为研究一株鸭源H9N2亚型禽流感病毒(JN1株)全基因组的分子特征和遗传进化情况以及感染雏鸭后的排毒规律,对其生物学特性、全基因组序列以及遗传进化进行了分析,通过点眼滴鼻方式感染3周龄健康雏鸭,利用建立的实时荧光定量PCR(RT-PCR)检测鸭的排毒规律,并测定血清抗体效价。JNI株的鸡胚半数感染量为10~(-7.54)/0.2mL,鸡胚最小致死量的平均死亡时间为72h,静脉致病指数为0.266,抗原相关系数为0.47。系统发育分析表明,JN1株的HA与CK/HK/G9/97和DK/HK/Y280/97在同一分支上,NA、M、NP、PA、PB1、PB2基因与SH/F/98同源性较高,NS基因则与H5N1亚型禽流感病毒进化关系较近。HA的裂解位点为PSRSSR↓GL,存在6个潜在糖基化位点,在234位的氨基酸残基为L,NA基因颈部存在缺失。雏鸭在感染该病毒后,饮食正常,精神良好,3~17d均有排毒,咽拭子和泄殖腔棉拭子排毒规律基本一致,分别在第3天和第13天出现排毒高峰。该H9N2亚型禽流感病毒属欧亚大陆分支,为低致病性禽流感,病毒可通过呼吸道和泄殖腔向外界排毒,排毒时间较长。  相似文献

从发病商品肉鸽组织中分离到3株新城疫病毒,对其基因序列和感染力进行测定和分析。用RT-PCR对3株病毒F基因进行了扩增,测序,3株新城疫病毒的F基因同源性比较及进化分析结果表明,3株病毒均属于Ⅵb亚型,且与2011年比利时分离株亲缘关系较近,F蛋白裂解位点符合强毒特征;肉鸽攻毒试验进一步证实了3株新城疫病毒的高致病性,肉鸽感染新城疫病毒后通过呼吸道及消化道途径排毒,具有高度接触传染性。试验结果证明了鸽感染新城疫病毒后的排毒时间及方式,为鸽新城疫的及早发现与防控提供了依据。  相似文献

The influence of three diets, comprising dam's milk (DM) from the same farm, commercial milk replacer with (CMRL) or without 3.2% lactose (CMR), on the duration and intensity of Candida glabrata shedding in the faeces of preweaned calves following experimental oral infection was examined. Shedding of other potential enteric pathogens was also monitored. The duration and intensity of C. glabrata shedding in DM-fed calves were reduced significantly compared with the calves fed the two diets based on milk replacers. Consequently, feeding calves with DM might disrupt the infective cycle, resulting in the yeast's elimination from a farm. In the CMR and CMRL groups, the periods of intensive shedding of C. glabrata and rotavirus overlapped but no diarrhoea was associated with the shedding of either microorganism. There was no evidence that lactose diminished colonization in vivo.  相似文献

Reproductive efficiency was successfully manipulated by the application of different concentrations of triadimefon at pre‐flowering, flowering and post‐flowering stages in chickpea (Cicer arietinum L.). Triadimefon improved the parameters determining reproductive efficiency. Yield was significantly improved due to reductions in flower and pod abscission by the application of triadimefon, which also lowered the ethylene levels in leaves, flowers and pods and delayed senescence. These findings suggest a relationship between ethylene level and reproductive efficiency in chickpea. Further, they indicate that triadimefon can be used for chemical manipulation of the reproductive efficiency of chickpea under field conditions.  相似文献

Salsola australis, like other agricultural weed species of the Salsola genus, produces a mobile seedbank. Aspects of this mobile seedbank were investigated in three field trials, including total seed production, rate of seed shedding, rate at which seeds lose germinability and the distance and directionality of plant movement. Total seed production was highly variable (ranging from 138 to 7734 seeds per plant), but was directly related to aboveground plant biomass at maturity. Following senescence, mature plants broke free of their root system and the wind driven plants moved considerable distances (1.6–1247.2 m). Half of the mobile plants moved <100 m, as they became entangled with other S. australis plants within the stand. Seed shedding commenced before the plants became mobile and increased with movement, but was also related to the aging and weathering processes experienced by stationary or mobile plants. All plants retained a proportion of their seed in spite of movement, weathering and ageing of the plants, although germinability of retained seed dropped to <2% after 2 months. Salsola australis engages in broad scale seed dispersal similar to that observed in other species of the Salsola genus, allowing this species to maintain a high rate of invasion and range expansion.  相似文献

This study was to evaluate the effects of lactulose supplementation on performance, blood profiles, excreta microbial shedding of Lactobacillus and Escherichia coli, relative organ weight and excreta noxious gas contents in broilers. A total of 720 ROSS 308 broilers with a body weight of 46 ± 0.1 g (1 day of age) were used in a 28‐d experiment. Broilers were randomly allotted to 4 experiment diets with 12 replicate pens and 15 birds per pen. Dietary treatments were as follows: NC, negative control (without antibiotic); PC, NC + 0.1% tiamulin; L1, NC + 0.1% lactulose; and L2, NC + 0.2% lactulose. Broilers were fed with phase 1 (1–8 day), phase 2 (9–18 day) and phase 3 (19–28 day) diets in the form of mash. During day 1–8, broilers fed the PC and L2 diets had higher (p < 0.05) body weight gain than those fed the NC diet. During day 19–28, broilers fed the L1 and L2 diets had lower (p < 0.05) feed intake than those fed the NC diet. The feed conversion ratio (FCR) was decreased (p < 0.05) in L1 treatment compared with NC treatment. Overall, the FCR was improved (p < 0.05) in all supplementation treatments compared with NC treatment. The apparently metabolizable nitrogen in L1 treatment was higher (p < 0.05) than that in NC treatment at day 28. The excreta Lactobacillus was increased and E. coli was decreased in PC and L2 treatments compared with NC treatment at day 28 (p < 0.05). The excreta NH3, H2S and acetic acid contents were decreased (p < 0.05) in L1 and L2 treatments compared with NC treatment. The relative weight of abdominal fat of broilers fed the PC diet was lowest (p < 0.05) compared with other treatments. In conclusion, this study indicated that dietary supplementation of 0.1% or 0.2% lactulose could improve growth performance, decrease excreta E. coli and excreta NH3 and H2S contents.  相似文献

为了分析免疫鸡群中新城疫强毒感染流行的原因,明确抗体效价与流行株感染排毒率之间的关系,本研究以LaSota为抗原制备新城疫灭活疫苗,并以0.02mL和0.4mL的量分别免疫3周龄的SPF鸡10只。免疫后7、14、21d分别测定免疫鸡血清中的抗体HI效价。免疫后21d以基因Ⅶd亚型新城疫流行株JS5/05进行攻毒,攻毒后每天观察试验鸡的临床症状,并于攻毒后3、5、7d采集试验鸡的喉气管与泄殖腔棉拭样品进行病毒分离,结果显示,免疫3周后0.02mL和0.4mL疫苗免疫组鸡的血清HI抗体平均效价分别为5.4log2和8.2log2;0.02mL免疫组在攻毒后的排毒率达到100%,且排毒时间较长,而0.4mL免疫组在攻毒后的排毒率明显降低,且排毒时间较短。上述结果表明新城疫抗体效价与流行株感染排毒率之间存在明显的负相关。  相似文献

Abstract

Sixty tartary buckwheat (Fagopyrum tataricum) strains were investigated for grain shedding habit, plant height, grain yield, dry weight of plant, weight of grain, days to flowering after sowing and DNA content of nuclei. The grain shedding habit was evaluated from the breaking strength of pedicel and the percentage of the grains dropped by threshing. DNA content of nuclei was detected by flow cytometry to classify the polyploidy level. The fluorescence intensity of the flow cytometry demonstrated no difference between common buckwheat (F. esculentum) and tartary buckwheat, but a clear difference was detected between the diploid and the autotetraploid strains. The survey of DNA content of the 60 tartary buckwheat strains revealed that most of the strains were diploids, but one strain was autotetraploid. The autotetraploid strain exhibited less grain shedding, and produced the largest grains among the strains tested. However, the other characters of the autotetraploid strain, such as plant height, grain yield, dry weight of a plant, and days to flowering, were similar to the mode of the traits of the diploid strains.  相似文献

为研究库尔勒香梨突萼、脱萼、宿萼3种类型果实的品质差异,对其单果质量、可食率等12种果实品质指标、矿质元素及香气成分含量进行测定。结果显示:突萼果的单果质量及果皮花青素含量显著优于脱萼果与宿萼果;脱萼果的石细胞含量显著低于突萼果与宿萼果,且可溶性固形物含量与可溶性糖含量均优于其他两种类型的果实;宿萼果的可食率及果皮叶绿素含量显著高于突萼果与脱萼果。突萼果N元素含量显著高于脱萼果与宿萼果,脱萼果的Ca、Mg、Zn元素含量显著高于突萼果与宿萼果、P元素含量显著低于突萼果与宿萼果。从库尔勒香梨中检测到39种香气成分,以醛类最高,香气物质总含量排序为脱萼果>突萼果>宿萼果,且脱萼果中香梨的主要特征香气成分含量也显著高于突萼果与宿萼果;突萼果和脱萼果果实香气成分中以(E)-2-己烯醛含量最高,宿萼果果实中含量最高的主要香气成分为正己醛。  相似文献
