Improvement of an enzyme-linked immunosorbent assay for equine herpesvirus type 4 by using a synthetic-peptide 24-mer repeat sequence of glycoprotein G as an antigen

To increase the sensitivity of an enzyme-linked immunosorbent assay (ELISA) for equine herpesvirus type 4 (EHV-4) that uses a 12-mer peptide of glycoprotein G (gG4-12-mer: MKNNPIYSEGSL) [4], we used a longer peptide consisting of a 24-mer repeat sequence (gG4-24-mer: MKNNPIYSEGSLMLNVQHDDSIHT) as an antigen. Sera of horses experimentally infected with EHV-4 reacted much more strongly to the gG4-24-mer peptide than to the gG4-12-mer peptide. We used peptide ELISAs to test paired sera from horses naturally infected with EHV-4 (n=40). gG4-24-mer ELISA detected 37 positive samples (92.5%), whereas gG4-12-mer ELISA detected only 28 (70.0%). gG4-24-mer ELISA was much more sensitive than gG4-12-mer ELISA.     

THE REIMPROVEMENT FOR IMPULSE OF G. I. TAYLOR THEORY OF PLASTIC DEFORMATION IN IMPACT OF CYL1NDICAL PROJECTILES

In this paper the reimprovement for G. I. Taylor theory of plastic deformation in impact of cylindical projectiles is done. The impulse in the reimproved theory is G. I. Taylor' s 1/2, and 3/4 of G. I. Taylor' s improved theory. The analytical solution of the reimproved theory shows that it gives better agreement with exprimental results than those of the original Taylor' s theory and, the improved theory.     

Caterpillar G3412型燃气发电机排障实践

介绍了Caterpillar G3412型燃气发电机的基本工作原理,分析了涩宁兰输气管道涩北首站该型燃气发电机拉缸的故障情况。通过更换2号缸的缸桶、活塞、气环和油环,以及高压线圈、O形圈、火花塞、水湿传感器等附件,使发电机故障得以排除,同时提出了发电机组日常运行的维护方法。     

基于3G通信与GPS技术的农机视频监控系统研究

针对现代化大农机发展的需求,研究了如何将3G通信技术与GPS定位技术相结合,建立农机视频监控系统,用于现代化大农机管理。采用WebGIS技术为基础,以B/S结构进行开发建设,为农机综合管理监控提供了新思路、新手段。     

Effect of pyrethroids on carbohydrate metabolic pathways in common carp,Cyprinus carpio

The activity levels of succinate dehydrogenase (SDH) and glucose‐6‐phosphate dehydrogenase (G6PD) were assessed in various tissues of Cyprinus carpio var communis which had been exposed to lethal concentrations of group‐II pyrethroids (deltamethrin, cypermethrin, fenvalerate and fluvalinate) for a period of 72 h. The results indicated a steady decrease in SDH activity with a concomitant increase in G6PD activity. The decreased SDH activity indicated inhibition of SDH at mitochondrial level and the increased G6PD activity an enhancement of an alternative pathway of carbohydrate metabolism, viz the hexose monophosphate shunt (HMP) or pentose phosphate pathway as a biochemical adaptation to overcome the toxic stress. © 2001 Society of Chemical Industry     

青海湖裸鲤和鲤鱼胰脏和肠管淀粉酶活性的研究

采用碘———淀粉比色法对青海湖裸鲤和鲤鱼胰脏及肠管淀粉酶的活性进行了定量分析。结果表明:青海湖裸鲤胰脏及肠管淀粉酶活性随体重的增加呈现下降趋势(P>0.05);青海湖裸鲤后肠淀粉酶活性明显高于前肠淀粉酶活性(P<0.01);青海湖裸鲤胰脏及肠道的淀粉酶活性均高于鲤鱼的淀粉酶活性(P>0.05)。     

Modifying Myxococcus xanthus protoporphyrinogen oxidase to plant codon usage and high level of oxyfluorfen resistance in transgenic rice

Protoporphyrinogen oxidase (Protox) of Myxococcus xanthus (Mx Protox) is a 49-kDa membrane protein that catalyzes conversion of protoporphyrinogen IX (Protogen IX) into protoporphyrin IX (Proto IX). Upon heterologous expression in transgenic rice plants, Mx Protox is dually targeted into plastids and mitochondria, increasing resistance against the herbicidal Protox inhibitor oxyfluorfen. Here, we describe the chemical synthesis of the Mx Protox gene by assembling several small synthetic DNA fragments derived by ligation-PCR. Codon usage in the resulting 1416-bp gene was modified to correspond to that of the Arabidopsis Protox gene, a change that resulted in a decrease in G+C content from 71 to 49%. The modified Mx Protox gene was used to generate transgenic rice plants via Agrobacterium-mediated transformation. Integration, expression, and inheritance of the transgenes were demonstrated by Southern, Northern, and Western blot analyses. In plants transformed with the modified, low G+C-content Mx Protox gene, levels of Protox expression and enzyme activity were low compared to the levels observed for plants transformed with the native Mx Protox gene. Nonetheless, like the native gene, the modified gene conferred a high level of resistance to the herbicide oxyfluorfen in a seedling growth test.     

水稻三系不育系G98A的选育

挖掘利用地方稻种资源,利用贵州高原粳稻与矮秆籼稻杂交改良,经过2次杂交(纳雍大红谷/南京11//8902S),多代单株选择,育成耐寒保持材料95F076,用其与野败胞质不育系珍汕97A杂交,再连续8代回交,育成新不育系G98A.     

翘嘴红肝脏G6Pase催化亚基的克隆以及摄食和饲料中碳水化合物对其表达的影响

采用RT-PCR和RACE法分离、克隆了翘嘴红G6Pase催化亚基基因全长cDNA，共1900 bp ［不含poly(A)］, 包括49 bp 5′非翻译区，1068 bp阅读框以及含Poly(A)信号AATAAA的778 bp 3′非翻译区［不包括Poly(A)］。阅读框共编码355个氨基酸，分子量为39.89 ku。序列比对分析表明翘嘴红G6Pase与斑马鱼G6Pase的相似性高达95%，与鼠、狗、人G6Pase的相似性为63%，和光滑爪蟾、金头鲷、河豚等G6Pase的相似性分别为69%、55%、76%，并具有G6Pase特有的3个保守基序。为了研究摄食以及饲料中碳水化合物对G6Pase的影响，使用实时定量RT-PCR分别测定了饲喂等能但不含碳水化合物和含23.98%碳水化合物的饲料的翘嘴红肝脏G6Pase基因的表达水平，在使用上述饲料饲喂8周后，禁食48 h, 然后测定禁食和摄食后3、6、12、24 h G6Pase mRNA的表达量，结果显示摄食后12 h，两组G6Pase的表达明显增加，说明摄食影响G6Pase基因的表达，禁食和摄食后3～12 h，含糖组G6Pase基因的表达量为无糖组的2～4倍，这表明碳水化合物可以诱导G6Pase mRNA的表达。图5参21 关键词：快速扩增cDNA末端；葡萄糖6磷酸酶；碳水化合物；翘嘴红；实时定量PCR E-mail:gexp@ffrc.cn
