In the past five decades, constant research has been directed towards yield improvement in pigeonpea resulting in the deployment of several commercially acceptable cultivars in India. Though, the genesis of hybrid technology, the biggest breakthrough, enigma of stagnant productivity still remains unsolved. To sort this productivity disparity, genomic research along with conventional breeding was successfully initiated at ICRISAT. It endowed ample genomic resource providing insight in the pigeonpea genome combating production constraints in a precise and speedy manner. The availability of the draft genome sequence with a large‐scale marker resource, oriented the research towards trait mapping for flowering time, determinacy, fertility restoration, yield attributing traits and photo‐insensitivity. Defined core and mini‐core collection, still eased the pigeonpea breeding being accessible for existing genetic diversity and developing stress resistance. Modern genomic tools like next‐generation sequencing, genome‐wide selection helping in the appraisal of selection efficiency is leading towards next‐generation breeding, an awaited milestone in pigeonpea genetic enhancement. This paper emphasizes the ongoing genetic improvement in pigeonpea with an amalgam of conventional breeding as well as genomic research.  相似文献

The present investigation aimed to evaluate the effect of dietary chitosan supplementation on growth performance, body composition, immune response and histopathology of Nile tilapia, and also the in vitro antibacterial activity of chitosan against Streptococcus agalactiae (S. agalactiae). About 180 fish (average body weight 39.3 ± 0.3 g) were randomly divided into three groups according to chitosan supplementation: control group (basal diet without chitosan), Ch3 group (3 g chitosan/kg diet) and Ch5 group (5 g chitosan/kg diet). Growth performance parameters and body proximate composition were measured before infection but biochemical parameters and lysozyme and antibacterial activities before and after experimental infection. Results of the present investigation showed dietary chitosan (5 g chitosan/kg diet) significantly (p < .05) improved growth performance parameters, body composition (dry matter, crude protein, ether extract, ash, and carbohydrate) and serum biochemistry (total protein, albumin, globulin, with no effect on AST, ALT, urea and creatinine) before infection in Ch5 group than the control. After infection, liver enzymes (serum AST and ALT) were maintained lower in fish fed Ch3 or Ch5 than the control. Serum lysozyme and bactericidal activities significantly increased (p < .05) in chitosan groups before and after the challenge. The mortality rate was markedly reduced in the Ch3 group and prohibited in the Ch5 group after the experimental infection. In conclusion, feeding 3 or 5 g chitosan/kg diet increased the growth rate and improved FCR of Nile tilapia. In addition, it reduced mortality by its antibacterial and immunostimulant effects.  相似文献

Summary Powdery mildew development was assessed on squash (Cucurbita pepo) plants of a susceptible cultivar, a resistant accession, their F1, and their F2 in an early summer planting in the field, covered or not covered with a shading net. Three reaction types were observed: susceptible, powdery mildew on stems and on both, the upper and lower leaf surfaces, as in the susceptible parent; resistant, no powdery mildew on leaves or stems, as in the resistant parent; and partially resistant, powdery mildew on upper leaf surfaces only, as in the F1. Disease presence on the stem was associated with susceptibility. Shading hastened the appearance of powdery mildew and increased the severity of infection on partially resistant and susceptible plants, facilitating identification of resistant individuals in the F2 population.Contribution No. 1613-E from the Agricultural Research Organization, Bet Dagan, Israel  相似文献

Significant tomato matrix effects on the volatility of certain fresh tomato odorants were found. The concentrations of odorants such as (E,E)-2,4-decadienal, beta-damascenone, and beta-ionone, in crushed fresh tomato fruit obtained by solid-phase microextraction (SPME), resulting from a tomato matrix calibration curve were 5.5-, 2-, and 12-fold higher, respectively, than those calculated by calibration based on buffer solutions. Static headspace analyses indicated that, in most cases, the tomato matrix significantly retains the odorants relative to the buffer solution. Thus, the concentration of odorants in the headspace of tomato is lower than expected compared to a simple matrix such as buffer. CaCl(2), although needed in crushed fruit tissue to block enzymatic activity, was found to interact specifically with 2-isobutylthiazole, reducing its content in the headspace by at least 6-fold. If a matrix effect is found, analysis of the odorant molecule contents in the headspace rather than in the food is recommended in order to better evaluate their access to the olfactory receptors.  相似文献

The antiproliferative effects of 100% methanol crude extract and of Toyopearl and silica gel fractions from the seed coats of black Jamapa beans (Phaseolus vulgaris L.) were evaluated using HeLa, human adenocarcinoma cells, and HaCaT, human premalignant keratinocytes. The 100% methanol crude extract [172.2 microM equiv of (+)-catechin] increased adhesion of HeLa cells; however, 3- and 5-fold higher concentrations decreased the number of cells attached as a function of the treatment time. The highest concentration tested diminished the cell adhesion until 40% (after 24 h) to almost 80% (after 72 h). The IC50 values showed that the 100% methanol crude extract was the most effective inhibitor of HeLa cell proliferation, even when it was dissolved in dimethylsulfoxide (DMSO) [34.5 microM equiv of (+)-catechin] or in medium [97.7 microM equiv of (+)-catechin]. The Toyopearl 5 (TP5) fraction and silica gel 2 (SG2) fraction inhibited 60% of the HeLa cell proliferation. The IC50 was 154 microM equiv of (+)-catechin of the 100% methanol crude extract on HaCaT cells. Toyopearl fractions TP4 and TP6 significantly inhibited HaCaT cell proliferation, but the silica gel fractions did not have a significant effect. The 100% methanol crude extract (35 microg of dry material/mL) decreased the number of HeLa cells in the G0/G1 phase from 68.9% (for control cells) to 51.4% (for treated cells) and increased apoptosis (2.9 and 21.2% for control and treated cells, respectively). The results indicated that black Jamapa beans could be a source of polyphenolic compounds, which have an inhibitory effect toward HeLa cancer cells but are less aggressive on HaCaT premalignant cells.  相似文献

The prevalence of Bartonella spp. in wild rodents was studied in 19 geographical locations in Israel. One hundred and twelve rodents belonging to five species (Mus musculus, Rattus rattus, Microtus socialis, Acomys cahirinus and Apodemus sylvaticus) were included in the survey. In addition, 156 ectoparasites were collected from the rodents. Spleen sample from each rodent and the ectoparasites were examined for the presence of Bartonella DNA using high resolution melt (HRM) real-time PCR. The method was designed for the simultaneous detection and differentiation of eight Bartonella spp. according to the nucleotide variation in each of two gene fragments (rpoB and gltA) and the 16S–23S intergenic spacer (ITS) locus, using the same PCR protocol which allowed the simultaneous amplification of the three different loci. Bartonella DNA was detected in spleen samples of 19 out of 79 (24%) black rats (R. rattus) and in 1 of 4 (25%) Cairo spiny mice (A. cahirinus). In addition, 15 of 34 (44%) flea pools harbored Bartonella DNA. Only rat flea (Xenopsyla cheopis) pools collected from black rats (R. rattus) were positive for Bartonella DNA. The Bartonella sp. detected in spleen samples from black rats (R. rattus) was closely related to both B. tribocorum and B. elizabethae. The species detected in the Cairo spiny mouse (A. cahirinus) spleen sample was closely related to the zoonotic pathogen, B. elizabethae. These results indicate that Bartonella species are highly prevalent in suburban rodent populations and their ectoparasites in Israel. Further investigation of the prevalence and zoonotic potential of the Bartonella species detected in the black rats and the Cairo spiny mouse is warranted.  相似文献

Abstract.— In a microcosm study, sodium sulfate was mixed with a controlled composition soil to yield sulfur concentrations of 100 to 3,000 mg/kg and the mixture was inundated to a depth of 35 cm with 18-ppt salinity water. One juvenile Litopenaeus vannamei with initial weight averaging 0.3 g was placed in each soil-water microcosm and fed for 96 d. Dissolved oxygen concentration was maintained at approximately 70% air saturation. Calculated interstitial hydrogen sulfide concentration 0.5 cm below the soil-water interface increased with time, reaching maximum values ranging from 57 to 113 mg/L. Interstitial sulfide concentrations 0.5 cm below the soil-water interface increased as a function of soil S. Average weight gain of shrimp ranged from 7.4 to 8.7 g across treatment groups. Differences in shrimp weight gain and survival among treatments were not significant. A higher dissolved oxygen demand due to sulfide oxidation implies greater risk of shrimp mortality if the soil-water boundary is disturbed. Further, this study suggests that shrimp growth is not adversely affected by high soil sulfide concentration provided the sediment surface remains oxidized and that dissolved oxygen concentration in the overlying water is maintained at 70% saturation.  相似文献

Globally, cocoa is famous for the production of chocolates and beverages. However, it also serves as a source of raw material for the manufacturing of high-quality cosmetic and pharmaceutical products. Due to its high demand, cocoa is regarded as a major commodity for poverty alleviation in deprived cocoa farming communities. Government has therefore instigated relevant measures to address issues facing the sector by investing in extension programs. These programs act as information dissemination tools for teaching modern farming technologies and enhancing learning among farmers. This study highlighted the effects of the Cocoa Life Project(CLP) by Cooperative for Assistance and Relief Everywhere(CARE) International on farm productivity and income of 200 cocoa farmers in three districts of the Eastern Region, Ghana. Regression on covariates, Heckman's treatment effects model, and propensity scores were used to test the robustness of the estimates. A positive association between extension program participation, farm productivity, and household income was discovered. Specifically, after correcting for selection bias, the estimated results of the Heckman's model showed that the extension program significantly increased participating farmers' cocoa yield by 14.3%. The effects on total farm income showed a significant increase of 25.1 and 42.9%, respectively from regression on propensity scores and the Heckman's model. Across all three models, program participation led to an increase in total household income by 46.1, 31.7, and 69.3%, respectively. Other variables such as farming characteristics, institutional and demographic factors affected farmers' income, depending on the estimation method used. This study reiterated the supporting role played by extension programs in enhancing cocoa production and increasing household income in Ghana. Conclusively, government and development partners should invest in extension programs by providing enough logistics, training qualified agents, and creating a conducive atmosphere to support learning and technology adoption.  相似文献

The pond bottom soil and the accumulated sediments are integral parts of ponds. Concentrations of nutrients, organic matter and microorganism density in the pond bottom are several orders of magnitude greater than in the water. The accumulation of organic sediments may limit pond intensification. The intensive organic matter degradation at the pond bottom and high sediment oxygen demand exceeds the oxygen renewal rate. This leads to the development of anoxic conditions in the sediments and at the sediment–water interface. A series of anaerobic processes, affected by the redox potential of the system, are taking place. A large number of potentially toxic materials are generated. Among those are organic acids, reduced organic sulfur compounds, reduced manganese and sulfides.

Shrimp, as animals that normally live on or near the bottom, are exposed to conditions on the pond bottom. Exposure to toxic materials endanger the well being of the cultured shrimp. Reduced feeding, slower growth, mortality and possibly higher sensitivity to disease are reported.

The rational use of aerators to minimize the area of sludge accumulation, construction of ponds to trap sludge, stirring sediments, chemical poising of the redox system and environmentally accepted treatment and reuse of drained sediments are means to control the conditions at the pond bottom.  相似文献
