病原微生物荚膜多糖的生物学功能   总被引:2,自引:1,他引:1  
荚膜多糖(capsular polysaccharide,CPS)是一种广泛存在于细菌、支原体、部分真菌等菌体表面的碳水化合物。同时,荚膜多糖有助于菌体抵抗干燥和低温等不利环境,并通过在菌体表面形成物理屏障阻碍宿主补体的杀伤与吞噬作用。在长期多种应激-压力环境下,病原菌已进化出多种免疫逃避机制并促进宿主感染;在非病原微生物中,荚膜多糖可正向调节宿主免疫作用,并拮抗机体免疫因子,保护宿主免受病原菌引起的炎症性疾病。本文将结合本团队的相关研究工作,对荚膜多糖的结构、合成调控机制、生物学功能、免疫逃避机制和致病机制,特别是荚膜多糖正向调节宿主免疫系统及其应用潜力等方面作一综述,为病原菌致病机制的研究和疫病的有效防控提供参考依据。  相似文献

吕梁山地区地形垂直差异明显,植被对气候变化反应敏感,研究吕梁山地区植被物候变化,探索植被物候变化与气候的响应关系,旨在为高海拔山区植被物候研究和生态治理提供借鉴。基于2000—2015年MODIS NDVI时间序列数据,通过动态阈值法提取吕梁山地区的植被物候,对气温、降水进行空间插值,并对植被2个关键物候期与气候因素进行偏相关分析。结果表明:(1)植被生长季开始日期(the start of the growing season,SOS)提前的区域约占85.7%,其中16.2%显著提前;植被生长季结束日期(the end of the growing season,EOS)推迟的区域约占90.6%,其中33.3%显著推迟。(2)区内74.8%、87.7%植被SOS分别与气温、降水呈负相关,气温升高或降水增加,植被SOS提前。植被SOS在高海拔山区受4月气温影响显著,而低海拔地区受4月降水影响显著。(3)区内72.6%、65.1%植被EOS分别与气温、降水呈正相关,气温升高或降水增加,植被EOS推迟。植被EOS在北部和西部地区受11月气温影响显著,而高海拔地区受9月降水影响显著。2000—2015年吕梁山地区植被物候发生显著变化,各地区对气温、降水的响应不同,研究结果可为区域物候、气候变化研究和陆地生态治理提供科学依据。  相似文献

以感官评价为指标,通过单因素试验和响应面试验设计优化朝鲜蓟发酵茶泡制工艺。结果表明,朝鲜蓟发酵茶的最佳泡制工艺为冲泡时间22 min,加水量243 mL,冲泡水温86℃,粉碎度40目,在此条件下感官评分为90分。通过ABTS法和DPPH法检测表明,朝鲜蓟袋泡茶具有明显的抗氧化能力。  相似文献

Peritrophic matrix/membrane (PM) critically prevents the midgut of insects from external invasion by microbes. The proteins in the peritrophic membrane are its major structural components. Additionally, they determine the formation and function of this membrane. However, the role of PM proteins in immune regulation is unclear. Herein, we isolated a novel PM protein (MdPM-17) from Musca domestica larvae. Further, the function of MdPM-17 in regulating host innate immunity was identified. Results showed that the cDNA of MdPM-17 full is 635 bp in length. Moreover, it consists of a 477-bp open reading frame encoding 158 amino acid residues. These amino acid residues are composed of two Chitin-binding type-2 domain (ChtBD2) and 19 amino acids as a signal peptide. Moreover, tissue distribution analysis indicates that MdPM-17 was enriched expressed in midgut, and moderate levels in the fat body, foregut, and malpighian tubule. Notably, MdPM-17 recombinant protein showed high chitin-binding capacity, thus belongs to the Class III PM protein group. MdPM-17 protein silencing via RNA interference resulted in the expression of antimicrobial peptide (defensin, cecropins, and diptericin) genes, and this occurred after oral inoculation with exogenous microbes Escherichia coli (Enterobacteriales:Enterobacteriaceae), Staphylococcus aureus (Bacillales:Staphylococcaceae), and Candida albicans (Endomycetales:Saccharomycetaceae)). Therefore, all the antimicrobial peptide (AMP) gene expression levels are high in MdPM-17-depleted larvae during microbial infection compared to controls. Consequently, these findings indicate that MdPM-17 protein is associated with the antibacterial response from the housefly.  相似文献

Tree-ring width chronologies of Larix chinensis were developed from the northern and southern slopes of the Qinling Mountains in Shaanxi Province,and climatic factors affecting the tree-ring widths of L.chinensis were examined.Correlation analysis showed that similar correlations between tree-ring width chronologies and climatic factors demonstrated that radial growth responded to climate change on both slopes.The radial growth of L.chinensis was mainly limited by temperature,especially the growing season.In contrast,both chronologies were negatively correlated with precipitation in May of the previous year and April of the current year.Spatial climate-correlation analyses with gridded land-surface climate data revealed that our tree-ring width chronologies contained a strong regional temperature signal over much of northcentral and eastern China.Spatial correlation with seasurface temperature fields highlights the influence of the Pacific Ocean,Indian Ocean,and North Atlantic Ocean.Wavelet coherence analysis indicated the existence of some decadal and interannual cycles in the two tree-ring width chronologies.This may suggest the influences of El Nin˜o-Southern Oscillation and solar activity on tree growth in the Qinling Mountains.These findings will help us understand the growth response of L.chinensis to climate change in the Qinling region,and they provide critical information for future climate reconstructions based on this species in semi-humid regions.  相似文献

谷胱甘肽(GSH)是一种普遍存在于植物中的抗氧化剂,在维持组织抗氧化防御和调节氧化还原敏感信号转导中起着关键作用。深入研究GSH在非生物胁迫中的作用,对从分子水平揭示植物GSH积累的调控机制具有重要意义。本研究从植物GSH代谢途径及其相关酶、GSH在植物应激反应中的调节、GSH参与植物激素代谢等方面进行综述,并对谷胱甘肽在植物生长发育、与其它信号通路间交互作用的研究前景进行展望,以期为植物谷胱甘肽代谢以及其在非生物胁迫方面的研究提供一定的理论参考。  相似文献

 稻曲病菌在侵染过程中会分泌大量的效应蛋白来帮助其侵染。本研究鉴定到一个稻曲病菌效应蛋白UvSix1-1,同源序列比对及系统发育分析发现其具有Six1蛋白保守的氨基酸位点,并与多个病原菌中的Six1蛋白具有同源性。进一步研究发现UvSix1-1可以抑制Bax、XEG1和INF1引起的烟草叶片细胞坏死,并且其预测的信号肽区域具有分泌功能,在烟草上瞬时表达UvSix1-1可以促进辣椒疫霉的定殖。对不同稻曲菌株中的序列进行分析,没有发现序列多态性,说明该基因非常保守。以上研究结果表明,UvSix1-1在病原菌侵染过程中发挥作用,研究结果为稻曲病菌效应蛋白的研究提供参考。  相似文献

为明确龟纹瓢虫Propylaea japonica对草地贪夜蛾Spodoptera frugiperda的控害潜力,在(25±1)℃、L∥D=16 h//8 h、RH (60±5)%的条件下研究了龟纹瓢虫雌、雄成虫和4龄幼虫对草地贪夜蛾卵、1龄和2龄幼虫的捕食能力。结果表明,龟纹瓢虫4龄幼虫和雌、雄成虫对草地贪夜蛾卵、1龄和2龄幼虫的捕食反应均符合HollingⅡ模型。龟纹瓢虫雌、雄成虫和4龄幼虫对草地贪夜蛾卵的日最大捕食量分别为204.6、110.9粒和124.7粒,瞬时攻击率分别为1.125、1.658和1.429,处理时间分别为0.004 9、0.009 0 d和0.008 0 d;对草地贪夜蛾1龄幼虫的日最大捕食量分别为242.4、265.5头和109.4头,瞬时攻击率分别为1.233、1.038和1.411,处理时间分别为0.004 1、0.003 8 d和0.009 1 d;对草地贪夜蛾2龄幼虫的日最大捕食量分别为41.2、40.3头和27.6头,瞬时攻击率分别为1.386、1.226和1.685,处理时间分别为0.024 3、0.024 8 d和0.036 2 d。雌、雄成虫和4龄幼虫对草地贪夜蛾卵和低龄幼虫的搜寻效应均随着猎物密度的增加而降低。龟纹瓢虫对草地贪夜蛾卵和1龄幼虫的捕食量高于2龄幼虫。结果表明龟纹瓢虫对草地贪夜蛾卵和低龄幼虫具有较好的控制作用,为利用龟纹瓢虫控制草地贪夜蛾提供了理论依据。  相似文献

AIM: To investigate the therapeutical effect of suramin on hypertrophic scar (HPS) and its mechanism. METHODS: After the mouse model of HPS was established by mechanical stretching, the suramin solution at low dose (5 mg/kg) and high dose (10 mg/kg) was applied onto the scar site caused by mechanical load in mice by transdermal administration once a day for 10 d. The degree of scar hyperplasia was observed by macroscopy. The scar cross-sectional area and scar elevation index in the HPS tissues were evaluated by hematoxylin-eosin (HE) staining. The expression levels of transforming growth factor-β1 (TGF-β1) and interleukin-6 (IL-6) in HPS tissues were detected by immumohistochemical staining. The expression level of α-smooth muscle actin (α-SMA) in HPS tissues was detected by immunofluorescence staining, RT-qPCR and Western blot. The levels of tumor necrosis factor-α (TNF-α), IL-6, IL-10 and TGF-β1 in the HPS tissues were measured by ELISA. RESULTS: Macroscopic observation showed that the surface areas of scar in the HPS mice after treatment with suramin at low and high doses were significantly reduced (P<0.01). HE staining results showed that the scar cross-sectional area and the scar elevation index of HPS mice after treatment with suramin at low and high doses were significantly reduced (P<0.05 or P<0.01). The results of immunofluorescence staining, RT-qPCR and Western blot showed that the number of α-SMA positive cells and the mRNA and protein expression of α-SMA in scar tissues of HPS mice after treatment with suramin at low and high doses were significantly decreased (P<0.05 or P<0.01). The results of immunohistochemical staining showed that the expression levels of TGF-β1 and IL-6 in scar tissues of HPS mice after treatment with suramin at low and high doses were significantly reduced (P<0.01). The results of ELISA showed that the levels of TNF-α, IL-6, IL-10 and TGF-β1 in the scar tissues of HPS mice after treatment with suramin at low and high doses were significantly reduced (P<0.01). CONCLUSION: Suramin inhibits the formation of HPS, which may be related to the inhibition of fibroplasia and reduction of local inflammatory response.  相似文献
