This study determined the effect of triploidy on the survival, growth and gonadal development of turbot from 6 to 48 months of age. From 6 to 24 months of age (first sexual maturity), survival was similar in both ploidies (P > 0.05). From 24 to 48 months of age, after the first sexual maturity, survival was 91.9% in diploids and 100% in triploids, which did not exhibit the post-spawning-associated mortality. Growth was similar for both ploidies during the first year of life. After that, triploids grew significantly (P < 0.05) more that diploids, with more marked differences after each spawning season. From 24 to 48 months, the average weight difference between both ploidies was 11.4 ± 1.9%, ranging from 4.3 to 23.0%. At 47 months of age, the biomass of triploids was 10.3% greater in total weight and 14.3% greater in eviscerated weight. Gonads of triploid males were similar to that of diploids, whereas in triploid females, they were significantly smaller and rudimentary. A histological analysis carried out at 47 months of age showed complete sterility of triploids in both sexes. Sex ratio was 1 male (M):0.6 female (F), for diploids, significantly (P < 0.05) different from 1:1, and 1 M:3.3 F for triploids, significantly (P < 0.05) different from 1:1 and from the diploids. Since females grow more than males, culture of triploids benefited from the high female ratio, which helped to reduce size dispersion. In addition, their sterility allowed better performance by avoiding the reduction in growth that takes place during the spawning periods. Together, these observations indicate that triploidy induction can be an interesting option for turbot aquaculture, especially for the production of large-size fish of more than 2 years of age.  相似文献

Before transfer to larval incubators, water was membrane filtered to remove >95% of the bacteria and then transiently maintained in a biofilter that promoted recolonization of the water by non-opportunistic bacteria. The process is termed microbial maturation of the water. Hypothetically the bacterial flora in the matured water should protect the marine larvae from colonization and proliferation by opportunistic bacteria. Testing of the hypothesis demonstrated 76% higher survival of yolk sac larvae of Atlantic halibut (Hippoglossus hippoglossus) in matured than in membrane filtered water. Proliferation of opportunistic bacteria was observed in the rearing water after hatching of turbot eggs (Scophthalmus maximus), but to a less extent in the microbially matured water. In the early phase of first feeding of turbot larvae, the matured water induced qualitative differences in the gut microflora. Significantly higher initial growth rate of the turbot larvae in the matured water affected 51% higher average weight of 13 days old larvae than in membrane filtered water. Algal addition to the matured water enhanced the larval growth further. The experiments conducted supported the proposed hypothesis that microbial maturation selects for non-opportunistic bacteria, which protects the marine larvae from proliferation of detrimental opportunistic bacteria.  相似文献

取0—63日龄的大菱鲆(Sxophthalmus maximus L.)进行固定并显微组织切片观察。根据观察结果特大菱鲆鳔器官发育周期分为形成、充气、退化和消失4个阶段。在水温15—16℃下,初孵仔鱼的鳔原基末出现;孵化后第2天,鳔原基形成;孵化后第5天,气腺形成并首次充气;孵化后第8天,鳔腔中充满气体。到第19天大部分仔鱼的鳔腔中充满气体,气腺最发达。仔鱼的变态发育和鳔器官的退化几乎同步进行。孵化后第25天,鳔前端的气腺上皮首先退化,只余腹部的气腺上皮;至第31天,整个气腺完全退化,开始变态为幼鱼;至第63天鳔完全消失。过度膨胀的鳔器官内腔被来自内壁增生和肥大的上皮细胞占据。末充气的鳔器官较小,鳔腔内有中等程度的炎症。本实验目的旨为大菱鲆人工育苗中提高鳔的充气率从而提高其成活率提供基础依据。  相似文献

The effect of a pathogenic Vibrio pelagius, isolated during a mass mortality of turbot larvae, on the non-specific immune response of turbot, Scophthalmus maximus (L.), macrophages was studied both in vitro and in vivo. The in vitro treatment of head kidney (HK) macrophages with viable V. pelagius caused a significant inhibition of the chemiluminescence (CL) response in comparison with untreated macrophages, while incubation with heat-killed bacteria did not affect this response. In vivo, the intraperitoneal injection of V. pelagius resulted in a significant inhibition of the CL response in infected fish at days 1 and 4 post-infection compared with the control fish response. The HK macrophage nitric oxide (NO) production was enhanced by in vitro incubation with intermediate doses of viable V. pelagius (5 x 10(3) and 5 x 10(4) bacteria mL(-1)) and higher doses of the heat-killed bacteria (5 x 10(4)-5 x 10(6) bacteria mL(-1)). In both cases, the NO inhibitor N-omega -nitro-L-arginine was capable of down-regulating the specific NO induction caused by incubation with the bacterial treatments. In contrast, incubation with ECPs at higher doses caused a reduction in NO production. In vivo, a significant enhancement in NO production was also observed in macrophage supernatants at day 10 post-infection. Lysozyme concentration in the serum was also significantly increased in the experimentally infected fish at days 4 and 10 post-injection. In addition, viable V. pelagius and its ECPs significantly reduced HK macrophage viability in vitro, whereas no significant differences in viability were observed during the incubation with heat-killed bacteria. As NO production was enhanced in the experimentally infected fish, the inhibitory effect of the NO donor, S-nitroso-acetyl-penicillamine (SNAP), was tested in vitro in a cell-free assay. The results showed that growth of V. pelagius was significantly inhibited using SNAP at a high concentration (1 mM).  相似文献

The recent demonstration that reared turbot (Scophthalmus maximus L) is a natural host for salmonid rhabdoviruses has made their rapid detection relevant to these fish species. A unique protocol to select and use non-competitive monoclonal antibodies (Mabs) for two high-sensitivity sandwich ELISAs has been developed to detect both infectious pancreatic necrosis virus (IPNV) and viral haemorrhagic septicaemia virus (VHSV) in turbot kidney extracts to assess the possibility of using them in field diagnosis. For maximum sensitivity, turbot kidney extracts can be two-fold diluted with high-ionic strength buffers and assayed for the presence of the major viral proteins (VMS rhabdovirus nucleoprotein N/Nx and/or IPN birnavirus protein VP3). The use of control plates coated with irrelevant mouse antibodies (IgG1 and IgG2a) in parallel ELISAs allows for a precise estimation of possible false positives. Turbot kidney extracts with low levels of virus might now be assayed directly without using cell culture, with high precision and in a short time during the acute phase of these viral diseases in reared turbot.  相似文献

In order to enhance growth, survival and quality during early juvenile stages of marine fish it is important to avoid lipid oxidation problems that are known to cause pathologies and disease. The aim of the present study was to characterize and compare the antioxidant systems in juvenile marine fish of commercial importance in European aquaculture, namely turbot (Scophthalmus maximus), halibut (Hippoglossus hippoglossus) and gilthead sea bream (Sparus aurata). The experiment investigated the interaction of the dietary antioxidant micronutrient, vitamin E, with antioxidant defence systems. Fish were fed with diets of identical unsaturation index supplemented with graded amounts of vitamin E. The relationships between dietary and subsequent tissue vitamin E levels were determined as well as the effects of vitamin E supplementation on lipid and fatty acid compositions of both liver and whole fish, on the activities of the liver antioxidant defence enzymes, and on the levels of liver and whole body lipid peroxidation products, malondialdehyde (thiobarbituric acid reactive substances, TBARS) and isoprostanes. Growth and survival was only significantly affected in sea bream where feeding the diet with the lowest vitamin E resulted in decreased survival and growth. A gradation was observed in tissue vitamin E and polyunsaturated fatty acid (PUFA)/vitamin E levels in response to dietary vitamin E levels in all species. The activities of the main radical scavenging enzymes in the liver, catalase, superoxide dismutase and glutathione peroxidase generally reflected dietary and tissue vitamin E levels being highest in fish fed with the lowest level of vitamin E. The indicators of lipid peroxidation gave consistent results in all three species, generally being highest in fish fed with the unsupplemented diet and generally lowest in fish fed with the diet with highest vitamin E. In this respect, isoprostane levels generally paralleled TBARS levels supporting their value as indicators of oxidative stress in fish. Overall the relationships observed were logical in that decreased dietary vitamin E led to decreased levels of tissue vitamin E, and generally higher activities of the liver antioxidant enzymes and higher levels of lipid peroxides.  相似文献

采用完全双列杂交法对2个大菱鲆(Scophthalmus maximus)养殖群体:西班牙群体(S)和英国群体(E)进行群体间杂交及群体内自繁,得到了S(♂)×S(♀)、E(♂)×E(♀)、S(♂)×E(♀)和E(♂)×S(♀)4个组合的子一代。对各交配组合的受精率、孵化率、畸形率、30日龄和70日龄稚鱼的体长、体质量等指标进行比较,结果表明,杂交组与自繁组在受精率、孵化率及畸形率等方面均不存在显著性差异;但杂交组30日龄稚鱼和70日龄稚鱼的体长、体质量与自繁组相比,表现出不同程度的杂种优势,其中S(♂)×E(♀)组30日龄稚鱼的体长杂种优势达到5.43%,E(♂)×S(♀)组70日龄稚鱼体质量杂种优势达到25.00%。实验初步显示,不同养殖群体间的杂交有可能是大菱鲆遗传改良的一条有效途径。  相似文献

The effects of dietary α‐tocopheryl acetate supplementation and different slaughtering methods were investigated on the flesh quality of farmed market‐size turbot Scophthalmus maximus (L). Turbot were divided into three groups and fed commercial diets, supplemented with different levels of α‐tocopheryl acetate at the following dietary inclusion levels: 72 (100), 547 (500), 969 (1000) (mg of α‐tocopheryl acetate kg?1 diet, analytical values with diet codes in brackets). After 5 months, fish (mean weight 1056 ± 19.7 g) from each dietary treatment were sampled, applying three different slaughtering methods: (A) bleeding in ice water; (B) thermal shock, no bleeding; (C) percussion followed by bleeding in ice water. The time course of rigor mortis was evaluated, using pH, rigor index and mechanical compression tests. The results showed that the three parameters corresponded very well. Percussive stunning resulted in higher initial post‐mortem pH (P < 0.01) and a significantly delayed onset of rigor mortis (P < 0.05). Diet significantly affected shelf‐life, with fillets from fish fed diets 500 and 1000 having lower TBARS (thiobarbituric acid reactive substances) numbers from day 2 (P < 0.001) and less colour deterioration from day 7 of storage on ice onwards (P < 0.05). These results suggest that an increase in dietary α‐tocopheryl acetate before slaughter as well as careful selection of the slaughtering method may greatly enhance the flesh quality of market‐size turbot.  相似文献

In a commercial scallop hatchery spat production depends on a culture system which ensures high survival and good growth. Reuse of water with algae may increase the food exploitation and hence reduce the costs. Post-larvae of great scallop (Pecten maximus) were studied in a commercial hatchery using a partial open and continuous feeding tank system. Three different water recirculation rates (67, 83 and 92%) were tried out in two experiments with post-larvae originating from three spawning groups of ages between 43 and 57 days post-spawn, 316–886 μm shell-height and 1.1–9.6 μg ash-free dry weight. The post-larvae were held in sieves in tanks of 2500 l where a downwelling flow was maintained by airlifts. New water with a mix of monocultured algae was continuously added to the tanks at algal concentrations of 10 and 15 cells μl−1 in experiment 1 (groups 1 and 2) and 2 (group 3), respectively. The algal supply to each sieve was reduced along with increased recirculation rate, but was kept between 6 and 13 cells μl−1. Generally no significant differences in survival, growth or chemical content were found between the three recirculation rates, while few differences were found between and within groups. Large variation in survival was found between and within groups (1–81%). Highest survival was found in experiment 1, and where post-larvae from two settlements were used, the first settlement survived better than the second. The daily growth ranged from 15 to 62 μm shell-height and from 0.3 to 2.6 μg ash-free dry weight. The scallop post-larvae could well be reared at all three recirculation rates studied as an increase from 67 to 92% did not seem to affect the post-larval performance seriously. The algal supply, however, had to be compensated by an increasing number of cells (>10 cells μl−1) when increasing the recirculation rate.  相似文献
