Proanthocyanidins (PAs) in the hot water extract prepared from Pinus radiata bark were isolated by a Sephadex LH-20 chromatography. The PAs were characterized using UV/Visible, FT-IR and 13C nuclear magnetic resonance (NMR) spectroscopic techniques, matrix-assisted laser desorption/ionization time of flight (MALDI-TOF) mass spectrometry (MS), and gel permeation chromatography (GPC) analysis. Bate-Smith acidic alcoholysis and FT-IR spectrum of the PAs indicated the predominant presence of the procyanidin (PC) structure. The 13C NMR spectrum revealed that the PAs comprise PC (94%) together with a small amount of prodelphinidin (PD, 6%), and also that they contain the higher trans configuration rather than the cis configuration of C2–C3. The MALDI-TOF MS results obviously proved the presence of PD units, but the maximum degree of polymerization (DP) was limited to octamer. The average molar mass (Mw, Mn), polydispersity (Mw/Mn), and DP of the pine bark PAs were 3,800 (Mw), 1,200 (Mn), 3.2, and 13, respectively.  相似文献

Forty‐eight castrated male goats were used to determine the effects of feeding green tea by‐products (GTB) on growth performance, meat quality, blood metabolites and immune cell proliferation. Experimental treatments consisted of basal diets supplemented with four levels of GTB (0%, 0.5%, 1.0% or 2.0%). Four replicate pens were assigned to each treatment with three goats per replicate. Increasing dietary GTB tended to linearly increase the overall average weight gain and feed intake (p = 0.09). Water holding capacity, pH and sensory attributes of meat were not affected by GTB supplementation, while cooking loss was reduced both linearly and quadratically (p < 0.01). The redness (linear; p = 0.02, quadratic; p < 0.01) and yellowness (quadratic; p < 0.01) values of goat meat were improved by GTB supplementation. Increasing dietary GTB quadratically increased protein and decreased crude fat (p < 0.05), while linearly decreased cholesterol (p = 0.03) content of goat meat. The proportions of monounsaturated fatty acid, polyunsaturated fatty acid (PUFA) and n‐6 PUFA increased linearly (p < 0.01) and n‐3 PUFA increased quadratically (p < 0.05) as GTB increased in diets. Increasing dietary GTB linearly increased the PUFA/SFA (saturated fatty acid) and tended to linearly and quadratically increase (p ≤ 0.10) the n‐6/n‐3 ratio. The thiobarbituric acid‐reactive substances values of meat were lower in the 2.0% GTB‐supplemented group in all storage periods (p < 0.05). Dietary GTB linearly decreased plasma glucose and cholesterol (p < 0.01) and quadratically decreased urea nitrogen concentrations (p = 0.001). The growth of spleen cells incubated in concanavalin A and lipopolysaccharides medium increased significantly (p < 0.05) in response to GTB supplementation. Our results suggest that GTB may positively affect the growth performance, meat quality, blood metabolites and immune cell proliferation when supplemented as a feed additive in goat diet.  相似文献

To evaluate the performance of new rice establishment methods viz., system of rice intensification (SRI) and integrated crop management (ICM), a field study was conducted during 2008–11 in South Garo Hills, Meghalaya, foot hills of Eastern Himalayas, India. Field demonstrations were undertaken during wet seasons of 2008–11 and socio-economic information in the context of farmer’s realities were obtained during 2010–11 using a well structured questionnaire administered to 134 farmers. The results indicated that the average higher productivity of rice under SRI and ICM demonstration was 209.9 and 185.4 %, respectively, over conventional rice culture (CRC). The SRI and ICM methods of rice cultivation could save seeds (97.56 and 60.98 %), saving water (78.05 and 63.66 %), reduce cost (70.33 %), higher yield etc. compared to CRC. The main reasons for non-adoption of SRI/ICM was related to involvement of more efforts, faith towards traditional practices, ignorance and lack of knowledge on scientific water management. The net-return of $816.69, $706.63 and $51.48/ha was realized under SRI, ICM and CRC, respectively. The co-efficient of multiple determinations (R 2) of the production function was 0.695 in SRI, 0.714 in ICM and 0.734 in CRC which indicated that about 69.5, 71.4 and 73.4 % of the variation in rice productivity under SRI, ICM and CRC, respectively were explained by the independent variable and remaining 30.5, 28.6 and 26.6 %., respectively in SRI, ICM and CRC were as a result of non-inclusion of some explanatory variables as well as other factors outside the farmers control.  相似文献

Electron spin resonance (ESR) spectroscopy and near-infrared (NIR) fluorescence spectroscopy were performed to observe singlet oxygen quenching by resveratrol. Resveratrol greatly decreased the 2,2,6,6-Tetramethyl-4-piperidone-N-oxyl radical signal as determined by ESR spectroscopy. Resveratrol also efficiently decreased luminescence emission at 1268 nm as studied with a NIR spectrofluorometer, showing positive evidence of singlet oxygen quenching by resveratrol. The total singlet oxygen quenching rate constant (kr+kq) of resveratrol in methanol was determined to be 2.55×10(7) M(-1) s(-1). The singlet oxygen chemical quenching rate constant (kr) of resveratrol was calculated by measuring its reaction rate with singlet oxygen relative to that of α-terpinene in the same solution under light illumination. The kr value of resveratrol was 1.15×10(6) M(-1) s(-1). The percent partition of chemical quenching over total singlet oxygen quenching (kr×100)/(kr+kq) for resveratrol was 5.11%. The results showed that resveratrol quenches singlet oxygen almost exclusively through the mechanism of physical quenching. Resveratrol showed a protective activity similar to that of BHA on the methylene blue sensitized photooxidation of α-terpinene. This unambiguously explains the mechanism of how resveratrol protects tissues and cells in biological systems or important nutrients in food systems against their photosensitized oxidations.  相似文献

The pear cultivar ‘Osa-Nijisseiki’ (S2Ssm 4; sm = stylar-part mutant) has been used as a parent to breed self-compatible cultivars that produce excellent fruits. However, determination of the self-compatibility of ‘Osa-Nijisseiki’ offspring requires a lot of time, 6 years or more, by conventional cross breeding. We have designed a rapid reliable method for the identification of self-compatible varieties of ‘Osa-Nijisseiki’ offspring based on the polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) with S-allele specific restriction endonucleases. By using this method, 8 self-compatible varieties were selected among 16 selections resulting from a cross between the self-compatible cultivar ‘Osa-Nijisseiki’ (S2Ssm 4) and the self-incompatible cultivars ‘Niitaka’ (S3S9), ‘Whasan’ (S3S5), ‘Chuwhangbae’ (S4S6). The S-genotypes of 16 ‘Osa-Nijisseiki’ offsprings were also determined. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献

The effect of acidic deposition on the soil under red pine forest in Chunchon, Korea was investigated. Precipitation, stream water, and soil solution chemistry were monitored at the watershed from 1997 to 1998. Acidity of the open-bulk precipitation was often neutralized by large amounts of ammonia (NH3) that might have originated from livestock farming and fertilization. Estimated elemental budget at the watershed showed a positive correlation between loss of base cations and proton (H+) production due to nitrogen transformation in soil (ΔH+ NT: ([NH4 +]in-[NH4 +]out)- ([NO3 ?]in-[NO3 ?]out)). When ΔH+ NT increased, concentrations of nitrate in soil solutions also increased. Consequently, pH values of soil solutions decreased, although ion exchange with base cations contributed to buffer reaction. Since acid buffering capacity of the red pine forest soil was small, it was concluded that the input of ammonium nitrogen enhanced nitrification in soil thus causing soil acidification represented by loss of base cations from the watershed.  相似文献

We studied the spatial and temporal patterns of decomposition of roots of a desert sub-shrub, a herbaceous annual, and four species of perennial grasses at several locations on nitrogen fertilized and unfertilized transects on a Chihuahuan Desert watershed for 3.5 years. There were few significant differences between the decomposition rates of roots on the NH4NO3 fertilized and unfertilized transects. Decomposition of all roots followed a two-phase pattern: early rapid mass loss followed by a long period of low mass loss. Rates of decomposition were negatively correlated with the initial lignin content of the roots (r=0.90). Mass loss rates of the roots of the herbaceous annual, Baileya multiradiata, were significantly higher than those of the grasses and the shrub, probably as a result of subterranean termites feeding on B. multiradiata root material. The only location where mass loss rates were significantly different was the dry lake bed, where mass loss rates were lower than those recorded on the upper watershed. The spatial differences in mass loss rates in the dry lake were attributable to the high clay content of the soils, which reduced water availability, and to the absence of termites. Non-polar substances in decomposing roots decreased rapidly during the first year, then decreased at a low but fairly constant rate. Water-soluble compounds decreased rapidly (50–60% of initial concentration) during the first 3–6 months. Lignin concentrations of roots of perennial grasses were higher than those of herbaceous annual plants and woody shrubs. Lignin concentrations increased in all species during decomposition. OBJECTIVE: This experiment evaluated the perinatal hypoxic effect on the retina of offspring of the ovoviviparous fish. ANIMAL STUDIED: The ovoviviparous fish Xiphophorous maculates was used for the experiment. PROCEDURE: The mothers were kept in a hypoxic environment of 3.5% oxygen for 6 h, starting 30 h before hatching. Subsequently, the retinae of the offspring were fixed, sectioned at 6 microm and evaluated microscopically from the age of 1 to 35 days. RESULTS: Degeneration of the outer nuclear layer of the retina was noted on the 3rd day and severe retinal degeneration was observed on the 35th day. Immunocytochemistry confirmed apoptosis by TUNEL reaction. There was no difference in neovascularization, as revealed by vascular endothelial growth factor, between controls (group 1) and hypoxic fish (group 2). CONCLUSIONS: Perinatal hypoxia could have long-lasting effects on the central nervous system in some species.  相似文献

Approximately 7,000 accessions of Korean soybean (Glycine max (L.) Merrill) landraces, largely composed of three collections, the Korea Atomic Energy Research Institute’s soybean (KAS), the Korean Crop Experiment Station’s soybean (KLS) and the Korean Agricultural Development and Technology Center’s soybean (KADTC) collections, have been conserved at the Rural Development Administration (RDA) genebank in Korea. The accessions within collections were classified based on their traditional uses such as sauce soybean (SA), sprouted soybean (SP), soybean for cooking with rice (SCR), and OTHERS. A total of 2,758 accessions of Korean soybean landraces were used to profile and to evaluate genetic structure using six SSR loci. A total of 110 alleles were revealed by at the six SSR loci. The number of alleles per SSR locus ranged from 9 to 39 in Satt187 and Satt_074, respectively. The number of alleles ranged from 87 in the KADTC collection to 96 in the KLS collection, and from 63 in the SCR group to 95 in the SP group. Nei’s average genetic diversity ranged from 0.68 to 0.70 across three collections, and 0.64 to 0.69 across the usage groups. The average between-group differentiation (G st) was 0.9 among collections, and 4.1 among the usage groups. The similar average diversity among three collections implies that the genetic background of the three collections was quite similar or that there were a large number of duplicate accessions in three collections. The selection from the four groups classified based upon usage may be a useful way to select accessions for developing a Korean soybean landrace core collection at the RDA genebank. DNA profile information of accessions will provide indications of redundancies or omissions and aid in managing the soybean collection held at the RDA genebank. The information on diversity analysis could help to enlarge the genetic diversity of materials in breeding programs and could be used to develop a core collection.  相似文献
