The aim of this study was to evaluate the prognostic value of concurrent measurement of
serum phosphorylated neurofilament heavy subunit (pNF-H) concentration and intramedullary
T2W hyperintensity in paraplegic to paraplegic dogs. Our hypothesis was that concurrent
measurement of these would provide a more accurate prediction of functional outcome in
dogs with thoracolumbar intervertebral disc herniation (IVDH). A prospective case-control
clinical study was designed using 94 dogs with acute onset of thoracolumbar IVDH. The
association of serum pNF-H concentration, T2W hyperintensity on sagittal MRI (T2H/L2),
deep pain perception and surgical outcome were evaluated with logistic regression analysis
after three months for all 94 surgically treated dogs. Sensitivity to predict
non-ambulatory outcome was compared among pNF-H and T2H/L2 and combination of both.
Logistic regression analysis indicated that serum pNF-H concentration and T2H/L2 were
significantly correlated with surgical outcome (P<0.05); however, deep
pain perception was not (P=0.41). The results of logistic regression
analysis indicated that the odds ratios of unsuccessful long-term outcome were 2.6 for
serum pNF-H concentration, 1.9 for T2H/L2 and 2.3 for deep pain sensation. The sensitivity
and specificity to predict non-ambulatory outcome for using serum parameter pNF-H>2.6
ng/ml, using T2H/L2 value of>0.84 and using both
serum pNF-H and T2H/L2, were 95% and 75.7%, 65% and 86.5%, and 90.0% and 97.5%,
respectively. Therefore, combined measurements of serum pNF-H and T2H/L2 might be useful
for predicting long-term outcome in dogs with thoracolumbar IVDH. 相似文献

Embryos obtained via superovulation are necessary for mammalian artificial reproduction, and viability is a key determinant of success. Nonfreezing storage at 4 C is possible, but currently used storage solutions can maintain embryo viability for only 24–48 h. Here we found that 10 mg/ml antifreeze protein (AFP) dissolved in culture medium 199 with 20% (v/v) fetal bovine serum and 25 mM HEPES could keep bovine embryos alive for 10 days at 4 C. We used a recombinant AFP isolated from the notched-fin eelpout (Zoarces elongatus Kner). Photomicroscopy indicated that the AFP–embryo interaction was enhanced at 37 C. Embryos pre-warmed with the AFP solution at 37 C for 60 min maintained high viability, whereas those that were not pre-warmed could live no longer than 7 days. This study compared the reproductive and growth performances of pigs before and after depopulation and
restocking after a foot-and-mouth disease outbreak in Japan. Data for the time period before and after
depopulation and restocking were obtained from three farrow-to-finish farms. As a result of depopulation and
restocking, hygiene levels were improved, and common infectious diseases, such as porcine reproductive and
respiratory syndrome and Aujeszky’s disease, remained undetected on the farms. Compared with before
depopulation, reproductive and growth performances improved after depopulation; the number of total pigs born
was higher, the postweaning mortality rate was lower, and the age at slaughter was lower
(P<0.05). In summary, depopulation and restocking improved the reproductive and growth

The effects of lipid-rich bovine serum albumin (LR-BSA) on the development of porcine blastocysts produced
in vitro were examined. Addition of 0.5 to 5 mg/ml LR-BSA to porcine blastocyst medium
(PBM) from Day 5 (Day 0 = in vitro fertilization) significantly increased the hatching rates
of blastocysts on Day 7 and the total cell numbers in Day-7 blastocysts. When Day-5 blastocysts were cultured
with PBM alone, PBM containing LR-BSA, recombinant human serum albumin or fatty acid-free BSA, addition of
LR-BSA significantly enhanced hatching rates and the cell number in blastocysts that survived compared with
other treatments. The diameter, ATP content and numbers of both inner cell mass and total cells in Day-6 and
Day-7 blastocysts cultured with PBM containing LR-BSA were significantly higher than in blastocysts cultured
with PBM alone, whereas LR-BSA had no effect on mitochondrial membrane potential. The mRNA levels of enzymes
involved in fatty acid metabolism and β-oxidation (ACSL1, ACSL3,
CPT1, CPT2 and KAT) in Day-7 blastocysts were
significantly upregulated by the addition of LR-BSA. The results indicated that LR-BSA enhanced hatching
ability and quality of porcine blastocysts produced in vitro, as determined by ATP content,
blastocyst diameter and expression levels of the specific genes, suggesting that the stimulatory effects of
LR-BSA arise from lipids bound to albumin. 相似文献

In situ hybridization was used to analyze the distribution pattern of Tomato chlorotic dwarf viroid (TCDVd) in floral organs of tomato plants. Following TCDVd invasion of floral organs, it became localized only in sepals
at an early developmental stage, then reached other floral organs at the flower opening stage, with the exception of part
of the placenta and ovules. When distribution of TCDVd was compared with that of Potato spindle tuber viroid (PSTVd), TCDVd was not detected in the outer integument around the embryo sac even though PSTVd was able to invade there,
suggesting that such specific distribution might reflect the frequent occurrence of viroid disease on crops caused by PSTVd-seed

Biwa salmon Oncorhynchus sp. is endemic to Lake Biwa, Japan, where it is an important commercial and recreational fisheries species. However, no information is currently available on its population structure and migration ecology. Therefore, here we evaluated whether otolith Sr/Ca, Ba/Ca and 87Sr/86Sr ratios can be used as natural signatures in Biwa salmon and then used these to determine the natal origins of lake-migration-phase individuals and spawning adults, and the homing ability of spawning adults in the Lake Biwa water system. Quadratic discriminant function analysis demonstrated that the lake-migration school comprised individuals with multiple origins, including rivers to the east, west and north of Lake Biwa, and that the homing rate of spawning adults was low (18 out of 80 individuals), with ca. 78% of fish straying into non-natal rivers. However, this straying behaviour was not spatially random, with fish tending to migrate upstream in rivers neighbouring their natal rivers. The high rate of straying in spawning adults is considered important for establishing and maintaining this species, which is highly adapted to life in the Lake Biwa water system where environmental disturbances often occur. 相似文献

We characterised the host range and physical properties of Tomato chlorotic dwarf viroid. Among the 46 plant species inoculated with the viroid, two in the family Compositae and 23 in the family Solanaceae were
found to be systemic hosts. The viroids in the crude sap from diseased tomato plants were thermally inactivated by heating
to 100°C for at least 40 min. These viroids also lost their infectivity when diluted in phosphate buffer to at least 10−6, or after 3 days of incubation at room temperature. However, the infectivity of the viroids in dried crude sap from the plants

The major digestive enzymes in Pacific bluefin tuna Thunnus orientalis larvae were characterized, and the physiological characteristics of the enzymes during early ontogeny were clarified using biochemical and molecular approaches. The maximum activity of trypsin (Try), chymotrypsin (Ct) and amylase (Amy) was observed at pH 6–11, 8–11 and 6–9, respectively. Maximum activity of Try, Ct and Amy occurred at 50 °C, that of lipase (Lip) was at 60 °C and that of pepsin (Pep) was at 40–50 °C. These pH and thermal profiles were similar to those for other fish species but differed from those previously reported for adult bluefin tuna. Enzyme activity for all enzymes assayed was found to decrease at high temperatures (Try, Ct, Amy and Pep: 50 °C; Lip: 40 °C), which is similar to findings for other fish species with one marked exception—increased Try activity was observed at 40 °C. Lip activity appeared to be dependent on bile salts under our assay conditions, resulting in a significant increase in activity in the presence of bile salts. Ontogenetic changes in pancreatic digestive enzymes showed similar gene expression patterns to those of other fish species, whereas marked temporal increases in enzyme activities were observed at 10–12 days post hatching (dph), coinciding with previously reported timing of the development of the pyloric caeca in bluefin tuna larvae. However, complete development of digestive function was indicated by the high pep gene expression from 19 dph, which contradicts the profile of Pep activity and previously reported development timing of the gastric gland.
