出所有青花菜基因型。聚类分析结果表明同一来源的基因型间具有相近的遗传基础,且聚类与熟性具有一定的相关性。本研究为今后青花菜种质资源的收集、利用及分子标记辅助育种提供了新的SSR标记和参考。 相似文献
