为明确引起云南文山州广南县西葫芦发生蕨叶、斑驳、泡状斑等症状的病原,从采集的西葫芦病样中提取总RNA,利用RT-PCR扩增后经电泳检测及测序获得大小为887bp的片段;经过BLAST序列同源性比对分析,该病原与番木瓜环斑病毒Papaya ringspot virus(PRSV)(KY996464)基因序列同源性达87%。由此可知,侵染文山州西葫芦的病毒病原是番木瓜环斑病毒。  相似文献

为防治番木瓜环斑病毒(Papaya ringspot virus,PRSV)在我国葫芦科作物上引起的病毒病害,以PRSV西瓜株系山东分离物(PRSV-SD)侵染性cDNA克隆为基础,采用定点突变方法将辅助成分-蛋白酶保守氨基酸137位和346位的天冬酰胺(N)和417位的缬氨酸(V)突变为丙氨酸(A),应用农杆菌浸润法接种西葫芦叶片并分析突变对PRSV-SD致病力的影响,筛选弱毒突变体,进而评价其交叉保护效果。结果表明,与野生型PRSV-SD相比,获得的3个突变体N137A、N346A和V417A,接种后在西葫芦植株上的症状明显减轻,衣壳蛋白在叶片中的积累水平分别为野生型PRSV-SD的24.0%、13.0%和4.0%,均为弱毒突变体。当保护间隔期为10 d时,弱毒突变体N137A具有完全的交叉保护效果,N346A可延迟发病15 d,而V417A无交叉保护效果。当间隔保护期为15 d时,弱毒突变体N137A和N346A的保护效率分别为100.0%和26.7%,而V417A无交叉保护效果。  相似文献

 The multiplex RT-PCR approach was developed for simultaneous detections of Arabis mosaic virus(ArMV),Strawberry latent ringspot virus(SLRSV)and Lily symptomless virus(LSV)from the imported lily bulbs.The results indicated that good specificity and sensitivity for simultaneous detection were obtained.The ultimate of RNA detection with three viruses mixture was 305 pg.The ultimate of RNA detection with ArMV,SLRSV and LSV were 156.0,13.4 pg and 1.12 ng respectively.This approach has potential to be used in quarantine of imports and exports.  相似文献

南方水稻黑条矮缩病毒一步双重RT-PCR 检测技术及其应用   总被引:5,自引:0,他引:5  
 An one-step dual RT-PCR method was developed for Southern rice black-streaked dwarf virus (SRBSDV) detection from host plants and insect vector white-backed planthopper (Sogatella furcifera Horvath,Hemiptera: Delphacidae), from which two cDNA fragments of the viral genome S5 and S10 were amplified
simultaneously. Two primer pairs, S5-F1 / S5-R2 (5′-ttacaactggagaagcattaacacg-3′ / 5′-atgaggtattgcgtaactgagcc -3′) and S10-oF / S10-oR(5cgcgtcatctcaaactacag -3′ / 5′-tttgtcagcatctaaagcgc -3′), were selectedfrom 40 primer pairs based on SRBSDV genome sequences, and amplified viral S5 ORF1 fragment (819 bp) and cp gene fragment ( 682 bp), respectively. Using total RNA extracts from infected plant leaf tissue or individual planthopper adult as templates, one step RT-PCR reaction in the conditions of annealing temperature of 53℃ with the final concentrations of 240 nmol / L S5-F1 / S5-R2 and 120 nmol / L S10-oF / S10-oR generated a similar yield of two amplicons in argrose electrophoresis analysis. Sequence analysis confirmed the correct
result of the amplification. Additionally, several commercal RNA extraction kits was proven to be fit for the template preparation, and the protocol for total RNA extraction from plant leaf tissue and individual planthopper was simplified by sample grinding direct in eppendorf tube. This method can be used for SRBSDV detection of dried or 70% alcochol soaked planthopper samples stored over one month in room temprature. 本研究在我国主要冬瓜产区采集具有典型病毒病症状的病叶材料105份,根据葫芦科作物上常见的5种病毒病原的CP基因设计特异性引物,对105份待检冬瓜材料进行RT-PCR检测。检测结果表明:5对特异引物可分别在105份待检材料的95份中检测到小西葫芦黄花叶病毒(Zucchini yellow mosaic virus,ZYMV)、西瓜花叶病毒(Watermelon mosaic virus,WMV)、黄瓜花叶病毒(Cucumber mosaic virus,CMV)、番木瓜环斑病毒(Papaya ringspot virus,PRSV)4种病毒,未检测到南瓜花叶病毒(Squash mosaic virus,SqMV);并且发现不同的冬瓜主产区致病的病毒种类有较大差异;同时还发现,在这些待检样品中4种病毒复合侵染现象较普遍,其中以PRSV与WMV组合最常见,占复合侵染现象的31.25%;未发现有4种病毒复合侵染。  相似文献

 Tobacco and potato samples showing symptoms of PVY were collected from different regions in Anhui Province, and the ELISA results of partial samples were positive. The total RNA was extracted from the positive samples by TRIZOL methods. Specific primer pair was designed to amplify cp gene of PVY by RT-PCR. Sequencing results indicated that the full length of cp gene of PVY from tobacco (PVY-CP-4) and pota-to (PVY-CP-7) is 801 nts, and each of them encodes 266 amino acids. A phylogenetic tree based on alignment of cp nucleotide sequences was constructed and the sequence comparing of cp gene was conducted. The results showed that PVY-CP-4 could be grouped into one branch with PVYO and PVYN:O and shared the highest sequence similarity (99.4%) with PVYO (EF026074). It was suggested that PVY-CP-4 derived from tobacco in Hefei might belong to PVYO. PVY-CP-7 clustered together with PVYN and PVYNTN and formed another branch. Furthermore, PVY-CP-7 shared the highest sequence similarity (98.3%) with PVYNTN(AJ890347), PVYNTN (EF026075) and PVYNTN(AJ585342). It was supposed that PVY-CP-7 derived from potato in Wuhe probably belonged to PVYNTN.  相似文献

防治苹果树腐烂病杀菌剂的室内筛选   总被引:6,自引:0,他引:6  
 The inhibitory effects of 10 fungicides on apple tree valsa canker were compared on dishes and twigs. The results showed that conidia germination and mycelial growth were inhibited by all fungicedes tested. The EC50 value of Difenoconazole was 6. 1×10-3μg·mL-1, the lowest among tested fungicide in inhibiting conidia germination. Tebuconazole and Imazalil also showed obvious inhibition effect. Thiophanatemethyl was the least efficient with the EC50 value 2.6×101 μg·mL-1. Difenoconazole also showed the highest activity for inhibiting mycelial growth with ECho value of 2.3×10-2 μg·mL-1. Tebuconazole was better than other fungicide. Thiophanate-methyl and Propineb were the least efficient with quite high EC50 value. Furthermore, the size of the lesion after inoculation on excised twigs also revealed that Difenoconazole was most efficient because it showed the smallest lesion of 394 mm2 compared with other tested fungicides. So Difenoconazole and Tebuconazole could be used to control apple tree valsa canker in field to subsititute forbided fungicides such as asomate.  相似文献

Qinghai Lake is the largest saline lake in China.The change in the lake volume is an indicator of the variation in water resources and their response to climate change on the Qinghai-Tibetan Plateau(QTP)in China.The present study quantitatively evaluated the effects of climate change and land use/cover change(LUCC)on the lake volume of the Qinghai Lake in China from 1958 to 2018,which is crucial for water resources management in the Qinghai Lake Basin.To explore the effects of climate change and LUCC on the Qinghai Lake volume,we analyzed the lake level observation data and multi-period land use/land cover(LULC)data by using an improved lake volume estimation method and Integrated Valuation of Ecosystem Services and Trade-offs(InVEST)model.Our results showed that the lake level decreased at the rate of 0.08 m/a from 1958 to 2004 and increased at the rate of 0.16 m/a from 2004 to 2018.The lake volume decreased by 105.40×108 m3 from 1958 to 2004,with the rate of 2.24×108 m3/a,whereas it increased by 74.02×108 m3 from 2004 to 2018,with the rate of 4.66×108 m3/a.Further,the climate of the Qinghai Lake Basin changed from warm-dry to warm-humid.From 1958 to 2018,the increase in precipitation and the decrease in evaporation controlled the change of the lake volume,which were the main climatic factors affecting the lake volume change.From 1977 to 2018,the measured water yield showed an"increase-decrease-increase"fluctuation in the Qinghai Lake Basin.The effects of climate change and LUCC on the measured water yield were obviously different.From 1977 to 2018,the contribution rate of LUCC was -0.76% and that of climate change was 100.76%;the corresponding rates were 8.57% and 91.43% from 1977 to 2004,respectively,and -4.25% and 104.25% from 2004 to 2018,respectively.Quantitative analysis of the effects and contribution rates of climate change and LUCC on the Qinghai Lake volume revealed the scientific significance of climate change and LUCC,as well as their individual and combined effects in the Qinghai Lake Basin and on the QTP.This study can contribute to the water resources management and regional sustainable development of the Qinghai Lake Basin.  相似文献

 Factors of influence on conidium germination and appressorium formation of Setosphaeria turcica,such as light condition,conidial concentration,nutrient resources and pH value of conidial suspension were studied.There was no significant difference among light treatments.The optimal pH for conidium germination and appressorium formation was 5.0 to 7.0.The exogenous nutrient sources were not the indispensable factors for conidium germination,but 5% sugar solution were more favorable for appressorium formation than the others.Low conidial concentration in suspension (conidia ≤ 104/mL) was propitious to conidium germination and appressorium formation,which were inhibited significantly in higher concentration.It was suggested that the phenomenon was due to the self-inhibitor,a kind of lipophilic substance,existing in the site of conidium germination.  相似文献

 以番木瓜环斑病毒黄斑分离物(PRV-YS) RNA为模板,在人工合成的两端引物引导下,反转录合成了外壳蛋白(CP)基因cDNA第一链并通过PCR扩增获得双链cDNA。用重组有cDNA的pUC19转化E.coli DH52,采用双脱氧链终止法进行了cDNA序列分析,结果表明CP基因为861nt,与文献[1]报道的相比,同源率90%,且少连续的6nt。  相似文献

采用标准地法和线路调查法,对湖北恩施市新塘乡双河厚朴基地的厚朴病虫害进行了系统调查,记录主要虫害13种,其中叶部害虫9种,枝干害虫2种,根部害虫2种。厚朴主要病害5种。藤壶蚧、厚朴枝角叶蜂和厚朴新丽斑蚜为湖北省首次报道,小绿叶蝉为厚朴新寄主记录种。同时记录了藤壶蚧的天敌6种,其中寄生小蜂2种,瓢虫4种;厚朴新丽斑蚜的天敌昆虫8种。对藤壶蚧、厚朴枝角叶蜂和厚朴苗木根腐病等重要病虫害的发生规律进行了初步调查,同时提出了防治建议。  相似文献

调查了甘肃省临洮县为害百合的地下害虫种类, 并以主要发生为害种类为供试对象, 采用拌土法测定了几种生物(源)农药对主要害虫的室内毒力和致死中时。调查发现小云斑鳃金龟Polyphylla gracilicornis、棕色鳃金龟Holotrichia titanis为主要发生及为害种类, 不同时间发生种类存在差异, 9月份以小云斑鳃金龟幼虫发生为害为主, 发生量占地下害虫总数的66.16%, 平均种群密度为10.48头/m2, 10月份, 则以棕色鳃金龟幼虫发生为主, 发生量占地下害虫总数的96.86%, 平均种群密度为7.79头/m2, 主要取食百合根系和鳞茎, 造成根系数量减少, 鳞茎出现不规则褐色缺口或凹陷斑。药剂筛选结果表明, 1.8%阿维菌素乳油、200亿孢子/g球孢白僵菌粉剂对两种蛴螬均具有较高的杀虫活性和速效性, 药后7 d时, 校正死亡率在82.22%以上, 对小云斑鳃金龟幼虫和棕色鳃金龟幼虫的LC50分别为0.161、0.060 mg/g和5.558×107、0.362×107孢子/g, LT50分别为2.237 d(2.2 mg/g)、1.393 d(2.2 mg/g)和6.645 d(40.0×107孢子/g)、4.940 d(2.0×107孢子/g), 这两种生物(源)农药对两种蛴螬的LC50和LT50值均略大于两种化学农药25%二嗪磷乳油、40%辛硫磷乳油处理的, 且对小云斑鳃金龟幼虫的杀虫活性均低于对棕色鳃金龟幼虫的杀虫活性, 致死中时则长于对棕色鳃金龟幼虫的致死中时。  相似文献

为了研究青稞种子外部和内部携带真菌情况,比较不同杀菌剂对青稞种子的带菌消毒效果和对幼苗生长的影响,为青稞种子播前包衣处理和种传真菌病害防控提供依据,采用离体平皿法对云南迪庆‘云青1号’、‘云青2号’和‘短白青稞’3个主栽品种进行带菌检测,并对种子进行拌种或浸种处理测定6种杀菌剂对种子消毒效果,分析杀菌剂对种子发芽和幼苗生长的影响。结果表明:供试青稞种子表面携带的优势菌群为青霉(Penicilliumspp.)、镰刀菌(Fusariumspp.);种子内部寄藏的真菌主要为镰刀菌、核腔菌(Pyrenophoraspp.)、附球菌(Epicoccumspp.)、丝核菌(Rhizoctoniaspp.)、链格孢(Alternariaspp.)和木霉(Trichoderma spp.)。青稞不同品种的种子表面及内部携带的真菌种类差异较大。致病性测定表明,镰刀菌对种子萌发和幼苗生长影响最大,后期出现幼苗坏死现象。45%咪鲜胺EW、75%百菌清WP、50%福美双WP对青稞种子携带真菌均有显著抑制作用和消毒效果,50%福美双WP消毒效果最优,达100%;45%咪鲜胺EW、75%百菌清WP、50%福美双WP处理对青稞种子发芽和幼苗生长均无显著影响。  相似文献

The effects of timing and method of application of Penicillium oxalicum on the control of fusarium wilt of tomato were investigated. Application of P. oxalicum to tomato seedlings in seedbeds reduced disease caused by Fusarium oxysporum f.sp. lycopersici in a growth chamber by 45–49% and in glasshouse experiments by 22–69%. Disease suppression was maintained for 60–100 days after inoculation with the pathogen in the glasshouse. No disease reduction was observed in tomato plants where P. oxalicum was applied to seeds. Treatment with P. oxalicum did not affect the population of F. oxysporum f.sp. lycopersici in the rhizosphere.  相似文献

The mode of action of the 2,4-diphenyl-1,3-oxazoline acaricide/insecticide etoxazole has been argued to be moulting inhibition, but experimental results supporting this hypothesis are lacking. This study investigated the effect of etoxazole on chitin biosynthesis in the fall armyworm, Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae). Etoxazole induced moulting defects in fall armyworm larvae similar, if not identical, to those caused by benzoylphenylureas, a well-known class of insecticidal chitin biosynthesis inhibitors. Furthermore, in contrast to untreated larvae, the chitin content in the integuments of larvae several days after treatment did not differ from that in freshly ecdysed individuals, thus suggesting strong chitin biosynthesis inhibition in vivo. A more detailed investigation of the inhibitory potential by incubating cultured integument pieces from larvae of S. frugiperda with [14C]N-acetyl-D-glucosamine, a radiolabelled chitin precursor, revealed I50 values of 2.95 and 0.071 microM for etoxazole and triflumuron respectively. The incorporation of radiolabel into potassium hydroxide-resistant material was inhibited by etoxazole in a dose-dependent manner. Based on these results, it is concluded that the acaricidal and insecticidal mode of action of etoxazole is chitin biosynthesis inhibition.  相似文献

棉花黄萎病菌致病型的AFLP分析   总被引:16,自引:0,他引:16  
 选用41个棉花黄萎病菌(Verticillium dahliae)代表菌系,在温室条件下,对4个棉花品种鄂荆1号(感)、中棉所12(耐)、文-5(抗)和唐棉2号(抗)进行致病性测定,结果可将供试菌系分为落叶型与非落叶型2类。选取8对AFLP引物PCR扩增的结果中,统计带型稳定、清晰且有多态性的条带,共169条作系统聚类分析,将上述菌系分为2大类,第一类为非落叶型菌系,包括10个非落叶型菌系和1个过渡菌系;第二类为30个落叶型菌系。根据聚类分析建立树状图,发现菌系与地理来源存在一定的相关性,而依据菌系致病力强弱分类则相关关系不大。选用25对EcoRⅠ和MseⅠ引物组合,对供试的41个V.dahliae进行AFLP扩增,筛选到2对引物E64(GACTGCGTACCAATTCGAC)、M53(GATGAGTCCTGAGTAACCG)和E49(GACTGCGTACCAATTCCAG)、M65(GAT-GAGTCCTGAGTAAGAG),能分别扩增出433bp和110bp2条仅为V.dahliae非落叶型菌系独有的特异片段,可将落叶型与非落叶型菌系分开,这2条特异片段被命名为EM433和EM110。  相似文献

The variability of dormancy of sclerotia of ten isolates ofSclerotium cepivorum was investigated. Of all isolates tested, the freshly harvested sclerotia were dormant. After drying for 48 hours the sclerotia of six isolates were able to germinate, two isolates stayed dormant and two isolates were infested by hyperparasitic fungi. After storage in soil at 5°C or 20°C, the sclerotia of the different isolates exhibited considerable differences in respect to germination capability, but all isolates showed highest germination after a treatment of 8 weeks at 20°C followed by 8 weeks at 5°C. The sclerotia of all isolates showed an increased capacity to germinate withoutAllium extracts at 10°C after pretreatment at 30°C for 28 days.  相似文献

在半干旱地区内蒙古中部3个不同造林立地类型对当地的8个乡土树种进行亚区造林对比试验,调查了被试树种在自然状态条件下的生长特征,应用PV技术测定了其水分生理指标,测定了抗寒及抗春季干旱风的等级,运用综合指标(成活、保存率、抗旱水分生理、耐寒性等)分析各树种的抗逆性和对不同立地条件的适应能力。结果表明:柄扁桃、桃叶卫矛、荆条和山杏(对照)抗逆性强,毛樱桃、山桃抗逆性较强,酸枣(山西种源)、欧李(山西栽培种)为中等,臭椿较差。  相似文献

以毒麦、田毒麦、多花黑麦草、多年生黑麦草、硬直黑麦草、高羊茅与狗牙根等禾本科的7种植物为材料,拟采用 DNA 测序、特异性位点比对、种间遗传距离测定、建立系统树等分析候选 DNA 条形码 psbA-trnH 鉴别黑麦草属常见植物的能力。实验结果表明,以 psbA-trnH 为 DNA 条形码时,建立的系统发育树能较好区分毒麦和田毒麦与其他几种植物。psbA-trnH 序列可以作为黑麦草属植物的潜在条形码。  相似文献
