Progress in autoantibodies to platelet and idiopathic thrombocytopenic purpura

The onset of idiopathic thrombocytopenic purpura (ITP) is related to excessive destruction of platelet resulting from antiplatelet autoantibody. The immunity with ITP is imbalance, so the hypofunction of suppressor T cell (Ts) can't restrain B lymphocytes producing antibody, as a result, autoant ibody is produced, the clearance of platelet in blood circulation is accelerated. The different antibodies have different functions on ITP. The measuring of platelet antibody has its significance in diagnosis, therapeutic evaluat ion and expectation of prognosis.     

Rat urotensin-II-induced main pulmonary artery contractions are mediated by MAPK

AIM: To investigate rat Urotensin-II(rat U-II)-induced vasoconstriction of rat main pulmonary arteries and the role of mitogen-activated protein kinase(MAPK). METHODS: The main pulmonary artery was dissected from the male Sprague-Dawley rats and artery ring width was 3-4 mm. Concentration-response curves were generated to rat U-II(0.03 nmol/L-30 nmol/L).Inhibitor of MAPK, PD 98059(0.1 μmol/L-10 μmol/L) were added into the medium after rat U-II(30 nmol/L)induced vasoconstriction had reached plateau to construct the relaxant concentration-response curves and their EC₅₀ and E_max. RESULTS：Rat U-II was a potent vasoconstrictor of isolated rat main pulmonary arteries [EC₅₀=7.95±0.40, E_max=(14.28±6.34)% of the response to 60 mmol/L KCl]; PD 98059 caused concentration-dependent relaxations of rat U-II precontracted arteries [EC₅₀=5.91±0.45, E_max=(81.39±13.65)%]. CONCLUSION: Rat U-II was a potent vasoconstrictor of rat main pulmonary arteries and this response was mediated through MAPK.     

单对汰选方法在加速抗高效氯氟氰菊酯棉铃虫品系汰选中的应用

通过设计杂合种群单对杂交方式,研究了加速获得抗性品系的方法。以高效氯氟氰菊酯群体汰选后抗性倍数为4.9倍的棉铃虫种群及其同源对照种群为材料,同时设置常规群体汰选方法与单对汰选方法,研究单对汰选方法在加速抗高效氯氟氰菊酯棉铃虫品系汰选中的作用。结果表明,群体汰选两代后抗性倍数由4.9倍提高到7.4倍, 而单对汰选两代后抗性倍数由4.9倍提高到27.3倍。表明在常规群体汰选中穿插几代单对汰选方法可明显加快棉铃虫种群对高效氯氟氰菊酯的抗性汰选进程。     

番木瓜环斑病毒株系的分子生物学方法鉴定

 以PRSV株系特异性引物对PRSV的PRSV126(PRSV日本分离物)、Ys、Vb和Sm等株系进行RT-PCR方法鉴定,引物PR21/PR22能把Ys从Vb和Sm中鉴定出来,PR300/PR301则能把Vb从Ys、Sm和PRSV126中鉴定出来;用限制性内切酶Hae Ⅱ、Sau3A I和Hinf I对PRSV的PRSV126、Ys、Vb和Sm等株系进行单酶切RT-PCR-RFLP分析,Hinf I能把PRSV126与Ys、Vb和Sm鉴别开来,Sau3A I能把Ys与Vb和Sm鉴别开来,Hae Ⅱ则能把Ys与PRSV126、Vb和Sm鉴别开来;以P1/P2为引物,对Vb、Ys和Sm株系进行RT-PCR-RFLP-SSCP分析,结果能一次把三者较好地区别开来。     

IL-12 induces T lymphocytes apoptosis and influences the expression and signal transduction of Fas/FasL

AIM: IL-12 acts upon Tlymphocytes and activates its receptor complexes of β1/β2,and so IL-12 can regulate TH1/TH2 balance. Our study is aimed at IL-12-inducing apoptosis of Tcells and expression and signal transduction of Fas/FasLduring Tcell apoptosis. METHODS: The apoptosis of Tcells was detected by Annexin Vstaining cytometry and the expression of Fas/FasLunder different inhibitors were detected by semi-quantitative PCR. RESULTS: IL-12 induced the human leukemic Tcell line(TIB-152) and the human lymphoma Tcell line(HTB-176) and the normal human Tcells to undergo apoptosis. The FasLexpression at 6 hours after treatment with IL-12 increased apparently, and reached the max at 24 hours, and FasLexpression induced by IL-12 was inhibited by PKCinhibitor. But IL-12 did not influence Fas expression. CONCLUSIONS: IL-12 can induce Tcells to undergo apoptosis which is characterized by early membrane changes, the inducing effect is correlated with the concentration of IL-12 and the maturation of Tcells. FasLparticipates in the progression of Tcell apoptosis as a apoptosis mediator, and the effect of IL-12 on FasLexpression may be related with PKCpathway.     

利用RT-PCR与PVP-ELISA检测水稻瘤矮病单头叶蝉

 目前国内外对水稻瘤矮病毒(Rice gall dwarf virus,RGDV)的研究主要集中在其核衣壳蛋白和外衣壳蛋白的性质、结构及其与介体的亲和性、与介体传毒能力的关系等方面,本室在已有室内检测研究的基础上,利用RT-PCR和PVP-ELISA法对田间带毒叶蝉进行检测,并对2种方法的检测结果进行了比较。     

12-(取代苯氧异丁酰氧亚氨基)-1,15-十五内酯的合成及除草活性

通过取代苯氧异丁酸与12-(羟基亚氨基)十五内酯的反应,合成了取代苯氧异丁酰氧亚氨基十五内酯,并测定了其对苋菜的除草活性。所有化合物均经过1H NMR、13C NMR和元素分析确证。初步的生物活性测定结果表明,目标化合物 Ⅲa~Ⅲd 的EC50值分别为34.570、46.492、55.385、50.114 mg/L,其活性比对照药剂2,4-D(117.325 mg/L)高,而比苯磺隆(22.381 mg/L)低。     

影响微孢子虫对棉铃虫幼虫致病力的因子

1997年田间调查时，发现一种寄生于棉铃虫的微孢子虫，对棉铃虫具有很强的致病力。为明确环境因子对该微孢子虫及其致病力的影响，测定了温度与紫外线对该微孢子虫及其杀虫效果的影响。结果表明，该微孢子虫孢子耐受温度范围较宽，最高温度上限为55℃。温度对其致病力有显著影响，在一定温度范围内，随着生境温度的升高，微孢子虫对棉铃虫幼虫致病力增强。该微孢子虫对紫外线较敏感，在紫外线照射下易失活而丧失致病力。     

从海南省杂草胜红蓟和假马鞭上检测到粉虱传双生病毒

 利用三抗体夹心ELISA(TAS-ELISA)及PCR检测的方法对采自海南的胜红蓟(Ageratum conyzoides)和假马鞭(Stachytarphetajamaicensis)的5个病样进行了检测,表明均为粉虱传双生病毒。PCR扩增产物克隆后进行序列测定,结果表明存在2类双生病毒,其中样品Hn2存在2类病毒的复合侵染。这是在海南首次报道存在有粉虱传双生病毒。     

侵染西瓜的小西葫芦花叶病毒的生物学特性鉴定

西瓜病毒病是制约西瓜产量及品质的顽疾.据报道,该病害造成的经济损失达到30%～50%.1981～1999年皖、苏、鲁、豫西瓜的病毒病曾几次大发生.危害西瓜的病毒病有西瓜花叶病毒1号(WMV-1)、西瓜花叶病毒2号(WMV-2)、黄瓜花叶病毒(CMV)、烟草花叶病毒(TMV)、烟草环斑病毒(TRSV)、南瓜花叶病毒(SqMV)等32种[1].小西葫芦花叶病毒(ZYMV)是侵染葫芦科的主要病毒之一,1973年首次在意大利北部发现此病毒,至今已成为一个全球性的植物病毒[2].笔者对ZYMV合肥分离物的生物学特性进行了研究.
