AIM To investigate the role of microRNA-142-3p (miR-142-3p) in endothelial cell apoptosis during atherosclerosis (AS) and the underlying mechanism. METHODS Human aortic endothelial cells (HAECs) were treated with oxidized low-density lipoprotein (ox-LDL). The expression level of miR-142-3p was detected by RT-qPCR. Apoptosis was determined via flow cytometry (FCM) and caspase-3 activity assay. Prediction of the binding site between miR-142-3p and 3’-UTR of Rictor mRNA was performed by bioinformatics analysis and confirmed by dual-luciferase reporter assay. RESULTS The expression of miR-142-3p was substantially up-regulated during the ox-LDL-elicited apoptosis in HAECs (P<0.05,P<0.01). Forced expression of miR-142-3p exacerbated apoptosis in HAECs whereas inhibition of miR-142-3p partly alleviated apoptotic cell death mediated by ox-LDL. Further analysis identified Rictor as a direct target gene of miR-142-3p, and Rictor knock-down abolished the anti-apoptotic effect of miR-142-3p inhibitor. Moreover, the Akt/endothelial nitric oxide synthase (eNOS) signaling pathway was found to mediate the beneficial effect of miR-142-3p inhibitor on endothelial cells apoptosis. CONCLUSION Down-regulation of miR-142-3p inhibits endothelial cell apoptosis and atherosclerotic development by up-regulating the expression of Rictor and activating the Akt/eNOS signaling pathway.  相似文献

Little effect of forest age on oribatid mites on the bark of trees   总被引:1,自引:0,他引:1  
This study investigates the effect of forest age (20-, 50-, 160-year-old, and primeval forest) on oribatid mite communities on the bark of oak trees in the National Park Pusza Białowieża in eastern Poland. We hypothesized that oribatid mite diversity on bark peaks at forests of intermediate age and that the number of parthenogenetic species of oribatid mites is highest in young stands. In contrast to these hypotheses, the diversity, density, number of juveniles, community structure and the mode of reproduction of oribatid mite species were not significantly affected by forest age. None of the oribatid mite species occurred exclusively on trees of a specific age. The results suggest that oribatid mite communities on the bark of trees are minimally affected by tree harvesting regimes. In contrast to oribatid mites in soil, communities on bark appear to be less sensitive to disturbances.  相似文献

为探究miR-142-3p在MCF-7细胞中作用机理,采用RNA免疫共沉淀技术和双荧光素酶报告基因技术筛选及验证miR-142-3p作用靶基因;蛋白质免疫印迹技术验证靶基因及其介导的PI3K-AKT-mTOR信号通路蛋白表达量及通路活性;应用实时荧光定量PCR验证靶基因PTEN对miR-142-3p调节关系。结果表明,miR-142-3p靶向调节AKT和PTEN表达;miR-142-3p过表达组中PI3K-AKT-mTOR通路活性显著降低;miR-142-3p抑制组中PI3K-AKT-mTOR通路活性显著上升;抑制PTEN表达显著提高miR-142-3p表达量。因此miR-142-3p靶向调节AKT和PTEN表达继而抑制PI3K-AKT-mTOR信号通路活性,PTEN与miR-142-3p存在调节关系。  相似文献

To determine the regulative role of miR-142-3p in lactation of dairy goat mammary epithelia cells (DGMECs),we selected the DGMECs in lactating dairy goat for material,used liposomal transfection techniques to silence miR-142-3p expression,and used Real-time quantitative PCR,Western blotting and Assay Kits to explore the changes of lactation after miR-142-3p inhibition.The results showed that when miR-142-3p was inhibited,the cell proliferation ability of DGMECs was increased,the secretion amount of β-casein and triglyceride were up-regulated.So we concluded that miR-142-3p could influence the lactation function of DGMECs,such as cell proliferation,β-casein and triglyceride secretion,by inhibiting the function of its target genes.  相似文献

MicroRNAs (miRNAs) are a group of small,non-coding RNA molecules about 22 nucleotides to regulate a wide variety of important biological processes,including cell proliferation,differentiation,apoptosis as well as the progression of tumors.Liposome transfection was used to detect the expression of miR-142-3p in human mammary epithelial cells,the effects of miR-142-3p on the cell proliferation,apoptosis and milk protein synthesis were detected by Real-time PCR,Western blotting,cell proliferation analysis.The results indicated that after miR-142-3p being silenced,prolactin receptor (PRLR) protein was increased,at the same time the expressions of related pathways protein AKT,mTOR,STAT5 and cyclinD1 were increased,the ability of cell proliferation was increased.The results suggested that in human mammary epithelial cells,the silence of miR-142-3p could increase the expression of PRLR protein,miR-142-3p could promote the synthesis of milk protein and increase the proliferation of mammary epithelial cells by regulating related pathways proteins AKT,mTOR,STAT5 and cyclinD1.  相似文献

为了建立柞蚕核型多角体病毒(Antheraea pernyi nucleopolyhedrovirus,ApNPV)的早期检测技术,采用NCBI提供的BLAST软件在线检索ApNPV特异基因ORF142的序列后设计引物AP1,对ApNPV基因组DNA进行PCR扩增,得到约130 bp的片段,最低检测量为0.5 fg/mL病毒DNA。分别以感染ApNPV的柞蚕幼虫总DNA和ApNPV多角体悬液为模板,相同扩增条件下可扩增出大小一致的一条特异性片段,最低检测量分别为1 fg/mL幼虫总DNA和20~25 PIBs/mL ApNPV多角体。以柞蚕4龄幼虫为材料,人工模拟感染ApNPV后取幼虫血淋巴为检测物可以直接检出ApNPV,当添毒量为2.3×108PIBs/mL时,添毒36 h后即可检出,且病毒感染后的检出时间与添毒浓度呈正相关。采用该方法的检测过程只需3 h,快速而方便。  相似文献

【目的】探索miR-142-3p对奶山羊化学诱导乳腺上皮细胞(chemical induced mammary epithelial cells, CiMECs)体外泌乳功能的调节作用,旨在为促进转基因乳腺生物反应器的发展以及“培养皿奶”的商业化生产奠定基础和提供新的思路。【方法】选取奶山羊耳缘成纤维细胞(GEFs)为研究材料,经单一小分子化合物TGFβR-1/ALK5的抑制剂(RepSox)诱导8 d后,分别从细胞的形态变化、特异性标记物免疫荧光染色以及油红O染色对其进行鉴定。之后运用脂质体转染技术在诱导成功的奶山羊CiMECs中分别转染miR-142-3p的抑制物(miR-142-3p inhibitor)及抑制物对照(inhibitor-NC),不转染的细胞作为空白对照(BC),转染24 h后,采用实时荧光定量PCR检测miR-142-3p的表达水平,CCK-8法检测细胞的增殖率,Western blotting检测β-酪蛋白(β-casein)的表达量,甘油三酯酶法测定甘油三酯的含量。【结果】GEFs诱导8 d后形成岛屿状多核仁样的上皮样聚集;免疫荧光染色结果显示,诱导后的GE...  相似文献

闽糖92-142与闽糖92-505系福建省农业科学院甘蔗研究所分别从杂交组合桂糖57-624×湛蔗74-141、选15×CP73-1574中选育而成的甘蔗新品种。2005年中试结果表明,闽糖92-142公顷蔗茎产量132.228t、11~1月蔗糖分为14.92%,公顷含糖量为19.725t,比福建省主栽品种闽糖70-611增产22.47%、蔗糖分提高0.29个百分点、含糖量提高25.41%,比新台糖10号增产32.60%、含糖量提高31.66%。该品种具丰产高糖、抗逆性强等特点。闽糖92-505平均蔗茎产量125.841t/hm2,11~1月份平均蔗糖分达15.93%,含糖量为20.049t/hm2,分别比闽糖70-611增产16.55%,蔗糖分提高1.30个百分点、增糖27.47%;比新台糖10号增产26.19%、蔗糖分提高0.91个百分点、增糖33.82%,是一个具高产高糖、中早熟、适应性广的甘蔗新品种。经t测验,闽糖92-142与闽糖92-505的蔗茎产量与两对照相比,增产均达显著水平。  相似文献

MicroRNAs(miRNAs)是一类长度约为22 nt的非编码的调控性小RNA,在诸多生命活动中发挥重要作用,如参与调控细胞的增殖、分化、凋亡及肿瘤的发生发展。本试验应用脂质体转染技术抑制miR-142-3p在人乳腺上皮细胞的表达。试验采用实时荧光定量PCR、Western blotting、细胞增殖分析等技术,探索miR-142-3p对人乳腺上皮细胞增殖及乳蛋白合成的影响。结果显示,miR-142-3p沉默后,催乳素受体(prolactin receptor,PRLR)蛋白表达增强,同时,相关通路蛋白AKT、mTOR、STAT5、cyclinD1表达量均增加,细胞增殖能力增强。结果表明,在人乳腺上皮细胞中,miR-142-3p的沉默使PRLR蛋白表达量升高,通过调控AKT、mTOR、STAT5、cyclinD1相关通路蛋白而促进乳蛋白质的合成和乳腺上皮细胞的增殖。  相似文献

为确定miR-142-3p对奶山羊乳腺上皮细胞泌乳功能的调节作用,试验选取泌乳期奶山羊乳腺上皮细胞为研究材料,利用脂质体转染技术抑制miR-142-3p的表达,采用实时荧光定量PCR、Western blotting、试剂盒等检测miR-142-3p基因沉寂后其对奶山羊乳腺上皮细胞泌乳功能的影响。结果显示,miR-142-3p基因沉寂后,奶山羊乳腺上皮细胞增殖能力增强,β-酪蛋白及甘油三酯分泌增加。由此可知,miR-142-3p通过抑制靶基因作用,影响奶山羊乳腺上皮细胞增殖、分泌β-酪蛋白及甘油三酯等泌乳功能。  相似文献
