Abstract

Shelf life of cultured gilt-head sea bream fillets (Sparus aurata) in over-wrap, vacuum and gas mixture packages stored during 27 days at 1±1°C was compared. The gas mixtures used contained CO2, O2, and N2 at different percentages (0% O2 + 50% CO2 + 50% N2; 10% O2 +50%CO2 +40%N2;20%O2 +50%CO2 +30%N2;30%O2 +50% CO2 + 20% N2).

The evolution of the freshness degree of the packaged fillets was carried out by measurements of total volatile bases (TVB), trimethylamine (TMA), Tiobarbituric Acid Reactives Substances (TBARS), K i and H values, psychrotrophic bacterial counts and sensory evaluation (color, odor, flavor).

Fish fillets were obtained 3 hours after harvesting with the best handling practices. As a consequence of this, the initial bacterial load was very low. The inhibitory effect of CO2 on bacterial growth was very effective, keeping very low counts throughout all the storage period. Gas packages without O2 and vacuum packages presented very low oxidation, whereas TBARS values in over-wrap and 10%, 20% and 30% O2packaged fillets increased according to the oxygen content in the package. Ki value and specially H value differentiated very well among aero-bically stored fillets, vacuum and modified atmosphere packaging. TVB and TMA productions were poor freshness indicators, since no differences were found among treatments. Sensory quality of fillets deteriorated faster in over-wrap than in vacuum and gas packages. Modified atmospheres containing 20 and 30% O2 were given lower sensory scores than vacuum packages, since they gave rise to yellowness of the fillets and off-odors and off-flavors.

In summary, modified atmosphere packaging of filleted gilt-head sea breams with a gas mixture consisted of 50% CO2 + 50% N2 gave rise to an important extension of shelf life compared to over-wrap packaging, vacuum and modified atmosphere packaging with gas mixtures containing O2. This was due to the very low oxidation levels produced during the storage, the lower H value as a consequence of a minor production of Hx and the great inhibition on microbial growth.  相似文献

ABSTRACT

The effects of different atmospheric conditions on the quality and the shelf life of pasteurized oyster meat in chilled storage were investigated. The alternatives tested for pasteurized oyster meat were normal air (Control), 75% CO2/25% N2 (MAP-1), 75% CO2/25% O2 (MAP-2), 75% CO2/5% O2/20% N2 (MAP-3), and vacuum packing (VP). The various observations of microbiological (total viable count (TVC), Enterobacteriaceae, Pseudomonas spp., lactic acid bacteria (LAB), and hydrogen sulfide–producing bacteria), physical (color coordinates L*, a*, and b*, and cutting strength), chemical (moisture, pH, total volatile basic nitrogen (TVB-N), peroxide value (PV), and thiobarbituric acid reactive substances (TBARS)) and sensory properties were performed every 3 days over a total of 27 days. During storage across all the treatments, L*, a*, cutting strength, pH, moisture content, and sensory scores decreased; whereas, b*, TVB-N, PV, TBARS, and microorganisms increased. However, among the various alternatives, MAP-1 had the best retention of oyster quality during storage. Based on sensory and microbiological qualities, the shelf lives in chilled storage were 9 days for control, 15 days for VP, 21 days for MAP-2 and MAP-3, and 24 days for MAP-1-treated cases.  相似文献

In this study, the fresh meat of bonito (Sarda sarda) was used to make lakerda, which is a type of salted fish (Turkish traditional product). The lakerda was packaged in plastic film (A), vacuumed (VP), placed in modified atmosphere packaging (MAP; M1 = 80%/20% CO2/N2, M2 = 65%/35% CO2/N2, M3 = 30%/60%/10% CO2/N2/O2), and stored at 2 ± 1°C. Microbiological, chemical, sensory, and color analyses were conducted to determine the changes in quality. Total viable bacteria counts did not exceed the limit of consumption (7 log cfu/g) for all groups on Day 31. Total volatile basic nitrogen (TVB-N) and trimethylamine- nitrogen (TMA-N) values of M1, M2, and M3 remained lower than the proposed acceptability limits of 35 and 12 mg N/100 g, respectively, up to 31 days of storage. However, VP lakerda samples exceeded TVB-N limits after 31 days of storage, whereas A lakerda samples exceeded after 8 days of storage. In our study, sensory analysis determined the shelf life of modified atmosphere packaged lakerda. When Group A is compared with the other groups, MAP and VP lengthened the shelf life of lakerda by 23 days.  相似文献

The stuffed mussel is a traditional, ready-to-eat food in Turkey which is produced by cooking rice and various spices filled inside the mussel. This article examines the effect on sensory, chemical, and microbiological parameters of stuffed mussels of two gas mixtures—MAP1 50% N2/50% CO2 and MAP2 100% CO2—used as modified atmospheres for cold storage at +4°C. Air packaged samples were chosen as the control group. Based primarily on sensory characteristics, MAP1 gas mixture was the most effective for stuffed mussel. According to sensory results, it has been determined that the control samples kept until the 11th day, while the MAP1 and the MAP2 groups kept until the 13th day. Of the chemical indices determined, the TVB-N values of MAP1 and MAP2 remained lower than the proposed acceptability limits of 20 mg N/100 g, up to 13 days of storage. Microbiological analysis results did not find any differences between the groups. It was found that the shelf life of air and MAP1-MAP2 packaged samples were 7 and 11 days, respectively.  相似文献

Combined impact of ethanolic coconut husk extract (ECHE) (200 ppm) and modified atmosphere packaging (MAP) with two separate gas mixtures [60% CO2/30% N2/10% O2 (M1) and 80% CO2/10% N2/10% O2 (M2)] on the shelf life of Asian sea bass slices during 4°C storage was investigated. Microbial load of slices was lowered when packaged under MAP, regardless of ECHE treatment, compared to the control (no ECHE treatment, packed in air). For control, slices treated with ECHE packaged in air, and samples packaged under M1, TVBC (total viable bacteria count) exceeded 6.0 log CFU/g at day 6, 9, and 12, respectively. TVBC of samples treated with 200-ppm ECHE and packaged under M1 and those packaged under M2 without and with ECHE treatment were lower than 6.0 log CFU/g at day 15. Peroxide and thiobarbituric acid reactive substances values in samples treated with ECHE, regardless of the packaging method, were lower than those kept under MAP and the control. Thus, shelf life extension of Asian sea bass slices could be achieved by treatment with 200-ppm ECHE in combination with M1 packaging, in which they could be stored longer than 15 days at 4°C.  相似文献

The objective of this work was to study the combined effect of gamma irradiation and coatings containing 0.5% rosemary (Rosmarinus officinalis) essential oil (RO) on the chemical, microbiological, and sensorial qualities of silver carp (Hypophthalmichthys molitrix) fish fillets (SFF) during cold storage (4°C). SFF were divided into three groups: uncoated (control), coated with edible coating (without additives), and γ-irradiated (0, 1, 3, and 5 kGy) coated with coating containing 0.5% rosemary. Gamma irradiation at 1, 3, and 5 kGy with coating reduced the initial total bacterial count, psychrophilic bacteria, and lactic acid bacteria and prolonged shelf life of the samples. Coated samples irradiated at 1 kGy reduced the counts of Enterobacteriaceae, Staphylococcus aureus, and Bacillus cereus, as well as eliminating Vibrio spp. and Salmonella spp., while coated samples irradiated at 3 and 5 kGy completely eliminated these bacteria. Combined treatment showed a slight increase in thiobarbituric acid-reactive substances postirradiation during cold storage but had no effect on the total volatile basic nitrogen and trimethylamine contents, while a gradual increase in these chemical quality indices was observed during cold storage. Combined treatment had no adverse effects on the sensory properties of SFF. The increase in the bacterial inhibitory effect is caused by both rosemary oil and irradiation.  相似文献

The combined effect of superchilling and modi?ed atmosphere packaging (MAP) treatment on the quality of swimming crab during storage was investigated. As a result, signi?cantly (p < 0.05) higher inhibitor effects on total aerobic plate counts (TPCs) were observed in MAP treatments than in control (air package). Additionally, chemical analysis results showed that MAP treatments with 40–80% carbon dioxide (CO2) were highly effective in maintaining lower total volatile basic-nitrogen (TVB-N), trimethylamine-nitrogen (TMA-N), pH, and thiobarbituric acid reactive substances (TBARS) values in swimming crab (p < 0.05). On the other hand, the brightness (L*) results also con?rmed that this combined treatment showed a positive effect on the stability of color during storage. Results also indicated that the combination of superchilling and 60% CO2 MAP treatment prolonged the shelf life of swimming crab to 15–20 days. However, 100% CO2 MAP treatment showed a negative effect on the drip loss and overall acceptability, indicating CO2 content should be considered as a critical factor in this promising technology.  相似文献

Abstract

The shelf life of iced redfish is known to be 16–19 days. This study evaluated the effects of modified atmosphere (CO2/N2: 60/40) in bulk storage of redfish and subsequent modified atmosphere packaging (MAP) of fillets. Fish were evaluated by sensory, microbiological and chemical methods. Modified atmosphere (MA) bulk storage of whole fish for more than 5 to 10 days did not improve their sensory quality or significantly increase shelf life. In fact, the study showed negative effects in texture and overall appearance. MAP of fillets processed from 10-day MA bulk stored fish showed a modest increase in shelf life, but negatively affected their texture. Sensory qualities, i.e., tenderness, that decreased under MA bulk storage were improved upon aerobic storage for fish stored under MA for 5 to 10 days, but not 14 days. Lower microbial levels were found in MA bulk stored fish and MAP fillets compared to traditionally iced fish, while higher TMA levels were found in MAP fillets.  相似文献

[目的]为探究不同包装对丁香鱼干常温贮藏期间的品质特性影响,[方法] 以空气包装为对照,与真空包装、气调包装和CO2包装做比较,测定贮藏过程中菌落总数、TVB-N、POV、TBA、色差、挥发性风味成分及感官评分的变化。[结果]结果显示,真空、气调、CO2三种包装均能有效抑制丁香鱼干微生物生长,常温25 ℃贮藏8 d后,真空包装组样品TVB-N、POV和TBA值分别较空气包装组降低47.08%、4.13%和25.49%,并且真空包装组样品在减缓样品色泽变化、保护肌纤维结构完整性、保留挥发性风味成分和保持感官特性方面均具有较佳效果,确定了真空包装为丁香鱼干的最佳包装方式。基于Arhenius方程建立了丁香鱼干货架期预测模型,25 ℃和35 ℃下真空包装丁香鱼干的货架期分别为6.07和3.23 d。[意义]本研究成果可为丁香鱼干常温物流过程中的品质控制提供理论依据。  相似文献

Solutions of chitosan (2%, w/v), garlic essential oil (5%, v/v), and their combination (2%, w/v chitosan +5%, v/v garlic essential oil) were applied to the surface of grass carp fillets. Subsequently, sensory, microbiological analyses (total viable count (TVC), Pseudomonas spp., Aeromonas spp., and H2S-producing bacteria), and physicochemical changes (total volatile basic nitrogen (TVB-N), thiobarbituric acid reactive substances (TBARS), and centrifugation loss) of grass carp fillets were evaluated over a 16-day storage at 4 ± 1°C. The treated samples resulted in significantly (< 0.05) higher overall acceptance scores than the control from day 8. The TVC of grass carp fillets was reduced from the initial level (4.0 log CFU/g) to 2–3 log CFU/g when the fillets were treated with either garlic essential oil or chitosan solution. After 4 days of storage, the Pseudomonas spp. counts in the chitosan-treated samples were significantly lower (< 0.05) than the counts in control and samples treated with garlic essential oil. A lag phase of 6 days for the counts of H2S-producing bacteria of fish fillets that were treated with chitosan or the combination solution was observed. The combination of chitosan with garlic essential oil did not show synergistic effects, although it displayed a higher degree of antimicrobial efficacy with respect to the spoiled bacteria. However, the combination of chitosan with garlic essential oil was the most effective at inhibiting the formation of TVB-N. Samples that were treated with garlic essential oil displayed the highest TBARS levels on days 4 and 6 (< 0.05).  相似文献

Chilled storage of spermatophores from white shrimp (Litopenaeus vannamei) is needed to generate a consistent and reliable supply of spermatozoa for domestication purposes. The objective of this study was to develop a protocol for the chilled storage of white shrimp spermatophores and to evaluate bacterial propagation during such storage. In the first experiment, spermatophores were immersed in four extenders, mineral oil, Ringer's solution, phosphate buffer and 0.85% NaCl, and stored at low temperature (2-4 °C) for 35 days. Characteristics of preserved spermatophores changed the least and viable sperm was highest when spermatophores were stored in mineral oil. Spermatophores preserved with mineral oil appeared morphologically normal. Bacillus circulans, Staphylococcus hominis and S. lugdunensis, S. sciuri, S. xylosus and Micrococcus spp. were identified as the predominant bacteria during chilled storage, and total bacterial counts gradually increased during the experiment. A second experiment investigated the effect of antibiotic on chilled storage. Spermatophores were preserved in only mineral oil or mineral oil with 0.1% penicillin-streptomycin. These were evaluated for changes in external morphology of spermatophores, sperm viability and total bacteria count every week during a 35-day experimental period. Percentages of viable sperm (69.5 ± 3.9%) were significantly higher (P < 0.05) among spermatophores preserved in mineral oil with 0.1% antibiotic compared with those preserved only in mineral oil (57.7 ± 3.4%) over 35 days. The number of total bacteria in the treatment with mineral oil ranged between 28.3 ± 4.8 and 2416.7 ± 299.4 CFU/g, but in mineral oil containing antibiotic bacteria were undetectable. This study suggests that chilled storage of spermatophores is a feasible approach for the management and spawning of white shrimp broodstock.  相似文献

The impact of shrimp larvae development, as well as water and food inputs upon the increase of bacterial populations within the bacterial community of hatchery tank biofilms, was studied. For this study, a total of 68 biofilm samples were collected from concrete tanks at three larvae production times in a commercial shrimp hatchery. Seventeen samples were taken at each larval development stage (Zoea I, Mysis I, postlarvae 1 and postlarvae 16), as well as 37 samples from water, shrimp nauplii and food, introduced into the shrimp hatchery tanks. Culturable and direct bacterial counts were performed and 16S‐rRNA‐targeted oligonucleotide probes were used to quantify the presence of specific bacterial groups. An average of 27–70% of DAPI total cell counts were detected with the EUB338 probe, while the GAM42a probe signal ranged from 1% to 11%. Vibrio‐like bacteria (VLB) counts in TCBS agar ranged from <10 to 101 VLB/cm−2, with a tendency to increase at the last postlarvae stage. The most significant external source of bacteria registered with GAM42a probe and TCBS agar were found in live Artemia nauplii, used as food; nevertheless, biofilms remain with low counts of these groups.  相似文献

Chilled storage of spermatozoa in fish has been extensively investigated for many years, but limited research was focused on crustacean species. Chilled storage of spermatophores of black tiger shrimp Penaeus monodon is needed to generate consistent and reliable supply of spermatozoa for subsequent use. The objective of this study was to develop a protocol for the chilled storage of black tiger shrimp spermatophores and to evaluate bacterial propagation during chilled storage of spermatophores. In the first experiment, spermatophores were selected and preserved using four different extenders, namely mineral oil, Ringer's solution, phosphate buffer and 0.85% sodium chloride, and stored at low temperature (2‐4 C) for 42 d without antibiotic supplementation. Results showed that mineral oil was the best extender for chilled storage of spermatophores, since the highest percentage of viable sperm (58.3 ± 2.9%) was observed with this extender at the end of experiment (day 42). Bacillus sp., Staphylococcus sp., and Pseudomonas aeruginosa were identified as the predominant bacteria occurring during chilled storage, and the total bacteria count gradually increased during the experiment. In the second experiment, spermatophores were preserved in the mineral oil with four concentrations of the antibiotic, penicillin‐streptomycin (0.1%,1%, 2%, and 3%). There was no significant difference (P 0.05) in the percentage of viable sperm among treatments with 0.1%,1 %, 2%, and 3% antibiotics. The total count of Bacillus sp., Staphylococcus sp., and P. aeruginosa in the antibiotic treated groups significantly decreased (P < 0.05) to undetectable levels by day 14 of the experiment. Fertility studies from artificial insemination indicated that P. monodon spermatophores preserved with mineral oil for 7‐8 d at 2‐4 C were capable of fertilizing eggs with hatching rates similar to the controls. This study suggests that chilled storage of spermatophores is a feasible approach for the management and spawning of black tiger prawn broodstock or other Invertebrate species that produce spermatophores.  相似文献

ABSTRACT

To understand biochemical characteristics, storage stability, and freshness indicators of white shrimp (Litopenaeus vannamei), changes in extractable nitrogenous compounds, microbial count, and sensory rating of white shrimp during storage at 25 and 4°C were investigated. Free amino acids showed a slow increase during storage at 25°C, but no obvious change was found at 4°C. Adenosine triphosphate (ATP) and adenosine diphosphate (ADP) were found at initial stage and decreased rapidly after storage. Both inosine 5′-monophosphate (IMP) and adenosine monophosphate (AMP) increased and then decreased during storage. Inosine, hypoxanthine, and the K-value gradually increased with time. The levels of total volatile basic nitrogen (TVB-N), NH3, and trimethylamine (TMA) of white shrimp increased with storage time at 25 and 4°C. The TVB-N, NH3, TMA, inosine, hypoxanthine, and K-value could be considered as freshness indicators of white shrimp during storage. However, the total plate count did not corroborate the acceptability recommended limits for white shrimp during storage. The sensory evaluation, associated with TVB-N, TMA, and K-value, showed the quality was unacceptable after 6 h storage at 25°C and 7 days at 4°C.  相似文献

A series of experiments to study the preservation of rainbow trout semen in the liquid and frozen state was carried out.For the preservation of liquid semen both undiluted and diluted semen was stored in total darkness at 20° or 4°C under a number of different gasses (air, O2, O2 + N2 (1:1), N2, CO2, N2 + H2 + CO2 (8:1:1)). The best results were obtained when storing semen in an undiluted state at 4°C under O2 (hatching rate after 15 days was 80.6%, controls were 98.2%). Good results were also obtained under air (after 9 days 94.7% vs 94.9% for the controls). Anaerobic conditions, dilution or an increase in temperature all rendered less favourable results.For the purpose of cryopreservation a diluent was developed on the basis of the composition of rainbow trout seminal plasma. The semen was mixed with the diluent immediately before freezing at a ratio of 1:1 or 1:3. The pellet-technique (Nagase, 1964) was found to be more convenient and successful than freezing in straws. The pellets were thawed in a 1% NaHCO3-solution. The hatching results after storage periods of 1–4 months were very variable (2.6–80.3%).  相似文献

Diseases are one of the most critical limiting factors in aquaculture. Recirculating aquaculture systems (RAS) are one of the most functionally viable sustainable aquaculture production systems adopted world over. In the event of any eventuality caused by bacterial pathogens, antibiotics or other antibacterial agents cannot be applied due to the vulnerability of biological filters which form an integral part of the RAS. Because of this, newer drugs are required for the effective control of diseases in RAS which would not interfere with the activity of nitrifying bacteria used in the biological filters. The antagonistic activity of pyocyanin, a bioactive compound produced by Pseudomonas aeruginosa, against vibrios is well established. The purpose of this study was to prove the effectiveness of pyocyanin as an aquaculture drug for application in RAS by employing a pilot-scale shrimp culture under RAS. It was noted that at the concentration of 5 mg L?1, pyocyanin could significantly bring down the population of Vibrio spp. in RAS without affecting noticeably the other natural heterotrophic bacteria. Also, pyocyanin at this concentration did not inhibit the activity of nitrifying bacterial consortia used in the SBSBR (stringed bed suspended bioreactor) of RAS. The reared shrimp (Penaeus monodon) showed 100% survival after the application of pyocyanin, besides exhibiting normal health signs. Pyocyanin was non-toxic to the shrimp hybrid cell line (PmLyO-Sf9) at the concentration required for its application in disease management (IC50?=?419.26 mg L?1). The present study has demonstrated that pyocyanin is effective as an environment-friendly and safe aquaculture drug for the application in RAS-based shrimp culture to control Vibrio spp. without impeding nitrification established through the deployment of nitrifying bioreactors.

Abstract

Lightly salted lumpfish roe (3.5–4.8% fw/w] salt in the water-phase, pH 5.4, vacuum-packed) was stored at 5°C. After 2 1/2 or 3 months of storage, different degrees of spoilage, caused by bacterial activity, occurred in eleven roe batches. Off-odors ranged from no or very weak odors to strong sulphury, sour odors. The microflora consisted of lactic acid bacteria, Enterobacteriaceae and Vibrio spp. Concentration of lactic acid, acetic acid, trimethylamine and total volatile bases were unrelated to spoilage odors. Volatile sulfur compounds (H2S, probably CS2, CH3SH and CH3CH2SH or CH3SCH3), produced during storage, appeared to be contributors to spoilage odors.  相似文献

为系统解析凡纳滨对虾白便综合征 (white feces syndrome,WFS)的发生与环境因子、微生物因子、宿主免疫力和水体微生物群落组成的关系。实验利用水体理化因子、可培养细菌、对虾机体免疫酶活性以及高通量测序等检测技术对健康与患WFS的池塘养殖凡纳滨对虾进行比较分析。结果显示,与健康组相比,患病池塘的水温、溶解氧 (DO)、pH、盐度等水质理化因子波动趋势相似,波动范围分别为26.1~29.0 °C、4.26~6.08 mg/L、8.39~8.73和40~49,患病组DO和盐度比健康组高;健康组对虾肝胰腺内可培养细菌和弧菌含量为1.19×105~7.70×105和8.8×103~1.96×104 CFU/g,弧菌占比为2%~16%,患病组对虾肝胰腺内可培养细菌和弧菌含量在3.80×105~2.51×106和2.02×105~1.49×106 CFU/g范围内,比健康组高15~113倍,弧菌占比在55%~70%。碱性磷酸酶 (AKP)、酸性磷酸酶 (ACP)、溶菌酶 (LZM)、超氧化物歧化酶 (SOD)和酚氧化酶 (PO)活性在健康组内为1.21~5.64、9.17~15.25、3.56~7.43、4.83~6.70及3.10~4.55 U/mg,在患病组内为2.12~5.39、19.22~26.96、19.73~26.85、3.00~4.14及7.76~9.21 U/mg。比较分析表明,WFS的发生与可培养细菌含量、弧菌占比、ACP、LZM、PO的相关性较强。高通量测序分析表明,患病组水体菌群结构的Ace和Chao指数呈一定程度下降趋势,PCoA指数偏离度较高,放线菌门、变形菌门相对丰度降低,拟杆菌门、蓝藻门相对丰度显著升高;RDA关联分析表明,盐度、溶解氧、虾体细菌、虾体弧菌、水体细菌是影响患病对虾水体菌群结构组成的重要因子。相关研究结果为解析养殖生产中对虾WFS发生机制提供数据支撑,并为WFS的临床防控奠定理论基础。  相似文献

The protective activity of a non-bacteriocinogenic Lactococcus piscium CNCM I-4031 strain against Listeria monocytogenes was investigated in tropical cooked peeled shrimp stored at 8°C in modified atmosphere packaging (50% N2–50% CO2). When inoculated alone (L. piscium 107 CFU g?1 and L. monocytogenes 104 CFU g?1), protective culture and target strain grew very well on shrimp reaching a maximum cell number of 109 CFU g?1 after 7 and 14 days, respectively. In the presence of L. piscium, growth of L. monocytogenes was totally prevented after 3 days of storage. The count was 3.4 log CFU g?1 lower than in the control after 10 days and until the end of storage (31 days). Using the Seafood Spoilage and Safety Predictor Software (http://sssp.dtuaqua.dk), it was shown that pH decrease from 6.58 to 5.94 and lactic acid concentration of 89.65 mM measured in the co-inoculated batch did not fully explain the inhibition observed.  相似文献
