To investigate the pathogen of bovine respiratory disease complex (BRDC) of a dairy farm in Guangxi, a strain of Mycoplasma and a strain of gram-negative pathogenic bacterium were isolated and identified by the means of field surveys, clinic observation, pathological examination, isolation studies and so on.Treatments were taken according to drug sensitivity test results.The Mycoplasma strain, growing on PPLO medium, formed typical "fried egg" colonies.A 448 bp of oppF fragment was amplified by PCR from the strain and had 98.4% nucleotide identity with Mycoplasma bovis reference isolate PG5 of USA.The biochemical features of the gram-negative bacterial isolate were same with Serratia marcescens.The PCR amplified 16S rDNA of the gram-negative pathogenic bacterium strain was 1 400 bp.It shared 99.0% nucleotide identity with other Serratia marcescens reference strains obtained from GenBank.Animal experiment showed that the gram-negative pathogenic bacterium isolate could cause the mice to die.The drug sensitivity tests showed all isolates were sensitive to spectinomycin, azithromycin, amikacin, gentamicin and neomycin.It was effective to treat with dexamethasone and spectinomycin.Pathogen analysis and drug treatment showed that the BRDC was caused by Mycoplasma bovis and Serratia marcescens. 相似文献

The present study was designed to construct recombinant plasmids,which could express porcine reproductive and respiratory syndrome virus (PRRSV) ORF5 gene.RNA was extracted from spleen and lung samples of the suspected pigs which were infected with PRRSV.According to PRRSV ORF5 gene,a pair of primers was designed for RT-PCR amplification.The ORF5 target gene was cloned into pMD19-T vector and then the recombinant pMD19-ORF5 was achieved.According to the sequencing results and the characteristics of expression vectors,a pair of primers with NcoⅠand XbaⅠenzyme cleavage sites was designed.Target fragment dORF5 was amplified and then connected to pProEXHTb and pNZ8149 vectors,respectively.And recombinant HTb-dORF5/DE3 and pNZ8149-dORF5/NZ3900 was induced by IPTG and Nisin,respectively,and analyzed by SDS-PAGE and Western blotting.Recombinant HTb-dORF5/DE3 induced by 1.5 mmol/L IPTG was expressed in the highest quantity.There were specific band at about 22 ku with reactionogenicity when it was tested by SDS-PAGE and Western blotting.Recombinant pNZ8149-dORF5/NZ3900 induced by 20 ng/mL Nisin was expressed in the highest quantity.There were specific band at about 19 ku with reactionogenicity when it was tested by SDS-PAGE and Western blotting.The IFA result showed specific green fluorescence.This study successfully constructed recombinant plasmids HTb-dORF5 and pNZ8149-dORF5 and expressed,the result laid a solid foundation for further development of PRRS vaccines. 相似文献

Background: Acclimatization to winter conditions is an essential prerequisite for survival of small passerines of the northern temperate zone. In the present study, we measured diurnal variations in body mass, body temperature and basal metabolic rate(BMR) for seasonally acclimatized Hwameis(Garrulax canorus).Methods: Body mass was determined with a Sartorius balance. Metabolic rates of Hwameis were measured with an open-circuit respirometry system.Results: Body masses varied with time of day and were higher in daytime for Hwameis in both summer and winter, and body masses in winter were higher compared to that in summer. Body temperatures of Hwameis were higher in daytime, and the summer acclimatized birds had significantly higher body temperatures compared to the winter acclimatized birds. BMRs of Hwameis were significantly higher during the daytime compared to the nighttime of the daily cycle in both summer and winter, and Hwameis in winter had significantly higher BMRs than that in summer.Conclusions: This result showed that Hwameis rely mostly on metabolic capacity to maintain their body temperature in cold weathers, and Hwameis exhibited daily and seasonal flexibility in morphology and physiology which is important under changing environmental conditions. 相似文献
