Mycosporine-like amino acids (MAAs), a group of small secondary metabolites found in algae, cyanobacteria, lichens and fungi, have become ecologically and pharmacologically relevant because of their pronounced UV-absorbing and photo-protective potential. Their analytical characterization is generally achieved by reversed phase HPLC and the compounds are often quantified based on molar extinction coefficients. As an alternative approach, in our study a fully validated hydrophilic interaction liquid chromatography (HILIC) method is presented. It enables the precise quantification of several analytes with adequate retention times in a single run, and can be coupled directly to MS. Excellent linear correlation coefficients (R2 > 0.9991) were obtained, with limit of detection (LOD) values ranging from 0.16 to 0.43 µg/mL. Furthermore, the assay was found to be accurate (recovery rates from 89.8% to 104.1%) and precise (intra-day precision: 5.6%, inter-day precision ≤6.6%). Several algae were assayed for their content of known MAAs like porphyra-334, shinorine, and palythine. Liquid chromatography-mass spectrometry (LC-MS) data indicated a novel compound in some of them, which could be isolated from the marine species Catenella repens and structurally elucidated by nuclear magnetic resonance spectroscopy (NMR) as (E)-3-hydroxy-2-((5-hydroxy-5-(hydroxymethyl)-2-methoxy-3-((2-sulfoethyl)amino)cyclohex-2-en-1-ylidene)amino) propanoic acid, a novel MAA called catenelline.  相似文献

Easy-to-capture and robust plant status indicators are important factors when implementing precision agriculture techniques on fields. In this study, aerial red, green and blue color space (RGB) photography and near-infrared (NIR) photography was performed on an experimental field site with nine different cover crops. A lightweight unmanned aerial system (UAS) served as platform, consumer cameras as sensors. Photos were photogrammetrically processed to orthophotos and digital surface models (DSMs). In a first validation step, the spatial precision of RGB orthophotos (x and y, ± 0.1 m) and DSMs (z, ± 0.1 m) was determined. Then, canopy cover (CC), plant height (PH), normalized differenced vegetation index (NDVI), red edge inflection point (REIP), and green red vegetation index (GRVI) were extracted. In a second validation step, the PHs derived from the DSMs were compared with ground truth ruler measurements. A strong linear relationship was observed (R 2 = 0.80?0.84). Finally, destructive biomass samples were taken and compared with the remotely-sensed characteristics. Biomass correlated best with plant height (PH), and good approximations with linear regressions were found (R 2 = 0.74 for four selected species, R 2 = 0.58 for all nine species). CC and the vegetation indices (VIs) showed less significant and less strong overall correlations, but performed well for certain species. It is therefore evident that the use of DSM-based PHs provides a feasible approach to a species-independent non-destructive biomass determination, where the performance of VIs is more species-dependent.  相似文献

Many mountain forests in the Swiss Alps are dense and overmature. The resulting lack of tree regeneration threatens their future ability to provide products and services for humans, e.g., protecting settlements and infrastructure against avalanches. To promote natural regeneration, slit-shaped gaps have been cut since the 1980s in many of Switzerland's Alpine forests dominated by Norway spruce (Picea abies). However, little is known about the success of this silvicultural technique. We sampled 38 gaps in 2001 and 2006 and analysed the density and vitality of P. abies seedlings in these gaps, and monitored the growth and survival of selected seedlings between the two inventories. The gaps analysed were located in upper montane and subalpine P. abies dominated forests in the Vorderrhein valley in the Grisons. The density and vitality of large (10–129 cm tall) and small (<10 cm tall) P. abies seedlings were assessed in three parallel transects running perpendicular to the longitudinal axis of each gap. The mean density of large seedlings increased significantly between 2001 and 2006 from 0.1 to 0.3 seedlings/m2, whereas that of small seedlings stayed constant at 0.7 seedlings/m2, even though it shifted locally between inventories. Significantly higher regeneration densities were found for gaps with NE–SW orientation (afternoon sun) and for those located at lower altitudes. While in gaps on North-facing slopes large P. abies seedlings were more frequent on the middle transect, in gaps on South-facing slopes they were denser near the lower gap edge which is usually less exposed to direct radiation. As expected regeneration density was significantly higher within the gaps than just outside underneath the adjacent stands for all P. abies seedlings. Damage caused by browsing did not turn out to be problematic in this study site. From the selected 280 small seedlings monitored in 2001, 53% died until 2006, 33% became large seedlings and 14% remained small (<10 cm). Our results suggest that creating slit-shaped gaps is a successful procedure to promote P. abies regeneration in the upper montane and subalpine belt of the northern intermediate Alps and helps to preserve protection forests and their goods and services for the coming generations.  相似文献

Cold-pressed, non-raffinated evening primrose oil was found to contain lipophilic radical scavengers. A highly enriched fraction of these compounds could be obtained from the oil by extraction with aqueous ethanol and subsequent liquid-liquid partitioning with petroleum. LC-DAD-MS analysis revealed that the fraction contained three aromatic compounds with identical UV and ESI-MS spectra. The compounds were isolated by RP-HPLC and their structures established by chemical and spectroscopic means as 3-O-trans-caffeoyl derivatives of betulinic, morolic, and oleanolic acid. The morolic acid derivative was a new compound. The three esters exhibited pronounced radical scavenging activity against the stable 2,2-diphenyl-1-picrylhydrazyl radical and were potent inhibitors of neutrophil elastase and cyclooxygenase-1 and -2 in vitro. Commercial samples of evening primrose oils contained only traces of these lipophilic antioxidants.  相似文献

Optimum plant densities are a key to maximise yields in most crops. However, such information is often lacking for more environmentally sound cropping systems, such as living mulches (LM) for small grains. In 2004 and 2005, three trials were conducted in the Swiss Midlands on fields managed in accordance with the Swiss organic farming guidelines. The objective of the study was to determine whether seeding density of winter wheat (Triticum aestivum L.) is a relevant factor for determining grain yield in a white clover (Trifolium repens L.) living mulch. The winter wheat cv. Titlis was directly sown in wide spaced rows (0.375 m) at densities of 300 (LM300), 450 (LM450) or 600 (LM600) viable grains m−2 in a white clover living mulch established at a seeding rate of 15 kg ha−1. A bare soil control treatment with a wheat density of 450 viable grains m−2 (BS450) was also included in the trials. Mean grain yields of LM300, LM450, and LM600 never reached the values observed in BS450. This was mainly due to a lower ear density, which, nevertheless, increased linearly with the seeding density within the living mulch in all trials, but the rate of increase depended on the environment. The decrease of the grain weight brought about by the increasing seeding density had only a marginal impact on the grain yield, which was increased from 1.31, 1.98, and 4.09 Mg ha−1 (LM300) to 1.97, 2.64, and 4.75 Mg ha−1 (LM600) for each of the three trials in the study. Significantly higher protein contents were observed for LM300 compared to the higher densities in the living mulch and to BS450. Our research showed that an increase of the seeding density is an effective mean to increase the grain yield in living mulch systems with white clover. However, it is likely that the control of the living mulch to reduce competition with the main crop is a more relevant factor.  相似文献
