日本血吸虫中国大陆株22.6kD膜相关蛋白基因的克隆与表达

以日本血吸虫 mRNA 为模板,用 RT-PCR 法快速克隆到一大小约600bp 的 DNA 片段,DNA 序列分析证实,所扩增到的 DNA 片段中含有日本血吸虫22.6kD 膜相关蛋白(Sj-22.6(Ch))基因,将该基因重组到表达型质粒 pGEX-4T 中,表达的 GST 融合蛋白分子量约48kD,用谷胱甘肽琼脂塘凝胶亲和层析柱纯化的重组蛋白不仅纯度好,而且得率高,纯化产量可达40mg·L~(-1)培养物,免疫试验结果表明该重组蛋白具有良好的抗原性,为其在血吸虫病抗感染中的免疫作用研究创造了条件.

Expression and functional analysis of the Follistatin-like 3 (FSTL3) gene in the sheep ovary during the oestrous cycle

Follistatin-like 3 (FSTL3) is a regulator of cellular apoptosis and was previously identified via RNA-Seq to be associated with follicular development in mammalian ovaries. However, the mechanism underlying the FSTL3 regulation of oestrus in sheep remained poorly understood. In this study, the oestrogen (E2) and progesterone (P4) concentrations in blood were detected, and the expression level and functional analysis of FSTL3 in the ovary were studied during the different reproductive stage in Aohan fine wool sheep (seasonal breeding breed in China). The concentrations of E2 and P4 at the anestrus were significantly lower compared to dioestrus, proestrus and oestrus stages. Higher expression levels of FSTL3 were observed in the sheep ovary, hypothalamus, and thyroid. During different reproductive stages, higher expression levels were found during the stages of dioestrus and proestrus, while lower levels were found during the oestrus and anestrus stages. Functional analysis of FSTL3 was performed in primary granulosa cells (GCs) of sheep. The concentration of E2 increased significantly after RNAi interference of FSTL3, while the P4 level decreased. FSTL3 can decrease P4 levels, which might be involved in mediating oestrous cycle in sheep.     

不同紫花苜蓿几个抗旱指标的灰色关联分析

运用灰色关联分析法,对5项抗旱指标与5个紫花苜蓿(Medicago Sativa)品种抗旱性的相关性进行了分析。关联度分析结果表明:各指标与苜蓿品种抗旱性的关联序为:游离脯氨酸含量>SOD>POD>相对含水量>叶片水势。各指标对苜蓿抗旱性的影响,以游离脯氨酸含量最大,其次为SOD及POD;再次相对含水量;而叶片水势的影响最小。

Inactivation of the Wnt/β-catenin signaling contributes to the epithelial barrier dysfunction induced by sodium oxalate in canine renal epithelial cells

High oxalate consumption has been recognized as a risk factor for renal calcium oxalate stones in companion animals (dogs and cats). However, the cellular signaling involved in oxalate-induced dysfunction in renal tubular epithelial cells remains not fully elucidated. In this study, Mardin–Darby canine kidney (MDCK) cells, an epithelial cell line derived from canine kidney tubule, were tested for cell proliferation activity and barrier function after being exposed to sodium oxalate (NaOx). Further, the involvement of Wnt/β-catenin in NaOx-induced renal epithelial barrier dysfunction was evaluated. MDCK cells treated with NaOx exhibited reduction in cell proliferation and migration. Besides, NaOx exposure led to a decrease in transepithelial electrical resistance and an increase in paracellular permeability. The deleterious effects of NaOx on epithelial barrier function were related to the suppressed abundance of tight junction proteins including zonula occludens, occludin, and claudin-1. Of note, protein levels of β-catenin and phosphorylated (p)-β-catenin (Ser552) in MDCK cells were repressed by NaOx, indicating inhibitory effects on Wnt/β-catenin signaling. An inhibition of glycogen synthase kinase-3β (GSK-3β) by SB216763 enhanced the abundance of β-catenin and p-β-catenin (Ser552), and protected against epithelial barrier dysfunction in NaOx-treated MDCK cells. The results revealed a critical role of Wnt/β-catenin signaling in the epithelial barrier function of MDCK cells. Activation of Wnt/β-catenin signaling might be a potential therapeutic target for the treatment of oxalate-linked renal stones.     

大修汽车可靠性分析中故障分布规律的研究

从分布函数拟合的基本原理入手,把回归分析方法和优化技术有机结合起来,提出了一种由观测数据拟合其分布函数的优化方法,并以威布尔分布为例,说明其具体过程,通过试验数据应用表明,该方法正确可行。     

脾虚证模型家兔神经内分泌机能的变化及四君子汤对其调整作用

给家兔胃管投服苦寒中药大黄水煎液,复制出家兔脾虚证动物模型,研究了脾虚证模型家兔神经内分泌机能的变化和健脾益气四 君子汤对其的调 整作用。结果表 明,脾 虚证模型家兔血 浆胃泌素、ｃ Ａ Ｍ Ｐ 和ｃ Ａ Ｍ Ｐ／ｃ Ｇ Ｍ Ｐ 较试验前和对照组明显下降 （ Ｐ ＜ ００５ 或 Ｐ ＜ ００１）, 血浆胰高血糖素和血清胆碱酯酶活性较对照组和试验前极显著升高 （ Ｐ ＜ ００１）。以上各指标在经四君子汤治疗后均恢复至试验前和对照组水平,与模型期比较变化明显 （ Ｐ ＜ ００５ 或 Ｐ ＜ ００１）。由此说明,脾虚证时神经内分泌机能发生了改变,表现为交感神经机能降低和副交感神经机能相对亢进,以及胃肠道激素如胃泌素和胰高血糖素的分泌发生变化,四君子汤对其有良好的调整作用或拮抗作用。
