Deep sequencing of small RNAs(sR NA) is widely used in sR NAs studies in plants. In order to investigate the sequencing frequency variation of sR NAs, the same sR NA samples from rice grains were sequenced twice using deep sequencing technique. The sR NAs were classified into three categories, high abundance( 100 RPM), medium abundance(10–100 RPM) and low abundance(1–10 RPM). According to the repeat sequencing data of the same sample, highly expressed sR NAs( 100 RPM) were less subject to random drift, and 95% of the sR NAs Log2 ratio between two samples fell between-0.649 and 0.558. The same trend was observed in mediumly expressed sR NAs(10–100 RPM), and 95% of the Log2 ratio fell between-0.535 and 0.759. As to lowly expressed sR NAs(1–10 RPM), 95% of the Log2 ratio varied between-1.009 and 1.011. These results can be used as a theoretical guide to find differentially expressed s RNAs in sR NA studies in plants. 相似文献

Bronchial hyper-responsiveness (BHR) describes a lung abnormality in which airways are easily triggered to constrict in response to normally harmless inhaled stimuli, and is a key element of human asthma pathophysiology. BHR contributes to equine respiratory diseases including inflammatory airway disease and recurrent airway obstruction. Collectively these diseases account for over 80% of poor performance in equine athletes, and at least 10% of veterinary admissions. BHR is also a contributing factor in ‘exercise induced pulmonary hemorrhage’. Increased sensitivity to airway constriction that characterizes BHR is a documented sequel to viral respiratory infections in several species, including horses and humans. Five respiratory viruses known to circulate extensively in equine populations place the horse at risk for BHR. Despite adverse effects of BHR on equine health, there remains a gap in our fundamental understanding of how gene products coordinate in the lung to cause BHR. Leveraging the equine genome sequence, we employ systems biology including proteomics and RNA sequencing to model the complex biology of BHR in the lungs of horses with pasture asthma. Using a self controlled experimental design, gene products that segregate with seasonal asthma exacerbation in diseased horses are being identified and their relevant physiology identified to address the need for better recognition and management of BHR in equine disease. 相似文献

AIM: To investigate the effect of endophilin A2 on synaptic vesicle endocytosis in rat hippocampal neurons.METHODS: The siRNAs for endophilin A2 isoform were designed, and the efficacy and specificity of these siRNAs were determined by Western blot. Immunostaining was performed to verify the interference efficiency of the siRNA for endogenous endophilin A2 in the hippocampal neurons. Excitatory postsynaptic currents (EPSCs) were recorded on cultured hippocampal neurons using dual whole-cell recordings by evoking the transfected presynaptic neurons in various stimulation patterns. RESULTS: The result of immunostaining in cultured hippocampal neurons demonstrated that the siRNA of endophilin A2 inhibited the expression of endogenous endophilin A2 significantly (P<0.05). The results of dual whole-cell recordings showed that no significant difference in the EPSCs amplitude evoked between endophilin A2 knockdown neurons and control was observed when stimulated at low frequency (0.1 Hz). The amplitude of normalized EPSCs evoked in endophilin A2 knockdown neurons decreased significantly for both single-train and multiple-train stimulations (P<0.05).CONCLUSION: An effective siRNA of endophilin A2 was screened out successfully. Knockdown of endophilin A2 isoform does not affect synaptic vesicle exocytosis, but inhibits synaptic vesicle endocytosis in rat hippocampal neurons. 相似文献

本试验对不同产乳量的健康奶山羊乳中环鸟苷酸(cGMP)和核糖核酸(RNA)的含量及葡萄糖-6-磷酸脱氢酶(G-6-PDH)的活性进行了测定,并分析了它们与产乳量之间的关系。结果表明:乳中cGMP和RNA的含量均与产乳量极显著相关(P<0.01)。高产羊与低产羊之间乳中cGMP含量存在显著差异(P<0.05)。因此认为,cGMP有促进乳生成作用。 相似文献

首次提出了一种适用于小佛肚竹(Bambusa ventricosa)和螺节竹(Pleioblastus gramineus)等竹类植物总RNA提取方法.用该方法提取叶片和笋尖的总RNA具有正常的光谱吸收,OD260/OD280比值为1.96~2.0,OD260/OD230比值为2.96~4.11;琼脂糖电泳后,28 SRNA的亮度基本为18 SRNA的亮度的2倍;这说明提取的RNA很完整,未降解,质量高.同时该方法操作简便、无DNA污染,且产率高;提取的RNA可用于RT-PCR、Northern杂交、cDNA文库的构建等. 相似文献
