Blackleg disease, caused by the hemibiotrophic fungal pathogen Leptosphaeria maculans, is one of the most devastating disease of Brassica species worldwide. To date, a total of 20 race-specific blackleg resistance (R) genes have been reported and all of those loci are located in either the A or B genomes of various Brassica species. The B. oleracea genome (CC) shares a high ancestral synteny with the A genome of B. rapa, suggesting the presence of qualitative (race specific) resistance to blackleg disease is also possible in B. oleracea germplasm. In the present study the C genome of Korean B. oleracea germplasm was screened for the presence of blackleg R genes. Thirty-two inbred cabbage lines with unknown resistance profiles, along with five control B. napus lines with well-characterised race-specific R genes, were assessed for cotyledon resistance against two L. maculans isolates with known and highly-contrasting avirulence gene (Avr) profiles. Two cabbage accessions were identified which produced a strong resistance when challenged with either isolate, demonstrating the presence of effective blackleg R genes in the cabbage C genome. Additionally, 16 microsatellite markers linked to seven different R genes of the B. napus A genome were converted into markers for their homologous regions on the B. oleracea C genome. These markers were used to screen all B. oleracea lines to assess if the novel C genome R genes were syntenous to known R gene-homologous regions of the A genome. The resistant cabbage lines offer C genome R genes for the protection of B. oleracea varieties against incursion of blackleg disease, as well as novel additional resistance sources for introgression into B. napus and B. carinata breeding material.  相似文献

Cotton(Gossypium hirsutum L.) is utilized commercially in the production of textile, clothing, and household goods. Its growth is affected by various environmental conditions such as soil, climate, and water supply. Irrigation is one of the most important factors for crop management. This research was designed with the aim of studying the biochemical and anatomical features of two genotypes of cottonseed(Latif and Golestan) in order to determine optimum irrigation. Protein, starch, gossypol gland, total soluble protein, starch content, and the anatomical structure of the cotton seeds were investigated. The data were analyzed using the SAS Software. The results showed that when rainfed, Golestan genotype seeds had a larger number of secretory cavities, soluble protein, and starch compared to Latif seeds. There was also a noticeable difference in the size of the seeds. Cellulosic cell walls and protein particles were also observed in the seed structure under induced water stress conditions. It can therefore be concluded that in response to water shortage, there was a noticeable change in the morphometric, anatomic, and biochemical features of the cottonseeds. It can be concluded that when rainfed, the Golestan genotype of cottonseed has more compatibility than the Latif genotype. In general, the application of different levels of irrigation showed that at moderate levels of irrigation, anatomical features appeared more normal. The observations indicate that at high levels of irrigation, some cells begin to dehisce due to osmotic stress, which results in a lack of accurate formation of tissue structures. The Golestan genotype is therefore the best suited for dryland farming.  相似文献

It can be a challenge to find suitable horse bedding materials that provide higher moisture absorption, better animal comfort, greater fertilizer values, and improved indoor environment. Our first objective was to determine the water absorption capacity (WAC) of two bedding materials, flax shive (FS) and pine wood shavings (PWS), commonly used by equine facilities. The second objective was to measure ammonia (NH3), hydrogen sulfide (H2S), and greenhouse gas (GHG) concentrations emitted from these bedded stall surfaces. In this study, the WAC of bedding materials were measured at 0.5, 1, 2, 3, 4, 6, 8, 12, and 24 hours in the laboratory. A total of eight horses were used for a 14-day study period. Of these, four horses (group-1) were bedded with FS and the other four (group-2) were bedded with PWS for week-1. In week-2, bedding materials were switched between the two groups. Ammonia and H2S were measured in situ. For GHG measurement, air samples (methane [CH4], carbon dioxide [CO2], and nitrous oxide [N2O]) were collected 152 mm above the bedded stall surface in Tedlar bags using a vacuum chamber and analyzed for GHG using a gas chromatograph. The WAC of FS was 56% greater than the PWS. There were no significant differences in NH3, H2S, CH4, CO2, and N2O concentrations between the two bedding materials (P > .05). Nutrient contents between fresh and soiled bedded samples for each bedding type were different (P < .05). Measured nutrient contents between fresh FS and PWS and bedded FS and PWS bedding materials were similar (P > .05).  相似文献

Movement of the ligand/receptor complex in rhodopsin (Rh) has been traced. Bleaching of diazoketo rhodopsin (DK-Rh) containing 11-cis-3-diazo-4-oxo-retinal yields batho-, lumi-, meta-I-, and meta-II-Rh intermediates corresponding to those of native Rh but at lower temperatures. Photoaffinity labeling of DK-Rh and these bleaching intermediates shows that the ionone ring cross-links to tryptophan-265 on helix F in DK-Rh and batho-Rh, and to alanine-169 on helix D in lumi-, meta-I-, and meta-II-Rh intermediates. It is likely that these movements involving a flip-over of the chromophoric ring trigger changes in cytoplasmic membrane loops resulting in heterotrimeric guanine nucleotide-binding protein (G protein) activation.  相似文献
