家蚕品种“明丰×春玉”改良系农村饲养初报

家蚕品种"明丰×春玉"抗Bm NPV改良系在农村试养中,表现出蔟中不利气候条件下结茧率偏低的现象。针对存在的问题,对品种特性进行进一步改良。改良品种的农村试养对比结果表明,改良系抗病毒病性状优良,各项经济性状优于对照品种。     

Effects of five cryoprotectants on proliferation and differentiation‐related gene expression of frozen‐thawed bovine calf testicular tissue

The cryopreservation of testicular tissue is a potential method for preserving male fertility. However, the effect of cryopreservation on bovine calf testicular tissue is scarce. This study investigated the effect of different cryoprotectants on bovine calf testicular tissue at the molecular level. Testicular tissue from ten immature bovine calves (6 months) was collected after slaughter and cryopreserved in an extender containing different concentrations of the following five cryopreservation solutions (CP): bovine serum albumin (BSA) with 5% dimethyl sulfoxide (DMSO), trehalose with 5% DMSO, DMSO and glycerol and ethylene glycol (EG). After 7‐day cryopreservation, the expression levels of three spermatogonial stem cell (SSC)‐related genes, octamer‐4 (OCT4), KIT ligand (MGF/SCF) and kit oncogene (C‐KIT), were investigated by quantitative PCR (qPCR). The cell viability was highest for the tissues preserved with 30 mg/ml BSA (77.82% ± 1.22) and 40 mg/ml trehalose (74.23% ± 1.16) compared with other groups (p < 0.05), and the level of expression of the three genes was highest with 30 mg/ml BSA (p < 0.05). Compared with other CPs, the 30 mg/ml BSA and 40 mg/ml trehalose have the better cryopreserve protection. The 30 mg/ml BSA is the most viable media for the cryopreservation of testicular tissue from cattle.     

湖南省某生猪批发市场疫病传播风险评估

为评估湖南省某生猪批发市场猪群疫病的发生风险,找出可能的风险因素,便于提出针对性预防措施,采用自行设计的调查问卷,结合访谈和现场调查,对27户经营户进行了调查,并将调查结果和调运数据相结合进行了疫病传播风险定性评估。结果显示:该市场疫病传入的可能性"高",内部传播的可能性"中等",疫病传出的可能性"高"。生猪调运频次多、数量大,经纪人行为复杂,运输车辆清洗消毒不彻底是可能的风险因素。评估结果提示,应尽量减少经纪人行为影响,重点加强对运输车辆的清洗消毒。     

6种人兽共患病病原核酸液相芯片检测体系的建立

本研究通过设计、合成特异性引物和探针并对其进行修饰,将探针和特定荧光编码微球共价交联后,利用Luminex检测仪读取荧光信号,从而建立了针对布鲁氏菌、沙门氏菌、弓形虫,狂犬病病毒、巴氏通体、弯曲杆菌等6种病原的液相芯片检测方法。数据显示,本研究建立的液相芯片检测体系具有良好的特异性,检测灵敏度达50拷贝数,能同时检测6种重要人兽共患病病原,对于这6种人兽共患病的快速筛查和控制提供了技术支撑。     

浙北地区早熟松花菜品种比较试验

在浙北松花菜秋季栽培区选择试点,对6个早熟松花菜新品种进行了比较试验。试验结果表明,浙农松花50天是浙北地区秋季抢早栽培的较为理想的品种,而农美50天产量潜力最大,但成熟期略迟,生产上可根据实际情况加以应用。     

饲草料半干青贮工艺及技术装备

饲草料是畜牧业发展的重要基础，浅述了饲草料半干青贮工艺，分析饲草料半干青贮装备的现有技术装备水平及发展趋势，为我国半干青贮装备的推广应用提供参考。      

生物菌肥对老桃园土壤和再植桃苗生长的影响

以生物菌肥和桃苗为试材,通过土壤检测和测量桃苗的生长,研究了生物菌肥对老桃园土壤和再植桃苗生长的影响。结果表明:每株浇灌2亿个·mL~(-1)光合菌剂600mL、2亿个·mL~(-1)光合菌剂300mL和3亿cfu·g~(-1)木霉可湿性粉剂1 500mL放线菌增幅较大。2亿个·mL~(-1)光合菌剂600mL和2亿个·g~(-1)木霉菌300g的速效钾含量增幅较大,2亿个·g~(-1)木霉菌300g能增加有效磷含量,3亿cfu·g~(-1)木霉可湿性粉剂1 500 mL的全氮含量有一定的增加,2亿个·mL~(-1)光合菌剂300mL全钾增幅较大。2亿个·mL~(-1)光合菌剂300mL和3亿cfu·g~(-1)木霉可湿性粉剂750mL对主干的增粗最明显而且也有利于三主枝伸长。侧重于桃苗生长情况,建议选用2亿个·mL~(-1)光合菌剂300mL。     

非调质钢在发动机曲轴上的应用研究

介绍了所选用的非调质钢材料在发动机曲轴上的应用研究。各项性能及工艺试验证明,所选用的S38Mn Si V非调质钢的锻造成形性、切削加工性、台架疲劳性能能够满足6110发动机曲轴技术要求,可替代调质钢制造柴油机曲轴。     

浙江高温热害气象下水稻孕穗阶段热害概率的拟合优度检验方法

为避免或降低高温热害对水稻孕穗阶段的影响，建立了一种热害概率预测模型，并采用卡方检验法判断其拟合度。结果表明，预测温度值与实际温度值的拟合度较高，该模型能够有效预测稻田的热害发生概率以及划分高温热害风险等级。      

Expression,Purification and Activity Evaluation of Mb1230 Gene of Mycobacterium bovis

To explore the value of Mb1230 protein of Mycobacterium bovis in the diagnosis of bovine tuberculosis,we obtained the Mb1230 gene by PCR and constructed recombinant plasmid pET-22b-Mb1230.Recombinant Mb1230 protein was obtained by IPTG induction and purified by affinity chromatography.The activity of the recombinant protein was evaluated by TST test,IGRA test and indirect ELISA.The size of the recombinant protein matched with the theoretical value proved by SDS-PAGE;Western blotting result showed that the recombinant protein could react with mouse anti-His antibody,and had specific band;The results of TST test,IGRA test and indirect ELISA test also showed the recombinant protein had antigenic activity.The results indicated the recombinant protein Mb1230 had good B cell and T cell activity,so,it had the potential application in the diagnosis of bovine tuberculosis.
