The digestive tract of termite(Microcerotermes diversus) contains a variety of lignocellulose-degrading bacteria with exocellulases enzyme activity, not found in the rumen, which could potentially improve fiber degradation in the rumen. The objectives of the current study were to determine the effect of inoculation of rumen fluid(RF) with three species of bacteria isolated from termite digestive tract, Bacillus licheniformis, Ochrobactrum intermedium, and Microbacterium paludicola, on in vitro gas production(IVGP), fermentation parameters, nutrient disappearance, microbial populations, and hydrolytic enzyme activities with fibrous wheat straw(WS) and date leaf(DL) as incubation substrate. Inoculation of RF with either of three termite bacteria increased(P0.05) ammonia-N concentration compared with the control group(free of termite gut bacteria). Termite bacteria inoculation had no effect(P0.05) on gas production characteristics, dry matter, organic matter and neutral detergent fiber disappearance, pH, and concentration and composition of volatile fatty acids. Population of proteolytic bacteria and protozoa, but not cellulolytic bacteria, were increased(P0.05) when RF was inoculated with termite bacteria with both WS and DL substrates. Inoculation of RF with termite bacteria increased protease activity, while activities of carboxymethyl-cellulase, microcrystalline-cellulase, α-amylase and filter paper degrading activity remained unchanged(P0.05). Overall, the results of this study indicated that transferring lignocellulose-degrading bacteria, isolated from digestive tract of termite, to rumen liquid increased protozoa and proteolytic bacteria population and consequently increased protease activity and ammonia-N concentration in vitro, however, no effect on fermentation and fiber degradation parameters were detected. These results suggest that the termite bacteria might be rapidly lysed by the rumen microbes before beneficial effects on the rumen fermentation process could occur.  相似文献

The effects of barley flour on the fermentation parameters of alfalfa silage and on the productivity of dairy cows were investigated. Alfalfa forage was ensiled either with or without barley flour. Barley flour was soaked in water for 24 h before being mixed with alfalfa (12 kg: 100 kg dry matter bases) at ensiling. Eighteen multi-parous cows were assigned to three equal treatment groups using a completely randomized design. Three isocaloric and isonitrogenous total mixed rations containing alfalfa hay, ordinary alfalfa silage or barley flour mixed alfalfa silage were then prepared. The concentrations of ammonia nitrogen, acetic acid and butyric acid were lower in barley flour mixed alfalfa silage compared to that in ordinary alfalfa silage but the concentration of lactic acid was lower in the ordinary alfalfa silage. Feeding behavior, milk yield and composition, ruminal fermentation and blood metabolites were measured. Although dry matter intake and milk production were not affected, the effect of preparation of alfalfa influenced feeding behavior and rumen fermentation parameters. Cows on alfalfa silage diets spent longer ruminating compared to those fed alfalfa hay. The ruminal ammonia nitrogen and blood urea were affected by ensiling (alfalfa hay versus alfalfa silages) while both parameters were lower in cows fed on barley flour mixed alfalfa silage than those fed on ordinary silage. Although similar blood glucose was recorded for cows fed on alfalfa silages, it was higher in cows fed on alfalfa hay. It is concluded that the addition of barely flour when making alfalfa silage may improve both the fermentation process during ensilage and the ruminal ammonia nitrogen utilization with no significant effects on productivity.  相似文献

Sixty-eight rice germplasms were used for cold tolerance evaluation at the germination stage in laboratory. Seeds of rice germplasms germinated at three different temperatures containing low temperature (constant 13°C for 28 days), alternative temperature (a temperature cycle of 12 h at 20°C and 12 h at 23°C for 14 days) and control (constant 26°C for 7 days). Analysis of variance revealed that temperature had a significant effect on germination rate, coleoptile length and radicle length. Germination rate was strongly affected by the low temperature and alternative treatments. Stress of low temperature at the germination stage on the tested rice germplasms caused a reduction in final germination rate and the lengths of coleoptile and radicle. The normal and healthy seeds began to germinate within 36 h after imbibition and germination were completed on the 7th day in the control, but delayed under the low and alternative temperature treatments. The low and alternative temperature treatments delayed the growth of coleoptile and radicle compared to the control, and the average lengths of coleoptile and radicule were strongly inhibited with the decreasing temperature in all of the tested germplasms. Taichung, a semi-dwarf, low amylose content and early maturing rice variety, showed a significantly higher germination rate at the three temperature treatments and its coleoptile length was significantly higher than other germplasms.  相似文献
