重要检疫性杂草刺萼龙葵分子生物学检测的研究   总被引:2,自引:0,他引:2  
刺萼龙葵属于茄属植物,是我国规定禁止进境的检疫性有害杂草。选取与刺萼龙葵种子形态非常近似的同科5种植物为研究对象,依据茄科植物GBSSI基因序列差异,设计刺萼龙葵的特异PCR引物SRSF1/SRSR1,扩增片段约为500bp,用于对刺萼龙葵的特异检测。本研究建立了刺萼龙葵快速简便、稳定可靠的分子生物学检测方法,一个工作日内完成检测。  相似文献

WinSEEDLE系统是专门用于研究种子或病斑形态的专业图像分析系统。以外来杂草刺萼龙葵(Solanum rostratum Dunal)种子为材料,探讨了应用该系统进行种子维度测定的最佳方案,以期为相关研究提供借鉴。结果表明,扫描分辨率显著影响WinSEEDLE系统测量种子的准确度和精确度,当分辨率达到300 dpi时可准确计数所有刺萼龙葵种子。测定刺萼龙葵种子维度的推荐分辨率为600 dpi,此条件下单次扫描用时较短(27 s),测量的准确度和精确度较其他分辨率高,测得种子平均长度2.61 mm、宽度2.11 mm,与人工实测值基本一致。  相似文献

刺萼龙葵(Solanum rostratum)是危害严重的外来入侵植物,基于种子形态的传统检疫方法有一定的局限性,不依赖于形态学特征的DNA条形码技术是基于DNA序列差异对物种进行鉴定的新方法,是传统形态学鉴定方法的有力补充。以刺萼龙葵及其8种龙葵亚属近缘植物为材料,从Gen Bank数据库中下载rbc L、mat K和内转录间隔区(ITS)序列,进行DNA条形码分析,结果显示ITS鉴定效果优于其他片段,可把刺萼龙葵从其近缘植物中鉴别出来。  相似文献

病毒诱导的基因沉默(VIGS)是通过插入目的基因片段的重组病毒来抑制植物内源基因表达的遗传技术,主要用于基因的功能分析。茄科植物刺萼龙葵(Solanum rostratum)是一种外来恶性杂草,研究证实,在农杆菌GV3101介导下,刺萼龙葵的八氢番茄红素脱氢酶(PDS)基因能被部分沉默,导致叶片和花朵出现白化表型。半定量RT-PCR检测显示,被侵染叶片和花朵的mRNA显著降解。VIGS沉默体系的建立可适用于研究刺萼龙葵的部分功能基因,有助于深入了解其生长发育调控的分子机制。  相似文献

茎叶除草剂对刺萼龙葵的防治效果评价   总被引:1,自引:0,他引:1  
采用茎叶喷雾法评价了7种防除阔叶杂草的除草剂对野外5~7叶期刺萼龙葵的防除效果。药后30d调查结果表明,7种除草剂对刺萼龙葵的生长均有一定的抑制作用,不同药剂对刺萼龙葵的防效差异显著。防除刺萼龙葵较好的药剂有48%三氯吡氧乙酸乳油、24%氨氯吡啶酸水剂和20%氯氟吡氧乙酸乳油,试验剂量下对刺萼龙葵的鲜重防效均在85%以上,25%氟磺胺草醚水剂对刺萼龙葵的防效较差。三氯吡氧乙酸、氨氯吡啶酸和氯氟吡氧乙酸能够有效防治刺萼龙葵,根据药剂的不同特性,探讨了在不同入侵地的应用可能性。  相似文献

β-甘露聚糖酶是种子萌发过程中降解胚乳细胞壁的关键酶,明确其活性的动态变化可为揭示杂草种子的休眠萌发机制提供重要依据。以外来杂草刺萼龙葵Solanum rostratum Dunal种子为材料,建立了种子中β-甘露聚糖酶活性的检测方法—凝胶扩散法。利用凝胶扩散法对不同贮存时间及贮存条件下刺萼龙葵种子中β-甘露聚糖酶的活性进行了检测,发现贮存3年以上的种子中该酶的活性为0.03 nmol/(min·mg),显著低于贮存3年以下的种子中的酶活性0.15 nmol/(min·mg),而湿润冷藏的种子中β-甘露聚糖酶的活性为0.12 nmol/(min·mg),显著高于干燥冷藏的种子的酶活性0.02 nmol/(min·mg)。实际应用结果表明,凝胶扩散法综合了传统方法的优势,检测特异性强、灵敏度高、重复性好,可同时检测大量种子样品,具有良好的实用性。  相似文献

刺萼龙葵(Solauum rostratum Dunal.)是一种世界性的恶性入侵杂草,扩散速度快,对环境为害严重。结合近年来的研究热点,从扩散机制、生物学特性、环境的可入侵性、植物与土壤的反馈作用以及防控与利用基础等多个方面解析其入侵扩张机制和控制策略。主要研究进展:1刺萼龙葵强大的繁殖和扩散能力、高度的抗逆性、入侵地植物种类的单一性以及入侵环境的脆弱性在其入侵过程中起着关键作用;2刺萼龙葵具有丰富的遗传多样性,在不同生境的长期作用下已产生一定程度的分化,形成了不同的地理种群;3刺萼龙葵不同生长阶段通过改变根际土壤微生物数量和种类来调节自身生长,并通过改变入侵地土壤微生物群落结构和功能,创造对自身生长有利的土壤环境,形成一个自我促进式的入侵机制;4采用人工物理清除和植物替代控制相结合的生态修复方法,可防止刺萼龙葵的再次入侵。刺萼龙葵的有效控制需要采取综合防治策略;5刺萼龙葵从药物开发和植物保护方面有很大的潜力可以挖掘。  相似文献

入侵杂草刺萼龙葵Solanum rostratum Dunal传播扩散的主要载体是种子,研究其种子休眠萌发基因的激素调控对于其防除具有重要意义,而选择合适的内参基因可以提高相关基因表达分析的准确性。本研究以赤霉素、脱落酸和水处理的刺萼龙葵种子为材料,利用geNorm、NormFinder、BestKeeper和RefFinder 4种软件对15个候选内参基因进行表达稳定性评价,并通过检测ABI5(abscisic acid-insensitive 5)的表达验证所筛选的内参基因的适用性。结果表明,对于赤霉素、脱落酸和水处理过的种子,最稳定的内参基因分别为eIF(eukaryotic initiation factor)、SAND(SAND protein family)和ACT(β-actin);对所有种子样本而言,PP2Acs(a catalytic subunit of protein phosphatase 2A)是最稳定的内参基因。研究结果将为刺萼龙葵种子休眠萌发的遗传调控研究提供重要参考。  相似文献

收集并整理2014~2016年从黄埔和南沙口岸进境美国高粱中截获的杂草种子,对截获频次进行统计,并对截获频次较高的9种检疫性杂草、14种非检疫性杂草进行萌发试验,结果表明,供试杂草种子几乎均能萌发,且萌发率较高,如长芒苋和裂叶牵牛萌发率达58%以上、糙果苋19.17%、假高粱和豚草的萌发率也达到7.50%和5.00%。因此要重视美国高粱中携带的杂草种子检疫以及杂草监测工作,防止这些杂草种子的定殖传播。  相似文献

Herbicides applied alone have failed to reduce weed populations in agricultural ecosystems. The use of this management strategy has followed other problems such as damage to ecosystems and organisms that live in it. Herbicide resistance in weeds has also occurred more quickly based on the application of herbicides. Today, most scientists are looking for new integrated weed management programs in agricultural ecosystems. Biological control is one of the most effective ways of weed management. In this way, living organisms are used to reduce and control weed species in arable lands. Weed seed predators, including predispersal and postdispersal predators as biological control agents, can be primary factors of weed seed mortality in agricultural ecosystems. This review has discussed factors influencing seed predator populations, activity, and granivory. Knowledge of the interrelations between factors and seed predators can contribute to designing future strategies that augment weed seed predator activity and enhance weed seed consumption as a component of an integrated weed management system.  相似文献

The timing of seed germination may determine the success of a weed species in an agroecosystem, and its expression is modulated by environmental conditions, but also by seed physiology and anatomy. The aims of this study were to investigate the roles of light, pericarp, dry storage and cold stratification on seed dormancy and germination in feral radish, a troublesome agricultural weed in temperate zones of the Americas that reduces crop yields. To this end, we used isolated intact pods and extracted seeds to test germination over time under contrasting temperature, light and storage conditions. Here, we showed that fresh seeds were non‐dormant, but that light and the presence of the pericarp reduced germination, especially under low temperatures. The pericarp reduced the final water content absorbed by seeds inside pods and decreased absorption/dehydration rates. The pericarp showed several small lignified cell layers in the endocarp, and x‐ray images displayed the lack of space between the partially embedded seed and the endocarp. Dry storage and cold stratification were ineffective in breaking the dormancy imposed by the pericarp. The apparent requirement for darkness and the mechanical restriction of the pericarp may have the potential to induce dormancy, spreading the timing of seed germination over a more extended period and hindering the control of feral radish.  相似文献

Seed predation by natural predators can contribute to the regulation of the weed seedbank, hence has been acknowledged as a promising way to significantly reduce the use of chemicals and mechanical weeding to manage arable weeds. However, the complex aspects that determine seed species preference are not yet fully understood, especially, the relationship between weed seed traits and the feeding behaviour of seed predator species. In this study, we examine predation on 42 weed species by four ground beetle species as a function of two major seed traits: seed mass and seed lipid content. We used an _original experimental design to create a statistically independent trait value distribution for evaluating the separate and interactive effects of these two traits. Our results show that all the ground beetles species consumed weed seeds, but at different rates. The smallest weed seeds with high lipid content were among the most consumed. While seed mass might be limiting for ground beetles, due to ease of handling or the thickness of the seed coat, they may preferentially consume seeds rich in lipids to increase energy uptake. To our knowledge, this study is the first to reveal that two major seed traits drive seed predation by ground beetles.  相似文献

Currently important alien weeds that have reached their final distributions are characterized by the high initial rates of spread with which they attained these distributions. Alien weeds, now seemingly unimportant but spreading at high rates, might be tomorrow's noxious weeds. It is suggested that rates of spread of species in a regional alien weed flora that have not yet reached final distributions may be used as indices for identifying future troublesome species. A literature search of several plant ecological attributes revealed that rapid seed germination, especially at low temperatures, may be associated with rapidly spreading weeds.  相似文献

Total weed control within a crop is both difficult and expensive to achieve, so that some weeds will often remain to set seed. The seed production resulting from these weeds will ultimately affect the sustainability of the weed control strategy. If too much is allowed to return each season there could be a gradual, but significant, increase in the potential weed flora over a number of seasons. Field trials were carried out in 2000 and 2001 to quantify the potential magnitude of this weed seed return from Chenopodium album L., grown at two planting densities either in pure stands or in competition with one of two crops (cabbage or onion). Crop and weed weights and weed seed production were notably greater in 2001. Both dry weight and seed production of C. album were suppressed by increasing planting density or by the presence of crop, with cabbage having a more suppressive effect. Despite the plasticity in seed production, a linear relationship was demonstrated between log weed seed production and log weed biomass that was robust over a range of competitive situations with onion and cabbage, at different planting densities and in growing seasons. The study also demonstrated that the relationship could be combined with an existing simple competition model to allow the consequences of incomplete weed control to be assessed in terms of potential weed seed return.  相似文献

Cambodia has experienced a rapid shift from transplanted to hand broadcast seeded rice, with a consequent increase in seeding rates from 25–30 to 100–200 kg ha?1. To reduce costs, farmers keep their own seed for sowing with the risk of greater weed seed contamination of the sowing seed. A survey of weed seed contamination in harvested rice paddy was conducted in two provinces of Cambodia (Battambang and Takeo) at the end of the wet season in 2016. Farmers were interviewed about rice‐seeding practices, and a total of 110 farmers' fresh paddy samples were inspected for weed seed contamination from the two provinces. Sowing seed samples collected from 28 seed producer lots and 71 samples of farmer‐kept seed were also analysed for weed seed contamination. In both provinces, the majority of farmers kept their own seed or bought seed from a neighbour. Farm‐kept seed for sowing accounted for 88% of sown seed in Battambang and 89% in Takeo. Seeds of 41 different weed species from 13 plant families were found in the farmers' freshly harvested paddy samples. Overall, farmers managed to reduce the number of weed propagules by 60% and seed producers by 95%. There was no significant difference between farmer‐kept seed and seed producer/seed company seed for the total number of weed seeds present. When shown photos, farmers' rankings of the 10 most common weed species found in freshly harvested paddy did not closely correspond to the actual weed seed frequency in the paddy. When farmers were asked to rank the frequency of weeds in their fields without the option to choose from a list, they ranked the weeds differently. Farmers ranked Ischaemum rugosum, Echinochloa spp. and Fimbristylis miliacea as the three most frequent weed species in their fields. The most frequent weeds in harvested paddy, apart from weedy rice, were Irugosum and Melochia corchorifolia. Farmers did not rank M. corchorifolia as a frequently occurring weed, and most farmers could not recognise M. corchorifolia from photographs. The priority for improved seed hygiene is to place the emphasis on assisting farmers to further improve their seed purification techniques and to caution them to inspect seed before purchasing from neighbours, seed producers and seed companies in the absence of the implementation of seed certification regulation.  相似文献

The onion, Allium cepa, is an important vegetable crop around the world. Globally, India ranks second in onion production, but yields are often much lower than in many other onion producing countries. This study reports on one major contributing factor to low onion yields in India: the presence of a weedy mimic that is morphologically similar to onion until the onset of flowering. We used morphological and molecular methods to identify this mimic as Asphodelus fistulosus, an exotic weed not previously reported in India. Our study indicated that molecular markers derived from the rbcL and/or matK chloroplast genes in a DNA barcoding approach allow accurate identification of this weed using any tissue and even at early developmental stages. Our results also showed that visual examination of seed lots coupled with DNA barcoding analysis of samples of 5–10 individual seeds can be used to confirm identification. The ability to easily identify contaminating material before and during cultivation can allow farmers to minimise production losses and prevent further spreading of this weed.  相似文献

Seed‐attacking microorganisms have an undefined potential for management of the weed seedbank, either directly through inundative inoculation of soils with effective pathogenic strains, or indirectly by managing soils in a manner that promotes native seed‐decaying microorganisms. However, research in this area is still limited and not consistently successful because of technological limitations in identifying the pathogens involved and their efficacy. We suggest that these limitations can now be overcome through application of new molecular techniques to identify the microorganisms interacting with weed seeds and to decipher their functionality. However, an interdisciplinary weed management approach that includes weed scientists, microbiologists, soil ecologists and molecular biologists is required to provide new insights into physical and chemical interactions between different seed species and microorganisms. Such insight is a prerequisite to identify the best candidate organisms to consider for seedbank management and to find ways to increase weed seed suppressive soil communities.  相似文献

Two field experiments investigated the influences of crop seed vigour on the effect of weed harrowing and crop:weed interactions in spring barley. Artificially reduced seed vigour, which was similar to the variation within commercial seed lots, caused a reduction in germination rate, delayed time of emergence and, consequently, caused reduced competitive ability against weeds. During both years, the reduced seed vigour increased the average weed biomass by 169% and 210%, and reduced the average crop yield by 16% and 21%. Without the influence of weeds, the yield reduction was estimated to be 8% and 10%. A three‐times harrowing strategy reduced the weed biomass by 75% and 72% on average. However, it also caused damage to the crop and reduced yield. There was no clear interaction between barley seed vigour and weed harrowing in the experiments but, in one year, reduced seed vigour tended to decrease the effect of weed harrowing and also increased crop damage. Results in both years, however, indicate potential possibilities for successful integrated weed control by adding the use of high seed quality to a weed harrowing strategy.  相似文献
