高良姜提取物对PC12细胞的神经保护作用

通过建立H2O2介导的PC12细胞损伤模型;采用MTT法检测细胞存活率,利用荧光显微镜观察细胞形态;检测细胞中乳酸脱氢酶漏出率,丙二醛(MDA)含量,超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GGSH-Px)活性,评价高良姜提取物对H2O2介导的PC12细胞损伤的保护作用。结果表明,高良姜提取物能明显降低细胞内乳酸脱氢酶漏出率,降低细胞内MDA含量,提高SOD和GGSH-Px的活性,且具有浓度依赖性,由此推断,高良姜提取物对H2O2介导的PC12细胞损伤有明显保护作用。     

高速逆流色谱法制备分离库拉索芦荟中的2’-对香豆酰芦荟宁

采用高速逆流色谱法分离纯化库拉索芦荟中的2’-对香豆酰芦荟宁。以正己烷-乙酸乙酯-丙酮-水(1∶5∶1∶5,V/V/V/V)为溶剂体系,上相为固定相,下相为流动相,线圈转速为860 r/min,流速为2 mL/min,紫外检测波长为300 nm,一次进样85 min内从487.5 mg库拉索芦荟丙酮提取物中分离得到19.5 mg单体组分,经高效液相色谱分析,测定纯度为95.6%(峰面积百分比),通过质谱、红外、紫外和核磁技术进行结构鉴定,确定组分为2’-对香豆酰芦荟宁。该法具有操作简便、速度快、样品制备量大、节省溶剂及回收率高等优点,可为芦荟宁类化合物的分离提供高效、稳定及可靠的方法。     

Ochracenoids A and B,Guaiazulene-Based Analogues from Gorgonian Anthogorgia ochracea Collected from the South China Sea

Two new guaiazulene-based analogues, ochracenoids A (1) and B (2), along with four known analogues (3–6), were isolated from the gorgonian Anthogorgia ochracea collected from the South China Sea. The planar structures of the new compounds were elucidated by comprehensive spectroscopic data. The absolute configuration of 1 was determined as 3R by the comparison of TDDFT calculated electronic circular dichroism with its experimental spectrum. Compound 1 is a rare guaiazulene-based analogue possessing a unique C₁₆ skeleton. The possible generation process of 1 through an intermolecular one-carbon-transfer reaction was also discussed. Compound 2 was previously described as a presumed intermediate involved in the biogenesis of anthogorgienes A and I. Compound 3 exhibited antiproliferative effects on the embryo development of zebrafish Danio rerio.     

Two New Cytotoxic Indole Alkaloids from a Deep-Sea Sediment Derived Metagenomic Clone

Two new indole alkaloids, metagenetriindole A (1) and metagenebiindole A (2), were identified from deep-sea sediment metagenomic clone derived Escherichia coli fermentation broth. The structures of new compounds were elucidated by spectroscopic methods. The two new indole alkaloids demonstrated moderately cytotoxic activity against CNE2, Bel7402 and HT1080 cancer cell lines in vitro.     

Bis(2,3-dibromo-4,5-dihydroxybenzyl) Ether,a Marine Algae Derived Bromophenol,Inhibits the Growth of Botrytis cinerea and Interacts with DNA Molecules

Bis(2,3-dibromo-4,5-dihydroxybenzyl) ether (BDDE) is a bromophenol isolated from marine algae. Previous reports have shown that BDDE possesses cytotoxic and antibacterial activity. In the present study, we demonstrate that BDDE displays broad-spectrum antifungal activities, especially on Botrytis cinerea. BDDE inhibits the growth of B. cinerea cultured on a solid medium of potato dextrose agar (PDA) as well as on the potato dextrose broth (PDB) medium. Moreover, BDDE decreases the incidence of fruit decay and severity of strawberries infected with B. cinerea. Further studies have revealed that BDDE decreases the germination rate and inhibits the mycelial growth of B. cinerea. The inhibition mechanisms are related to the disruption of the cell membrane integrity in B. cinerea spores and newly formed germ tubes. This study also suggests that BDDE possibly interacts with DNA via intercalation and minor groove binding. The studies provide evidence that BDDE has potential application in the control of gray mold after fruit harvest and the compound could serve as a candidate or lead template for rational drug design and for the development of antifungal agents.     

Pseudaboydins A and B: Novel Isobenzofuranone Derivatives from Marine Fungus Pseudallescheria boydii Associated with Starfish Acanthaster planci

Two novel isobenzofuranone derivatives, pseudaboydins A (1) and B (2), along with five known compounds, including (R)-2-(2-hydroxypropan-2-yl)-2,3-dihydro-5-hydroxybenzofuran (3), (R)-2-(2-hydroxypropan-2-yl)-2,3-dihydro-5-methoxybenzofuran (4), 3,3′-dihydroxy-5,5′-dimethyldiphenyl ether (5), 3-(3-methoxy-5-methylphenoxy)-5-methylphenol (6) and (−)-regiolone (7), were isolated from the culture broth of the marine fungus, Pseudallescheria boydii, associated with the starfish, Acanthaster planci. Their structures were elucidated primarily based on NMR and MS data. The absolute configurations of 1–4 were determined by CD spectroscopy and single-crystal X-ray diffraction studies. The cytotoxic and antibacterial activities of 1–4 were evaluated. Pseudaboydin A (1) showed moderate cytotoxic activity against human nasopharyngeal carcinoma cell line HONE1, human nasopharyngeal carcinoma cell line SUNE1 and human glandular lung cancer cell line GLC82 with IC₅₀ values of 37.1, 46.5 and 87.2 μM, respectively.     

微波辅助提取霍山石斛多糖的研究

[目的]运用微波辅助提取技术提取霍山石斛多糖,并对工艺条件进行优化。[方法]通过单因素实验研究了微波提取霍山石斛多糖过程中物料粒度、提取次数、提取时间、微波功率和料液比对多糖得率的影响;采用正交设计研究了影响多糖得率的关键因素,并与传统回流提取比较。[结果]所有单因素在水平较低时对石斛多糖得率影响较大,水平越高影响越小;正交分析筛选出微波法提取霍山石斛多糖的最优工艺为：提取4次,每次40 min,微波功率550 W,并验证了提取次数对多糖得率有显著影响。[结论]微波辅助提取能够提高霍山石斛多糖的提取效率,为开发新的霍山石斛多糖提取技术打下基础。     

Five new triterpenoidal saponins from the roots of Ilex cornuta and their protective effects against H2O2-induced cardiomyocytes injury

Five new ursane-type triterpenoidal saponins (1–5), together with five known ones (6–10), were isolated from the EtOH extract of the roots of Ilex cornuta. The structures of saponins 1–5 were elucidated as 19α-hydroxyurs-12-en-28-oic acid 3β-O-β-D-glucuronopyranoside (1), 19α-hydroxyurs-12-en-28-oic acid 3β-O-β-D-glucuronopyranoside-6-O-ethyl ester (2), 19α-hydroxyurs-12-en-28-oic acid 3β-O-α-L-arabinopyranosyl-(1 → 2)-β-D-glucuronopyranoside (3), 3β-O-[α-L-arabinopyranosyl-(1 → 2)-β-D-glucuronopyranosyl]-19α-hydroxyurs-12-en-28-oic acid 28-O-β-D-glucopyranosyl ester (4) and 3β-O-[α-L-arabinopyranosyl-(1 → 2)-β-D-glucuronopyranoside-6-O-methyl ester]-19α-hydroxyurs-12-en-28-oic acid 28-O-β-D-glucopyranosyl ester (5), on the basis of spectroscopic analyses (IR, ESI-MS, HR-ESI-MS, 1D and 2D NMR) and chemical reactions. Protective effects of compounds 1–10 against H₂O₂-induced H9c2 cardiomyocyte injury were tested. Compounds 1–5, 7, and 10 showed cell-protective effects. Among them compound 5 exhibited the highest activity. No significant DPPH radical scavenging activity was observed for compounds 1–10.     

Noreudesmane sesquiterpenoids from the leaves of Nicotiana tabacum

Six new 14-noreudesmane sesquiterpenoids, nicotabacosides A–F (1–6), along with five known sesquiterpenoids (7–11), were isolated from the leaves of Nicotiana tabacum. The structures of compounds 1–6 were elucidated as isorishitin 3-O-β-d-glucopyranoside (1), rishitin 3-O-β-d-glucopyranoside (2), rishitin 2-O-β-d-glucopyranoside (3), 1, 6-dehydro-rishitin 3-O-β-d-glucopyranoside (4), 2-hydroxyl-ligudentatol 3-O-β-d-glucopyranoside (5) and oxyglutinosone 3-O-β-d-glucopyranoside (6) based on extensive spectroscopic analyses (HRESIMS, UV, IR, 1D and 2D NMR). Their absolute configurations were determined by X-ray single-crystal diffraction and comparison of their electronic circular dichroism (ECD) spectra.     

白蔹挥发油化学成分分析

首次采用水蒸气蒸馏法制备白蔹挥发油,通过面积归一化法测定其相对百分含量,应用气相色谱-质谱联用技术研究分析其化学成分。最终分离鉴定出14种化学成分,占挥发油总量的99.997%,主要包括:芳姜黄酮(21.465%)、2-甲基-6-对甲苯基-2-庚烯醇(17.438%)和蒽(14.224%)等,为白蔹的进一步开发利用提供了研究依据。
