The objective of this study was to examine the effect of sampling technique (pluck or cut), storage duration (immediate analysis, 24‐h or 48‐h), storage temperature (ambient or chilled) and storage conditions (air present, air excluded or breathable) on the composition of fresh grass sampled from a sward managed to simulate grazing. Treatments were repeated across four sampling dates, with grass samples stored in grip seal bags prior to analysis using near‐infrared reflectance spectroscopy. Grass sampled by ‘pluck’ had a higher crude protein and ME content, and a lower acid detergent fibre (ADF) content, compared to that sampled by ‘cut’. Grass stored for 48 h had a lower water soluble carbohydrate (WSC) and ME content and a higher ADF content than for immediate analysis. Samples stored for 24 h did not differ from immediate analysis. Grass stored at ambient temperature had a lower WSC and ME content compared to immediate analysis. Grass stored under ‘breathable’ conditions had a lower ME content and higher ADF content than immediate analysis or samples stored with air present or air excluded. It is recommended that grass for analysis should be sampled by cutting, stored chilled (4°C) in a sealed bag to minimize exposure to oxygen and analysed within 24 h of harvest.  相似文献

Nitrogen fertilizer practices affect nitrous oxide (N2O) emissions from agricultural soils. The “4R” nutrient stewardship framework of using N fertilizer at the right rate, right source, right placement and right time can reduce N2O emissions while maintaining or improving yield of field crops, but understanding of how the various factors affect N2O emissions from irrigated processing potato is lacking. We examined the effects of selected 4R practices on emissions, using results from two irrigated processing potato studies each conducted in 2011 and 2012 in Manitoba, Canada. Experiment 1 examined combinations of source (urea, ESN), placement (pre-plant incorporation [PPI], banding), and rate (100 and 200 kg N ha-1) on a clay loam soil. Experiment 2 examined timing and source treatment combinations (urea PPI, ESN PPI, urea split, urea split/fertigation) on a loamy fine sandy soil. For Experiment 1, use of ESN at 200 kg ha-1 did not reduce area-, yield- and applied fertilizer N- based N2O emissions compared to urea at 200 kg ha-1, irrespective of placement. Emissions from pre-plant banding ESN at 200 kg ha?1, however, were 32% lower than from PPI ESN. For Experiment 2, compared to single pre-plant urea application, fertigation simulated by in-season application of urea ammonium nitrate (UAN) gave lower area-, yield- and applied fertilizer N- based emissions. Split urea ( \( \raisebox{1ex}{$2$}\!\left/ \!\raisebox{-1ex}{$3$}\right. \) pre-plant, \( \raisebox{1ex}{$1$}\!\left/ \!\raisebox{-1ex}{$3$}\right. \) hilling) also reduced area- and yield- based N2O emissions compared to single pre-plant urea application. Emissions were generally lower at the site with loamy fine sandy soil than the site with clay loam soil. These results demonstrate that combinations of “4R” practices rather than source alone are best to achieve reductions in N2O emissions from irrigated potato production.  相似文献

ABSTRACT

1. This study quantified xylanase-induced changes in soluble monosaccharides, xylooligosaccharides (XOS) and volatile fatty acid (VFA) contents of the different sections of the gastrointestinal tract (GIT) and whether these were related to altered bird performance.

2. An in vitro digestion of the wheat-based diet was carried out with the xylanase (Econase XT at 16,000BXU/kg diet) to compare the in vitro and in vivo generation of these XOS and monosaccharides. For the in vivo study, 80 male Ross 508 b roiler chicks were split into two groups fed a wheat-based diet with or without Econase XT (16,000BXU/kg diet) for 21 days.

3. There were no effects of Econase XT inclusion on growth performance characteristics, likely a result of the high-quality wheat diet, the corresponding high performance of the control group (FCR average of 1.45 in controls) and the relatively young age of the birds (from four to 26 days of age).

4. Econase XT supplementation increased the xylotetraose (X4) content in the colon (P = 0.046, enzyme x GIT section interaction) and the xylose contents in the colon and caeca (P < 0.001, enzyme x GIT section interaction).

5. The trend for increased acetate production in the caeca of Econase XT treated birds (P = 0.062) suggested that the XOS generated were subsequently fermented in the caeca, potentially impacting upon the types of microbiota present.

The present study suggested that wheat arabinoxylan degradation was enhanced by xylanase supplementation, which may have increased the production of beneficial volatile fatty acids (VFA) in the caeca, and thereby potentially modulated the caecal microbiome, but without affecting bird performance at this early age.  相似文献

Cryopreservation has become anessential tool for operational application offorest tree embryogenic cultures, due to thelong evaluation periods needed for treesregenerated from these cultures. Fiveyellow-poplar (Liriodendron tulipifera)and seven sweetgum (Liquidambar spp.)embryogenic culture lines werestored in liquid nitrogen for 48 hours, afterwhich they were thawed and tested for regrowthand ability to produce somatic seedlings.Combinations of two sorbitol pretreatments andthree dimethylsulfoxide (DMSO) cryoprotectantlevels were evaluated for their impact onrecovery following cryogenic storage. The bestresults were obtained with 0.4 M sorbitol and5% DMSO, which provided 100% recovery.Somatic seedlings were regenerated from allculture lines and treatments, except for atransgenic sweetgum line.  相似文献

A series of conventional distance-independent and distance-dependent competition indices, a highly flexible distance-dependent crowding index, and two light resource estimation indices were compared to predict individual tree diameter growth of five species of mature trees from natural-origin boreal mixed forests. The crowding index was the superior index for most species and ecosites. However, distance-independent indices, such as basal area of competing trees, were also effective. Distance-dependent light estimation indices, which estimate the fraction of seasonal photosynthetically-active radiation available to each tree, ranked intermediate to low. Determining separate competition indices for each competitor species accounted for more variation than ignoring species or classifying by ecological groups. Species’ competitive ability ranked (most competitive to least): paper birch ≈ white spruce ≈> trembling aspen > lodgepole pine > balsam poplar. Stratification by ecosite further improved model performance. However, the overall impact of competition on mature trees in these forests appears to be small.  相似文献

Dehydrins are proteins associated with conditions affecting the water status of plant cells, such as drought, salinity, freezing and seed maturation. Although the function of dehydrins remains unknown, it is hypothesized that they stabilize membranes and macromolecules during cellular dehydration. Red-osier dogwood (Cornus sericea L.), an extremely freeze-tolerant woody plant, accumulates dehydrin-like proteins during cold acclimation and the presence of these proteins is correlated with increased freeze tolerance (Karlson 2001, Sarnighausen et al. 2002, Karlson et al. 2003). Our objective was to determine the location of dehydrins in cold-acclimated C. sericea stems in an effort to provide insight into their potential role in the freeze tolerance of this extremely cold hardy species. Abundant labeling was observed in the nucleus and cytoplasm of cold-acclimated C. sericea stem cells. In addition, labeling was observed in association with plasmodesmata of cold-acclimated vascular cambium cells. The unique association of dehydrin-like proteins with plasmodesmata has not been reported previously.  相似文献

The response of a forest to nutrient and moisture stresses is reflected in nutritional, physiological, and structural changes that include efficiency of nutrient use, translocation and cycling of nutrients, transpiration, retention of foliage, below-ground and above-ground allocation of carbon, as well as the structural development of the forest stand and its growth characteristics. This article reviews the relationship of forest ecosystems to nutrient and moisture stresses and addresses the means by which productivity can be enhanced by altering nutrient and moisture regimes.

Considerable research has focused on optimizing productivity by minimizing nutrient and moisture stresses. Research involved in nutrient additions has led to the use of commercial fertilizers to improve forest productivity. The results suggest that many forests are deficient in N and P and, to a lesser extent, S, K, Mg and trace elements. The duration of response for most nutrient additions is, however, relatively brief and the efficiency of the tree in using fertilizer is relatively poor. Long-term correction of nutrient deficiencies is seldom achieved with chemical fertilizers. However, N added through symbiotic fixation or, on a more limited scale, through addition of municipal and industrial waste by-products, can provide an excellent long-term growth response.

It is seldom feasible to change the moisture regime of a forest ecosystem through irrigation. However, field trials involving irrigation have demonstrated that moisture stress can limit productivity. There are various ways of minimizing moisture stress without irrigation, including mulching, removing ground-cover vegetation, and changing the spatial characteristics of the forest cover.

Research trials show that forest ecosystems will respond to moisture and nutrient additions; however, these responses and interactions between nutrients and moisture are typically poorly understood, and many questions remain unanswered: Does fertilization increase moisture-use efficiency of a forest or simply improve the nutrition of the site? Does improving the moisture regime of a site improve productivity primarily by decreasing moisture stress or by increasing nutrient availability and the rate of nutrient uptake? Is there a synergism in growth response with the addition of both nutrients and moisture? The linkages between nutrients and moisture appear inseparable and confound experimentation in this field. Answers to these questions and issues need to be found for the future development of plantation forestry.  相似文献

Summary Although the bonds between the cellular components of pine epithelia (isolated from holocellulose) can be strengthened by the addition of multivalent ions such as those of calcium, iron, uranium, etc., it was observed that these bonds are only weakened and not destroyed by the removal of the ions from the tissues. Reagents, such as KOH and K2SO4, which do not yield soluble calcium salts, do not cause a discernible weakening of the bonds between the cellulose components of epithelia pretreated with Ca ion. Washing the weakened epithelia with water causes them to swell. The separation of epithelial cells occurs only after the application of mechanical forces to the tissues. Electron microscopic examination of the swollen and unswollen tissues shows that fibrils and lamellae extend between the individual cells which must be destroyed by mechanical action before cell separation can occur. The swelling of the tissues by washing after removal of multivalent ions was attributed to the expansion of electrical double layers and to the increasing osmotic pressure from the increasing Donnan potential, while the difference in swelling as a result of HCl treatment vs. KCl and K4Fe(CN)6 was attributed to differences in charge development and to hydrogen bonding between acidic components. The restrengthening of the tissue as a result of the readdition of multivalent ion was attributed to the collapse of the electrical double layers and the reduction of the Donnan potential, with the re-forming of bonds between the components of the epithelia. Evidence suggesting that multivalent ions actually participate in bonding was obtained and although the nature of the bonding cannot be determined, both salt formation and complex formation between the ions and the components of the tissues is suggested.  相似文献
