以哈密瓜汁中的大肠杆菌为研究对象,采用超临界CO_2技术(压力10、20、35 MPa,温度35、45、55℃),以温度、压力、保压时间为自变量,以大肠杆菌存活对数为因变量,对哈密瓜汁中大肠杆菌进行超临界CO_2(SCCO_2)杀菌效果的研究。采用一级反应动力学模型分析超临界CO_2对大肠杆菌的杀菌动力学;采用透射电镜观察超临界CO_2对大肠杆菌超微结构的影响。试验结果表明,在35~55℃、10~35 MPa的条件下对哈密瓜汁中大肠杆菌进行超临界二氧化碳处理,灭菌速率分为先快后慢两个阶段;随温度和压力的提高,大肠杆菌的灭菌速率均逐步上升;经超临界二氧化碳处理后,大肠杆菌细胞出现明显的缺陷,部分细胞壁消失。  相似文献

为了研究不同种子处理对种子传播病毒的灭活效果,试验以番茄、辣椒、甜瓜、南瓜种子样本中被番茄花叶病毒(ToMV)、烟草花叶病毒(TMV)、黄瓜花叶瓜病毒(CMV)感染的种子为研究对象,经过酶联免疫吸附测定(ELISA)和逆转录聚合酶链反应(RT-PCR)测试选择出研究材料,采用7种病毒失活处理方法,分别为醋酸、过氧化氢、盐酸、次氯酸钠浸种,干热处理,温汤浸种和臭氧熏蒸处理。结果发现,能有效减少病毒浓度的处理为盐酸浸种、干热处理、温汤浸种(65℃)和臭氧(10 g/m3)处理,分别能将种子传播病毒的浓度平均降低51%、50%、42%和32%。其他灭活方法治疗效果较差,病毒浓度平均降低了12%~27%。盐酸浸种和干热处理是最有效的方法,对种子出芽率和活力没有影响。  相似文献

Piscine orthoreovirus infects various salmonid fish species, and the infection is associated with diseases such as heart and skeletal muscle inflammation (HSMI) in farmed Atlantic salmon (Salmo salar). There are no vaccines available or genetically selected resistant hosts that can efficiently control piscine orthoreovirus (PRV) infection. Currently, the only prophylactic measure against PRV is general biosecurity measures aiming to break the transmission cycle. Methods to eradicate infectious virus from contaminated facilities are desirable, but the knowledge on how to inactivate PRV is lacking. A major bottleneck for inactivation studies is the lack of ability to propagate PRV in cell culture. Therefore, in this study we developed an in vivo model for detection of infectious PRV particles after treatment of the virus with inactivation tools such as heat, pH, iodine, UV and commercially available disinfectants. The results show that standard iodine treatment is efficient in inactivation of the virus, and similarly are high and low pH extremes and treatment with Virocid, a commercially available disinfectant. A UV dose of at least 50 mJ/cm2 is required for inactivation, and the virus has high resistance against heat treatment.  相似文献

高压脉冲电场对石榴汁杀菌的研究   总被引:8,自引:3,他引:8  
该文研究了高压脉冲电场(PEF)对石榴汁的杀菌效果和对石榴汁的理化性质的影响。结果表明,电场强度和处理时间是影响PEF杀菌效果的主要因素。在频率为100 Hz时,当电场强度达到35 kV/cm,处理时间达到200 μs时,石榴汁中接种的大肠杆菌、酵母菌和霉菌的数量分别下降3.62、2.34、0.74个lg值,处理过程中样品温度≤18℃,霉菌对PEF的抵抗力最强。当处理频率增加为333 Hz,电场强度为35 kV/cm,处理时间为200 μs时,霉菌数量的下降增加到1.82个lg值,此时处理温度(处理腔内温度)提高到50℃。经PEF处理的石榴汁的理化性质基本不发生改变。  相似文献

Abstract

Results of a field experiment, comprising elemental S and three commercial preparations of Fe, applied with and without several other factors showed that the application of S was significantly better than that of Fe supplying materials in preventing the incipient chlorosis of corn leaves on the alkaline calcareous soils. Prevention of chlorosis in corn due to sulphur application increased the grain yield of corn 25–31%.

Leaf samples from plants raised with Fe supplying materials were chlorotic even though they contained significantly higher amounts of Fe. Persistence of chlorosis in spite of high Fe content of leaves, vis a vis freedom from chlorosis in spite of low Fe content, suggest that deficiency of Fe was not a factor in chlorosis. Freedom from chlorosis accompanied by significant increases in corn yields due to S application, on the contrary, show that it was the lack of S rather than Fe as the cause of Chlorosis.

When the supply of S is low, Fe seems to be subjected to a great deal of chemical inactivation and under conditions of stress more and more absorption of Fe is necessitated. In all probability the rate of inactivation exceeds the rate of absorption and chlorosis develops. In this situation either the plant's ability to utilize iron is affected or the physiological availability of absorbed iron is very low. Increased S seems to arrest the process of Fe inactivation by providing a better nutritional environment.  相似文献

Postharvest diseases cause considerable losses to harvested fruits and vegetables worldwide. In addition to various treatments to control postharvest losses caused by pathogens, the global trend is shifting toward more ecofriendly alternatives safer to human health. Thus far, the main approach to postharvest disease control has mostly relied on the use of chemicals. However, the intense use of chemical fungicides has caused side effects such as environmental issues and evolution of fungicide‐resistant isolates. With increasing demand for the use of non‐chemicals, ionizing radiation has been investigated for application in many aspects of postharvest treatment, especially as an approach with significant potential for the control of postharvest disease. Recently, a number of more or less technologically advanced methodologies, e.g. irradiation combined with other types of treatments and induced disease‐resistance, have been developed to control postharvest diseases as well as to increase the quality and storage life of fresh commodities. Challenges for future application of irradiation in fresh produce are the cost, lack of irradiation facilities, lack of knowledge about optimal conditions for different commodities, and lack of acceptance of irradiated fresh produce. This review aims to document the advances in understanding of the effects of irradiation on postharvest disease, the possible modes of action, and the perspectives in commercial use.  相似文献

为探索从水母毒素中获取抗植物病毒物质的应用前景,采用半叶枯斑法、整株法和漂浮叶圆片法,对白色霞水母Cyanea nozakii Kishinouye刺细胞毒素的抗烟草花叶病毒(Tobacco mosaic virus,TMV)活性进行了检测。结果表明,白色霞水母刺细胞毒素对TMV具有很强的直接钝化作用,枯斑抑制率随钝化时间的延长而增大。0.99 mg/m L毒素体外钝化TMV 30 min,枯斑抑制率达98.85%,将TMV与0.80 mg/m L毒素体外混合后立即接种,枯斑抑制率仍达89.57%;20~70℃之间,毒素对TMV的钝化作用随温度升高而降低,但又表现出一定的高温耐受性,70℃下处理30min,1.33 mg/m L毒素对枯斑的抑制率为61.73%。毒素对病毒初侵染有一定的预防作用,并可抑制TMV的增殖;1.93 mg/m L毒素处理接种TMV的叶圆片2 d后,对TMV的增殖抑制率为49.41%;但该毒素对TMV所致病害的治疗效果不明显。该毒素还具有较强的蛋白水解酶活性,可能与其抗TMV活性相关。表明白色霞水母刺细胞毒素抗TMV作用明显,其抗植物病毒活性值得深入研究。  相似文献

This study investigated the use of alkaline hydrolysis at ambient temperature for inactivation of selected fish pathogens in fish tissues under conditions approximating those that are likely to be found in the aquaculture industry. Infectious salmon anaemia virus (ISAV) and Lactococcus garvieae have been determined in a previous study to be the most resistant virus and bacteria to pH 12 from a wide range of viruses and bacteria tested. They were spiked at high titres into fish extracts that were then treated with 1 m sodium hydroxide (NaOH). Viable L. garvieae was not detected in the treated fish extract after 1 h, and ISAV was not detected after 24‐h exposure. Field mortalities of Atlantic salmon, Salmo salar L., caused by infectious pancreatic necrosis virus were treated by alkaline hydrolysis at ambient temperature. The macerated fish mortalities contained a high titre of virus (3.38 × 108 TCID50 g?1) that was reduced to approximately 2.2 × 103 TCID50 g?1 after 24‐h exposure to NaOH, and virus was not detected after exposure for 48 h. The results suggest that alkaline hydrolysis at ambient temperature has potential as a biosecure treatment method for fish by‐products containing fish pathogens.  相似文献

This study evaluated the effects of sodium chlorite (SC) alone and its sequential treatment with edible coatings on browning inhibition and quality maintenance of fresh-cut d’Anjou pears. Edible coatings were prepared from chitosan (CH) and its water-soluble derivative carboxymethyl chitosan (CMCH), separately. Pear wedges were immersed in SC solution, followed by coating with CH or CMCH solutions. The samples were packed in unsealed bags and stored at 4 °C for subsequent color, firmness, and weight loss measurement. The effects of the SC and coating treatments on polyphenol oxidase (PPO) inhibition and microbial inactivation were also evaluated. Results indicated that SC exhibited significant (P < 0.05) inhibition of browning and PPO activity. The SC treatment was also strongly effective in inactivating Escherichia coli O157:H7 on pear slices. Coating SC-treated pear slices with CH adversely affected the quality of pear slices by accelerating the discoloration of cut surfaces and increasing the PPO activity. On the contrary, coating SC-treated samples with CMCH significantly prevented the browning reaction and inhibited PPO activity. In addition, SC and CH/CMCH coatings maintained tissue firmness and did not affect weight loss. Our study may provide a scientific basis for the use of SC + CMCH treatment as an alternative preservation treatment for fresh-cut fruits and vegetables.  相似文献
