Among the Phytophthora species that cause black pod of cacao, P. megakarya is the most virulent, posing a serious threat to cacao production in Africa. Correct identification of the species causing the black pod and understanding the virulence factors involved are important for developing sustainable disease management strategies. A simple PCR‐based species identification method was developed using the species‐specific sequences in the ITS regions of the rRNA gene. A phylogenetic tree generated for 119 Phytophthora isolates, based on the 60S ribosomal protein L10 gene and rDNA sequence, verified the PCR‐based identification assay and showed high interspecific variation among the species causing black pod. Phytophthora megakarya isolates were uniformly virulent in an assay using susceptible cacao pod husks inoculated with zoospores, while the P. palmivora isolates showed greater divergence in virulence. The virulence of P. megakarya was associated with earlier production of sporangia and an accelerated induction of necrosis. While zoospore germ tubes of both species penetrated pods through stomata, only P. megakarya produced significant numbers of appressoria. A hypersensitive‐like response was observed when attached SCA‐6 pods were inoculated with P. palmivora. SCA‐6 pods became vulnerable to P. palmivora when wounded prior to zoospore inoculation. Phytophthora megakarya was more aggressive than P. palmivora on attached SCA‐6 pods, causing expanding necrotic lesions with or without wounding. Phytophthora megakarya is predominant in the Volta region of Ghana and it remains to be seen whether it can displace P. palmivora from cacao plantations of Ghana as it has in Nigeria and Cameroon.  相似文献

Andrographis paniculata is a medicinal plant of immense therapeutic value. The present study was aimed to elucidate its genetic diversity based on morphochemical and RAPD markers from 53 accessions belonging to 5 eco-geographic regions. Analysis of variance and D 2 statistics revealed significant differences in all the metric traits and sufficient inter-cluster distances indicating considerable diversity among the accessions. The complementary approach of RAPD was used to evaluate the genetic dissimilarities among all the accessions using 6 highly polymorphic primers. The average proportion of polymorphic loci across primers was 96.28%. The molecular genetic diversity based on Shannon index per primer averaged 5.585 with values ranging from 3.08 to 8.70 indicating towards wide genetic base. RAPD based UPGMA and D 2 cluster analysis also revealed that various accessions available in different eco-geographic regions might have originated from native places of wild abundance. Similarity matrices were generated for molecular markers and morphometric data to determine the degree of congruence between the two. A highly significant but low correlation (r = 0.547, P < 0.001) was obtained thus implying the correspondence between the two. The species is hermaphroditic and a habitual inbreeder. The present study yielded a typical triangular congruence between its breeding system, morphometric traits and RAPD markers thus elucidating the usefulness of complementary approaches to make diversity analysis more explanatory and purposeful for optimum genetic amelioration and effective conservation of its genotypic variability.  相似文献

We report the results of a 3-year study in which we collected and analyzed soil and yield data from experimental plots in six commercial processing tomato fields in Yolo and Solano counties in California. Our objective was to assess the effect of soil chemistry and soil-delivered nutrients and fungicides on tomato plant health and fruit yield at harvest. Marketable yield, leaf necrosis, and fruit loss due to sunburn differed significantly between individual fields, with averages ranging from 81.2–138.5 Mg ha?1, 32–72% leaf necrosis and 1.9 to 8.8% sunburnt fruit, respectively. Higher-yielding fields showed significantly lower levels of leaf necrosis and sunburn damage and a positive correlation with pre-plant soil parameters such as potassium concentration and cation exchange capacity (CEC). Interestingly, soil amendments of composted poultry manure or other nutrient supplements in low-CEC fields, but not high-CEC fields, resulted in higher fruit yield and less leaf necrosis. While all fields showed symptoms typical of Verticillium wilt and some fields showed symptoms of Fusarium wilt, Fusarium crown and root rot, corky root, and root knot nematode, none of our soil amendments, including chemical and biological fungicides, significantly or consistently reduced incidence or severity of these diseases. We discuss our findings in the context of premature vine decline of tomato, an emerging phenomenon in production fields in the Sacramento Valley, which is characterized by the loss of plant vigor and canopy cover at the onset of fruit ripening and for which causative agent(s) and management options in California remain elusive. Effort rights‐based fisheries management (RBM) is less widely used than catch rights, whether for groups or individuals. Because RBM on catch or effort necessarily requires a total allowable catch (TAC) or total allowable effort (TAE), RBM is discussed in conjunction with issues in assessing fish populations and providing TACs or TAEs. Both approaches have advantages and disadvantages, and there are trade‐offs between the two approaches. In a narrow economic sense, catch rights are superior because of the type of incentives created, but once the costs of research to improve stock assessments and the associated risks of determining the TAC and costs of monitoring, control, surveillance and enforcement are taken into consideration, the choice between catch or effort RBM becomes more complex and less clear. The results will be case specific. Hybrid systems based on both catch and effort are increasingly employed to manage marine fisheries to capture the advantages of both approaches. In hybrid systems, catch or effort RBM dominates and controls on the other supplements. RBM using either catch or effort by itself addresses only the target species stock externality and not the remaining externalities associated with by‐catch and the ecosystem.  相似文献

A field-plot reclamation experiment was conducted on a virgin saline-sodic, sandy loam, permeable soil while growing rice with pre- and post-planting leaching under conditions of continuous and intermittent submergence. The soil studied contained very high amounts of soluble salts and exchangeable sodium throughout the profile. The chief salts were Cl? and SO2?4 of Na+, Ca2+ and Mg2+. The data obtained showed that post-transplanting leaching under intermittent submergence alone progressively decreased salinity and sodicity throughout the top 100 cm of the soil to levels safe for cultivation of relatively deep-rooted crops. The surface few centimetres of soil were essentially reclaimed within a few hours after leaching so that young rice seedlings established and survived to give good yield. It was concluded, therefore, that reclamation of these types of soil in arid and semi-arid regions, where good quality water is not available for leaching prior to transplanting rice, would not require any such pre-planting leaching. The results further indicated that there is no need to apply an amendment such as gypsum, mainly because Ca2+ and Mg2+ present in such soils are adequate to replace the initially high exchangeable sodium during leaching. Leaching efficiency was high under conditions of intermittent submergence. It was shown that leaching curves could be useful in determining the amount of leaching water required for a given mode of application in order to decrease harmful levels of salinity and sodicity to safer levels for a particular crop.  相似文献

A paramyxovirus with a thermostability of 60 min (typical of velogenic viruses) and a mean death time of > 90 h (typical of lentogenic viruses) was isolated from layers near Mooi River, South Africa. Our results, based on comparative nucleotide sequence data indicated that the virus is pigeon paramyxovirus 1 (PPMV-1), a variant of Newcastle disease virus. The F0 cleavage site contains a 112RRKKRF117 motif, and the virus had 98% sequence identity with PPMV-1 strains from the Far East. PPMV-1 was last reported in South Africa during the 1980s, with this being the first report of PPMV-1 isolated from chickens in South Africa.  相似文献

Rainwater samples (N = 51) were collected at Rampur, an areafree from anthropogenic activity during the monsoon of 1997 and1998. The concentration of ions follows a general pattern as Ca> NH4 > Mg > SO4 > Cl > F >Na > NO3 > K > HCOO >CH3 COO. The pH of precipitation ranges between 5.9 and 7.4. The levels of Ca and Mg at this site are higher than otherremote sites, probably dominated by particles of soil origin.Good correlation between Ca, NO3, SO4, HCOO and CH3COO indicate that a fraction of NO3, SO4, HCOOand CH3COO may be derived from soil or associated with Ca and Mg after neutralization. The order of neutralization factorCa (2.19) > NH4 (1.26) = Mg (1.26) indicates that majorneutralization occurred by Ca. Factor analysis suggested thatCa, Mg, Na, K, NO3, SO4, HCOO and CH3COO arecontributed by soil. NH3 is known to exist as(NH4)2SO4, NH4NO3 and NH4Cl. Theymay be formed in the atmospheric water droplets by scavenging ofaerosols and reaction of gaseous species.  相似文献
