国内外药典相对密度测定法比较研究

通过对比中国兽药典、中国药典、美国药典和欧洲药典中相对密度测定法,了解四部药典的异同点,旨为中国兽药典引入新方法提供参考。分别对四部药典的收载情况、测定法原理、应用范围等进行比较和讨论。四部药典收载的测定法略有差异,中国兽药典收载的测定法在测定黏稠液体时具有一定局限性。建议中国兽药典在充分调研基础上,引入振荡型密度计测定法。     

微胶囊红磷膨胀型阻燃剂对环氧树脂的阻燃研究

研究了微胶囊红磷膨胀型阻燃剂的组成、含量对环氧树脂阻燃和力学性能的影响规律.试验结果表明:当膨胀型阻燃剂3组份(微胶囊红磷、季戊四醇和三聚氰胺)的质量比为1.46:0.34: 3.15,添加量为30%时,阻燃环氧树脂可以获得较好的阻燃性能并对力学性能影响较小.热重分析表明膨胀型阻燃剂可以显著提高环氧树脂的高温残炭量.残炭的数码照片和扫描电镜照片表明,膨胀型阻燃剂可以在环氧树脂的表面生成致密的膨胀炭层而对内部基材起到隔热、隔氧,从而提高了环氧树脂的阻燃效果.     

Prokaryotic Expression and Polyclonal Antibody Preparation of African Swine Fever Virus Georgia 2007/1 Strain CD2v Protein

The study was aimed to express the EP402R gene of African swine fever virus (ASFV) Georgia 2007/1 strain via prokaryotic expression system,obtain the recombinant CD2v protein,and prepare polyclonal antibodies against the purified recombinant CD2v protein.After codon optimization,ASFV EP402R full-length gene was linked into pET-28a(+) expression vector to construct prokaryotic recombinant expression plasmid.After induction by 1 mmol/L IPTG at 16 ℃ for 12 h,the recombinant protein was identified by SDS-PAGE and Western blotting.The purified recombinant CD2v protein was used as immunogen to prepare mouse anti-CD2v polyclonal antibodies.The antibody titer was measured by indirect ELISA and the specificity was further analyzed by indirect immunofluorescence assay (IFA) and Western blotting.The results showed that ASFV EP402R gene was successfully cloned into pET-28a(+),and pET-28a-EP402R was obtained.The recombinant plasmid was transformed into E.coli BL21(DE3) for expression,the recombinant protein was expressed mainly in the form of inclusion bodies,with molecular mass at about 47 ku,while some of the recombinant protein could also exist in a soluble form.Western blotting results showed that the purified protein had good immunoreactivity.The indirect ELISA result showed that the polyclonal antibodies had a high titer of 1:512 000,IFA and Western blotting results indicated that it could specifically recognize recombinant CD2v protein.These results confirmed the recombinant CD2v protein expressed via prokaryotic system had good immunogenicity,and the prepared polyclonal antibodies had high titer and specificity.This research provided technical support for further study of ASFV EP402R biological function,as well as its gene-deletion based vaccine development.     

辽宁水稻直立穗型基因位点SSR标记变异分析

利用与辽粳5号直立穗型基因(EP)紧密连锁的SSR标记RM5833-11、RM5686-23及RM1189分析了64份辽宁省水稻种质,分别检测到93.48%、95.65%和100%直立、半直立穗型种质,其中55.56%,66.67%和83.33%的弯曲穗型种质与辽粳5号具有相同的等位变异。这些品种(系)经系谱分析表明,有43份直立、半直立穗型和10份弯曲穗型种质与辽粳5号亲缘关系密切。因此推测辽宁地区可能是辽粳5号直立穗型基因的主要来源地。     

新疆长输管道原油流动性改进剂的应用研究

新疆东部火烧山－北三台输油管道是我国西部一条高粘易凝原油输送管道，自投产以来一直处在超低输量下运行，１９９２年的输油量只有４０万ｔ（设计最小输量６８．３万ｔ／ａ）。由于输油量低，制约了原油外运，直接影响火烧山油田的生产，为了保证输油量，需要改变原低温流变性，针对火烧山原油特性，新疆石油局与浙江大学共同研制出ＥＰ源油流动性改进剂。通过室内试验及现场不同温度，不同加剂量的工业试验，对原油的主要物性进行     

水牛实验感染肝片吸虫后IGF-Ⅰ和β内啡肽含量变化及其对免疫功能的影响

头健康阉公水牛, ５ 头经口每日感染肝片吸虫囊蚴６０ 个／ 头, 连续２０ ｄ ; ３ 头不感染作对照。于感染前和感染后每周采集颈静脉血样１次, 共２７ 次, 分别测定血浆 Ｉ Ｇ Ｆ Ⅰ、β内啡肽 （β Ｅ Ｐ） 以及 Ｗ Ｂ Ｃ 、 Ｄ Ｃ ; Ｔ、 Ｂ 淋巴细胞比例和血清抗肝片吸虫 Ｉｇ Ｇ 水平。结果 Ｉ Ｇ Ｆ Ⅰ和β Ｅ Ｐ 分别在开始感染后第５ 周和第４ 周显著升高, 并持续到第２３ 周和第９ 周; Ｉｇ Ｇ 水平也在第４ ～２２ 周显著高于对照组, 嗜酸性粒细胞和 Ｂ 淋巴细胞比例也明显升高。结果表明肝片吸虫感染后水牛免疫功能的变化可能与 Ｉ Ｇ Ｆ Ⅰ及β Ｅ Ｐ 有关。     

伪狂犬病毒EP0基因的克隆及其在大肠杆菌中的表达研究

为了研究EP0蛋白在伪狂犬病毒粒子中的定位及其功能,试验利用PCR从伪狂犬病毒基因组中扩增PRV EP0基因的编码区,克隆至pMD18-T载体,构建重组质粒pMD-EP0。经测序鉴定正确后,用EcoRⅠ和Hind Ⅲ双酶切pMD-EP0,回收EP0基因与经同样双酶切处理的pET-32a(+)进行连接,构建重组质粒pET-EP0。将pET-EP0转化大肠杆菌BL21(DE3),经IPTG诱导后,通过SDS-PAGE和Western blotting检测融合蛋白的表达情况及反应原性。结果表明,成功扩增得到EP0基因编码区序列,大小为1227 bp;重组表达质粒 pET-EP0经诱导后,EP0融合蛋白获得了表达,大小约为75 ku,主要以包涵体形式存在,且能与伪狂犬病毒阳性血清反应。     

Pharmacokinetics of grapiprant,a selective EP4 prostaglandin PGE2 receptor antagonist,after 2 mg/kg oral and i.v. administrations in cats

Drugs that provide effective analgesia in cats are limited. The aim of the study was to assess the pharmacokinetics of grapiprant after 2 mg/kg administration via p.o. and i.v. routes in cats. Six healthy adult cats were used according to an open, single‐dose, two‐treatment, two‐period, randomized cross‐over design. Cats were assigned to two treatment groups and administered with 2 mg/kg of grapiprant (pure powder) through p.o. and i.v. administration. Blood samples were collected at preassigned times and analysed by a validated HPLC method. After both administrations, grapiprant concentrations were detectable in plasma for up to 24 hr in five of six animals. The critical parameters including clearance (173.2 ml hr^?1 kg^?1, range 120–326 ml hr^?1 kg^?1) and volume of distribution (918 ml/kg, range 611–1608 ml/kg) were calculated from the i.v. group. The mean oral F% was low (39.6% range 31.5%–45.2%). If the assumption that the minimal effective concentration in dogs (164 ng/ml) applies in cats too, grapiprant orally administered at 2 mg/kg might be effective for 10 hr. Further studies are necessary to establish the minimal effective concentration in this animal species.
