为了进一步研究甜菜谷胱甘肽转移酶BvGSTU9 (LOC104894060)在重金属胁迫过程中的功能。本研究以‘780016B/12优’为实验材料,对该基因序列特征、结构、功能进行预测分析,并利用qPCR检测该基因在不同浓度镉胁迫下的表达量变化。结果显示甜菜BvGSTU9基因全长925 bp,开放阅读框675 bp,编码了由224个氨基酸组成的不稳定膜外蛋白。BvGSTU9与菠菜、藜麦的氨基酸序列相似性较高,与系统发育进化树分析结果基本相符。二级和三级结构预测表明该基因主要由α-螺旋、β-折叠、延伸链及无规则卷曲组成。qPCR显示BvGSTU9基因在不同浓度的镉胁迫下均受到不同程度的诱导,因此可以推断甜菜BvGSTU9基因无论从结构还是功能上,与镉逆境胁迫存在着一定的应答关系。研究结果也为甜菜耐重金属镉机制研究提供参考依据。  相似文献

以大豆质核互作雄性不育系NJCMS1A及其保持系NJCMS1B的花芽为试验材料,利用试剂盒法和紫外分光光度计测定分析了糖分含量(可溶性糖和淀粉)和总ATPase、蔗糖磷酸合成酶(SPS)、磷酸烯醇式丙酮酸羧激酶(PEPCK)、抗坏血酸氧化酶(AO)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)以及过氧化物酶(POD)等酶活性的变化情况。结果表明:相较于保持系NJCMS1B,总ATPase、PEPCK、AO和CAT活性在不育系NJCMS1A中显著下降;糖分含量、SPS和SOD活性在不育系NJCMS1A中下降水平不显著;POD活性在不育系NJCMS1A中显著上升。根据结果分析推测,与能量代谢或胁迫响应等相关的物质或酶活性在不育系NJCMS1A中的亏损现象可能是NJCMS1A花粉败育的主要原因。  相似文献

为了了解fad基因在胡麻蒴果发育过程中对不饱和脂肪酸的调控,对高、中、低三个不同亚麻酸(C18:3)含量的胡麻品种(’CDC Gold’,‘内亚7号’,’Linola’)进行了不同时期的品质测定,以及脂肪酸去饱和酶2a基因(fad2a)、脂肪酸去饱和酶2b基因(fad2b)、脂肪酸去饱和酶2c基因(fad2c)、脂肪酸去饱和酶3a基因(fad3a)、脂肪酸去饱和酶3b基因(fad3b)的qRT-PCR定量分析。结果表明,随着蒴果成熟,可溶性糖含量呈降低趋势,粗脂肪与粗蛋白不断积累,且差异显著(p<0.05)。fad2a基因、fad3a基因以及fad3b基因在各个时期中的表达符合正态分布。以0 d的蒴果为对照,在胡麻种子形成过程中,‘内亚7号’的三个基因在5 d和15 d的表达量迅速增加,到30 d时急剧减少,15 d的fad2a基因表达量为5.23倍,fad3a基因表达量是fad2a基因表达量的14.52倍,fad3b基因的表达量是fad2a基因表达量的16.14倍,表明这三个基因参与不饱和脂肪酸积累过程。在低亚麻酸含量品种‘Linola’30 d中,fad3a基因是fad2a基因表达量的52.71倍,fad3b呈下调趋势;在高亚麻酸含量品种‘CDCGold’30d中,fad3a基因与fad2a基因的表达量均呈下调趋势,fad3b的表达量为3.92倍;在中等亚麻酸含量品种‘内亚7号’中,fad2a基因表达量降低了0.31倍,fad3a基因是fad3b基因表达量的1.87倍。fad2b基因、fad2c基因熔解曲线不稳定,峰值低,可以在后续试验中继续探索。  相似文献

对异硫氰酸丙烯酯毒杀松材线虫作用机制进行了初步研究。测定了其对线虫虫体形态、运动行为、体内总糖和蛋白质含量的影响,以及处理前后线虫体内过氧化氢酶(CAT)、乙酰胆碱酯酶(AChE)和谷胱甘肽-S-转移酶(GST)活性变化。结果表明:经异硫氰酸丙烯酯处理后的松材线虫虫体形态严重扭曲,运动形态出现由正常的S形变化为多节弯曲和螺旋状卷曲等异常状况,线虫行动随时间的推移变得迟缓,最终呈C形、新月形或直接死亡。随着异硫氰酸丙烯酯处理质量浓度的增大,线虫体内总糖和蛋白质含量显著上升,糖和蛋白质的代谢发生紊乱;线虫体内CAT、AChE和GST活性显著降低,且均低于对照组,表明线虫体内氧化和抗氧化作用失衡;细胞受到极大的影响,神经系统遭到破坏,解毒能力明显降低。研究结果可为研究利用异硫氰酸丙烯酯作为植物源杀线虫剂提供依据。  相似文献

Root development of sugar beet plants on a sandy loess site with regard to nitrogen nutrition.
Root development of sugar beet plants in a sandy loess soil (Haplic Phaeozem) was observed from the early seedling stage up to harvest by measuring at first the greatest vertical and lateral extension of the root systems of single plants and later the rooting density of the whole plant stands (auger method, profile wall method).
During the seedling stage not only the subsoil, but also large parts of the topsoil between the plants remained unoccupied by the root systems. In this phase the greatest lateral extension of single roots reaches nearly the length of the greatest leaf of the plant. With the closure of the canopy the rooting density in the topsoil accounts to 1–2 cm cm−3.
In summer roots penetrate to a depth of 100–150 cm with rooting densities of 0.1 to 1 cm - cm−3. Thus, the plants gain not only access to water reserves, but sometimes meet remarkable amounts of nitrate which under the relatively dry conditions of the region tends to accumulate in 60–120 cm depth and – when taken up by the beet plants in the late stage of growth – affects crop quality negatively.  相似文献

A field experiment was conducted to find out the critical physiological stages of irrigation schedules inducing better growth, physiological efficiency and seed yield potential of berseem ( Trifolium alexandrinum L., Var. S-99-1). For this purpose eight irrigation treatments were made comprised of four treatments of three irrigation (W1, W2, W3 and W4), three treatments of four irrigation (W5, W7 and W8) and one treatment of five irrigation (W6) at various physiological stages i. e. regeneration, flower initiation, full bloom, seed initiation and advance seed development stage.
Thus based on the experimental results the physiological role of watering in berseem seed production could be discussed as:
With-holding of irrigation either at regeneration or at full bloom stage developed potential water stress in plants as indicated by high proline content of irrigation treatments — W4, W1 and W7; and further brought out disturbance on the formation of carotene, synthesis of water soluble sugar and translocation of sugar towards reproductive organs during grain development stage. These stresses adversely affected the plant growth and flowering behaviour. The irrigation at seed initiation stage increased the seed yield. Continuous irrigation did not appear to be useful. Thus it can be concluded that irrigation at three critical physiological stages i. e. regeneration, full bloom and seed initiation was found to be essential for obtaining potential seed yield of berseem.  相似文献

用剩余初乳代替全乳培育犊牛是近年来奶牛业中的新课题之一。本文通过1986—1987年的犊牛饲养试验,对犊牛的饲料采食量、生长发育、成活率及经济效益等方面进行了比较系统的研究。研究结果表明,用发酵初乳培育的犊牛与用常乳培育的犊牛相比:(1)耗奶少培育成本低,每头犊牛节约鲜奶415公斤,节省饲料费233.15元。(2)生长发育良好,尤其早期补喂犊牛料,促进了瘤胃的提早发育。(3)抵抗力强、发病率低,成活率高。(4)经济社会效益显著。  相似文献

A field experiment was set up in 1988 to study the development of rhizomania disease of sugar beet at different inoculum levels of beet necrotic yellow vein virus (BNYVV) in soil. Five, tenfold different, inoculum levels were created by addition of the approximate amounts of 0, 0.5, 5, 50 and 500 kg infested soil per ha (the latter corresponding to 0.01% v/v calculated to the tillage layer). A drip irrigation treatment was applied to study the influence of soil moisture on disease. Susceptible sugar beet, cv. Regina, was grown for three consecutive years.In the first year, root symptoms were not observed, but BNYVV-infected plants were detected by ELISA in low numbers at all inoculum levels at harvest. After late drilling in 1989, high numbers of infected plants, up to 90–100% in plots with the highest inoculum level, were detected already in June. Root symptoms were also observed from June onwards. In both these years disease incidence increased in time and was significantly influenced by the initial inoculum level. In the third year, the whole field was heavily diseased, and only for the non-irrigated plots incidence differed for different initial inoculum levels. The expression of symptoms by BNYVV-infected plants was influenced by initial inoculum level, thus by the amount and timing of primary infection.Root weight at harvest was not affected, but sugar content decreased with increasing inoculum level already in 1988, leading to a reduction in sugar yield of 10% at the highest inoculum level. In 1989, both root weight and sugar content decreased progressively with increasing inoculum level, resulting in sugar yield reductions of 11–66% (down to approximately 3000 kg ha–1) for low to high inoculum levels compared to the control. As the control plots became contaminated, all yields were low in 1990, still showing a decrease with increasing inoculum level in the non-irrigated plots, but an overall mean sugar yield of 3323 kg ha–1 for the irrigated ones.Sodium and -amino nitrogen content of the root, additional quality parameters determining extractability of sucrose, showed an increase and decrease, respectively, with increasing initial inoculum level already in the first year. The relative differences in contents compared to those from the control were largest for Na content. A significant negative correlation was found between Na (mmol kg–1 root) and sugar content (% of fresh weight); linear for 1988, exponential for 1989 and 1990.In spring 1989, the infestation of individual plots was assessed using a quantitative bioassay estimating most probable numbers (MPNs) of infective units of BNYVV per 100 g dry soil. The relationship between the MPns determined and root weight, sugar content and sugar yield at harvest could be described by Gompertz curves. The increase in disease incidence with increasing MPN in 1989 was adequately fitted with a logistic equation.  相似文献

以6年生曙光油桃为试材,研究40℃、45℃、50℃3个梯度高温短时间处理对花芽和叶芽的存活率、萌芽级数以及淀粉和可溶性糖含量的影响,探讨短时间高温处理对桃芽自然休眠的调控效应。结果表明:随短时间高温处理时期的推迟,处理温度的升高,处理持续时间的延长,短时间高温处理对桃芽自然休眠的解除作用增强。11月30日高温处理中,40℃高温处理对桃芽自然休眠解除呈负调控效应,其萌芽级数和可溶性糖含量均低于对照,而淀粉含量高于对照;45℃和50℃高温处理对桃芽自然休眠解除呈正调控效应,其萌芽级数和可溶性糖含量与对照相比明显升高,而淀粉含量显著降低,虽然部分花芽和叶芽因高温处理而死亡,但是在存活芽中短时间高温对桃芽自然休眠的解除作用明显增强。12月10日高温处理中,40℃高温处理对桃芽自然休眠解除的调控效应不明显,45℃和50℃高温处理和11月30日处理相同,具有正调控效应,只是前者对桃芽自然休眠的解除效果优于后者。不同高温处理桃芽的萌芽级数和淀粉含量及可溶性糖含量之间的相关性分析表明,淀粉含量的降低,可溶性糖含量的升高即淀粉向可溶性糖的快速转变是高温解除休眠的部分原因。  相似文献
